应用NB4细胞株建立SCID beige小鼠急性早幼粒细胞白血病病理模型 被引量:4

目的:本研究旨在建立稳定、有效、重复性好的人急性早幼粒白血病重度联合免疫缺陷(SCID)小鼠移植瘤模型,观察模型的病程特点和生物学行为。方法:将3-5周龄SCID beige小鼠随机分为实验组和对照组;实验组鼠尾静脉注射对数生长期的NB4细胞5... 目的:本研究旨在建立稳定、有效、重复性好的人急性早幼粒白血病重度联合免疫缺陷(SCID)小鼠移植瘤模型,观察模型的病程特点和生物学行为。方法:将3-5周龄SCID beige小鼠随机分为实验组和对照组;实验组鼠尾静脉注射对数生长期的NB4细胞5×106个/只,动态监测小鼠外周血白细胞数和外周血涂片中人早幼粒细胞阳性率;应用形态学和组织病理学方法确认白血病细胞浸润肝、脾、肺、肾、脑等器官情况,Western blot检测组织中PML-RARa融合蛋白表达情况。结果:接种NB4细胞2周内的实验组SCID小鼠外周血白细胞计数与对照组相比差异无统计学意义(P>0.05);同时,血涂片姬姆萨染色后镜检未见到NB4细胞;接种第21天和第28天实验组SCID小鼠外周血白细胞计数分别为(4.79±1.13)×109/L和(7.62±2.24)×109/L,显著高于对照组(P<0.05);血涂片吉姆萨染色后可见NB4细胞比例分别占(2.14±0.63)%和(6.6±2.76)%;形态学观察显示,接种21 d后实验组SCID小鼠体内出现单个或多个肿瘤实体包块;同时,组织切片HE染色表明,肝、脾、肺、肾、脑组织均有不同程度的瘤细胞浸润;细胞免疫荧光结果显示,实验组小鼠骨髓细胞CD33表达呈阳性(P<0.05);Western blot数据证实,肝、肾、脑组织中检测到不同水平的PML-RARa融合蛋白表达。结论:SCID小鼠鼠尾静脉注射NB4细胞可成功构建人急性早幼粒细胞白血病小鼠模型,该模型能模拟临床白血病累及骨髓和弥漫生长特点,是研究人类白血病发病机理及实验治疗的良好模型。 展开更多

目的建立稳定有效的绒毛膜癌SCID beige小鼠移植瘤模型,观察模型的病程特点和生物学行为。方法将3-5周龄SCID beige雌性小鼠随机分为实验组和对照组;实验组12只小鼠分为A(皮下移植瘤模型)/B(肺转移瘤模型)两组,分别皮下注射或尾静... 目的建立稳定有效的绒毛膜癌SCID beige小鼠移植瘤模型,观察模型的病程特点和生物学行为。方法将3-5周龄SCID beige雌性小鼠随机分为实验组和对照组;实验组12只小鼠分为A(皮下移植瘤模型)/B(肺转移瘤模型)两组,分别皮下注射或尾静脉注射JAR细胞5×10^6/只,应用形态学、放射性免疫分析法测定β-HCG、小动物活体成像系统(IVIS)和组织学HE染色方法明确JAR细胞皮下移植瘤及肺转移瘤形成等情况。结果实验组接种28 d后,A组小鼠皮下形成质硬肿瘤结节,HE染色符合绒毛膜癌细胞形态学表现;B组小鼠应用IVIS检测显示体内出现单个或多个肿瘤实体包块,HE染色表明肺组织均有不同程度的瘤细胞浸润。接种第14天,实验组β-HCG水平显著高于对照组(P〈0.05);其中B组β-HCG水平明显高于A组(P〈0.05)。结论 SCID beige小鼠皮下/尾静脉注射JAR细胞可成功构建人绒毛膜癌移植瘤病理模型,该模型能模拟临床绒毛膜癌上皮性实体瘤特性及肺转移特点,是研究人类绒毛膜癌发病机理及实验治疗的良好模型。 展开更多

Adipose tissue is a major metabolic organ, and it has been traditionally classified as either white adipose tissue(WAT) or brown adipose tissue(BAT). WAT and BAT are characterized by different anatomical locations, morphological structures, functions, and regulations. WAT and BAT are both involved in energy balance. WAT is mainly involved in the storage and mobilization of energy in the form of triglycerides, whereas BAT specializes in dissipating energy as heat during cold- or diet-induced thermogenesis. Recently, brownlike adipocytes were discovered in WAT. These brownlike adipocytes that appear in WAT are called beige or brite adipocytes. Interestingly, these beige/brite cells resemble white fat cells in the basal state, but they respond to thermogenic stimuli with increased levels of thermogenic genes and increased respiration rates. In addition, beige/brite cells have a gene expressionpattern distinct from that of either white or brown fat cells. The current epidemic of obesity has increased the interest in studying adipocyte formation(adipogenesis), especially in beige/brite cells. This review summarizes the developmental process of adipose tissues that originate from the mesenchymal stem cells and the features of these three different types of adipocytes. 展开更多

Characteristics of Beige Adipocytes Induced from White Adipocytes by Kikyo Extract

As such, compounds capable of inducing the development of beige adipocytes may be useful as drugs for anti-obesity and anti-type 2 diabetes. However, the true nature of beige adipocytes remains unclear. The purpose of this study is to confirm whether or not white adipocyte can differentiate to beige adipocytes and to clarify the characteristics of beige adipocytes. We first searched for an inducer of beige adipocytes and found that kikyo extract, a component of bofu-tsusho-san, was a strong inducer. We then attempted to prove that beige adipocytes could be induced from white adipocytes. Second, we clarified the characteristics of beige adipocytes induced from white adipocytes. The results suggested that beige adipocytes were high-performance adipocytes with a greater ability to synthesize and consume triglyceride and take up glucose than white adipocytes. 展开更多

Nevertheless, several pig breeds are known to be cold resistant. The molecular mechanism(s) enabling such adaptation are currently unknown. Here, we show that this resist- ance is not dependent on shivering, but rather depends on UCP3 and white adipose tissue (WAT) browning. In two cold-resistant breeds (Tibetan and Min), but not a cold-sensitive breed (Bama), WAT browning was induced after cold exposure. Beige adipo- cytes from Tibetan pigs exhibited greater oxidative capacity than those from Bama pigs. Notably, UCP3 expression was signifi- cantly increased only in cold-resistant breeds, and knockdown of UCP3 expression in Tibetan adipocytes phenocopied Bama adipocytes in culture. Moreover, the eight dominant pig breeds found across China can be classified into cold-sensitive and cold- resistant breeds based on the UCP3 cDNA sequence. This study indicates that UCP3 has contributed to the evolution of cold resistance in the pig and overturns the orthodoxy that UCP1 is the only thermogenic uncoupling protein. 展开更多

Sympathetic arborizations act as the essential efferent signals in regulating the metabolism of peripheral organs including white adipose tissues (WAT). However, whether these local neural structures would be of plastic nature, and how such plasticity might participate in specific metabolic events of WAT, remains largely uncharacterized. In this study, we exploit the new vol- ume fluorescence-imaging technique to observe the significant, and also reversible, plasticity of intra-adi- pose sympathetic arborizations in mouse inguinal WAT in response to cold challenge. We demonstrate that this sympathetic plasticity depends on the cold-elicited sig- nal of nerve growth factor (NGF) and TrkA receptor. Blockage of NGF or TrkA signaling suppresses intra- adipose sympathetic plasticity, and moreover, the cold- induced beiging process of WAT. Furthermore, we show that NGF expression in WAT depends on the cate- cholamine signal in cold challenge. We therefore reveal the key physiological relevance, together with the reg- ulatory mechanism, of intra-adipose sympathetic plas- ticity in the WAT metabolism. 展开更多

Adipose tissue is an important organ for energy homeostasis. White adipose tissue stores energy in the form of triglycerides, whereas brown adipocytes and recently identified beige adipocytes are specialized in dissipating energy by thermogenesis or contribution to dispose glucose and clear triglycerides in blood. The inverse correlation between the brown adipose tissue activity and body mass suggests its protective role against body fat accumulation. Thus, recruitment and activation of brown or beige adipose tissue become particularly appealing targets for increasing energy expenditure. Angiogenesis and sympathetic nerve signals are the fundamental determinants for brown and beige adipose tissue development, as well as for their metabolic functions. Secretary factors including BMPs can induce the development, the activation of brown or beige adipose tissue, which seem to be promising for therapeutic development. 展开更多

电针通过cAMP-PKA/ATF2轴促进白色脂肪米色化产热预防肥胖机制研究

目的:旨在探讨电针(EA)疗法是否通过环磷酸腺苷(cAMP)-蛋白激酶A(PKA)/激活转录因子2(ATF2)信号轴促进白色脂肪米色化产热,进而预防肥胖的发生。方法:将40只C57BL/6J小鼠随机分成四组,每组10只:正常(Ctrl)组、Model组、EA组和EA+H-89组... 目的:旨在探讨电针(EA)疗法是否通过环磷酸腺苷(cAMP)-蛋白激酶A(PKA)/激活转录因子2(ATF2)信号轴促进白色脂肪米色化产热,进而预防肥胖的发生。方法:将40只C57BL/6J小鼠随机分成四组,每组10只:正常(Ctrl)组、Model组、EA组和EA+H-89组。检测各组小鼠体重、Lee’s指数以及血脂指标含量。HE染色观察腹部脂肪组织的形态学变化。蛋白免疫印迹检测PR结构域包含蛋白16(PRDM16)、解偶联蛋白1(UCP1)、过氧化物酶体增殖物激活受体γ共激活因子1α(PGC-1α)、PKA和ATF2蛋白表达水平。ELISA检测cAMP含量。结果:与Ctrl组比较,Model组的体重,Lee’s指数,总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)含量,白色脂肪质量显著增加;高密度脂蛋白胆固醇(HDL-C)含量、PRDM16、UCP1、PGC-1α、PKA和ATF2蛋白表达水平,cAMP含量显著降低(P<0.05)。与Model组比较,EA组的体重,Lee’s指数,TC、TG、LDL-C含量,白色脂肪质量显著降低;PRDM16、UCP1、PGC-1α、PKA和ATF2蛋白表达水平,cAMP含量显著增加(P<0.05)。与EA组比较,EA+H-89组的体重,Lee’s指数,TC、TG、LDL-C含量,白色脂肪质量显著增加;PRDM16、UCP1、PGC-1α、PKA和ATF2蛋白表达水平,cAMP含量显著降低(P<0.05)。结论:EA能够激活cAMP-PKA/ATF2信号通路,进而上调白色脂肪组织米色化进程中的关键调控因子PRDM16、UCP1以及PGC-1α的表达水平,推动白色脂肪向米色脂肪的转化,增强机体的产热效能,发挥减肥降脂作用。 展开更多

Background Thermogenic adipose tissue,both beige and brown,experiences whitening as animals are exposed to warmth and age,but the potential mechanisms are not fully understood.In this study,we employed singlenucleus RNA-seq to construct a cell atlas during whitening progression and identified the characteristics of thermogenic adipocytes.Results Our histological studies and bulk transcriptome gene expression analysis confirmed that both perirenal and omental adipose tissues(pAT and oAT)exhibited progressive whitening in goats.Compared to the classic brown adipocytes in mice,goat thermogenic adipocytes were more closely related in gene expression patterns to human beige adipocytes,which was also confirmed by adipocyte type-and lineage-specific marker expression analysis.Furthermore,trajectory analysis revealed beige-and white-like adipocytes deriving from a common origin,coexisting and undergoing the transdifferentiation.In addition,differences in gene expression profiles and cell communication patterns(e.g.,FGF and CALCR signaling)between oAT and pAT suggested a lower thermogenic capacity of oAT than that of pAT.Conclusions We constructed a cell atlas of goat pAT and oAT and descripted the characteristics of thermogenic adipocytes during whitening progression.Altogether,our results make a significant contribution to the molecular and cellular mechanisms behind the whitening of thermogenic adipocytes,and providing new insights into obesity prevention in humans and cold adaptation in animals. 展开更多

目的:探索激活α7烟碱型乙酰胆碱受体(nicotinic acetylcholine receptor,nAChR)促进寒冷条件下的肥胖小鼠白色脂肪米色化和产热的作用机制。方法:8周龄雄性C57BL/6J小鼠适应性饲养1周后,给予高脂饮食(high-fat diet,HFD)12周构建肥胖... 目的:探索激活α7烟碱型乙酰胆碱受体(nicotinic acetylcholine receptor,nAChR)促进寒冷条件下的肥胖小鼠白色脂肪米色化和产热的作用机制。方法:8周龄雄性C57BL/6J小鼠适应性饲养1周后,给予高脂饮食(high-fat diet,HFD)12周构建肥胖小鼠模型。将50只小鼠随机分为对照组(低脂饮食)、HFD组、模型组(HFD+寒冷刺激)、激动剂组(HFD+寒冷刺激+α7 nAChR激动剂GTS-21)和抑制剂组(HFD+寒冷刺激+α7 nAChR抑制剂α-银环蛇毒素),每组10只,干预4周。ELISA法检测血清总胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)和游离脂肪酸(free fatty acid,FFA)水平,以及白色脂肪组织中肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)、白细胞介素1β(interleukin-1β,IL-1β)、Toll样受体4(Toll-like receptor 4,TLR4)、核苷酸结合寡聚化结构域样受体蛋白3(nucleotide-binding oligomerization domain-like receptor protein 3,NLRP3)、IL-10、转化生长因子β(transforming growth factor-β,TGF-β)、环磷酸腺苷(cyclic adenosine monophosphate,cAMP)和去甲肾上腺素(norepinephrine,NE)水平;HE染色观察白色脂肪组织形态学变化;RT-qPCR检测白色脂肪组织中一氧化氮合酶2(nitric oxide synthase 2,NOS2)、精氨酸酶1(arginase 1,Arg1)、解偶联蛋白1(uncoupling protein 1,UCP1)、过氧化物酶体增殖物激活受体γ辅激活因子1α(peroxisome proliferator-activated receptorγcoactivator-1α,PGC-1α)和含PR结构域蛋白16(PR domain-containing 16,PRDM16)的mRNA表达水平;免疫组化检测白色脂肪组织中CD86、CD206、C/EBP同源蛋白(C/EBP homologous protein,CHOP)、免疫球蛋白重链结合蛋白(immunoglobulin heavy-chain binding protein,BiP)和UCP1表达水平;Western blot检测白色脂肪组织中CHOP、BiP、UCP1、α7 nAChR、核因子κB(nuclear factor-κB,NF-κB)p65、磷酸化Janus激酶2(phosphorylated Janus kinase 2,p-JAK2)和磷酸化信号转导及转录激活因子3(phosphorylated signal transducer and activator of transcription 3,p-STAT3)的蛋白水平。结果:与对照组比较,HFD组白色脂肪平均脂滴面积显著增大(P<0.01),白色脂肪组织中TLR4、NLRP3、TNF-α、IL-1β、BiP、CHOP和NOS2水平显著上升(P<0.01),TGF-β和IL-10水平及Arg1的mRNA表达水平显著下降(P<0.01);与HFD组比较,模型组血清TC、TG和FFA水平进一步升高(P<0.05或P<0.01),白色脂肪组织中TLR4、NLRP3、TNF-α和IL-1β水平显著升高(P<0.05或P<0.01),TGF-β和IL-10水平显著下降(P<0.05或P<0.01);与模型组比较,激动剂组脂滴面积显著缩小(P<0.05),血清TC、TG和FFA水平显著降低(P<0.01),白色脂肪组织中cAMP和NE水平及UCP1和PRDM16的mRNA表达水平显著升高(P<0.01),TLR4、NLRP3、TNF-α和IL-1β水平显著降低(P<0.01),TGF-β和IL-10水平显著升高(P<0.01),BiP和CHOP表达水平显著降低(P<0.05),NOS2的mRNA表达水平显著下降(P<0.05),Arg1的mRNA表达水平显著升高(P<0.01),p-STAT3和p-JAK2蛋白水平显著升高(P<0.05或P<0.01)。结论:激活α7 nAChR与寒冷刺激的联合作用不仅促进了肥胖小鼠白色脂肪的米色化,还缓解其代谢紊乱,进一步提高了白色脂肪的米色化产热效能。 展开更多
