[Objective] This study was to explore the effect of purple yam on the nutritional and physiological functions of rats.[Method] Forty SD rats were randomly divided into four groups with ten rats in each group which wer...[Objective] This study was to explore the effect of purple yam on the nutritional and physiological functions of rats.[Method] Forty SD rats were randomly divided into four groups with ten rats in each group which were reared with artificial semisynthetic feed added with 10% steamed yam powder.The four groups,the casein(CK) group which was used as the control,the purple yam(PY) group,the Tiegun yam(TY) group and common Huai yam(HY) group,were supplied with feed of equal content of energy,protein and fat.The rats were pair-fed for 56 days to observe changes of relevant nutritional and physiological indices.[Result] There were no significant differences of body weight,food conversion rate and organ indices between rats in PY group and CK group.Mean corpuscular hemoglobin in PY group was significantly lower than that in CK group;hemoglobin showed no significant difference with that in CK group;eosnophils was lower than that in CK group.Low density lipoprotein-cholesterol,total cholesterol and atherogenic index in PY group were lower than that in the other three groups;anti-atherogenic index was the highest among the four groups,while showing no significant difference;triglyceride content was lower than that in CK group.Among the antioxidant indices,the activity of catalase and superoxide dismutase were higher than that in CK group,while malondialdehyde content was lower than that in CK group.[Conclusion] Purple yam showed the trend of reducing the content of blood glucose,total cholesterol,low density lipoprotein-cholesterol,triglyceride,malondialdehyde,and raising the activity of glutathione peroxidase,superoxide dismutase and catalase,thus having certain antioxidative function.展开更多

Objective: To examine the effect of Senna alata(S. alata) leaf extracts on the regulation of lipid metabolism in high-fat diet-induced obese mice.Methods: The obesity condition was induced in the male ICR mice by feed...Objective: To examine the effect of Senna alata(S. alata) leaf extracts on the regulation of lipid metabolism in high-fat diet-induced obese mice.Methods: The obesity condition was induced in the male ICR mice by feeding them with high-fat diet(45 kcal% fat) for 12 weeks. At the 7th week of diet feeding, the obese mice were treated with the water extract of S. alata leaf at 250 and 500 mg/kg/day, respectively, that continued for six weeks. At the end of the treatment period, the biochemical parameters were determined. The hepatic histology and the gene and protein expressions were also examined.Results: In comparison with the obese control mice, the mice treated with S. alata showed a significant reduction in the elevated blood glucose levels and a decrease in the serum insulin and leptin levels. A reduction in the serum total cholesterol, triglyceride,non-esterified fatty acid, and hepatic triglyceride levels were also observed. The histological examination of the obese mice treated with S. alata showed a reduced lipid accumulation in the liver tissue. Hepatic lipogenic gene expression showed that S. alata decreased the activity of sterol regulatory element binding protein 1c, fatty acid synthase,and acetyl-Co A carboxylase. S. alata could suppress hepatic peroxisome proliferatoractivated receptor gamma(PPARg) protein. Moreover, the protein expression of PPARa in liver tissue was clearly increased by S. alata treatment.Conclusion: The treatment with S. alata could decrease several parameters of impaired lipid metabolism in the obese mice by downregulating sterol regulatory element binding protein 1c and PPARg and upregulating PPARa. This study is the first report on the role of S. alata leaf extract in alleviating the abnormal lipid metabolism in obese conditions.展开更多
Objective To study the abortifacient activity of aqueous extract of Senna alata leaves in female Wistar rats.Methods Pregnant rats weighing 143.65±13.11 g were completely randomized into 5 groups(A-E).Rats in gro...Objective To study the abortifacient activity of aqueous extract of Senna alata leaves in female Wistar rats.Methods Pregnant rats weighing 143.65±13.11 g were completely randomized into 5 groups(A-E).Rats in group A(negative control)were orally administered,once daily with 0.5 ml of distilled water on days 10-18 post-coitum while those in groups B,C,D,and E were treated exactly like the negative control except they received the same volume containing 2.85 mg/kg body weight of mifepristone(group B,reference drug),250,500 and 1000 mg/kg body weight of the extract respectively(groups C-E).Results Phytochemical screening of the extract showed positive results for saponins(1.22%),flavonoids(1.06%),cardiac glycosides(0.20%),cardenolides and dienolides(0.18%),phenolics(0.44%)and alkaloids(0.52%).Respiratory distress,salivation,diarrhoea,changes in the appearance of hair as well as maternal mortality were not observed at any time during the exposure period except in the mifepristone-treated animals where there was episode of diarrhoea and tiredness.The extract significantly reduced(P〈0.05)the number of life foetus,weight and survival ratio of the foetus,numbers of implantations and Corpora lutea,implantation index,progesterone,prolactin,estradiol,follicle stimulating and luteinizing hormones whereas the number of dead foetus,number and percentage of rats that aborted,percentage vaginal opening,resorption index,pre-and post-implantation losses increased significantly.In addition,mifepristone-treated animals produced resorption index that compared well with the distilled water control.There was also no dead or life foetus and serum progesterone concentration was increased in the animals treated with mifepristone.All cases of abortion were accompanied with vaginal bleeding.Although,the final weight of the rats increased significantly,the feed and water intake were not significantly altered in all the treatment groups.The weight of the uterus,uterine-body weight ratio,length of the right uterus horn and uterine cholesterol decreased significantly in all the treatment groups.The uterine alkaline phosphatase activity and glucose concentration increased in only the extract-treated animals whereas mifepristone decreased the uterine alkaline phosphatase activity and glucose content of the animals.Conclusion This study has provided evidence to the age-long claim of S.alata leaves in"washing the uterus".The abortifacient properties were most pronounced at 500 and 1000 mg/kg body weight of the extract and were similar to the animals treated with 2.85 mg/kg body weight of mifepristone.Hormonal influence,changes in implantation site,estrogenicity and uterogenicity are suggested as possible mechanism of abortifacient activity of aqueous extract of S.alata leaves.Overall,the extract may be used as an abortifacient especially at 500 and 1000 mg/kg body weight and therefore not safe for consumption as oral remedy during pregnancy.展开更多
Objective:To investigate the bone-resorbing effect of demethylbelamcandaquinone B(Dmcq B)extracted from Marantodes pumilum var.alata on osteoclast differentiation in RAW264.7 cells.Methods:RAW264.7 macrophages were di...Objective:To investigate the bone-resorbing effect of demethylbelamcandaquinone B(Dmcq B)extracted from Marantodes pumilum var.alata on osteoclast differentiation in RAW264.7 cells.Methods:RAW264.7 macrophages were differentiated using RANKL into osteoclast-like cells.Then,they were treated with 10μg/mL Marantodes pumilum var.alata crude aqueous extract,5μg/mL dichloromethane fraction,and 0.6μg/mL Dmcq B and 0.06μg/mL estradiol.Tartrate-resistant acid phosphatase 5b(TRACP 5b)as an osteoclast phenotypic marker was determined by TRACP staining and TRACP 5b colometric assay,and bone-resorbing pits were examined.The gene expression of pro-inflammatory cytokines(TNF-αand IL-6)was measured.Moreover,the protein expressions of pro-inflammatory cytokines(TNF-αand IL-6)and estrogen receptors were evaluated.Results:Marantodes pumilum var.alata crude aqueous extract and Dmcq B inhibited RANKL-stimulated osteoclast differentiation as evidenced by size reduction of giant multinucleated osteoclast cells,decreased TRACP 5b activity as well as the subsiding of resorbed pit area compared with normal control.In addition,they reduced the gene and protein expressions of TNF-αand IL-6.Marantodes pumilum var.alata,Dmcq B,and estradiol treatments increased the protein expressions of estrogen receptors alpha and beta in osteoclasts.Conclusions:Marantodes pumilum var.alata and its active compound,Dmcq B can inhibit osteoclast differentiation.展开更多

[Results] The caffeic acid showed a good linear relationship in the range of injection volume of 0. 025 92-0. 259 2 μg( R =0. 999 5). The average recovery rate was 98. 33%( RSD = 1. 85%). L. alata in different production areas of Guangxi contained the caffeic acid,and there was a great difference in the caffeic acid. L. alata in Baise had the highest content of caffeic acid,while that in Guilin had the lowest content of caffeic acid. To s...The use of yam vine cuttings as propagules with appropriate synthetic rooting hormones such as IBA (Indole-Butyric Acid) and NAA (Naphthalene Acetic Acid) has previously been reported as successful but expensive. To source for cheaper alternatives, this study compared the effect of some natural and synthetic hormones on rooting of vine cuttings in two varieties of Dioscorea alata. Vine cuttings from three months old water yam varieties were treated separately with 1%, 3% and 5% concentrations of each of the following: neem leaf ash, coconut water, 2,4-D and IBA with water as control. Treated cuttings were planted in triplicates in polythene bags using the completely randomised design and watered once in two days for 49 days. Data collected on rooting percentage, number of roots per plant, mean root length, number and length of new vines were subjected to analysis of variance tests and the means separated using least significant difference tests. A 100% rooting was observed in vines treated with 1% or 3% coconut water and neem ash. These vines also generated significantly greater number of new vines (P < 0.05) compared to those from synthetic hormones. Vines treated with 5% neem produced the highest number of roots (145.33 ± 9.21;P < 0.001) while those treated with 3% neem produced the longest roots (45.3 ± 9.23 cm) followed by those treated with 3% coconut water (24.3 ± 3.48). The study reveals that neem leaf ash and coconut water are better root promoting agents for water yam vines than 2,4-D and IBA and could be tested further for use as natural hormonal substitutes to the synthetic hormones.展开更多

It has high yield and stores longer than Dioscorea cayenensis/rotundata yam and therefore fills the hunger gap created when other yam types arc not available. However, very little research has been done on it. Several cultivars are susceptible to pests and diseases and lack the aesthetic values of smooth skin and elegant tuber shape ofDioscorea rotundata that appeal to consumers in the market. This study therefore sought to establish the genetic variability and relationships among 35 accessions of Ghanaian Dioscorea alata together with 14 accessions introduced from International Institute of Tropical Agriculture in Nigeria. Morphological variation was studied to establish genetic relationship among 49 accessions assembled as Dioscorea alata in Ghana. Principal component analysis (PCA) indicated that the first two principal components accounted for 25.27% of total variation from accessions with PCA1 and PCA2, contributing 13.92% and 11.35%, respectively. Cluster analysis performed clearly separated the 49 accessions into three main groupings using unweighted neighbour-joining method. Morphological descriptors were able to group the accessions into distinct clusters independent of place of collection.展开更多

Fresh leaves of S. alata were blended with a homogenizer in 1 litre of dist...The efficacy of crude extracts of Senna alata in the improvement of vegetative and reproductive growth in Celosia aregentea was investigated. Fresh leaves of S. alata were blended with a homogenizer in 1 litre of distilled water. The resultant green paste was filtered under suction. Different concentrations (75%, 50%, 40%, 30%, 25%, 12%, 10%, and 5%) were prepared from the 100% crude extract. Seeds of C. argentea were presoaked in these different concentrations including a control (0%) and planted out after 24 hours. Results obtained showed that seedling height, leaf area, dry weight and leaf area ratio were promoted and enhanced by presoaking seeds in the extract. At the end of the experimental period (six weeks), seedling height in 75% and 100% treatments were 109 ± 16.12 cm and 117 ± 19.32 cm, leaf area 128 ± 17.91 cm2 and 125 ± 18.12 cm2, dry weight 7.48 kg and 7.0 kg respectively. Seedlings raised from seeds presoaked in water (control) however, flowered earlier (8 weeks) than the treatments (10 weeks in 75% and 100%). Presoaking seeds of C. argentea in crude extracts of S. alata before planting is recommended for optimum production of the leafy vegetable. The procedure is cheap and easily implementable by resource-poor farmers who are the main growers of C. argentea.展开更多

The prevalent genera were Aspergillus (A. flavus and A. parasiticus), Fusarium (F. moniliforme and F. oxysporum) and Penicillium (Penicillium sp.). Such seedborne fungi differ in their colonization in different parts of seeds with most of them are colonized in seed coat, endosperm, and embryo of the seed. The usage of different storage containers for storing seeds indicated that the cotton cloth bags were the most favorable ones as they maintain seed moisture content (SMC) below the critical level resulting in minimum seed deterioration compared with other seed storage containers.展开更多

Method: Wistar rats received by ...Description of the Subject: Senna alata (L.) was a plant in the Benin pharmacopoeia used to treat skin infections. Objectives: The aim of our work was to test its harmlessness in vivo. Method: Wistar rats received by gavage a single dose of 2000 mg/kg of Senna alata leaves aqueous extract for the Acute Oral Toxicity (AOT) test. For the sub-Chronic Oral Toxicity (SCT) test, rats force-fed the extract at a daily dose of 300 mg/Kg of body weight for 28 days. The weight of the rats was taken and the blood samples were collected on Day 0, then respectively day 14 for the AOT and Day 28 for the SCT. The renal balance was carried out by dosage of the creatinine, the liver balance by the transaminases AST and ALT and the blood balance by the hemogram. The liver, kidneys and spleen were removed for histological analysis. The results were analyzed using the Student test, with the significance level set at 5%. Results: The weight of the rats did not change significantly in the acute or subchronic oral toxicity tests suggesting an absence of physical disturbance in the rats. Serum creatinine did not vary significantly, suggesting preservation of renal function. That was the same for ASAT and ALAT transaminases, indicating an absence of hepatic cytolysis. In hematology, the hemoglobin level and the number of blood platelets did not vary significantly, suggesting that the extract did not create anemia and did not influence blood coagulation. Hepatic, renal and splenic parenchyma showed no atypia. Conclusion: The aqueous extract of Senna alata (L.) leaves did not reveal any acute or subchronic toxicity and offered prospects for its use in the treatment of infections.展开更多

Methods: H...The purpose of this study was to investigate the effect of Laggera alata flavonen (LAF) on the inhibit- ing effect of human ovarian cancer HO-8910 cells proliferation and its possible mechanism in vitro. Methods: Human ovarian cancer HO-8910 cells were cultured in vitro. Inhibitory effect of LAF on the viability of HO-8910 cells was evaluated by the MTT assay. Apoptotic effect of different concentrations of LAF on HO-8910 cells was assessed by AO/EB staining and FCM with propidium iodide (PI) staining. Expression of proteins related to apoptosis was analyzed by Western blot. Results: LAF significantly inhibited the viability of HO-8910 cells proliferation in a dose-dependent and time-dependent manner, there were statistical significance compared with NS group (P 〈 0.05), and the ICso was 4.28 pg/mL for 48 h. The cells treated with LAF showed typical morphological change and apoptotic rate increased by FCM in a dose-dependent, and there was notable dif- ference compared with NS group (P 〈 0.05). Western blot showed that expression of Fas, caspase-8, tBid and Cyto-c proteins were up-regulated after treatment with LAF for 48 h in a concentration dependent. Conclusion: LAF could inhibit HO-8910 cells proliferation and induce apoptosis, which may be through the pathway of death receptor in vitro.展开更多

文摘Objective To study the abortifacient activity of aqueous extract of Senna alata leaves in female Wistar rats.Methods Pregnant rats weighing 143.65±13.11 g were completely randomized into 5 groups(A-E).Rats in group A(negative control)were orally administered,once daily with 0.5 ml of distilled water on days 10-18 post-coitum while those in groups B,C,D,and E were treated exactly like the negative control except they received the same volume containing 2.85 mg/kg body weight of mifepristone(group B,reference drug),250,500 and 1000 mg/kg body weight of the extract respectively(groups C-E).Results Phytochemical screening of the extract showed positive results for saponins(1.22%),flavonoids(1.06%),cardiac glycosides(0.20%),cardenolides and dienolides(0.18%),phenolics(0.44%)and alkaloids(0.52%).Respiratory distress,salivation,diarrhoea,changes in the appearance of hair as well as maternal mortality were not observed at any time during the exposure period except in the mifepristone-treated animals where there was episode of diarrhoea and tiredness.The extract significantly reduced(P〈0.05)the number of life foetus,weight and survival ratio of the foetus,numbers of implantations and Corpora lutea,implantation index,progesterone,prolactin,estradiol,follicle stimulating and luteinizing hormones whereas the number of dead foetus,number and percentage of rats that aborted,percentage vaginal opening,resorption index,pre-and post-implantation losses increased significantly.In addition,mifepristone-treated animals produced resorption index that compared well with the distilled water control.There was also no dead or life foetus and serum progesterone concentration was increased in the animals treated with mifepristone.All cases of abortion were accompanied with vaginal bleeding.Although,the final weight of the rats increased significantly,the feed and water intake were not significantly altered in all the treatment groups.The weight of the uterus,uterine-body weight ratio,length of the right uterus horn and uterine cholesterol decreased significantly in all the treatment groups.The uterine alkaline phosphatase activity and glucose concentration increased in only the extract-treated animals whereas mifepristone decreased the uterine alkaline phosphatase activity and glucose content of the animals.Conclusion This study has provided evidence to the age-long claim of S.alata leaves in"washing the uterus".The abortifacient properties were most pronounced at 500 and 1000 mg/kg body weight of the extract and were similar to the animals treated with 2.85 mg/kg body weight of mifepristone.Hormonal influence,changes in implantation site,estrogenicity and uterogenicity are suggested as possible mechanism of abortifacient activity of aqueous extract of S.alata leaves.Overall,the extract may be used as an abortifacient especially at 500 and 1000 mg/kg body weight and therefore not safe for consumption as oral remedy during pregnancy.

文摘Objective:To investigate the bone-resorbing effect of demethylbelamcandaquinone B(Dmcq B)extracted from Marantodes pumilum var.alata on osteoclast differentiation in RAW264.7 cells.Methods:RAW264.7 macrophages were differentiated using RANKL into osteoclast-like cells.Then,they were treated with 10μg/mL Marantodes pumilum var.alata crude aqueous extract,5μg/mL dichloromethane fraction,and 0.6μg/mL Dmcq B and 0.06μg/mL estradiol.Tartrate-resistant acid phosphatase 5b(TRACP 5b)as an osteoclast phenotypic marker was determined by TRACP staining and TRACP 5b colometric assay,and bone-resorbing pits were examined.The gene expression of pro-inflammatory cytokines(TNF-αand IL-6)was measured.Moreover,the protein expressions of pro-inflammatory cytokines(TNF-αand IL-6)and estrogen receptors were evaluated.Results:Marantodes pumilum var.alata crude aqueous extract and Dmcq B inhibited RANKL-stimulated osteoclast differentiation as evidenced by size reduction of giant multinucleated osteoclast cells,decreased TRACP 5b activity as well as the subsiding of resorbed pit area compared with normal control.In addition,they reduced the gene and protein expressions of TNF-αand IL-6.Marantodes pumilum var.alata,Dmcq B,and estradiol treatments increased the protein expressions of estrogen receptors alpha and beta in osteoclasts.Conclusions:Marantodes pumilum var.alata and its active compound,Dmcq B can inhibit osteoclast differentiation.

文摘[Objectives] To establish a method for determining the content of Laggera alata( D. Don) Sch. Bip. Ex Oliv. using caffeic acid the target component,and to compare the content of caffeic acid in the medicinal materials of L. alata in different production areas of Guangxi.[Methods]The content was determined by Inertsil~ODS-3 chromatographic column C_(18)( 4. 60 mm × 250 mm,5 μm,mobile phase: acetonitrile-0. 1% phosphoric acid( 22∶ 78),detection wavelength: 320 nm,flow rate: 1. 0 m L/min,column temperature: 30℃,and injection volume: 10 μL. [Results] The caffeic acid showed a good linear relationship in the range of injection volume of 0. 025 92-0. 259 2 μg( R =0. 999 5). The average recovery rate was 98. 33%( RSD = 1. 85%). L. alata in different production areas of Guangxi contained the caffeic acid,and there was a great difference in the caffeic acid. L. alata in Baise had the highest content of caffeic acid,while that in Guilin had the lowest content of caffeic acid. [Conclusions]This method can accurately determine the content of caffeic acid and is expected provide a scientific basis for the development and utilization of herbal medicine L. alata.
文摘The use of yam vine cuttings as propagules with appropriate synthetic rooting hormones such as IBA (Indole-Butyric Acid) and NAA (Naphthalene Acetic Acid) has previously been reported as successful but expensive. To source for cheaper alternatives, this study compared the effect of some natural and synthetic hormones on rooting of vine cuttings in two varieties of Dioscorea alata. Vine cuttings from three months old water yam varieties were treated separately with 1%, 3% and 5% concentrations of each of the following: neem leaf ash, coconut water, 2,4-D and IBA with water as control. Treated cuttings were planted in triplicates in polythene bags using the completely randomised design and watered once in two days for 49 days. Data collected on rooting percentage, number of roots per plant, mean root length, number and length of new vines were subjected to analysis of variance tests and the means separated using least significant difference tests. A 100% rooting was observed in vines treated with 1% or 3% coconut water and neem ash. These vines also generated significantly greater number of new vines (P < 0.05) compared to those from synthetic hormones. Vines treated with 5% neem produced the highest number of roots (145.33 ± 9.21;P < 0.001) while those treated with 3% neem produced the longest roots (45.3 ± 9.23 cm) followed by those treated with 3% coconut water (24.3 ± 3.48). The study reveals that neem leaf ash and coconut water are better root promoting agents for water yam vines than 2,4-D and IBA and could be tested further for use as natural hormonal substitutes to the synthetic hormones.
文摘The Asiatic Dioscorea alata yam species is an important food crop, especially in Cote d'lvoire. It has high yield and stores longer than Dioscorea cayenensis/rotundata yam and therefore fills the hunger gap created when other yam types arc not available. However, very little research has been done on it. Several cultivars are susceptible to pests and diseases and lack the aesthetic values of smooth skin and elegant tuber shape ofDioscorea rotundata that appeal to consumers in the market. This study therefore sought to establish the genetic variability and relationships among 35 accessions of Ghanaian Dioscorea alata together with 14 accessions introduced from International Institute of Tropical Agriculture in Nigeria. Morphological variation was studied to establish genetic relationship among 49 accessions assembled as Dioscorea alata in Ghana. Principal component analysis (PCA) indicated that the first two principal components accounted for 25.27% of total variation from accessions with PCA1 and PCA2, contributing 13.92% and 11.35%, respectively. Cluster analysis performed clearly separated the 49 accessions into three main groupings using unweighted neighbour-joining method. Morphological descriptors were able to group the accessions into distinct clusters independent of place of collection.

文摘The aim of the current study is to review the medicinal properties of the plants found in Brunei Darussalam namely Litsea elliptica, Dillenia suffruticosa, Dillenia excelsa, Aidia racemosa, Vitex pinnata and Senna alata. The known phytochemical constituents of these plants and their ability to bring about a range of biological activities are included in this review. These plants have been used traditionally for a multitude of diseases and illnesses. There is a lot of untapped potential in these medicinal plants which could cure multiple diseases.
文摘The efficacy of crude extracts of Senna alata in the improvement of vegetative and reproductive growth in Celosia aregentea was investigated. Fresh leaves of S. alata were blended with a homogenizer in 1 litre of distilled water. The resultant green paste was filtered under suction. Different concentrations (75%, 50%, 40%, 30%, 25%, 12%, 10%, and 5%) were prepared from the 100% crude extract. Seeds of C. argentea were presoaked in these different concentrations including a control (0%) and planted out after 24 hours. Results obtained showed that seedling height, leaf area, dry weight and leaf area ratio were promoted and enhanced by presoaking seeds in the extract. At the end of the experimental period (six weeks), seedling height in 75% and 100% treatments were 109 ± 16.12 cm and 117 ± 19.32 cm, leaf area 128 ± 17.91 cm2 and 125 ± 18.12 cm2, dry weight 7.48 kg and 7.0 kg respectively. Seedlings raised from seeds presoaked in water (control) however, flowered earlier (8 weeks) than the treatments (10 weeks in 75% and 100%). Presoaking seeds of C. argentea in crude extracts of S. alata before planting is recommended for optimum production of the leafy vegetable. The procedure is cheap and easily implementable by resource-poor farmers who are the main growers of C. argentea.
文摘Sequential incubation of seed samples yielded 20 fungal species belonging to 13 genera. The prevalent genera were Aspergillus (A. flavus and A. parasiticus), Fusarium (F. moniliforme and F. oxysporum) and Penicillium (Penicillium sp.). Such seedborne fungi differ in their colonization in different parts of seeds with most of them are colonized in seed coat, endosperm, and embryo of the seed. The usage of different storage containers for storing seeds indicated that the cotton cloth bags were the most favorable ones as they maintain seed moisture content (SMC) below the critical level resulting in minimum seed deterioration compared with other seed storage containers.
文摘Description of the Subject: Senna alata (L.) was a plant in the Benin pharmacopoeia used to treat skin infections. Objectives: The aim of our work was to test its harmlessness in vivo. Method: Wistar rats received by gavage a single dose of 2000 mg/kg of Senna alata leaves aqueous extract for the Acute Oral Toxicity (AOT) test. For the sub-Chronic Oral Toxicity (SCT) test, rats force-fed the extract at a daily dose of 300 mg/Kg of body weight for 28 days. The weight of the rats was taken and the blood samples were collected on Day 0, then respectively day 14 for the AOT and Day 28 for the SCT. The renal balance was carried out by dosage of the creatinine, the liver balance by the transaminases AST and ALT and the blood balance by the hemogram. The liver, kidneys and spleen were removed for histological analysis. The results were analyzed using the Student test, with the significance level set at 5%. Results: The weight of the rats did not change significantly in the acute or subchronic oral toxicity tests suggesting an absence of physical disturbance in the rats. Serum creatinine did not vary significantly, suggesting preservation of renal function. That was the same for ASAT and ALAT transaminases, indicating an absence of hepatic cytolysis. In hematology, the hemoglobin level and the number of blood platelets did not vary significantly, suggesting that the extract did not create anemia and did not influence blood coagulation. Hepatic, renal and splenic parenchyma showed no atypia. Conclusion: The aqueous extract of Senna alata (L.) leaves did not reveal any acute or subchronic toxicity and offered prospects for its use in the treatment of infections.

文摘[Objectives]To establish a method for quality analysis of Laggera alata.[Methods]The water content,total ash and alcohol-soluble extract of Laggera alata were determined according to the method of 2015 edition of Chinese Pharmacopoeia.[Results]Ten batches of Laggera alata from different producing areas and different collection time in Guangxi had the same plant morphology and medicinal properties.The experimental results were as follows:the water content was 5.19%-10.86%;the total ash content was 5.91%-10.74%;the acid-insoluble ash content was 0.44%-0.92%;the extract content was 14.64%-19.95%.[Conclusions]The experimental results can provide scientific basis for the development and utilization of Laggera alata and the establishment of its quality standard.
文摘The purpose of this study was to investigate the effect of Laggera alata flavonen (LAF) on the inhibit- ing effect of human ovarian cancer HO-8910 cells proliferation and its possible mechanism in vitro. Methods: Human ovarian cancer HO-8910 cells were cultured in vitro. Inhibitory effect of LAF on the viability of HO-8910 cells was evaluated by the MTT assay. Apoptotic effect of different concentrations of LAF on HO-8910 cells was assessed by AO/EB staining and FCM with propidium iodide (PI) staining. Expression of proteins related to apoptosis was analyzed by Western blot. Results: LAF significantly inhibited the viability of HO-8910 cells proliferation in a dose-dependent and time-dependent manner, there were statistical significance compared with NS group (P 〈 0.05), and the ICso was 4.28 pg/mL for 48 h. The cells treated with LAF showed typical morphological change and apoptotic rate increased by FCM in a dose-dependent, and there was notable dif- ference compared with NS group (P 〈 0.05). Western blot showed that expression of Fas, caspase-8, tBid and Cyto-c proteins were up-regulated after treatment with LAF for 48 h in a concentration dependent. Conclusion: LAF could inhibit HO-8910 cells proliferation and induce apoptosis, which may be through the pathway of death receptor in vitro.
