The purpose of this study is to analyze factors affecting the purchasing behavior of low-income Vietnamese on e-commerce platforms.700 questionnaires were dispersed among universities,coffee shops,and Facebook community groups in Vietnam.Respondents to the questionnaires were consumers of those places in Vietnam,which were randomly selected.Finally,SEM analysis was used on the data to test the hypotheses of the study.The study identified that“Subjective norms”and“Consumer attitudes”have no effect on shopping behavior on e-commerce platforms,while“Perceived behavioral control”is most affected and“Perceived risks”are least affected on shopping behavior. 展开更多

Combined with elastic network model(ENM),the perturbation response scanning(PRS)has emerged as a robust technique for pinpointing allosteric interactions within proteins.Here,we proposed the PRS analysis of drug-target networks(DTNs),which could provide a promising avenue in network medicine.We demonstrated the utility of the method by introducing a deep learning and network perturbation-based framework,for drug repurposing of multiple sclerosis(MS).First,the MS comorbidity network was constructed by performing a random walk with restart algorithm based on shared genes between MS and other diseases as seed nodes.Then,based on topological analysis and functional annotation,the neurotransmission module was identified as the“therapeutic module”of MS.Further,perturbation scores of drugs on the module were calculated by constructing the DTN and introducing the PRS analysis,giving a list of repurposable drugs for MS.Mechanism of action analysis both at pathway and structural levels screened dihydroergocristine as a candidate drug of MS by targeting a serotonin receptor of se-rotonin 2B receptor(HTR2B).Finally,we established a cuprizone-induced chronic mouse model to evaluate the alteration of HTR2B in mouse brain regions and observed that HTR2B was significantly reduced in the cuprizone-induced mouse cortex.These findings proved that the network perturbation modeling is a promising avenue for drug repurposing of MS.As a useful systematic method,our approach can also be used to discover the new molecular mechanism and provide effective candidate drugs for other complex diseases. 展开更多

The objective of this study is to compare the ranking of the quality of e-commerce B2C portals in Brazil according to explicit criteria described in the article with the ranking according to the INFO 100 ranking. Theoretical research is first described, followed by the construction of eight self-established criteria of B2C portal quality measures and identification and compilation of requirements used by the I-Best, Opinia, E-bit and other institutes. After that, a portal quality evaluation is made by the web site content analysis;finally, a comparison of the rankings is presented. The obtained results showed that in spite of the lack of knowledge of the criteria adopted by INFO100 [1], there could be agreement with the criteria adopted in this study, because there was agreement with the results for the Ponto Frio portal. Therefore, it was concluded that the eight criteria used in this study can be useful in a quality analysis of B2C electronic portals. 展开更多

E-commerce to cloud computing as the basic environment, the construction of public cloud services and private cloud resources, based on public cloud to the SaaS way to provide services to customers, focus on business search and business collaboration, make full use of the lnternet, modern communications technology to provide the real- service. The development of agricultural economy, no longer depends only on the number of some traditional agricultural resources, but also depends on the modern technology, information access and use. 目的探究中药丹参有效成分在治疗硅肺中的作用和作用机制。方法运用网络药理学和分子对接技术筛选出中药丹参治疗硅肺的有效成分和关键靶点。热转移实验检测有效成分与关键靶点的结合稳定性,细胞划痕实验检测上皮细胞的迁移能力,蛋白质... 目的探究中药丹参有效成分在治疗硅肺中的作用和作用机制。方法运用网络药理学和分子对接技术筛选出中药丹参治疗硅肺的有效成分和关键靶点。热转移实验检测有效成分与关键靶点的结合稳定性,细胞划痕实验检测上皮细胞的迁移能力,蛋白质印迹法检测上皮间质转化的标志蛋白、纤维化标志蛋白以及凋亡相关蛋白的表达水平。结果中药丹参的有效成分木犀草素与硅肺关键靶点B淋巴细胞瘤-2(BCL-2)具有最低的结合能,热转移实验验证木犀草素与BCL-2蛋白具有较好的结合稳定性。细胞划痕实验显示木犀草素能够抑制转化生长因子-β1刺激的上皮细胞迁移。蛋白质印迹法证明木犀草素能够抑制BCL-2蛋白的表达,与转化生长因子-β1刺激组相比,木犀草素给药及沉默BCL-2后能够抑制上皮间质转化及胶原蛋白的形成,进而抑制纤维化的进程。木犀草素给药及沉默BCL-2后能够促进细胞凋亡。结论木犀草素能够与BCL-2结合进而抑制硅肺纤维化的进程,其作用机制可能与木犀草素能够促进细胞凋亡有关。 展开更多

弥漫大B细胞淋巴瘤R-CHOP化疗的生存预后评估模型 被引量:1

目的建立弥漫大B细胞淋巴瘤(DLBCL)R-CHOP化疗的生存预后评估模型,以指导临床进行风险分层。方法选取2018年1月至2023年1月在金华市中心医院确诊的DLBCL患者210例为研究对象,根据2:1抽样比例随机分为建模集140例和验证集70例。所有患者... 目的建立弥漫大B细胞淋巴瘤(DLBCL)R-CHOP化疗的生存预后评估模型,以指导临床进行风险分层。方法选取2018年1月至2023年1月在金华市中心医院确诊的DLBCL患者210例为研究对象,根据2:1抽样比例随机分为建模集140例和验证集70例。所有患者均接受R-CHOP化疗方案至少4个疗程(21 d为1个疗程),常规随访至2023年6月。根据生存预后分为良好组和不良组。比较两组患者的临床资料[包括性别、年龄、BMI、基础疾病、血清乳酸脱氢酶(LDH)、美国国家综合癌症网络国际预后指数(NCCN-IPI)]、病理特征[包括Hans分型、Ann Arbor分期、原发部位、Ki-67阳性表达率、MYC和B细胞淋巴瘤(Bcl)-2蛋白阳性表达以及骨髓侵犯]、治疗(化疗疗程和并发症)和随访时间。结果建模集中良好组105例和不良组35例,预后不良发生率25.0%;验证集良好组55例和不良组15例,不良发生率21.4%。建模集中不良组年龄大于良好组,LDH水平、NCCN-IPI、Ann Arbor分期Ⅲ~Ⅳ比例、Ki-67阳性表达率、MYC和Bcl-2蛋白双阳性表达率和骨髓侵犯比例均高于良好组,差异均有统计学意义(均P<0.05)。多因素Cox回归分析显示,NCCN-IPI(HR=2.526,95%CI:2.001~3.125,P<0.001)、Ann Arbor分期Ⅲ~Ⅳ(HR=5.021,95%CI:4.125~5.998,P<0.001)、MYC和Bcl-2蛋白双阳性(HR=3.859,95%CI:3.256~4.754,P<0.001)均是DLBCL患者R-CHOP化疗预后不良的危险因素。建立预测模型Y=0.056+1.032×(NCCN-IPI)+1.986×(Ann Arbor分期)+1.434×(MYC和Bcl-2蛋白双阳性)。ROC曲线显示,模型预测建模集与验证集预后不良的AUC分别为0.923和0.866(均P<0.01)。结论DLBCL患者化疗前NCCN-IPI、Ann Arbor分期升高以及MYC和Bcl-2蛋白双阳性表达与R-CHOP化疗预后不良密切相关,通过建立量化预测模型能够辅助临床早期、准确识别预后不良的高危群体,有较好的应用价值。 展开更多

目的:基于磷脂酰肌醇3-激酶(PI3K)/蛋白激酶B(Akt)信号通路探讨黄连知母汤改善2型糖尿病(T2DM)糖脂代谢紊乱及肝脏胰岛素抵抗(IR)的作用及可能的分子机制。方法:采用高脂饮食喂养诱导Goto-Kakizaki(GK)大鼠制备T2DM模型,随机分为正常组... 目的:基于磷脂酰肌醇3-激酶(PI3K)/蛋白激酶B(Akt)信号通路探讨黄连知母汤改善2型糖尿病(T2DM)糖脂代谢紊乱及肝脏胰岛素抵抗(IR)的作用及可能的分子机制。方法:采用高脂饮食喂养诱导Goto-Kakizaki(GK)大鼠制备T2DM模型,随机分为正常组、模型组、二甲双胍组(0.10 g·kg^(-1))、黄连知母汤组(3.60 g·kg^(-1)),每组8只;给予药物连续干预8周,收集各组大鼠血清、肝脏组织。酶联免疫吸附测定法(ELISA)测定大鼠空腹胰岛素(FINS)水平,并计算胰岛素抵抗指数(HOMA-IR);采用全自动生化分析仪测定大鼠总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)水平;苏木素-伊红(HE)染色进行肝组织病理观察;ELISA检测大鼠血清中白细胞介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)含量。结合网络药理学及转录组学测序分析正常组、模型组、黄连知母汤组肝组织差异表达基因(DEGs),并进行京都基因和基因组百科全书(KEGG)通路富集分析获得黄连知母汤干预T2DM差异表达通路。实时荧光定量聚合酶链式反应法(Real-time PCR)检测肝组织胰岛素受体底物-1(IRS-1)、PI3K、Akt、过氧化物酶体增殖物激活受体γ(PPARγ) mRNA表达,蛋白免疫印迹法(Western blot)检测肝组织IRS-1、PI3K、Akt、PPARγ蛋白表达。结果:黄连知母汤干预8周后能够显著改善T2DM大鼠模型多饮、多食、多尿等典型症状(P<0.01),降低T2DM大鼠空腹血糖水平和胰岛素抵抗(P<0.01);病理切片结果显示黄连知母汤有效改善了肝组织的脂肪变性和炎症水肿程度,减少脂质空泡;生化检测结果表明黄连知母汤有效降低了TC、TG、LDL-C水平(P<0.01);ELISA检测结果显示黄连知母汤有效降低血清炎症因子IL-6、TNF-α的含量(P<0.05)。结合网络药理学预测结果及转录组学KEGG通路分析提示黄连知母汤组与模型组比较差异基因显著富集于PI3K/Akt等信号通路。Real-time PCR和Western blot结果确证黄连知母汤可有效上调肝组织中PI3K/Akt信号通路相关mRNA及蛋白表达(P<0.05,P<0.01),发挥抗炎、减少肝脏脂质蓄积、增加胰岛素敏感性的作用。结论:黄连知母汤能有效改善T2DM糖脂代谢紊乱,其机制可能与调控PI3K/Akt通路,改善炎症,减少肝脏脂质沉积,减轻肝脏胰岛素抵抗有关。 展开更多

目的 运用生物信息学、网络药理学及实验验证的方法探究熟地黄Rehmanniae Radix Praeparata治疗2型糖尿病(type 2 diabetes mellitus,T2DM)的作用机制。方法 熟地黄的水提物ig给予T2DM小鼠,并通过超高效液相色谱-高分辨质谱(UPLC-HRMS)... 目的 运用生物信息学、网络药理学及实验验证的方法探究熟地黄Rehmanniae Radix Praeparata治疗2型糖尿病(type 2 diabetes mellitus,T2DM)的作用机制。方法 熟地黄的水提物ig给予T2DM小鼠,并通过超高效液相色谱-高分辨质谱(UPLC-HRMS)对入血及入胰腺成分进行辨识,在瑞士目标预测数据库(Swiss Target Prediction)预测潜在靶点,并与基因表达综合数据库(GeneExpressionOmnibus,GEO)中GSE15932、GSE25724数据集及比较毒物基因组学数据库(Comparative Toxicogenomics Database,CTD)、人类基因数据库(GeneCards)取交集获取共同基因,并在Cytoscape中进行聚类分析,得到熟地黄入血和入胰腺靶点。对熟地黄入血和入胰腺的靶点取交集分析。在ADMETab3.0数据库中进行毒性预测、分子对接及信使核糖核酸(messenger ribonucleic acid,m RNA)-微小核糖核酸(microRNA,miRNA)-长链非编码核糖核酸(long non-coding RNA,lncRNA)互作网络分析,运用基因和蛋白相互作用和功能数据库(GeneMANIA)分析主要生物功能,并结合实时荧光定量PCR实验验证。结果 熟地黄可改善T2DM小鼠异常葡萄糖水平和糖耐受量。熟地黄入血化合物138个,相对应的潜在基因靶点分别为18个和1个聚类;入胰腺化合物为359个,相对应的潜在基因靶点分别为141个和6个聚类。甲状腺素转运蛋白基因(transthyretin,TTR)和ATP结合盒转运蛋白B1(ATP binding cassette subfamily B member 1,ABCB1)为熟地黄入血及胰腺的交集基因靶点;得到包含6个相同miRNA及14个相同lncRNA的m RNA-miRNAlncRNA互作网络。主要生物功能涉及脂质运输和代谢、药物运输、离子和神经递质运输、血管过程及炎症反应的调节等。熟地黄可影响T2DM小鼠血液和胰腺中TTR、ABCB1的m RNA表达水平。结论 TTR和ABCB1可被视为熟地黄治疗T2DM的潜在作用靶点,为T2DM治疗提供依据的同时为后续深入研究熟地黄作用于T2DM的相关问题提供思路。 展开更多

Purpose: This research attempts to examine the relationship between B2C interaction and customer loyalty in Business-to-Customer(B2C) context from a new perspective of the interactive tool.Design/methodology/approach: The scale for B2C interactive tools is of seven dimensions: efficiency, security, fulfillment, mobility, community, cultivation, and customization. A model reflecting the influences of these attributes on customer loyalty is developed and empirically examined based on data collected from 265 B2C customers. Findings: Results reveal that the fulfillment, mobility, community, and customization of B2C interactive tools can enhance customer loyalty directly and significantly. Efficiency and security, serving as the premise for possible purchase behavior, facilitate fulfillment. In addition, cultivation promotes the formation of customization, which directly strengthens customer loyalty.Research limitations: Models considering individual-level indicators and combined with classic loyalty mechanisms in B2C context may lead to a deeper understanding of the tested effects of interaction on customer loyalty.Practical implications: To strengthen B2C interaction and further cultivate loyal customers, making interactive tools more fundamental, flexible, and personalized is critical for B2C enterprises. Originality/value: This study proposes a new perspective from interactive tools when measuring the relationship between B2C interaction and customer loyalty, and offers a useful theoretical lens and reasonable explanations for investigating customer loyalty in B2C e-commerce context. 展开更多

To overcome this barrier, businesses need to implement comprehensive consumer protection systems that protect consumers during every stage of the purchasing process. This paper used the consumer behaviour model as the basis for analysing risks in Bussiness-to-Consumer (B2C) transactions. Four categories of risks were identified: information, agreement, payment and delivery risk. By combining these risk categories with the three dimensions of management, technology and legislation, a comprehensive B2C consumer protection framework is developed. 展开更多

目的:阐明虾青素逆转2型糖尿病(type 2 diabetes mellitus,T2DM)小鼠胰岛素抵抗的作用机制。方法:以高热量饲料喂养小鼠建立T2DM为模型,通过口服葡萄糖耐量实验和胰岛素耐量实验观察虾青素对T2DM小鼠糖耐量和胰岛素抵抗的影响;通过酶联... 目的:阐明虾青素逆转2型糖尿病(type 2 diabetes mellitus,T2DM)小鼠胰岛素抵抗的作用机制。方法:以高热量饲料喂养小鼠建立T2DM为模型,通过口服葡萄糖耐量实验和胰岛素耐量实验观察虾青素对T2DM小鼠糖耐量和胰岛素抵抗的影响;通过酶联免疫吸附试剂盒测定肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)和果糖胺(fructosamine,FRA)的含量。在此基础上,以网络药理学分析结果为指导,通过Western blot检测小鼠肝脏Janus激酶2(Janus kinase 2,JAK2)、信号传导及转录激活蛋白3(signal transducer and activator of transcription 3,STAT3)、核因子κB(nuclear factor kappa B,NF-κB)的表达,观察炎症信号在虾青素逆转胰岛素抵抗中的作用。结果:虾青素可显著降低T2DM小鼠的血糖、TNF-α和FRA水平,改善胰岛功能和胰岛素抵抗;Western blot结果表明虾青素显著抑制了JAK、STAT3、NF-κB蛋白磷酸化水平。结论:虾青素可通过抑制JAK2/STAT3和NF-κB信号通路发挥降糖、逆转胰岛素抵抗的作用。 展开更多

BACKGROUND Ulcerative colitis(UC),a chronic and challenging condition,necessitates the development of more effective treatments owing to the unsatisfactory efficacy and side effects associated with current medications.Traditional Chinese medicine(TCM),known for its multi-stage and multi-targeted approach,has a long history in treating gastrointestinal diseases and offering a promising alternative UC treatment.Panax ginseng(P.ginseng),a commonly used remedy for UC in TCM,exemplifies this potential,although the specific components and mechanisms through which its therapeutic effects are exerted remain to be fully elucidated,highlighting the need for further research to unlock its full potential as a treatment option.AIM To investigate the key constituents and biological pathways through which P.ginseng exerts therapeutic effects on UC.METHODS Network pharmacology investigated the UC-alleviating mechanism of P.ginseng,including“active ingredient-target-disease”network analysis,and Gene Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analyses.Panaxadiol(PD;active ingredient of P.ginseng)was tested in a mouse model of 3%dextran sulfate sodium-induced UC,with assessments of body weight,Disease Activity Index scores,and colon length.Colitis and intestinal barrier integrity were analyzed via hematoxylin-eosin and Alcian blue and periodic acid-Schiff staining,immunohistochemistry,real time-quantitative PCR,and western blotting.RESULTS By integrating and analyzing the targets of P.ginseng and UC,15 critical hub genes were discovered.Kyoto Encyclopedia of Genes and Genomes pathway analysis revealed the mechanisms involved to be linked to MAPK and PI3K-Akt signaling.Among the 10 main active ingredients identified as potentially effective,PD was most abundant and was validated in vivo to mitigate weight loss,reduce Disease Activity Index scores,and prevent colon shortening.PD also reduced inflammation and suppressed expression of pro-inflammatory cytokines and mediators.In addition,PD increased expression of mucin and tight junction proteins.Ultimately,PD counteracted effects of dextran sulfate sodium by inhibiting phosphorylation of NF-кB and MAPK,while increasing phosphorylation of AMPK and expression of NRF2 and NQO1.CONCLUSION PD alleviates colitis and aids intestinal barrier repair,partly via modulation of the MAPK/NF-кB and AMPK/NRF2/NQO1 pathways.These findings also suggest new research methods for treatment of UC with TCM. 展开更多
