Prenatal alcohol exposure disrupts the development of normal fetal respiratory function, but whether it perturbs respiratory rhythmical discharge activity is unclear. Furthermore, it is unknown whether the 5-hydroxytryptamine 2A receptor(5-HT2AR) is involved in the effects of prenatal alcohol exposure. In the present study, pregnant female rats received drinking water containing alcohol at concentrations of 0%, 1%, 2%, 4%, 8% or 10%(v/v) throughout the gestation period. Slices of the medulla from 2-day-old neonatal rats were obtained to record respiratory rhythmical discharge activity. 5-HT2 AR protein and m RNA levels in the pre-B?tzinger complex of the respiratory center were measured by western blot analysis and quantitative RT-PCR, respectively. Compared with the 0% alcohol group, respiratory rhythmical discharge activity in medullary slices in the 4%, 8% and 10% alcohol groups was decreased, and the reduction was greatest in the 8% alcohol group. Respiratory rhythmical discharge activity in the 10% alcohol group was irregular. Thus, 8% was the most effective alcohol concentration at attenuating respiratory rhythmical discharge activity. These findings suggest that prenatal alcohol exposure attenuates respiratory rhythmical discharge activity in neonatal rats by downregulating 5-HT2 AR protein and m RNA levels. 展开更多

BACKGROUND Hepatocellular carcinoma(HCC)is a leading cause of cancer-related mortality worldwide,with hepatitis B virus(HBV)infection serving as a significant etiological factor in endemic regions.Alpha-fetoprotein(AFP),the most commonly used biomarker,has limited sensitivity,particularly in AFP-negative HCC.Recent studies have identified origin recognition complex subunit 1(ORC1)and extra spindle pole bodies-like 1(ESPL1)as promising serum biomarkers,both linked to HBV DNA integration,a mechanism known to drive hepatocarcinogenesis.AIM To assess serum ORC1’s diagnostic value for HBV-HCC and its link to S gene integration.METHODS In this case-control study,479 HBV-infected patients were enrolled,including 20 hepatitis B,154 with HBV-related cirrhosis,and 96 with HBV-HCC.The control group comprised 73 individuals:29 with non-HBV-HCC and 44 healthy participants.Serum ORC1 and ESPL1 were measured by enzyme-linked immunosorbent assay.HBV integration sites were identified via whole-genome sequencing.Diagnostic performance was assessed using receiver operating characteristic analysis,including in AFP-negative patients.RESULTS HBV integration near the ORC1 locus(chromosome 1p32.3)was detected in 71.4%of HBV-HCC tissues.Serum ORC1 levels were significantly higher in HBV-infected patients than in non-HBV-infected controls(980.11 ng/L vs 746.82 ng/L,P<0.05)and in HBV-HCC compared with non-HBV-HCC(1077.07 ng/L vs 749.54 ng/L,P<0.05).Serum ORC1 and ESPL1 were elevated in HBV-HCC regardless of AFP status,and detected 64.8%and 73.2%of AFP-negative cases,respectively.The combined panel of ORC1[Area under receiver operating characteristic curve(AUC)=0.587],ESPL1(AUC=0.776),and AFP(AUC=0.844)achieved an AUC of 0.887,significantly higher than any single marker(P<0.05),with a sensitivity of 84.44%,specificity of 84.19%,and a negative predictive value of 94.91%.CONCLUSION Serum ORC1,driven by HBV integration,is a promising biomarker especially for AFP-negative HBV-HCC.Its combination with ESPL1 and AFP significantly improves early detection. 展开更多

The coordination reactions of Cu(Ⅱ) and Ni(Ⅱ) with acid alizarine blue B (AABB) in the presence of cetyltrimethylammonium bromide (CTAB) micelle were investigated using the microsurface adsorptionspectral correction technique (MSASC). The aggregation of AABB on CTAB followed the Langmuir isothermal adsorption law. The enrichment of AABB on CTAB sensitized the complexation between Cu(Ⅱ) or Ni(Ⅱ)and AABB. The binding ratio of AABB to CTAB was 1:2.5, and monomeric aggregate, AABB2CTAB5, was formed with an adsorption constant of 5.95×10^5 at 20 ℃ or 2.48×10^5 at 40 ℃. In the ternary complexation, the ratio of AABB:Cu and AABB:Ni were 1:1 and 1:2.5, respectively. Two types of aggregates, Cu2.AABB2·CTAB80 and Ni5.AABB2.CTAB80, were formed. 展开更多

The aim of the present study was to evaluate whether tissue levels of vitamin B complex and vitamin B12 were altered after crush-induced peripheral nerve injury in an experimental rat model. A total of 80 male Wistar rats were randomized into one control(n = 8) and six study groups(1, 6, 12, 24 hours, 3, and 7 days after experimental nerve injury; n = 12 for each group). Crush-induced peripheral nerve injury was performed on the sciatic nerves of rats in six study groups. Tissue samples from the sites of peripheral nerve injury were obtained at 1, 6, 12, 24 hours, 3 and 7 days after experimental nerve injury. Enzyme-linked immunosorbent assay results showed that tissue levels of vitamin B complex and vitamin B12 in the injured sciatic nerve were significantly greater at 1 and 12 hours after experimental nerve injury, while they were significantly lower at 7 days than in control group. Tissue level of vitamin B_(12) in the injured sciatic nerve was significantly lower at 1, 6, 12 and 24 hours than in the control group. These results suggest that tissue levels of vitamin B complex and vitamin B12 vary with progression of crush-induced peripheral nerve injury, and supplementation of these vitamins in the acute period may be beneficial for acceleration of nerve regeneration. 展开更多

BACKGROUND Ulcerative colitis(UC)is a chronic,nonspecific intestinal inflammatory disease with undefined pathogenesis.Non-SMC condensin I complex subunit D2(NCAPD2)and non-SMC condensin II complex subunit D3(NCAPD3)play pivotal roles in chromosome assembly and segregation during both mitosis and meiosis.To date,there has been no relevant report about the functional role of NCAPD2 and NCAPD3 in UC.AIM To determine the level of NCAPD2/3 in intestinal mucosa and explore the mechanisms of NCAPD2/3 in UC.METHODS Levels of NCAPD2/3 in intestinal tissue were detected in 30 UC patients and 30 healthy individuals with in situ hybridization(ISH).In vitro,NCM60 cells were divided into the NC group,model group,si-NCAPD2 group,si-NCAPD3 group and si-NCAPD2+si-NCAPD3 group.Inflammatory cytokines were measured by ELISA,IKK and NF-κB were evaluated by western blot,and IKK nucleation and NF-κB volume were analyzed by immunofluorescence assay.RESULTS Compared with expression in healthy individuals,NCAPD2 and NCAPD3 expression in intestinal tissue was significantly upregulated(P<0.001)in UC patients.Compared with levels in the model group,IL-1β,IL-6 and TNF-αin the si-NCAPD2,si-NCAPD3 and si-NCAPD2+si-NCAPD3 groups were significantly downregulated(P<0.01).IKK and NF-κB protein expression in the si-NCAPD2,si-NCAPD3 and si-NCAPD2+si-NCAPD3 groups was significantly decreased(P<0.01).Moreover,IKK nucleation and NF-κB volume were suppressed upon si-NCAPD2,si-NCAPD3 and si-NCAPD2+si-NCAPD3 transfection.CONCLUSION NCAPD2/3 is highly expressed in the intestinal mucosa of patients with active UC.Overexpression of NCAPD2/3 promotes the release of pro-inflammatory cytokines by modulating the IKK/NF-κB signaling pathway. 展开更多

A monitoring system is an important guarantee of safety in a production mine. However, not all monitoring substations pass the electrical fast transient/burst (EFT/B) immunity test and the explosion-proof test simultaneously. To enhance the immunity, the values of capacitance and inductance should be increased, which are actually limited by coal mine explosion-proof standards. Hence, for the first time, an active filter was applied in an electromagnetic interference (EMI) output filter. As a result, the interference within 30 MHz clearly weakened, but the frequency spectrum had a wide range. An EMI input filter and ferrite beads were adopted to restrain higher frequency interference. An output interference spectrogram of the substation was obtained with an analyzer. The results indicate that the improved complex filtering markedly help to control interference. With the support of improved complex filtering and other enhancing immunity means about I/O ports, the substation managed to pass both the EFT/B immunity test and the explosion-proof test synchronously. We conclude that improved complex filtering is of vital importance in enhancing the electromagnetic compatibilitv (EMC) of the coal mine monitoring system. 展开更多

Quantum-chemical Study on the Structures and Properties of Uracil-BX_3 (X = F,Cl) Complexes 被引量:1

For the uracil-BX3 (X = F, Cl) systems, geometries and binding energies have been calculated by using the Lee-Young-Parr correlation functionals (B3LYP) method of density functional theory (DFT) and the second-order Moller-Plesset (MP2) method of ab initio at the 6- 311 +G^* or 6-311 ++G^* basis set. Four isomers were found for each system, and then the single-point energy evaluations were performed using the larger basis sets of (6-311 +G(2df, p) and aug-cc-pVDZ with DFF method. In the most stable isomer of uracil-BF3 or uracil-BCl3, the boron atom of BX3 (X = F, Cl) connects to the carbonyl oxygen O7 of uracil with a stabilization energy of -46.56 or -31.10 kJ/mol at the B3LYP/6-31 1+G^* level (BSSE corrected). The analyses for combining interaction between BX3 and uracil with the atom-in-molecule theory (AIM) and natural bond orbital method (NBO) have been performed. The results indicate that all isomers were formed with σ-p type interactions between uracil and BX3, in which the carbonyl oxygen offers its lone pair electrons to the empty p orbital of boron atom and the concomitances of charge transfer from uracil to BX3 occur. Moreover, there exists one or two hydrogen bonds in most isomers of uracil-BX3 system and these hydrogen bonds contribute to the stability of the complex systems. Frequency analysis suggests that the stretching vibration of BX3 undergoes a red shift in complexes. Uracil-BF3 complex is more stable than uracil-BCl3 although the distance of B-O is shorter in the latter. Besides, the conversion mechanisms between different isomers of uracil-BF3 have been obtained. 展开更多

Complex I crystallizes in the orthorhombic system, space group P212121, with a = 8.9040(18), b = 9.1530(18), c = 24.891(5)A^°, V = 2028.6(7) A^°3, Z = 4, C20H26CuN2O9, Mr = 501.97, Dc = 1.644g·cm^3, μ = 1.135 mm^-1, F(000) = 1044, GOOF = 1.194, the final R = 0.0484 and wR = 0.1420 for 6186 observed reflections (I 〉 2σ(I)). In I, two L^- anions are coordinated to the copper ion in tridentate and bidentate chelating modes, respectively, resulting in the coordinated geometry of copper ion to be a distorted square pyramid. The intermolecular hydrogen bonds between the complexes, complexes and lattice water molecules lead to a 2D supramolecular network. The bioactivity of the complex as a potential PTPs inhibitory agent in vitro was investigated, displaying potent inhibition against PTP1B (IC50, 0.27 μM) and TCPTP (IC50, 0.57 μM) with a moderate selectivity. 展开更多

The top limit of. the experimental temperature range of deuterium oxide solutions was extended from 100℃ to 167 ℃ instead of the previous 100 ℃ by using sealed, thick-walled tubes to observe experimental phenomena further. It was found that another intramolecular process, i. e. B, R (Bonded State, Rotational State) conversion, exists in these complexes formed by polyaminepolycarboxylic ligands in addition to △,A conversion and nitrogen inversion. 目的利用优化的CRISPR/Cas9(Clustered Regularly Interspaced Short Palindromic Repeats,CRISPR)技术,高效建立一种通过递送新型Cas9-gRNA核糖核蛋白复合物(RNP)制备人类重组激活基因1(RAG1)c.946T>G点突变的前B淋巴细胞系的方法,... 目的利用优化的CRISPR/Cas9(Clustered Regularly Interspaced Short Palindromic Repeats,CRISPR)技术,高效建立一种通过递送新型Cas9-gRNA核糖核蛋白复合物(RNP)制备人类重组激活基因1(RAG1)c.946T>G点突变的前B淋巴细胞系的方法,并对突变体进行功能分析。方法实验以人前B淋巴细胞系(Nalm6细胞)为研究对象,在体外将化学修饰的gRNA(guide RNA)和高保真HIFI Cas9蛋白孵育形成RNP复合物,通过细胞核转染技术将RNP和含RAG1(T>G)目标突变与同义突变(C>A)的超分子单链寡核苷酸(Ultramer-ssODNs)同源模板带入细胞内,诱导RAG1基因发生切割与重组,T7EⅠ酶切检测基因编辑效率,并经计数稀释法进行单克隆筛选,Sanger测序鉴定基因型。结果成功构建RAG1(c.946T>G)纯合点突变的细胞系,切割效率高达78.21%,单克隆筛选获得较高比例33.33%(4/12)的目标单克隆。通过功能实验发现c.946T>G点突变导致前B淋巴细胞RAG1、RAG2蛋白表达下调,细胞凋亡比例增多,可能是导致最终成熟B淋巴细胞的生成减少的原因。结论该研究通过优化的Cas9 RNP技术高效获得RAG1基因c.946T>G纯合点突变的前B淋巴细胞系,为单碱基突变疾病模型的建立提供实验依据,更为提升剪切精确性、提高转染效率提供了新的方法。 展开更多

A high performance liquid chromatographic assay has been developed for the simultaneous determination of five water soluble vitamins:thimine, riboflavin, niacin, pyridoxine and folic acid in cereal products and fresh vegetables. Food samples were hydrolyzed in 0.4 mol/L HCl, autoclaved at 120℃ 15 psi for 20 minutes, using a μ-Bondapak column (3.9×300mm, Waters Co.), a mobile phase of methanol-water (30:70) (0.005 mol/L heptanesulfonic acid) and a flow rate of 1.0 ml/min gave the most satisfactory separation of the five water soluble vitamins. A double channel deteetion was used: four vitamins (B_5, folic acid, B_2, B_1) were detected hy UV spectrophotometry (254 nm) first, pyridoxine (B_6) was detected by fluoromelry (EX 290nm, EM 395 nm) afterwards. Detection limits were 2, 5, 5, 2and 5ng, linear ranges were 5-10ng, 5-50ng, 5-40ng, 5-50ng and 10-50ng for B_1, B_2, B_5, B_6 and folic acid respectively. Recoveries were 92-100%(B_5), 51-52% (folio acid), 103—105% (B_2), 99—100% (B_6) and 91.3—102% (B_1) respectively. In comparison with a reference method and checking with food composition tables, very satisfactory results were obtained by this method. 展开更多
