In the in vivo experiment, 1 m L of 5% sodium thioglycollate was injected into the abdomen of the mice on Day 0 and macrophages were harvested on Day 4. The macrophages were cultured in plates and treated with different concentrations of BCPs and stimulus. Effects of BCPs on macrophage functions were assessed by chemotaxis assay, phagocytosis assay and Enzyme-Linked Immunosorbent Assay(ELISA). Our results showed the enhanced chemotaxis, phagocytosis and secretion of nitric oxide(NO) and inflammatory cytokines by macrophages when treated with BCPs. However, when chemotaxis and phagocytosis were up-regulated by complement components or opsonized particles, BCPs inhibited these effects. Also, the NO production induced by lipopolysaccharides(LPS) was suppressed by BCPs mildly. Moreover, BCPs had an inhibitory effect on the [Ca^(2+)]i elevation of macrophages. These results suggested that BCPs exerted modulatory effects on macrophage functions, which may contribute to developing novel approaches to treating inflammatory diseases.展开更多

METHOD: Silica gel, Sephadex LH-20, MPLC Rp-C18 column chromatography, and HPLC were used for isolation of compounds. The structures were elucidated on the basis of 1D- and 2D-NMR technology and HRESI-MS. Compounds were evaluated in vitro for their inhibitory ability against the proliferation of rat mesangial cells by the MTT method. RESULTS: Twelve compounds were isolated, and their structures were identified on the basis of their spectroscopic and ico-chemical properties as 13, 28-epoxy-olean-11-en-3-one(1), saikogenin E(2), saikogenin G(3), 11α-methoxy-3β, 16β, 23, 28-tetrahydroxyolean-12-ene(4), saikogenin D(5), prosaikogenin F(6), prosaikogenin A(7), prosaikogenin G(8), prosaikogenin D(9), laccaic acid(10), methyl gallate(11), and ethyl gallate(12). Compounds 1, 2, 7, 8, and 10 were observed to have inhibitory activity against mesangial cell proliferationin to different degrees. CONCLUSION: Compound 1, 8, and 10 exhibit significant inhibitory effects on rat mesangial cell proliferation induced by Ang II.展开更多

Forty lactating Holstein cows were randomly assigned to 1 of 4 treatments. The treatments consisted of 0, 0.25, 0.5, and 1.0 g of BE kg-1 dry matter. Supplementation with BE decreased (P〈0.05) blood urea nitrogen (BUN) contents and increased blood total protein (TP) and albumin (ALB) levels compared with control cows, but it had no effects (P〉0.05) on blood glucose (GLU), nonesterified fatty acid (NEFA), total triglycedde (TG), low-density lipoprotein cholesterol (LDL-C), and high density lipoprotein cholesterol (HDL-C). Compared with control cows, cows fed BE had higher (P〈0.05) superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activity. However, supplementation with BE had no effect (P〉0.05) on total antioxidant capacity (T-AOC) or malondialdehyde (MDA) levels. The immunoglobulin (Ig) A and G contents increased (P〈0.05) in cows fed 0.25 or 0.5 g of BE kg-1. Interleukin (IL)-2 and IL-4 levels were higher (P〈0.05) in cows fed 0.5 and 1.0 g of BE kg-1, and IL-6 was significantly elevated (P〈0.05) in cows fed 0.5 g of BE kg-1. There were no treatment effects (P〉0.05) on the CD4+ and CD8+ T lymphocyte ratios, CD4+/CD8+ratio, or tumor necrosis factor-α (TNF-α) level among the groups. These findings suggest that BE supplementation may improve protein metabolism, in addition to enhancing antioxidant activity and immune function in heat-stressed dairy cows.展开更多

In the present study, the 70% EtO H extract of the roots of Bupleurum chinense was revealed to have agonistic activity on 5-HT2 C receptor, and the subsequent bioassay-guided isolation led to identification of several saikosaponins as the active constituents with 5-HT2 C receptor agonistic activity in vitro and anti-obesity activity in vivo. The new compound, 22-oxosaikosaponin d(1), was determined by extensive spectroscopic analyses(HR-ESI-MS, IR, and 1D and 2D NMR). The primary structure-activity relationship study suggested that the intramolecular ether bond between C-13 and C-28 and the number of sugars at C-3 position were closely related to the 5-HT2 C receptor agonistic activity. Saikosaponin a(3), the main saponin in B. chinense, showed obviously agonistic activity on 5-HT2 C receptor with an EC50 value of 21.08 ± 0.33 μmol×L^(–1) in vitro and could reduce food intake by 39.1% and 69.2%, and weight gain by 13.6% and 16.4%, respectively, at 3.0 and 6.0 mg×kg^(–1) in vivo. This investigation provided valuable information for the potential use of B. chinense as anti-obesity agent.展开更多
