To investigate the chemical components from the stems of Casearia velutina Bl.,the constituents were isolated by repeated chromatography with silica gel,Sephadex LH-20,and ODS columns.The structures were elucidated by...To investigate the chemical components from the stems of Casearia velutina Bl.,the constituents were isolated by repeated chromatography with silica gel,Sephadex LH-20,and ODS columns.The structures were elucidated by spectroscopic analysis.Eleven triterpenoids and its glycosides were isolated from the crude extract of C.velutina,and their structures were identified as friedelin-2,3-lactone(1),friedelane(2),epifriedelanol(3),friedelin(4),2α,3α,19α-trihydroxy-urs-12-en-28-oic acid(5),2α,3β,19α-trihydroxy-urs-12-en-28-oic acid(6),2α,3α,23-trihydroxy-urs-12-en-28-oic acid(7),2α,3α,23-trihydroxy-olean-12-en-28-oic acid(8),2α,3α,19α,23-tetrahydroxy-urs-12-en-28-oic acid(9),2α,3α,19α,23-tetrahydroxy-urs-12-en-28-oic acid-28-O-β-D-glucopyranosyl ester(10),and 3β,19α-dihydroxy-urs-12-en-28-oic acid 3-O-α-L-arabinopyranoside(11).All the compounds described above were isolated from this species for the first time.Compound 1 is a rarely occurred seco-friedelolactone in Flacourtiaceae.展开更多
Gastrodia elata Bl.is a traditional Chinese herbal medicine with special pharmacological effects on headache and numbness.However,its wild sources are now in short supply because of over-excavation.A total of 14 endop...Gastrodia elata Bl.is a traditional Chinese herbal medicine with special pharmacological effects on headache and numbness.However,its wild sources are now in short supply because of over-excavation.A total of 14 endophytic fungi were isolated from Gastrodia elata Bl collected from Yinjing,in which 10 strains were identified,including Neonectria,Ascomycota,Fusarium,Oidiodendron and Dothiorella.All crude extracts showed some degrees of antioxidant activities by T-AOC method and DPPH methods,especially those from Oidiodendron GeR6 and Ascomycota sp.GeR2,which might be attributed due to their high phenolic contents.However,only Oidiodendron GeR6 contained gastrodin(39.2μg/g)as the host plant in the crude extracts.In conclusion,the notable activities of GeR6 indicated its medical use as a natural antioxidant agent,as well as substitutable use for Gastrodia elata Bl.in the future.展开更多

This article discusses the specific molecular mechanism and amelior...Objective: Sarcopenia causes loss of skeletal muscle and function, thus seriously affecting the physical function and quality of life in the elderly. This article discusses the specific molecular mechanism and ameliorating effects of Tudangshen(TDS) on sarcopenia in elderly rats with type 2 diabetes mellitus(T2DM).Methods: Elderly Sprague-Dawley(SD) rats were randomly selected and fed with a high-fat diet combined with an intraperitoneal injection of streptozotocin to establish T2DM model. The model rats were stratified and randomly divided into the model group,metformin group, TDS high-dose group, TDS medium-dose group, and TDS low-dose group according to blood glucose combined with body weight, and the same batch of old SD rats was set as the normal control group. The effects of TDS in an elderly T2DM sarcopenia rat model were evaluated by observing the body positions of the rats, analyzing blood biochemistry, testing exercise capacity, and pathologically staining sectioned gastrocnemius muscle tissues. The molecular mechanisms of the effects were analyzed using quantitative real-time polymerase chain reaction and western blotting.Results: TDS has no statistically significant effect on blood glucose, insulin and glycosylated serum protein in aged rats with T2DM, but it can reduce levels of glycosylated serum protein, total cholesterol, triglycerides, and low-density lipoprotein;it improves pathological changes in rat gastrocnemius muscle tissues, and increases muscle cell activity in elderly rats with T2DM and sarcopenia. TDS also promoted the upregulation of the expression of mammalian target of rapamycin(mTOR)/protein kinase B(PKB/AKT)/phosphatidylinositol 3-kinase(PI3K)/ribosomal protein S6 kinase/eukaryotic initiation factor 4 E binding rotein1 mRNA in rats and triggered an increase in corresponding protein levels.Conclusions: TDS alleviated muscle decline in elderly rats with T2DM by activating the PI3 K/AKT/mTOR signaling pathway and regulating the synthesis of corresponding proteins.展开更多
Acrylamide(AA)is ubiquitous in packaging materials,paints,cosmetics,food and drinking water.However,AA has obvious neurotoxicity,reproductive toxicity and carcinogenicity,which seriously endangers human health.As one ...Acrylamide(AA)is ubiquitous in packaging materials,paints,cosmetics,food and drinking water.However,AA has obvious neurotoxicity,reproductive toxicity and carcinogenicity,which seriously endangers human health.As one of precious traditional Chinese medicines,Gastrodia elata Bl.has been mainly used to lower blood pressure and resist myocardial ischemia.However,the antioxidant activity of Gastrodia elata Bl.on AA-induced DNA damage has not been reported.We found that AA(25,30 and 50 mg/kg)could cause DNA damage and the damage was time/dose dependent.After AA administration,Tail moment,micronuclei formation rate,8-OHdG and MDA level were significantly increased and SOD activity was decreased.Gastrodia elata Bl.decoction,gastrodin and vitamin C could reduce AA-induced DNA damage by decreasing AA-induced increment of tail moment and micronuclei formation rate.What’s more,Gastrodia elata Bl.decoction,gastrodin and vitamin C could lower 8-OHdG and MDA level and improve SOD activity.In conclusion,our research showed that Gastrodia elata Bl.decoction and gastrodin had protective effects on AA-induced DNA damage,and the potential protective mechanism might be related to anti-oxidative stress.展开更多
[Objectives] To screen the active components and core targets of Gastrodia elata Bl. based on network pharmacology, and further explore its potential molecular mechanism in preventing and treating chronic heart failur...[Objectives] To screen the active components and core targets of Gastrodia elata Bl. based on network pharmacology, and further explore its potential molecular mechanism in preventing and treating chronic heart failure (CHF).[Methods] The candidate active constituents of G. elata Bl. were screened by HERB, ETCM (Encyclopedia of Traditional Chinese Medicine) database, and bioinformatics analysis tool BATMAN-TCM platform, Swiss Target Prediction platform was used to predict compound targets, and CHF disease targets were searched in GeneCards and OMIM databases;the intersection targets were taken, and the String database and Cytoscape 3.10.0 software were used for protein-protein interaction (PPI) and topological analysis to obtain key active constituents and core target genes;the online tool bioinformatics platform was used for gene ontology (GO) function and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis of common targets. Using the active constituent of G. elata Bl. as ligand and its core target as receptor, molecular docking visualization was carried out.[Results] 136 common targets of active constituents of G. elata Bl. and CHF were screened out, which mainly involved AGE-RAGE signaling pathway, lipid and atherosclerosis, interleukin-17 signaling pathway, non-alcoholic fatty liver, diabetic cardiomyopathy, cancer and other signaling pathways. The core target proteins mainly included albumin, catalytic subunit α activated by protein kinase CAMP, insulin gene, interleukin-1β, tumor necrosis factor, estrogen receptor 1, interferon γ, etc. The results of molecular docking showed that the ligand of the compound had good affinity with the target receptor.[Conclusions] G. elata Bl. prevents and treats CHF through multiple components, multiple targets, and multiple pathways. The potential mechanism of G. elata Bl. in treating CHF was preliminarily explored, providing a certain theoretical basis for subsequent research on its pharmacological material basis in treating CHF.展开更多
With the rapid expansion of the Internet of Things(IoT),user data has experienced exponential growth,leading to increasing concerns about the security and integrity of data stored in the cloud.Traditional schemes rely...With the rapid expansion of the Internet of Things(IoT),user data has experienced exponential growth,leading to increasing concerns about the security and integrity of data stored in the cloud.Traditional schemes relying on untrusted third-party auditors suffer from both security and efficiency issues,while existing decentralized blockchain-based auditing solutions still face shortcomings in correctness and security.This paper proposes an improved blockchain-based cloud auditing scheme,with the following core contributions:Identifying critical logical contradictions in the original scheme,thereby establishing the foundation for the correctness of cloud auditing;Designing an enhanced mechanism that integrates multiple hashing with dynamic aggregate signatures,binding encrypted blocks through bilinear pairings and BLS signatures,and improving the scheme by setting parameters based on the Computational Diffie-Hellman(CDH)problem,significantly strengthening data integrity protection and anti-forgery capabilities;Introducing a random challenge mechanism and dynamic parameter adjustment strategy,effectively resisting various attacks such as forgery,tampering,and deletion,significantly improving the detection probability of malicious Cloud Service Providers(CSPs),and significantly reducing the proof generation overhead for CSPswhilemaintaining the same computational cost forDataOwners.Theoretical analysis and performance evaluation experiments demonstrate that the proposed scheme achieves significant improvements in both security and efficiency.Finally,the paper explores potential applications of the Enhanced Security Scheme in fields such as healthcare,drone swarms,and government office attendance systems,providing an effective approach for building secure,efficient,and decentralized cloud auditing systems.展开更多
