Background:This research aims to investigate potential gene targets and mechanisms through which the traditional Chinese medicine(TCM)Bupleuri Radix(Chaihu)may impact on clear cell renal cell carcinoma(ccRCC)treatment.Methods:Public databases were employed to identify Cedrenol,an active component of Bupleuri Radix,and its associated gene targets.Among these,the gene RORC(also known as RORγ)was selected for detailed analysis due to its high expression correlation.Expression levels of RORC across various cancers were assessed using the TCGA pan-cancer dataset.Further analyses,including differential expression,prognostic relevance,immune infiltration,single-gene,and functional enrichment analyses,were conducted using the TCGA ccRCC dataset.Additionally,potential drug sensitivities and molecular docking interactions with RORC were explored using the GSCALite and CellMiner databases.The effects of RORC on ccRCC were also validated through cellular experiments.Results:RORC exhibited elevated expression in clear cell renal carcinoma tissue in contrast to normal tissues,and lower RORC expression was related to better prognosis.Immune cell infiltration analysis suggested that RORC may influence the penetration of cells that inhibit immune responses,such as regulatory T cells,thereby affecting ccRCC progression.Furthermore,molecular docking studies revealed that several drugs,including Axitinib,Docetaxel,Methotrexate,and Temsirolimus,have a high affinity for RORC and exhibit strong molecular binding.In cellular experiment experiments confirmed that RORC knockdown led to reduced proliferation,metastasis,and ccRCC cell invasion.Conclusion:The study implies that RORC may be a potential gene target for Bupleuri Radix in the management of ccRCC. 展开更多

To facilitate the species identification and quality assessment of Chaihu (Bupleuri Radix), a simple and valid chromatographic fingerprint method was developed. The method uses high-performance liquid chromatography coupled with evaporative light scattering detector (HPLC-ELSD) and the data analysis is assisted by professional analytical software recommended by the State Food and Drug Administration (SFDA). The results indicate that Nan Chaihu raw materials and Chaihu decoction pieces vary markedly in chemical quality, while Bei Chaihu raw materials are relatively more stable. Furthermore, it is obvious that Nan Chaihu is chemically very different from Bei Chaihu, suggesting that Nan Chaihu may not be suitable for medicinal use. In addition, the obvious differences between the chromatograms of decoction pieces and raw materials, especially the peaks between 30 and 40 rain and after 45 rain, indicate possible effects of the processing procedures on the chemicals. By analyzing the fingerprints of all samples, 12 main saponin-like fingerprint peaks, of which at least three are characteristic peaks of saikosaponins a, c, and d, are proposed to be considered for further characterization and quality evaluation of Chaihu. 展开更多

Active components of Bupleuri Radix in the treatment of schizophrenia analyzed by network pharmacology and clinical verification

Background:Bupleuri Radix is a common Chinese medicinal material in traditional Chinese medicine.Currently,the therapeutic effect of treating schizophrenia is relatively well understood.However,there are fewer studies examining the underlying mechanisms of its treatment.The objective of the study was to investigate the primary mechanisms of Bupleuri Radix in treating schizophrenia through network pharmacology and clinical validation.Method:Network pharmacology revealed possible molecular mechanisms,followed by clinical verification.Sixty-seven schizophrenia patients undergoing treatment at the Hunan Brain Hospital between October and November 2022 were recruited and randomly divided into the olanzapine group and the olanzapine+Bupleuri Radix group.Additionally,32 healthy people undergoing physical examinations during the same period were included as the control group.The patient’s positive and negative symptom scale scores were compared.qPCR was used to detect the mRNA expression levels of ESR1,mTOR,EIF4E,and SMAD4 in peripheral blood.Results:Through network pharmacological analysis,it was concluded in this study that Bupleuri Radix might regulate the mTOR,PI3K-Akt,and HIF-1 signaling pathways.Clinical experiments indicated that compared with before treatment,the positive and negative symptom scale scores and total scores of the two treatment groups were significantly decreased after treatment(P<0.01).In addition,the positive and negative symptom scale scores and total scores in the olanzapine+Bupleuri Radix group were significantly decreased(P<0.01)compared to the olanzapine group after treatment.Before treatment,ESR1 mRNA expression levels in peripheral blood were significantly higher in the two treatment groups than in the control group,whereas the mRNA expression levels of mTOR,EIF4E,and SMAD4 in peripheral blood were significantly lower(P<0.01).The mRNA expression levels of mTOR,EIF4E,and SMAD4 in peripheral blood were significantly higher after therapy than before treatment,whereas the mRNA expression levels of ESR1 in peripheral blood were significantly lower(P<0.01).After therapy,the olanzapine+Bupleuri Radix group’s mRNA expression levels of mTOR,EIF4E,and SMAD4 were significantly higher than those of the olanzapine group,whereas the mRNA expression levels of ESR1 were significantly lower(P<0.01).Conclusion:The mechanism of Bupleuri Radix’s therapeutic efficacy in schizophrenia may involve the up-regulation of mTOR,EIF4E,and SMAD4 mRNA expression and the down-regulation of ESR1 mRNA expression in peripheral blood. 展开更多

Radix Bupleuri(RB)is commonly used to treat depression,but it can also lead to hepatotoxicity after longterm use.In many anti-depression prescriptions,RB is often used in combination with Radix Paeoniae Alba(RPA)as an herb pair.However,whether RPA can alleviate RB-induced hepatotoxicity remain unclear.In this work,the results confirmed that RB had a dose-dependent antidepressant effect,but the optimal antidepressant dose caused hepatotoxicity.Notably,RPA effectively reversed RB-induced hepatotoxicity.Afterward,the mechanism of RB-induced hepatotoxicity was confirmed.The results showed that saikosaponin A and saikosaponin D could inhibit GSH synthase(GSS)activity in the liver,and further cause liver injury through oxidative stress and nuclear factor kappa B(NF-kB)/NOD-like receptor thermal protein domain associated protein 3(NLRP3)pathway.Furthermore,the mechanisms by which RPA attenuates RBinduced hepatotoxicity were investigated.The results demonstrated that RPA increased the abundance of intestinal bacteria with glycosidase activity,thereby promoting the conversion of saikosaponins to saikogenins in vivo.Different from saikosaponin A and saikosaponin D,which are directly combined with GSS as an inhibitor,their deglycosylation conversion products saikogenin F and saikogenin G exhibited no GSS binding activity.Based on this,RPA can alleviate the inhibitory effect of saikosaponins on GSS activity to reshape the liver redox balance and further reverse the RB-induced liver inflammatory response by the NFkB/NLRP3 pathway.In conclusion,the present study suggests that promoting the conversion of saikosaponins by modulating gut microbiota to attenuate the inhibition of GSS is the potential mechanism by which RPA prevents RB-induced hepatotoxicity. 展开更多

We investigated the potential hepatoprotective effect of Radix Bupleuri(RB) by inducing acute liver injury(ALI) in an animal model using acetaminophen(APAP) after pretreatment with RB aqueous extract for three consecutive days. Compared to those of the APAP group, the biochemical and histological results of the RB pretreatment group showed lower serum aspartate transaminase(AST) and alanine transaminase(ALT) levels as well as less liver damage. Pharmacokinetic study of the toxicity related marker acetaminophen-cysteine(APC) revealed a lower exposure level in rats, suggesting that RB alleviated APAP-induced liver damage by preventing glutathione(GSH) depletion. The results of cocktail approach showed significant inhibition of CYP2 E1 and CYP3 A activity. Further investigation revealed the increasing of CYP2 E1 and CYP3 A protein was significantly inhibited in pretreatment group,while no obvious effect on gene expression was found. Therefore, this study clearly demonstrates that RB exhibited significant protective action against APAP-induced acute live injury via pretreatment, and which is partly through inhibiting the increase of activity and translation of cytochrome P450 enzymes, rather than gene transcription. 展开更多

OBJECTIVE: To analyze the essential oils from flowers, leaves, stems, roots, and fruits of Chaihu(Radix Bupleuri Chinensis).METHODS: We extracted essential oils from different parts of Chaihu(Radix Bupleuri Chinensis) using a steam distillation method. The essential oils were analyzed by gas chromatography-mass spectrometry(GC-MS). Data were collected in full scan mode(m/z 60-600). Volatile components were identified based on their retention indices and by comparing their mass spectra with those in the National Institute of Standards and Technology 2005 database assisted by tandem mass spectrometry information. The relative content of each constituent wasdetermined by area normalization.RESULTS: We identified 111 components, of which12 were common to all 5 parts, 30 were found only in roots, 14 were found only in flowers, 6 were found only in leaves, 4 were found only in stems,and 17 were found only in fruits.CONCLUSION: Our results show that the stems,flowers, leaves, and fruits of Chaihu(Radix Bupleuri Chinensis)contain a high concentration of essential oils, and that the exact composition of the essential oils differs among the plant parts. To develop new medicines and make full use of the Chaihu(Radix Bupleuri Chinensis)resource, it is important to characterize the essential oils from different parts of the plant. In future research, it will be important to determine the pharmacological effects of the various components and the essential oil mixtures. 展开更多

OBJECTIVE:To investigate the bioactive compounds of Chaihu(Radix Bupleuri Chinensis)(RB)on glaucomatous optic atrophy(GOA),and to study the pharmacological mechanism.METHODS:We collected information on the bioactive compounds of RB from the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP).Targets related to bioactive compounds and GOA were also obtained.Gene Ontology(GO),Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway and network analyses were performed to investigate the potential mechanism of RB against GOA.Subsequently,the main bioactive compounds of RB and targets of GOA were docked by Autodock software.Moreover,a GOA model of retinal ganglion cells(RGCs)induced by cobalt chloride was established to verify the effect of RB on GOA.RESULTS:There were 17 main bioactive compounds and 46 key targets were screened as potential players in GOA.The compound-target network mainly contained 17 compounds and 46 corresponding targets,and the key targets consisted of interleukin-6(IL-6),hypoxia inducible factor-1α(HIF1A),Caspase-3,estrogen receptor alpha(ESR1),MYC proto-oncogene(MYC),and vascular endothelial growth factor A(VEGFA).Forty-nine significantly enriched GO terms,and 134 KEGG signaling pathways were identified(P<0.05),including HIF-1,tumor necrosis factor,VEGF,prolaction,and other signaling pathways.Molecular docking results showed that the main bioactive compounds of RB exhibited the strongest binding activity with IL-6.Furthermore,experimental validation showed that the RB extract inhibited the activity and promoted apoptosis of RGCs in a dose-dependent manner.The RB extract also suppressed the expression of Bax,Caspase-3,and Caspase-9 and regulated malonaldehyde,superoxide dismutase,and glutathione peroxide by inhibiting the IL-6/HIF-1αsignaling pathway.CONCLUSIONS:The present study provided insights into the mechanism of RB on GOA.RB mainly reverses GOA by inhibiting the IL-6/HIF-1αsignaling pathway. 展开更多

Objective: To explore the mechanism of Radix Bupleuri Treatment of Breast Cancer through network pharmacology. Methods: TCMSP Database was used and related literature was collected to screen out the active constituents of Radix Bupleuri. Multiple databases were used to search the targets of the constituents and the disease. Next, a visual map of the compound-target-path network were constructed. The protein-protein interaction network was visually analyzed. Finally, the enrichment analysis of GO biological process and pathway enrichment analysis were carried out. Results: Active compounds of Radix Bupleuri related to disease targets have been obtained,and they play therapeutic roles for breast cancer through mainly regulating target proteins such as PTGS2, NOS2, AR and ESR. Meanwhile, the active compounds of Radix Bupleuri have an impact on biological processes such as steroid receptor activity and endocrine resistance, platinum resistance, breast cancer-related signaling pathways. Hence, it plays a role in the treatment of breast cancer. Conclusion: The results of this study preliminarily validate the target and mode of Radix Bupleuri in the treatment of Breast Cancer, and lay a foundation for further revealing its mechanism of action. 展开更多

目的 通过分析醋润蜜麸炒柴胡炮制过程中成分差异,探究建昌帮特色炮制方法对柴胡成分变化的影响,筛选差异性成分,为揭示其炮制机制奠定基础。方法 采用超高效液相色谱-线性离子阱-静电场轨道阱质谱联用(UPLC-LTQ-Orbitrap MS)技术分析... 目的 通过分析醋润蜜麸炒柴胡炮制过程中成分差异,探究建昌帮特色炮制方法对柴胡成分变化的影响,筛选差异性成分,为揭示其炮制机制奠定基础。方法 采用超高效液相色谱-线性离子阱-静电场轨道阱质谱联用(UPLC-LTQ-Orbitrap MS)技术分析生柴胡和醋润蜜麸炒柴胡及其炮制过程中的化学成分,采用SIMCA14.1软件对样品数据进行主成分分析(principal component analysis,PCA)及正交偏最小二乘法-判别分析(orthogonal partial least squares-discriminant analysis,OPLS-DA),以变量重要性投影(variableimportanceprojection,VIP)值>1为指标筛选差异性成分。结果 从生柴胡中鉴定出63个化学成分,从醋润蜜麸炒柴胡中鉴定出66种化学成分,包括柴胡皂苷类、黄酮类、有机酸类、香豆素类及其他类,筛选出炮制前后差异性成分21种[黄芩苷、L-(-)-阿拉伯糖醇、葡萄糖酸、芹菜素、柠檬酸、L-(-)-苹果酸、(15Z)-9,12,13-三羟基-15-十八碳烯酸、柴胡皂苷a、壬二酸、柴胡皂苷b2、12,13-二羟基-9-十八碳烯酸、D-(-)-奎宁酸、反-6-羟基十八碳-4-烯酸、(±)-9-氢过氧基十八碳-10,12-二羧酸、罗汉松脂素、2″-O-乙酰柴胡皂苷a、绿原酸、9,10,13-三羟基-11-十八碳烯酸、柴胡次皂苷f、柴胡皂苷d和汉黄芩素]。通过峰面积比较发现,芹菜素、L-(-)-苹果酸、(15Z)-9,12,13-三羟基-15-十八碳烯酸、壬二酸、柴胡皂苷b2、12,13-二羟基-9-十八碳烯酸、反-6-羟基十八碳-4-烯酸、(±)-9-氢过氧基十八碳-10,12-二羧酸、绿原酸、9,10,13-三羟基-11-十八碳烯酸、柴胡次皂苷f和汉黄芩素相对含量升高,黄芩苷、L-(-)-阿拉伯糖醇、柴胡皂苷a、罗汉松脂素、2″-O-乙酰柴胡皂苷a和柴胡皂苷d相对含量降低,葡糖酸、柠檬酸和D-(-)-奎宁酸为炮制后新增成分。柴胡炮制过程中,经分析发现柴胡生品、醋润后、蜜麸炒后差异显著,共筛选出23个差异性成分[柠檬酸、葡萄糖酸、(15Z)-9,12,13-三羟基-15-十八碳烯酸、黄芩苷、9,10,13-三羟基-11-十八碳烯酸、罗汉松脂素、柴胡皂苷a、芹菜素、(±)-9-氢过氧基十八碳-10,12-二羧酸、异鼠李素、柴胡次皂苷f、12,13-二羟基-9-十八碳烯酸、马来酸、绿原酸、麻黄脂肪酸f、2,4-二羟基-2,3-双[(4-羟基-3-甲氧苯基)甲基]丁酸、柴胡皂苷d、3-羟基-3,5,5-三甲基-4-(3-氧代-1-丁烯-1-亚基)环己基-I(2)-D-葡萄糖硫苷、鸟苷、柴胡皂苷e、15,16-二羟基-10,13-十八碳二烯酸、D-(+)-阿拉伯糖醇和1-O-[(3β,5ξ,9ξ)-3-(β-D-葡糖基)-27-羟基-27,28-二氧代齐墩果-12-烯-28-基]-β-D-葡萄糖]。结论 柴胡经醋润蜜麸炒后,皂苷类成分发生转换,酚酸类成分种类和含量增多,刺激性成分含量降低,且不同炮制阶段成分差异较为显著,为深入研究醋润蜜麸炒柴胡的炮制作用机制奠定基础。 展开更多

运用网络药理学和分子对接技术分析柴胡抗氧化应激的作用机制,通过网络药理学方法分析得到柴胡抗氧化应激的主要活性成分与核心靶点、柴胡与氧化应激的交集靶点以及可能的信号通路,预测柴胡主要活性成分与氧化应激核心靶点的结合能力。... 运用网络药理学和分子对接技术分析柴胡抗氧化应激的作用机制,通过网络药理学方法分析得到柴胡抗氧化应激的主要活性成分与核心靶点、柴胡与氧化应激的交集靶点以及可能的信号通路,预测柴胡主要活性成分与氧化应激核心靶点的结合能力。结果显示:从柴胡中筛选得到14种活性成分及378个作用靶点,后与1 934个氧化应激相关靶点取交集,得到191个共同靶点。基因本体(Gene Ontology,GO)功能和京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomes,KEGG)通路富集分析发现,柴胡的抗氧化应激作用主要通过影响癌症通路、阿尔茨海默病通路、磷脂酰肌醇-3-激酶/蛋白激酶B信号通路、Rap1信号通路、白细胞介素-17信号通路等实现。分子对接结果显示,从柴胡中筛选出的主要活性成分与抗氧化应激核心靶点间均具有较好的结合活性。综上,柴胡可通过多成分、多靶点、多通路调节机体氧化应激。 展开更多

抑郁症是当今社会一种高发、高致残、高经济负担的慢性精神类疾病,以情绪低落、快感缺失为核心症状,其病因及发病机制尚未明确,症状甚广,病情持续,严重损害患者身心健康。西药目前以对症治疗为主,但是存在诸多不良反应及使用禁忌。中医... 抑郁症是当今社会一种高发、高致残、高经济负担的慢性精神类疾病,以情绪低落、快感缺失为核心症状,其病因及发病机制尚未明确,症状甚广,病情持续,严重损害患者身心健康。西药目前以对症治疗为主,但是存在诸多不良反应及使用禁忌。中医药治疗以解郁为主,常伴疏肝,要药为柴胡,柴胡类方是指以柴胡为君或重用柴胡的一类方剂,进而柴胡类方广泛应用于抗抑郁领域。前期研究从动物实验、临床研究及现代药理研究等方面证实柴胡类方多途径、多层面抗抑郁疗效确切,但缺乏较为全面系统地总结与分析,故该文专注于对柴胡类方在抗抑郁治疗中的临床应用及其作用机制进行相关文献的分析和综述。结果表明,柴胡类方如小柴胡汤、逍遥散、四逆散、柴胡疏肝散、柴胡加龙骨牡蛎散等抗抑郁机制与调节单胺类神经递质和脑源性神经营养因子(BDNF),抑制炎症反应,调节肠道菌群及下丘脑-垂体-肾上腺(HPA)轴,相关信号通路等作用相关,应用于临床能有效改善患者临床症状,减轻抑郁程度,降低中医证候积分,并且能够降低使用西药的不良反应发生率,有望发挥中医药治疗抑郁症优势,为患者提供安全有效、更具个体化的新型治疗方案。在此基础上分析当前研究存在的不足及未来发展趋势,以期为抗抑郁新药研发提供循证医学依据。 展开更多

慢性心力衰竭(CHF)作为多种心血管疾病的终末阶段,具有病程长、病情反复的特点。CHF伴焦虑抑郁属于“双心疾病”范畴,研究发现焦虑抑郁与心血管事件发生风险和心衰死亡率升高等不良结果密切相关,其复杂病机导致现代医学治疗面临疗效局... 慢性心力衰竭(CHF)作为多种心血管疾病的终末阶段,具有病程长、病情反复的特点。CHF伴焦虑抑郁属于“双心疾病”范畴,研究发现焦虑抑郁与心血管事件发生风险和心衰死亡率升高等不良结果密切相关,其复杂病机导致现代医学治疗面临疗效局限与不良反应并存的问题。该文以少阳枢机理论为核心,阐明少阳枢机不利引起的气化失司、水液代谢障碍、相火失布是CHF伴焦虑抑郁的重要病机。和解少阳的经典名方——柴胡类方(小柴胡汤、柴胡桂枝汤、柴胡加龙骨牡蛎汤)通过和解枢机,能达到气化复衡、通阳利水、输布相火的作用,有效改善CHF伴焦虑抑郁气滞血瘀、水饮内停及痰火扰心等病理状态,从而缓解双心共病症状。机制研究表明,柴胡类方具调节神经递质、抑制炎症反应、调节脂质代谢、保护心肌细胞及改善内皮功能等多靶点效应。该研究基于少阳枢机理论阐释CHF伴焦虑抑郁的中医病机,深入分析柴胡类方的疗效原理和作用机制,为优化CHF伴焦虑抑郁的中医诊疗方案提供理论依据和治疗参考。 展开更多

目的:通过鳖血柴胡的指纹图谱,分析鳖血柴胡治疗阴虚药效活性并进行谱效关系研究。方法:采用高效液相色谱法,利用《中药色谱指纹图谱相似度评价系统(2012版)》绘制15批鳖血柴胡的指纹图谱,并进行相似度评价;采用甲状腺素致小鼠阴虚模型... 目的:通过鳖血柴胡的指纹图谱,分析鳖血柴胡治疗阴虚药效活性并进行谱效关系研究。方法:采用高效液相色谱法,利用《中药色谱指纹图谱相似度评价系统(2012版)》绘制15批鳖血柴胡的指纹图谱,并进行相似度评价;采用甲状腺素致小鼠阴虚模型,结合血清生化指标三碘甲状腺原氨酸(T3)、小鼠甲状腺素(T4)、环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)水平的变化,运用灰色关联度法联合偏最小二乘法分析其治疗阴虚作用成分与药效之间的关联性。以谱效关系筛选的成分为研究对象,通过中药系统药理学(TCMSP)、ETCM及TCMIP数据库检索活性成分靶点,联合在线人类孟德尔遗传数据库(OMIM)、GeneCards、TTD、DisGeNET、Drugbank等数据库,建立鳖血柴胡药效物质-抗阴虚靶点网络,分析作用网络,对核心靶点进基因本体(GO)分析和京都基因与基因组百科全书(KEGG)通路富集分析,并通过实时荧光定量聚合酶链式反应(Realtime PCR)对潜在关键通路和机制进行验证。结果:15批鳖血柴胡指纹图谱中,对鳖血柴胡指纹图谱与对照指纹图谱相似度组内对比发现,相似度在0.976~0.999;与模型组比较,给药组T3、T4水平均升高,而cAMP、cGMP、cAMP/cGMP水平均降低;灰色关联度法分析结果表明,按照关联度由大到小排序依次为柴胡皂苷B1>柴胡皂苷B2>柴胡皂苷C>柴胡皂苷D>柴胡皂苷A;偏最小二乘法分析发现柴胡皂苷A、柴胡皂苷D、柴胡皂苷B1、柴胡皂苷B2的VIP值较大。网络药理学发现共获得30个交集靶点。对30个靶点蛋白进行GO功能和KEGG通路富集分析,共得到276条GO条目和115条KEGG相关条目。Real-time PCR结果表明,与模型组比较,空白组和给药组的胱天蛋白酶(Caspase)-9、酪氨酸激酶受体(KDR)、哺乳动物雷帕霉素靶蛋白(mTOR)mRNA表达上调;鼠双微基因2(MDM2)的mRNA表达下调。结论:鳖血柴胡对阴虚证有治疗作用,可为鳖血柴胡的研究提供新的思路。 展开更多

目的 探讨柴胡的抗抑郁作用及其分子机制。方法 采用慢性不可预测温和应激法(CUMS)联合孤养法建立大鼠抑郁模型,将36只SD大鼠随机分为空白对照组、模型组、柴胡低剂量组、柴胡中剂量组、柴胡高剂量组及氟西汀组,每组6只。比较各组的体... 目的 探讨柴胡的抗抑郁作用及其分子机制。方法 采用慢性不可预测温和应激法(CUMS)联合孤养法建立大鼠抑郁模型,将36只SD大鼠随机分为空白对照组、模型组、柴胡低剂量组、柴胡中剂量组、柴胡高剂量组及氟西汀组,每组6只。比较各组的体重、蔗糖偏好百分比、旷场实验得分、血清皮质酮(CORT)、促肾上腺皮质激素(ACTH)、促肾上腺皮质激素释放激素(CRH)水平及海马组织糖皮质激素受体(GR)、盐皮质激素受体(MR)、谷氨酸(Glu)、γ-氨基丁酸(γ-GABA)含量。结果 第28天,模型组大鼠的体重、蔗糖偏好百分比、旷场实验得分低于空白对照组(P<0.05)。第49天,柴胡各剂量组及氟西汀组的体重、蔗糖偏好百分比、旷场实验得分高于模型组(P<0.05)。柴胡中剂量组与氟西汀组的CORT水平低于模型组(P<0.05);柴胡高剂量组与氟西汀组的ACTH水平低于模型组(P<0.05);柴胡各剂量组及氟西汀组的CRH水平低于模型组,但差异无统计学意义(P>0.05)。柴胡中剂量组、柴胡高剂量组的GR含量高于模型组(P<0.05);柴胡各剂量组及氟西汀组的MR含量低于空白对照组、模型组,但差异无统计学意义(P>0.05)。柴胡各剂量组及氟西汀组的Glu含量低于模型组,γ-GABA含量高于模型组(P<0.05)。结论 柴胡通过下调CUMS模型大鼠血清中的CORT、ACTH和CRH水平,提高海马组织GR表达,并调节Glu与γ-GABA等神经递质的平衡,以发挥抗抑郁作用。 展开更多
