In most cases this infection is a subclinical event with fe...In 2010 the World Health Organisation estimated the number of infections with Entamoeba histolytica at about 50 million cases including 100000 fatal courses. In most cases this infection is a subclinical event with few or none symptoms noticeable for the patient. Courses of this disease and incidence of this parasite in industrialised nations are not yet fully investigated. Our case reports about a 68-year-old male patient from Turkey who lives for more than 30 years in Germany and had not been abroad during the past 2 years. Resistant asymptomatic amoebic dormant bodies caused an emergency-laparoscopy and revealed the seldom complication of a colon perforation. In the age of globalisation all providers in the health care systems are urged to acquaint themselves also with non-typical syndromes for the countries they work in order to reduce preventable morbidity respectively mortality rates.展开更多
Amoebiasis is a parasitic intestinal infection caused by the highly pathogenic amoeba Entamoeba histolytica.It is spread through person-toperson contact or by eating or drinking food or water contaminated with feces.I...Amoebiasis is a parasitic intestinal infection caused by the highly pathogenic amoeba Entamoeba histolytica.It is spread through person-toperson contact or by eating or drinking food or water contaminated with feces.Its transmission rate depends on the number of cysts present in the environment.The traditional models assumed a homogeneous and contradictory transmission with reality.The heterogeneity of its transmission rate is a significant factor when modeling disease dynamics.The heterogeneity of disease transmission can be described mathematically by introducing fuzzy theory.In this context,a fuzzy SEIR Amoebiasis disease model is considered in this study.The equilibrium analysis and reproductive number are studied with fuzziness.Two numerical schemes forward Euler method and a nonstandard finite difference(NSFD)approach,are developed for the learned model,and the results of numerical simulations are presented.The numerical and simulation results reveal that the proposed NSFD method provides an adequate representation of the dynamics of the disease despite the uncertainty and heterogeneity.Moreover,the obtained method generates plausible predictions that regulators can use to support decision-making to design and develop control strategies.展开更多

Subsequently, peritonitis wi...A 52-year-old man had bloody stools during chemo-therapy for gastric cancer. A colonoscopy revealed necrotizing ulcer-like changes. A biopsy confirmed the presence of amoebic trophozoites. Subsequently, peritonitis with intestinal perforation developed, and emergency peritoneal lavage and colostomy were performed. After surgery, endotoxin adsorption therapy was performed and metronidazole was given. Symptoms of peritonitis and colonitis resolved. However, the patient's general condition worsened with the progression of gastric cancer. The patient died 50 d after surgery. Fulminant amoebic colitis is very rarely associated with chemotherapy. Amoebic colitis should be considered in the differential diagnosis of patients who have bloody stools during chemotherapy.展开更多

With the resurgence of resistant pathogenic strains as well as ancestral considerations in developing countries such as Cameroon, many people rely on medicinal plants to treat a plethora of diseases. This work aimed to highlight the anti-amoebic and anti-oxidant potential of Codiaeum variegatum extracts and fractions. The anti-amoebic potential of C. variegatum was assayed on the polyxenic culture of the clinical isolates of E. histolytica. Then, the anti-oxidant potential of the ethanolic/hydroethanolic extracts and fractions was evaluated through DPPH radical scavenging, iron reduction (FRAP), lipid peroxidation inhibitory potential and total antioxidant capacity tests followed by the determination of phenolic compound and flavonoid content. It was found that the fractionation process decreased the amoebicidal activities of C. variegatum leaf extracts. However hydroethanolic extract (CI<sub>50</sub>: 10.08 ± 0.42, 5.18 ± 0.09, 5.18 ± 0.09 μg/mL respectively after 24, 48 and 72 hours) was more active than ethanolic extract (CI<sub>50</sub>: 15.59 ± 6.17;9.61 ± 2.37;6.26 ± 3.22 μg/mL respectively after 24, 48 and 72 hours). Interestingly, the activities of hydroethanolic extract were significantly non-different compared to metronidazole CI<sub>50</sub>: 8.42 ± 0.44, 6.45 ± 0.22 and 3.42 ± 0.33 μg/mL, respectively after 24, 48 and 72 hours). Ethanolic extract and EF5 showed higher Phenolic compound contents and higher antioxidant activity than hydroethanolic extract and other fractions through DPPH radical scavenging power (EC50 = 311.50 ± 4.12 μg/mL) and total antioxidant capacity (44 ± 0.07 mgEAA/gF). However, these activities are significantly lower than those of ascorbic acid (EC50 = 31.20 ± 4.39 μg/mL, and 61.34 ± 4.42 μg/mL respectively). This low antioxidant activity was confirmed by poor phenolic and flavonoid compounds contents found in the extracts and fractions. The present result brings a new approach to the ethnopharmacological uses of C. variegatum against dysentery in cases associated with Amoebiasis in Cameroun.展开更多

METHODS E. histolytica were isolated from stool samples and maintained in Robinson's biphasic culture medium. Clinial isolates were maintained in xenic culture medium,and harvested for determination of minimum inhibitory concentrations to the two antiamoebic drugs,Metronidazole and Auranofin using microtiter plate tests. The percent survival of the two isolates were determined using the trypan blue cell count. Isolate 980 was treated with 70 μmol/L and 2 μmol/L while isolate 989 was treated with 20 μmol/L and 0.5 μmol/L of metronidazole and auranofin respectively for 24 h. Fifty thousand cells of each isolate were harvested after 24 h of treatment for analysis of the mRNA expressions of the antioxidant enzymes,thioredoxin reductase,peroxiredoxin and FeSOD using the specific primers. Cell lysate was used for determination of enzyme activity of thioredoxin reductase by measuring DTNB reduction spectrophotometrically at412 nm.RESULTS Minimum inhibitory concentration of the clinical isolates 980 and 989 for auranofin was 3 μmol/L and 1 μmol/L respectively while that for metronidazole was 80 μmol/L and 30 μmol/L respectively. Thioredoxin reductase,peroxiredoxin and FeSOD expression levels were significantly reduced in the isolate 980 when treated with Auranofin. Metronidazole treatment showed a down regulation of thioredoxin reductase. Though not significant both at the mRNA and the enzyme activity levels. Peroxiredoxin and FeSOD however remained unchanged. Auranofin treatment of isolate 989,showed an upregulation in expression of thioredoxin reductase while Peroxiredoxin and FeSOD did not show any change in expression. Upon treatment with metronidazole,isolate 989 showed an increase in thioredoxin reductase expression. Peroxiredoxin and FeSOD expressions however remain unchanged both at mRNA and enzyme activity level.CONCLUSION Clinical isolates from New Delhi NCR region show different sensitivities to antiamoebic drugs. Auranofin is effective against isolate showing higher tolerance to metronidazole as shown by its inhibition in thioredoxin reductase activity.展开更多
