In this study, ori-entation of antibody was retained by protecting the antigen-binding site of the antibody prior to immo-bilization to -functionalized mixed self-assembled monolayer (SAM) of 12-mercaptododecanoic acid and 1-heptanethiol. More importantly, the number of immobilization bonds formed between each an-tigen-binding site protected antibody molecule and the solid surface was controlled by optimizing the mole fraction of the activated carboxyl group of the linker molecules in the mixed SAM. The amount of antibody used in this study was approximately equivalent to the amount for one monolayer surface coverage. The resulting activity of protected immo-bilized antibody was about 10 fold higher than that of random immobilized展开更多
With Mosuo's sacred natural sites of mountainous forest in Yongning-Lugu Lake area as the research subject,by using the method of ecology,the vegetation type in this area was initially identified.The control sampl...With Mosuo's sacred natural sites of mountainous forest in Yongning-Lugu Lake area as the research subject,by using the method of ecology,the vegetation type in this area was initially identified.The control sample plot of the same vegetation type at similar altitude and in similar soil condition would be selected and compared with the sample plot in sacred natural sites at length,so as to quantitatively analyze the protection of biological diversity in the perspective of ecological system.It showed that under the circumstance of the damage of ecological environment,complete natural vertical pattern was preserved thanks to sacred natural sites of mountainous forest established by reason of traditional religion and culture.The results of research and quantitative analysis showed that compared with the control sample plot of the same vegetation type,sacred natural sites were high in biological diversity and the species composition of plant community in sacred natural sites was closer to that of primitive forest.The result of quantitative analysis also indicated that sacred natural sites played a remarkable role in protecting vegetation and this protective function became more remarkable if the altitude was higher.And human were reminded to pay close attention to the preservation and protection function of sacred natural sites to natural ecological system at high altitude.展开更多

This study was undertaken to highli...Two protected sites located on the outskirts of the Sena Oura National Park (PNSO) in West Mayo-Kebbi cover an area of 1800 m2 is pattern choose in pastoral enclave in both village. This study was undertaken to highlight the floristic diversity of the herbaceous and woody vegetation on these sites in the offing to know the ability of charge into UBT that most support the park peripheral. It took place on two experimental sites on a natural course in two villages: Wazetelan and Massang. The approach used for the study was a survey using the systematic sampling method and a 30 m × 30 m plot. The survey consisted in listing all the taxa in a floristically homogeneous plot, each assigned the Braun-Blanquet dominance abundance coefficient. These surveys revealed a floristic richness of 73 species, 58 genera, including 46 herbaceous and 26 woody species. The Shannon result gives H1' = 0.12 bit for herbaceous species and H2' = 0.44 bit for woody species, meaning that the herbaceous and woody populations of all the surveys have a very low species diversity, as H' < 3 according to the Shannon index assessment threshold. Herbaceous species are divided into 13 families and 33 genera. The most represented families are Fabaceae (27.3%) and Poaceae (21.9%). Most of the other families (1.3%) have only one species, if any. Herbaceous species are divided into 33 genera grouped into 13 families. The most represented families are Fabaces (16 species), 34%, and Poaceae (12 species), 26%. The 26 woody species, most of which come from itinerant surveys, are distributed across 24 genera and 12 families, the most important of which is Fabaceae with 09 species (34.6%). This floristic assessment, in terms of quantity and quality, has enabled us to estimate the carrying capacity of the two pastoral enclaves in Dari and Goumadji cantons, and to guide the government’s actions with regard to rangeland management.展开更多

In the context of territorial spatial planning,the overall coordinated protection has become a consensus in the field of cultural heritage protection,and the study on the spatial and temporal pattern of trans-regional...In the context of territorial spatial planning,the overall coordinated protection has become a consensus in the field of cultural heritage protection,and the study on the spatial and temporal pattern of trans-regional cultural heritage is of great significance to the overall protection.Based on the cultural relics protection units in Jingzhou area of national great site protection,the temporal and spatial distribution characteristics and influencing factors of cultural heritage resources were studied with the help of ArcGIS.The data show that on the whole,the cultural heritage in Jingzhou area of national great site protection presented the time series of “a large span,more at two ends” and the spatial distribution pattern of “one main,two secondary and three cores”.Cultural heritage in different historical periods had the characteristics of differentiated regional aggregation.The formation of the spatial pattern of different types of cultural heritage was closely related to the local historical and cultural background.Cultural heritage was affected by rivers,terrain,transportation,land,culture and other factors.It was mainly distributed along rivers in areas with little topographic fluctuation and low altitudes.The development and construction of cultural heritage sites were weak,and the ecological foundation was poor.However,the traffic accessibility was good,and the cultural heritage was deep.The research results are expected to provide a scientific basis for guiding the large site protection area to strengthen the integrated protection of cultural resources.展开更多

In present article the author offers a variation in which a damaged nuclear station can be quickly covered by such a cheap inflatable dome. By containing the radioactive dust from the damaged nuclear station, the danger zone is reduced to about 2 km2 rather than large regions which requires the resettlement of huge masses of people and which stops indus-try in large areas. If there is a big city (as Tokyo) near the nuclear disaster or there is already a dangerous amount of radioactive dust near a city, the city may also be covered by a large inflatable transparent Dome. The building of a gi-gantic inflatable AB Dome over an empty flat surface is not difficult. The cover is spread on a flat surface and a venti-lator (fan system) pumps air under the film cover and lifts the new dome into place but inflation takes many hours. However, to cover a city, garden, forest or other obstacle course in contrast to an empty, mowed field, the thin film cannot be easily deployed over building or trees without risking damage to it by snagging and other complications. This article proposes a new method which solves this problem. The design is a double film blanket filled by light gas such as, methane, hydrogen, or helium - although of these, methane will be the most practical and least likely to leak. Sections of this AB Blanket are lighter than air and will rise in the atmosphere. They can be made on a flat area serving as an as-sembly area and delivered by dirigible or helicopter to station at altitude over the city. Here they connect to the already assembled AB Blanket subassemblies, cover the city in an AB Dome and protect it from bad weather, chemical, bio-logical and radioactive fallout or particulates. After assembly of the dome is completed, the light gas can be replaced by (heavier but cheaper) air. Two projects for Tokyo (Japan) and Moscow (Russia) are used in this paper for sample computation.展开更多

Avian metapneumovirus(aMPV),a paramyxovirus,causes acute respiratory diseases in turkeys and swollen head syndrome in chickens.This study established a reverse genetics system for aMPV subtype B LN16-A strain based on...Avian metapneumovirus(aMPV),a paramyxovirus,causes acute respiratory diseases in turkeys and swollen head syndrome in chickens.This study established a reverse genetics system for aMPV subtype B LN16-A strain based on T7 RNA polymerase.Full-length cDNA of the LN16-A strain was constructed by assembling 5 cDNA fragments between the T7 promoter and hepatitis delta virus ribozyme.Transfection of this plasmid,along with the supporting plasmids encoding the N,P,M2-1,and L proteins of LN16-A into BSR-T7/5 cells,resulted in the recovery of aMPV subtype B.To identify an effective insertion site,the enhanced green fluorescent protein(EGFP)gene was inserted into different sites of the LN16-A genome to generate recombinant LN16-As.The results showed that the expression levels of EGFP at the site between the G and L genes of LN16-A were significantly higher than those at the other two sites(between the leader and N genes or replacing the SH gene).To verify the availability of the site between G and L for foreign gene expression,the VP2 gene of very virulent infectious bursal disease virus(vvIBDV)was inserted into this site,and recombinant LN16-A(rLN16A-vvVP2)was successfully rescued.Single immunization of specificpathogen-free chickens with rLN16A-vvVP2 induced high levels of neutralizing antibodies and provided 100%protection against the virulent aMPV subtype B and vvIBDV.Establishing a reverse genetics system here provides an important foundation for understanding aMPV pathogenesis and developing novel vector vaccines.展开更多
Background:Nesting trees and habitat represent the key factor underpinning stand selection by forest-dwelling birds.While two large European species–the black stork(Ciconia nigra L.)and white-tailed eagle(Haliaaetus ...Background:Nesting trees and habitat represent the key factor underpinning stand selection by forest-dwelling birds.While two large European species–the black stork(Ciconia nigra L.)and white-tailed eagle(Haliaaetus albicilla L.)–are known to require old,large trees for nesting,we sought to investigate further by comparing species requirements at the levels of the nesting tree,nesting stand,and landscape.This entailed a detailed examination of forest features within circles of radius 15 m surrounding 16 and 19 trees holding the nests of storks and eagles respectively.The same parameters were also checked in the vicinity of 50 randomly-selected mature trees.Results:Our results indicate different nesting preferences,with the eagles entirely confining themselves to Scots pine(Pinus sylvestris L.)–a species also chosen by black storks,which nevertheless regularly favour pedunculate oak(Quercus robur L.).Both species select trees of greater girth than the random ones,but white-tailed eagles choose to nest in the vicinity of patches of mature old stands with a loose canopy,to the extent that nesting trees and surrounding trees are of similar ages.In contrast,black storks prefer“veteran”trees with low-set crowns that are much older than any others in their vicinity.Nesting trees of the eagles are away from roads and close to lakes,while black storks do not avoid roads.Conclusions:As the ages of nesting trees of both species are greater than ages at final cutting in this region’s managed forest,silvicultural measures will need to be adjusted,with small patches of forest spared,or larger areas as“islands”of old-growth.Likewise,as tending and thinning are engaged in,certain trees with horizontal branches will need to remain,with relict trees also left untouched.As it happens,all of these recommendations are anyway key elements of close-to nature silviculture and multifunctional forestry.展开更多

Assessment of the exposure dose for workers is crucial to protecting workers from the radiological risk.This preliminary study estimates the potential radiological exposure for a soil remediation worker at a nuclear d...Assessment of the exposure dose for workers is crucial to protecting workers from the radiological risk.This preliminary study estimates the potential radiological exposure for a soil remediation worker at a nuclear decommissioning site contaminated with Cs-137 in Korea,and then calculates the maximum workable soil concentration to comply with the occupational dose constraint of 20 mSv per year.The Korean characteristic data,detailed exposure scenarios for workers by the type of work,and relevant exposure pathways were used in the dose estimation.As a result,the most severe exposure-induced work type was identified as the excavator operation with an annual individual dose of 5.92×10^-5 mSv for a unit concentration of soil,from which the derived maximum workable soil concentration was 3.38×105 Bq/kg.Furthermore,dose contribution by each exposure pathway was found to be decreased in the following order:external radiation exposure,soil ingestion,dust inhalation,and skin contamination.The results of this study are expected to be used effectively to optimize radiation protection for workers and establish appropriate work procedures for future site remediation.展开更多

If there is a big city (as Tokyo) near the nuclear disaster or there is already a dangerous amount of radioactive dust near a city, the city may also be covered by a large inflatable transparent Dome. The building of a gi-gantic inflatable AB Dome over an empty flat surface is not difficult. The cover is spread on a flat surface and a venti-lator (fan system) pumps air under the film cover and lifts the new dome into place but inflation takes many hours. However, to cover a city, garden, forest or other obstacle course in contrast to an empty, mowed field, the thin film cannot be easily deployed over building or trees without risking damage to it by snagging and other complications. This article proposes a new method which solves this problem. The design is a double film blanket filled by light gas such as, methane, hydrogen, or helium - although of these, methane will be the most practical and least likely to leak. Sections of this AB Blanket are lighter than air and will rise in the atmosphere. They can be made on a flat area serving as an as-sembly area and delivered by dirigible or helicopter to station at altitude over the city. Here they connect to the already assembled AB Blanket subassemblies, cover the city in an AB Dome and protect it from bad weather, chemical, bio-logical and radioactive fallout or particulates. After assembly of the dome is completed, the light gas can be replaced by (heavier but cheaper) air. Two projects for Tokyo (Japan) and Moscow (Russia) are used in this paper for sample computation.
文摘Land for protective forest on the coast has special site conditions, and site classification is the scientific basis for seaboard afforestation. The site classification system on the coast zone and islands of China may be classified into five levels-site region (sub - region), district, class, group, and type. The land division for afforestation is carried out by the principle of enviornmental heterogeneity among regions, sub-region and district on large scale, according to the difference of air temperature, moisture and type of coast geomorphy. It may be classified into 7 regions, 12 sub-regions and 55 districts. The medium and small scaled division for site class, group and type, subdivided in a site district, are based on medium topography, topographic climate, micro - relief and soil conditions.

文摘Avian metapneumovirus(aMPV),a paramyxovirus,causes acute respiratory diseases in turkeys and swollen head syndrome in chickens.This study established a reverse genetics system for aMPV subtype B LN16-A strain based on T7 RNA polymerase.Full-length cDNA of the LN16-A strain was constructed by assembling 5 cDNA fragments between the T7 promoter and hepatitis delta virus ribozyme.Transfection of this plasmid,along with the supporting plasmids encoding the N,P,M2-1,and L proteins of LN16-A into BSR-T7/5 cells,resulted in the recovery of aMPV subtype B.To identify an effective insertion site,the enhanced green fluorescent protein(EGFP)gene was inserted into different sites of the LN16-A genome to generate recombinant LN16-As.The results showed that the expression levels of EGFP at the site between the G and L genes of LN16-A were significantly higher than those at the other two sites(between the leader and N genes or replacing the SH gene).To verify the availability of the site between G and L for foreign gene expression,the VP2 gene of very virulent infectious bursal disease virus(vvIBDV)was inserted into this site,and recombinant LN16-A(rLN16A-vvVP2)was successfully rescued.Single immunization of specificpathogen-free chickens with rLN16A-vvVP2 induced high levels of neutralizing antibodies and provided 100%protection against the virulent aMPV subtype B and vvIBDV.Establishing a reverse genetics system here provides an important foundation for understanding aMPV pathogenesis and developing novel vector vaccines.
文摘Background:Nesting trees and habitat represent the key factor underpinning stand selection by forest-dwelling birds.While two large European species–the black stork(Ciconia nigra L.)and white-tailed eagle(Haliaaetus albicilla L.)–are known to require old,large trees for nesting,we sought to investigate further by comparing species requirements at the levels of the nesting tree,nesting stand,and landscape.This entailed a detailed examination of forest features within circles of radius 15 m surrounding 16 and 19 trees holding the nests of storks and eagles respectively.The same parameters were also checked in the vicinity of 50 randomly-selected mature trees.Results:Our results indicate different nesting preferences,with the eagles entirely confining themselves to Scots pine(Pinus sylvestris L.)–a species also chosen by black storks,which nevertheless regularly favour pedunculate oak(Quercus robur L.).Both species select trees of greater girth than the random ones,but white-tailed eagles choose to nest in the vicinity of patches of mature old stands with a loose canopy,to the extent that nesting trees and surrounding trees are of similar ages.In contrast,black storks prefer“veteran”trees with low-set crowns that are much older than any others in their vicinity.Nesting trees of the eagles are away from roads and close to lakes,while black storks do not avoid roads.Conclusions:As the ages of nesting trees of both species are greater than ages at final cutting in this region’s managed forest,silvicultural measures will need to be adjusted,with small patches of forest spared,or larger areas as“islands”of old-growth.Likewise,as tending and thinning are engaged in,certain trees with horizontal branches will need to remain,with relict trees also left untouched.As it happens,all of these recommendations are anyway key elements of close-to nature silviculture and multifunctional forestry.
文摘The groyne has been used widely in bank protection and river regulation. The plane layout and structural style of groyne are closely related to the stability of the groyne itself and the effect of bank protection. In this paper, based on the preliminary summary of the design of bank protection in the bore surging area of Qiantang River, some problems regarding the structure of groyne are pointed out and an improved plan is put forward. Site experiment was carried out combined with an emergency repair. The results of the experiment indicate that the improved project is reliable and successful.

文摘Assessment of the exposure dose for workers is crucial to protecting workers from the radiological risk.This preliminary study estimates the potential radiological exposure for a soil remediation worker at a nuclear decommissioning site contaminated with Cs-137 in Korea,and then calculates the maximum workable soil concentration to comply with the occupational dose constraint of 20 mSv per year.The Korean characteristic data,detailed exposure scenarios for workers by the type of work,and relevant exposure pathways were used in the dose estimation.As a result,the most severe exposure-induced work type was identified as the excavator operation with an annual individual dose of 5.92×10^-5 mSv for a unit concentration of soil,from which the derived maximum workable soil concentration was 3.38×105 Bq/kg.Furthermore,dose contribution by each exposure pathway was found to be decreased in the following order:external radiation exposure,soil ingestion,dust inhalation,and skin contamination.The results of this study are expected to be used effectively to optimize radiation protection for workers and establish appropriate work procedures for future site remediation.
