Current studies on the interactions between microor... Dissolved organic matter(DOM) in the ocean is one of the largest carbon pools on Earth. Microbial metabolism is an important process that shapes the marine DOM pool. Current studies on the interactions between microorganisms and DOM focus mainly on oxic environments. Few studies have addressed the molecular characteristics of DOM in microbial-mediated transformation under anoxic/hypoxic conditions. As a result of deteriorating water quality due to eutrophication and global warming, anoxia occurs frequently in coastal waters. In this study, we performed an experiment to investigate changes in microbial community responses and the molecular characteristics of DOM in microbial-mediated transformation under hypoxic conditions. We compared microbial-mediated DOM transformation at different dissolved oxygen levels(7, 5, and 2 mg L^(-1)) and in different media(natural and artificial seawater with and without laminarin). We also investigated differences in DOM composition between groups using spectroscopic analysis and ultrahigh-resolution Fourier transform ion cyclotron resonance mass spectrometry. The results showed decreased microbial metabolic activity and delayed community succession at low oxygen(≤2 mg L^(-1)) in natural seawater supplemented with laminarin. The growth of strictly aerobic bacteria such as Pseudomonadaceae and Sphingomonadaceae was inhibited and the total organic carbon utilization rate was reduced by 36.9–46.7% from 4 to 32days. Moreover, tyrosine-like and tryptophan-like components were preserved, while DOM humification and modified aromaticity indices were significantly reduced under low oxygen conditions. This experiment provides justification for further study of the processes and mechanisms of improved labile DOM preservation in anoxic estuarine and coastal waters. 展开更多

Background:Under hypoxia,exaggerated compensatory responses may lead to acute mountain sickness.The excessive vasodilatory effect of nitric oxide(NO)can lower the hypoxic pulmonary vasoconstriction(HPV)and peripheral blood pressure.While NO is catalyzed by various nitric oxide synthase(NOS)isoforms,the regulatory roles of these types in the hemodynamics of pulmonary and systemic circulation in living hypoxic animals remain unclear.Therefore,this study aims to investigate the regu-latory effects of different NOS isoforms on pulmonary and systemic circulation in hypoxic rats by employing selective NOS inhibitors and continuously monitoring hemodynamic parameters of both pulmonary and systemic circulation.Methods:Forty healthy male Sprague–Dawley(SD)rats were randomly divided into four groups:Control group(NG-nitro-D-arginine methyl ester,D-NAME),L-NAME group(non-selective NOS inhibitor,NG-nitro-L-arginine methyl ester),AG group(in-ducible NOS inhibitor group,aminoguanidine),and 7-NI group(neurological NOS in-hibitor,7-nitroindazole).Hemodynamic parameters of rats were monitored for 10 min after inhibitor administration and 5 min after induction of hypoxia[15%O2,2200 m a.sl.,582 mmHg(76.5 kPa),Xining,China]using the real-time dynamic monitoring model for pulmonary and systemic circulation hemodynamics in vivo.Serum NO concentra-tions and blood gas analysis were measured.Results:Under normoxia,mean arterial pressure and total peripheral vascular resist-ance were increased,and ascending aortic blood flow and serum NO concentration were decreased in the L-NAME and AG groups.During hypoxia,pulmonary arterial pressure and pulmonary vascular resistance were significantly increased in the L-NAME and AG groups.Conclusions:This compensatory mechanism activated by inducible NOS and en-dothelial NOS effectively counteracts the pulmonary hemodynamic changes induced by hypoxic stress.It plays a crucial role in alleviating hypoxia-induced pulmonary arte-rial hypertension. 展开更多

BACKGROUND Anoxic brain injury is a potentially lethal condition characterized by cerebral hypoperfusion and irreversible neuronal injury.Arterial spin-labeling(ASL)perfusion and diffusion-weighted imaging(DWI)magnetic resonance imaging(MRI)have been proposed as tools to detect cerebral ischemic changes and may aid in the assessment of anoxic injury.AIM To explore the relationship between regional ASL perfusion patterns and clinical outcomes following cardiac arrest.METHODS We performed a retrospective review to identify patients with clinical suspicion of anoxic brain injury who underwent MRI within 15 days of cardiac arrest.Receiver operator characteristic(ROC)analysis and univariate logistic regression were used to evaluate associations between ASL perfusion scores,DWI signal intensity,and the following clinical features:(1)Myoclonus status epilepticus(MSE)within 24 hours;(2)Absent extensor or motor reflexes(EMR)at day 3 post-arrest;and(3)Absent brainstem reflexes(BSR)within 15 days.RESULTS Twenty-eight patients met inclusion criteria.Increased ASL signal in the left occipital lobe was significantly associated with MSE(P=0.038),while a trend was observed between right frontal ASL signal and EMR(P=0.078).ROC analysis showed that ASL scores≥7 were associated with higher odds of absent BSR(OR 2.14,P=0.53),though this did not reach statistical significance.DWI signal intensity did not show significant associations with clinical outcomes.The overall discriminatory performance of ASL for predicting outcomes was limited(AUC≈0.52).CONCLUSION This exploratory study suggests that regional ASL hyperperfusion,particularly in the left occipital and right frontal lobes,may be associated with adverse clinical signs following cardiac arrest.However,most findings did not reach statistical significance,and the study was underpowered to detect small-to-moderate effects.These preliminary results should be interpreted with caution and considered hypothesis-generating.Larger,prospective studies are warranted to clarify the prognostic value of ASL perfusion imaging in anoxic brain injury. 展开更多

Pulmonary hypertension(PH)is a pulmonary vascular disease caused by multiple etiologies,characterized by increased pulmonary vascular resistance and elevated pulmonary artery pressure,which can lead to severe cardiovascular complications.The third type of PH,hypoxic pulmonary hypertension(HPH)caused by chronic lung disease and/or hypoxia,has complex and incomplete pathological mechanism.Current clinical treatment for HPH primarily focus on alleviating symptoms,with limited effectiveness in improving pulmonary vascular remodeling(PVR).Recent studies by various scholars have indicated that certain traditional Chinese medicine(TCM)monomers,extracts,and formulations can inhibit multiple signaling pathways,thereby suppressing pulmonary vascular remodeling and demonstrating favorable efficacy against HPH.This article reviews the pathogenesis of HPH,including pulmonary arterial wall thickening,immune inflammation,and thrombogenesis,and discusses the latest research advancements regarding the pharmacodynamic mechanisms of TCM in treating HPH. 展开更多

[Objective] The aim of this study was to provide a fast, stable and efficient piggery wastewater processing technology. [Method] The start-up process was studied through the experiment of piggery anaerobic fermentation slurry treated by Anoxic/Oxic (A/O) reactor. The process was divided into two stages: at the first stage, dominant micro flora were cultivated in Anoxic and Oxic reaction tanks respectively; at the second stage. Anoxic and Oxic reaction tanks were initiated jointly to gradually enhance water load and continued to cultivate and domesticate microorganisms, and finally the start-up process was completed. [ Result] The results showed that return mixture ratio and return sludge ratio was 2 and 1 respectively when the temperature reached 32 ±2 ℃. However. when aeration rate of Oxic reaction amounted to 0.5 m^3/h, the re- moval rate of COD and NH4^+ -H were 89.87% and 89.31% respectively through practical operation within 50 days, which indicated that the start- up process through A/O reactor was successful. Conclusion This study can provide a scientific basis and reference for innocuous technique of piggery anaerobic fermentation slurry treatment. 展开更多

A kind of functional liquid was extracted from compound Chinese traditional medicine over 20 different varieties such as ginseng, danggui and so on. The anti-anoxic effects were observed by the ways of oxygen lack in normal atmosphere of mice, decapitation and erythrocyte count and hemoglobin in the mice. Experiments done in this study included observing the best dose and the best time of appearing anti-anoxic effects in mice. Results were found that the functional liquid could prolong not only the survival time and the brain survival time of the mouse, but also increasing the number of erythrocytes (RBC) and the content of hemoglobin (Hb); and the functional liquid has markedly protecting cardio-cerebral function; If the dosage of the functional liquid control on the 0.3ml/day for each mouse, the best effects should appear after four hour. And the functional liquid had no obvious effect on the weight of the mice within thirty days. 展开更多

BACKGROUND: Hepatocyte apoptosis is a severe form of cell death after hepatic ischemia-reperfusion injury (HIRI), and its relief is an important issue in liver transplantation. Hypoxic preconditioning (HP) is considered to have protective effects on HIRI. This study was designed to explore the impact of HP on apoptosis and its possible mechanism during orthotopic liver autotransplantation. METHODS: A modified orthotopic liver autotransplantation model was used to simulate HIRI. Sprague-Dawley rats were randomly divided into normal control, autotransplantation (AT) and HP groups. The HP group was subjected to an 8% oxygen atmosphere for 90 minutes before surgery. At 1, 6 and 24 hours after surgery, the rats were killed and their liver tissue was sampled to assess the expression of Bcl-2 protein. The samples were subjected to blood chemistry study, morphological study under a light or transmission electron microscope, and quantitative study of mitochondria. RESULTS: The serum levels of ALT and AST in the HP group were lower than those in the AT group at 1, 6 and 24 hours after orthotopic liver autotransplantation (P < 0.05). Bcl-2 protein expression was increased in the HP group at each measurement point (P < 0.05). Light microscopy showed that hepatic injury in the AT group was much more severe than in the HP group. Hepatocytes in the AT group showed typical apoptosis signs under a transmission electron microscope. The ultrastructural appearance of hepatocytes in the HP group was much better than in the AT group, and the area, perimeter and diameter of the mitochondria were smaller in the HP group than in the AT group (P < 0.05). CONCLUSIONS: Hepatocytes sense and respond to decreased tissue oxygenation. Stimulation by HP relieves apoptosis by upregulating expression of Bcl-2 protein and its protection of mitochondria after orthotopic liver autotransplantation. 展开更多

In order to investigate the protective effect of hypoxic preconditioning on the cerebral ischemia-reperfusion injury, the expression of Bcl-2 and Bax was detected by using immunohistochemical staining after 3 h cerebral ischemia followed by 1, 6, 12, 24 and 48 h reperfusion respectively in rats treated with or without hypoxic preconditioning before cerebral ischemia. In addition, the apoptosis of neural cells and the behavioral scores for neurological functions recovery were evaluated by TUNEL staining and "crawling method", respectively. Compared with control group (cerebral ischemia-reperfusion without hypoxic preconditioning), the expression of Bcl-2 was significantly increased, but that of Bax decreased in the hypoxic preconditioning group (cerebral ischemiareperfusion with hypoxie preconditioning), both P〈0.05. The pre-treatment with hypoxic preconditioning could reduce the apoptosis of neural cells and promote the neurological function recovery as compared to control group. It was suggested that hypoxic preconditioning may have protective effects on the cerebral ischemia-reperfusion injury by inhibiting the apoptosis of neural cells, increase the expression of Bcl-2 and decrease the expression of Bax. 展开更多

This paper presents new geological and geochemical data from the Shuanghu area in northern Tibet, which recorded the Early Toarcian Oceanic Anoxic Event. The stratigraphic succession in the Shuanghu area consists mostly of grey to dark-colored alternating oil shales, marls and mudstones. Ammonite beds are found at the top of the Shuanghu oil shale section, which are principally of early Toarcian age, roughly within the Harplocearas falciferrum Zone. Therefore, the oil shale strata at Shuanghu can be correlated with early Toarcian black shales distributing extensively in the European epicontinental seas that contain the records of an Oceanic Anoxic Event. Sedimentary organic matter of laminated shale anomalously rich in organic carbon across the Shuanghu area is characterized by high organic carbon contents, ranging from 1.8% to 26.1%. The carbon isotope curve displays the δ 13C values of the kerogen (δ 13Ckerogen) fluctuating from –26.22 to –23.53‰ PDB with a positive excursion close to 2.17‰, which, albeit significantly smaller, may also have been associated with other Early Toarcian Oceanic Anoxic Events (OAEs) in Europe. The organic atomic C/N ratios range between 6 and 43, and the curve of C/N ratios is consistent with that of the δ 13Ckerogen values. The biological assemblage, characterized by scarcity of benthic organisms and bloom of calcareous nannofossils (coccoliths), reveals high biological productivity in the surface water and an unfavorable environment for the benthic fauna in the bottom water during the Oceanic Anoxic Event. On the basis of organic geochemistry and characteristics of the biological assemblage, this study suggests that the carbon-isotope excursion is caused by the changes of sea level and productivity, and that the black shale deposition, especially oil shales, is related to the bloom and high productivity of coccoliths. 展开更多

Previous studies have demonstrated the protective effect of hypoxic preconditioning on acute cerebral infarction, but the mechanisms underlying this protection remain unclear. To investigate the protective mechanisms of hypoxic preconditioning in relation to its effects on angiogenesis, we in- duced a photochemical model of cerebral infarction in an inbred line of mice (BALB/c). Mice were then exposed to hypoxic preconditioning 30 minutes prior to model establishment. Results showed significantly increased vascular endothelial growth factor and CD31 expression in the ischemic penumbra at 24 and 72 hours post infarction, mainly in neurons and vascular endothelial cells. Hypoxic preconditioning increased vascular endothelial growth factor and CD31 expression in the ischemic penumbra and the expression of vascular endothelial growth factor was positively related to that of CD31. Moreover, hypoxic preconditioning reduced the infarct volume and improved neu- rological function in mice. These findings indicate that the protective role of hypoxic preconditioning in acute cerebral infarction may possibly be due to an increase in expression of vascular endothelial growth factor and CD31 in the ischemic penumbra, which promoted angiogenesis. 展开更多

Background: Vascular endothelial growth factor A (VEGFA) can induce endothelial cell proliferation, promote cell migration, and inhibit apoptosis. These processes play key roles in physiological blood vessel formation and pathological angiogenesis. Methods: In this study, we examined VEGFA gene expression in the heart, liver, and kidney of Tibetan pigs (-I-P), Yorkshire pigs that migrated to high altitudes (YH), and Yorkshire pigs that lived at low altitudes (YL). We used PCR and Sanger sequencing to screen for single nucleotide polymorphisms (SNPs) in 5'-flanking DNA and exons of the VEGFA gene. Quantitative real-time PCR and western blots were used to measure expression levels and PCR products were sequenced. Results: Results showed that the VEGFA mRNA and protein expression in heart, liver and kidney of TP was higher than that in YH and YL. In addition, the mRNA sequence of the pig VEGFA gene was conserved among pig breeds, and only five SNPs were found in the 5'-flanking region of the VEGFA gene, the allele frequency distributions of the 5 SNPs were not significantly different between the TP, Yorkshire (YL), and Diannan small-ear (DN) pig populations. Conclusion: In conclusion, the Tibetan pig showed high tissues, which suggests that the VEGFA gene may play a levels of VEGFA gene expression in several hypoxic major functional role in hypoxic adaptation. 展开更多

To investigate the effects of hypoxic exercise training on microRNA (miRNA) expression and the role of miRNA expression in regulating lipid metabolism, 20 dietary-induced obese SD rats were divided into a normoxic sedentary group (N, n=10) and a hypoxic exercise training group (H, n=10). After four weeks, measurements were taken of body weight, body length, fat mass, serum lipid concentration, miRNAs differentially expressed in rat liver, and gene and protein expression levels of perexisome proliferator activated receptor a (PPARα), fatty acid synthetase (FAS), and carnitine palmitoyl transferase 1A (CPTIA) in rat liver. Body weight, Lee's index, fat mass, fat/weight ratio, and serum levels of total cholesterol (TC) and high density lipoprotein cholesterol (HDL-C) were all significantly lower in the H group than in the N group (P〈0.01). Six miRNAs expressed significantly differently in the liver (P〈0.05). Specifically, expression levels of miR-378b were significantly lower in the H group than in the N group (P〈0.05). Compared with the normoxic sedentary group, hypoxic exercise training resulted in a lower ratio of FAS mRNA to CPTIA mRNA (P〈0.05), as well as lower CPT1A protein levels (P〈0.01), while a higher ratio of FAS to CPT1A protein levels (P〈0.01) was observed. In conclusion, hypoxic training may elevate the resistance of high fat diet induced obesity in rats by reducing the expression of miR-378b, and decrease the fatty acid mitochondrial oxidation in obese rat livers by decreasing the protein expression of CPTIA and increasing the protein expression ratio of FAS/CPTIA. 展开更多

In order to enhance phosphorus removal in traditional step-feed anoxic/oxic nitrogen removal process,a modified pilot-scale step-feed anaerobic/anoxic/oxic(SFA 2/O) system was developed,which combined a reactor similar to UCT-type configuration and two-stage anoxic/oxic process.The simultaneous nitrogen and phosphorus removal capacities and the potential of denitrifying phosphorus removal,in particular,were investigated with four different feeding patterns using real municipal wastewater.The results showed that the feeding ratios(Q1)in the first stage determined the nutrient removal performance in the SFA 2/O system.The average phosphorus removal efficiency increased from 19.17% to 96.25% as Q1 was gradually increased from run 1 to run 4,but the nitrogen removal efficiency exhibited a different tendency,which attained a maximum 73.61%in run 3 and then decreased to 59.62%in run 4.As a compromise between nitrogen and phosphorus removal,run 3 (Q1=0.45Qtotal) was identified as the optimal and stable case with the maximum anoxic phosphorus uptake rate of 1.58 mg·(g MLSS)-1 ·h-1.The results of batch tests showed that ratio of the anoxic phosphate uptake capacity to the aerobic phosphate uptake capacity increased from 11.96% to 36.85% with the optimal influent feeding ratio to the system in run 3,which demonstrated that the denitrifying polyP accumulating organisms could be accumulated and contributed more to the total phosphorus removal by optimizing the inflow ratio distribution.However,the nitrate recirculation to anoxic zone and influent feeding ratios should be carefully controlled for carbon source saving. 展开更多

To achieve stable and efficient nitrogen and phosphorus removal and to investigate the characteristics of the A/A SBR enriched with denitrifying phosphorus removal bacteria(DPB),the whole course of startup was studied with two reactors operated in different mode.The reactor I was operated under anaerobic/settling/anoxic/settling mode,and the reactor II was operated under anaerobic/anoxic/settling mode.Differences between the two reactors in removal efficiency of COD,nitrogen and phosphorus were examined.The results indicated that efficient performance could be achieved in both reactors with different startup operation mode,while the phosphorus removal efficiency was improved sooner in reactor I than in reactor II,which suggested that reactor I would supply a more favorable condition for DPB proliferation.Meanwhile,it was observed that the amount of organic substrates consumption had a linear correlation to that of phosphorus release in anaerobic phase when DPB was accumulated in the A/A SBR denitrifying phosphorus removal system. 展开更多

Hypoxic preconditioning refers to the exposure of organisms, systems, organs, tissues or cells to moderate hypoxia/ischemia that results in increased resistance to a subsequent episode of severe hypoxia/ischemia. In this article, we review recent research based on a mouse model of repeated exposure to autohypoxia. Pre-exposure markedly increases the tolerance to or protection against hypoxic insult, and preserves the cellular structure of the brain. Furthermore, the hippocampal activity amplitude and frequency of electroencephalogram, latency of cortical somatosensory-evoked potential and spinal somatosensory-evoked potential progressively decrease, while spatial learning and memory improve. In the brain, detrimental neurochemicals such as free radicals are down-regulated, while beneficial ones such as adenosine are up-regulated. Also, antihypoxia factor(s) and gene(s) are activated. We propose that the tolerance and protective effects depend on energy conservation and plasticity triggered by exposure to hypoxia via oxygen-sensing transduction pathways and hypoxia-inducible factor-initiated cascades. A potential path for further research is the development of devices and pharma-ceuticals acting on antihypoxia factor(s) and gene(s) for the prevention and treatment of hypoxia and related syndromes. 展开更多

The anaerobic/anoxic/oxic(A2O) process is globally one of the widely used biological sewage treatment processes. This is the first report of a metagenomic analysis using Illumina sequencing of full-scale A2O sludge from a municipal sewage treatment plant.With more than 530,000 clean reads from different taxa and metabolic categories, the metagenome results allow us to gain insight into the functioning of the biological community of the A2O sludge. There are 51 phyla and nearly 900 genera identified from the A2O activated sludge ecosystem. Proteobacteria, Bacteroidetes, Nitrospirae and Chloroflexi are predominant phyla in the activated sludge, suggesting that these organisms play key roles in the biodegradation processes in the A2O sewage treatment system.Nitrospira, Thauera, Dechloromonas and Ignavibacterium, which have abilities to metabolize nitrogen and aromatic compounds, are most prevalent genera. The percent of nitrogen and phosphorus metabolism in the A2O sludge is 2.72% and 1.48%, respectively. In the current A2O sludge, the proportion of Candidatus Accumulibacter is 1.37%, which is several times more than that reported in a recent study of A2O sludge. Among the four processes of nitrogen metabolism, denitrification related genes had the highest number of sequences(76.74%), followed by ammonification(15.77%), nitrogen fixation(3.88%) and nitrification(3.61%). In phylum Planctomycetes, four genera(Planctomyces, Pirellula, Gemmata and Singulisphaera) are included in the top 30 abundant genera, suggesting the key role of ANAMMOX in nitrogen metabolism in the A2O sludge. 展开更多
