Compact calli derived from immature spikelet of a foxtail millet variety—Jigu 11cann’t be directly used for protoplast isolation because of its firm physical structure,and must beloosened with subculturing in M<s...Compact calli derived from immature spikelet of a foxtail millet variety—Jigu 11cann’t be directly used for protoplast isolation because of its firm physical structure,and must beloosened with subculturing in M<sub>1</sub>,M<sub>2</sub> and M<sub>3</sub> media successively and altering these media compo-sitions.The loosened calli can be selected from the regulation and used for protoplast isolationsuccessfully.Rate of protoplast division in KM<sub>8</sub>P medium was 12.3—33.5%.Calli derivedthrough protoplast division are loose and cann’t be used directly for plan regeneration because ofits soft physical structure.When they were subcultured in N<sub>6</sub>—1,N<sub>6</sub>—2,N<sub>6</sub>—3 and N<sub>6</sub>—4 media,in which the media compositions were changed,the compact calli were obtained and 129 plantletswere regenerated from them.101 plants,which grew to maturity after transplanting the plantletsinto field,exhibited sterility in some degree.Most of the subsequent lines derived from the regen-erated plants were sterile and only two lines could get normal reproduction.展开更多
Plasma membrane vesicles were prepared from the developing cotyledons of common beau (Phaseolus vulgaris L cv Diyundou) by aqueous two-phase partitioning and characterized as to their purity by assaying marker enzymes...Plasma membrane vesicles were prepared from the developing cotyledons of common beau (Phaseolus vulgaris L cv Diyundou) by aqueous two-phase partitioning and characterized as to their purity by assaying marker enzymes for other membranes.The putative plasma membrane fraction was minimally contaminated by membranes Other than plasma membrane and hence was of high purity. It exhibited a Ca2+dependent ATPase activity, which was inhibited by 1μ mol/L EB and promoted by calcium ionophore A23187. Such an activity was responsible for the observed ATPdependent 45Ca2+ uptake into inside-out plasma membrane vesicles. This process was stimulated by 0.6μmol/L CaM and 20μmol/L IAA but inhibited by 2μmol/L ABA and abolished by A23187. Possible role of cytoplasmic Ca2+ in mediating phytohormones activity is discussed.展开更多

Under the synchronous propulsion of new four modernizations and the coordinated development of urban and rural areas,the important issues we face are to protect the Chinese traditional villages,inherit the agricultural heritage,and develop the farming culture resources. There are 60 Chinese traditional villages in the Wuling Mountain area. This article introduces the Chinese traditional villages in the area,analyzes its Chinese traditional village protection and beautiful village construction,and puts forward corresponding recommendations.展开更多

Besides the statistics of its vigor of germination, the M1 root-tip cells of these broad beans were systematically analyzed on their changes in mitotic percentage, morphology and behavior of chromosomes, along with the structure o f cytoskeletons, including microtubule and intermediate filament. Based on all results of these studies, our opinions have been expressed in the report on the mechanism of low-energy N+ beams effecting on higher dicotyledons such as broad beau.展开更多
