The present paper covers the actional mechanism of trifluoroacetic acid for the separation of biopolymers investigated by using the parameters of stoichiometric displacement model for retention(SDM-R) in reversed-phase liquid chromatography. It was found that the trifluoroacetic acid(TFA) may participate in, or stimulate the association among displacing agent molecules in mobile phase, and decrease the affinity of both the associate molecules of the displacing agent and the TFA-protein ion-pairing. The former dominates over the separation selectivity of biopolymers as the concentration of TFA is lower than a given value, and the two contrary functions partly offset to each other and the latter dominates as its concentration is greater than the given value. 展开更多

The paper aims at investigating the encoding of self-propelled motion events in Homeric Greek in the light of the typology of motion events, taking into account the case of to go.The verbal class of the self-propelled motion refers to those verbs expressing the idea of a simple translational motion, such as to go,to move, without any information about the manner of motion(see,by contrast, the class of the manner-of-motion verbs, such as to run,to swim)or about the path of motion(see,by contrast, the class of the path verbs, such as to enter,to exit).According to Talmy(2000),world languages can be distinguished depending on whether they prototypically express the semantic component of Path in or outside the motion verb.Languages belonging to the S(atellite)-Framed type tend to convey Path outside the motion verb,in a satellite element, such as a particle,an adposition(adpositional phrase),a preverb,an adverb(ial),a nominal case marker. The prototypical encoding pattern of the S-Framed languages, such as Homeric Greek, involves a motion verb conveying Manner and a satellite conveying Path,i.e.,[manner-of-motion verb+Path-satellite].Nonetheless,another pattern is used by this type of languages, albeit less prototypical, which involves a motion verb conveying only Motion and a satellite conveying Path,i.e.,[self-propelled motion verb+Path-satellite].Verb-inherent actionality, namely telicity, turns out to be a strong feature within the ancient Indo-European languages, such as Homeric Greek, playing a role not only in the development of aspectual/tense morphology, but also in the encoding of motion events,at least with reference to manner-of-motion verbs. The present paper aims at verifying the role of inherent telicity within self-propelled motion verbs, through the analysis of Homeric verbs for go.The study takes into account the Homeric suppletive paradigm for go,focusing on ερχομαι“go;come”and ηλθον(aorist)“go;come”(also with reference to the unclear actional opposition between ερχομαι and ειμι“go;come”).From the textual analysis of all the occurrences of both ερχομαι and ηλθον,as well as their co-occurring Path-encoding elements in the Iliad and the Odyssey, data show to what extent verbal inherent telicity plays an important role in motion event encoding also within the class of self-propelled motion verbs. 展开更多

Background:Diabetic foot,a severe complication of diabetes,is characterized by chronic refractory wounds.Sanhuang Oil,a topical herbal formula, demonstrates significant therapeutic effects including antibacterial,anti-inflammatory,and immunomodulatory activities. However, its active constituents and mechanisms of action against diabetic foot remain to be elucidated. Methods:In this study, the chemical constituents of Sanhuang Oil were identified using UPLC-QE-Orbitrap-MS.Subsequently,the mechanism by which Sanhuang Oil promotes diabetic foot ulcer healing was predicted by integrating network pharmacology and molecular docking.Additionally, diabetic mouse model was established in ICR mice using a combination of a high-fat diet(HFD)and streptozotocin(STZ)chemical induction.A full-thickness skin defect was created on the dorsum of the mice.Wound healing and the healing rate were observed following Sanhuang Oil intervention. The mechanism underlying Sanhuang Oil’s promotion of diabetic ulcer healing was further investigated using transcriptomics and histopathological examination(H&E staining). Results:A total of 97 active ingredients were identified from Sanhuang Oil.Network pharmacology analysis predicted 543 common targets, and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis identified 203 relevant pathways.Molecular docking further confirmed high binding affinity(binding energy≤−5.0 kcal/mol)between specific active components in Sanhuang Oil(e.g.,coptisine,phellodendrine,baicalein)and key targets associated with diabetic foot ulcers(e.g.,EGFR,AKT1,STAT3).In vivo experimental results demonstrated that the wound healing rate was significantly higher in Sanhuang Oil-treated groups compared to the model group(P<0.001).HE staining revealed that the high-dose Sanhuang Oil group exhibited more pronounced epithelial tissue coverage over the wound, reduced inflammatory cell infiltration, and increased collagen deposition and fibroblast proliferation.transcriptomic analysis identified Pdk4,Ttn,Csrp3,Actn2,Myoz2,Tnnc2,Myod1,Myog,Myot,and Myf6 as key regulatory proteins involved in promoting wound healing. Conclusion:Sanhuang Oil promotes wound healing in diabetic ulcer mice, potentially by mitigating inflammation and regulating key targets such as Pdk4 to enhance fibroblast function. These findings provide novel insights into the multi-target,multi-pathway mechanism of Sanhuang Oil for treating diabetic foot ulcers. 展开更多

Ulcerative colitis (UC) is a chronic inflammatory bowel disease characterized byclinical symptoms of diarrhea and mucopurulent bloody stools, and its incidenceis increasing globally. The etiology and pathogenesis of UC remain elusive. Currenttherapeutic approaches, including anti-inflammatory, immunosuppressiveand immunomodulating agents, are often limited in efficacy and frequently associatedwith adverse drug reactions. Therefore, there is an urgent need to developsafer and more effective treatment strategies to address the limitations of existingtherapies. Scutellaria baicalensis Georgi (HQ), a traditional Chinese medicinal herb, has been employed in the treatment of UC for over 2000 years. Recent studieshave demonstrated that HQ contains multiple active components capable oftreating UC through anti-inflammation, immune modulation, intestinal barrierprotection, antioxidant activity, and regulation of the gut microbiota. This paperreviews recent studies on the mechanism of action and clinical trials of HQ intreating UC based on relevant literature, with the aim of providing valuable insightsinto future treatment approaches. 展开更多

Oligodendrocytes and their cell-intrinsic gene regulatory network:Oligodendrocytes(OLs)are the myelinating glial cells of the vertebrate central nervous system. They are responsible for insulating neuronal axons with a lipid-rich myelin sheath, which enables the saltatory conduction of action potentials.During development, oligodendrocyte progenitor cells(OPCs)emerge from neural stem cells in the ventricular zone. They then proliferate, increase their number, and migrate to their final destination where they encounter unmyelinated neuronal axons and differentiate in a stepwise fashion into myelinating oligodendrocytes(mOLs)under the influence of environmental stimuli. 展开更多

In this paper,we consider the fractional critical Schrödinger equation(FCSE)(-Δ)^(s)u-|u|2^(*)s-2 u=0,where u∈˙H^(s)(R^(N)),N≥4,0<s<1 and 2^(*)s=2 N/N-2 s is the critical Sobolev exponent of order s.By virtue of the variational method and the concentration compactness principle with the equivariant group action,we obtain some new type of nonradial,sign-changing solutions of(FCSE)in the energy space˙H^(s)(R^(N)).The key component is that we take the equivariant group action to construct several subspace of˙H^(s)(R^(N))with trivial intersection, then combine the concentration compactness argument in the Sobolev space with fractional order to show the compactness property of Palais-Smale sequences in each subspace and obtain the multiple solutions of(FCSE)in˙H^(s)(R^(N)). 展开更多

Combined with elastic network model(ENM),the perturbation response scanning(PRS)has emerged as a robust technique for pinpointing allosteric interactions within proteins. Here,we proposed the PRS analysis of drug-target networks(DTNs),which could provide a promising avenue in network medicine. We demonstrated the utility of the method by introducing a deep learning and network perturbation-based framework, for drug repurposing of multiple sclerosis(MS).First, the MS comorbidity network was constructed by performing a random walk with restart algorithm based on shared genes between MS and other diseases as seed nodes. Then, based on topological analysis and functional annotation, the neurotransmission module was identified as the“therapeutic module”of MS.Further, perturbation scores of drugs on the module were calculated by constructing the DTN and introducing the PRS analysis, giving a list of repurposable drugs for MS.Mechanism of action analysis both at pathway and structural levels screened dihydroergocristine as a candidate drug of MS by targeting a serotonin receptor of se-rotonin 2B receptor(HTR2B).Finally,we established a cuprizone-induced chronic mouse model to evaluate the alteration of HTR2B in mouse brain regions and observed that HTR2B was significantly reduced in the cuprizone-induced mouse cortex. These findings proved that the network perturbation modeling is a promising avenue for drug repurposing of MS.As a useful systematic method, our approach can also be used to discover the new molecular mechanism and provide effective candidate drugs for other complex diseases. 展开更多

The evacuation of people under threat is an effective disaster prevention and mitigation measure in response to flash floods and geological hazards, and it is also an essential element of pre-disaster planning. However, the effect of the interactions between perception factors on residents'willingness to evacuate is an urgent problem to be solved. Therefore, this paper introduces risk, stakeholder, and protective action perceptions from the protective action decision model as the main explanatory variables. These three core perceptions are subdivided into affective risk perception, cognitive risk perception, government perception,other-stakeholder perception, resourcerelated attributes, and hazard-related attributes.A questionnaire survey was conducted from June to July 2023 among residents of mountainous communities in nine villages in three towns in Sichuan Province,China.359 cross-sectional data were analyzed using structural equation modeling to explore the effects of six perception factors on evacuation intentions. The results of the study showed that:(1)affective risk perception, government perception,other-stakeholder perception, and hazard-related attributes all directly and positively influence residents'intentions to evacuate;(2)cognitive risk perception is mediated by stakeholder and protective action perceptions, which indirectly and positively affect residents'intentions to evacuate.Based on the hypothesized paths, strategies to improve residents'willingness to evacuate are discussed from the perspective of three core perceptions:strengthening disaster risk education, improving residents'cohesion, and building government credibility. The results of this study can provide theoretical support and practical suggestions for emergency management departments to formulate emergency evacuation strategies, which can aid decision-makers in better understanding residents'intentions to evacuate, optimizing evacuation information dissemination pathways, and strengthening disaster risk management capabilities. 展开更多
