Electron microscopic autoradiographic studies on the dynamics and location of DNAsynthesis by means of incorporation of ~8H-thymidine during the replication of duck plaguevirus (DPV) revealed that the duration of DNA synthesis of DPV was rather long. The repli-cation of viral DNA occurred simultaneously with the assembly procedure of nucleocapsids, thematuration and release of viruses. DNA synthesis of DPV occurred in the matrix with lowerelectron density in the nucleus. The replicated viral DNA accumulated in the viroplast With highelectron density where the assembly of nucleocapsids occurred. The viroplast with high electrondensity was not the region of viral DNA synthesis. 展开更多

The distribution of subversive element sulfur in high temperature phases during solidification of cast iron was investigated by means of liquid quenching method and autoradiography.The results show that in grey cast iron,sulfur is concentrated in the liquid,flake graphite and in the peripherial area of D-type supercooled graphite eutectic cells,but less sulfur is in austenite. Besides,the content of sulfur in A-type graphite is higher than that in D-type graphite,and sulfur content in eutectic austenite is a little higher than that in primary austenite.While,in nodular graphite cast iron less sulfur is in liquid and austenite,but more in nodular graphite. The blackness of the spots on the emulsion film corresponds to the atomic percentage of radioactive substances located on the surface of the specimen. 展开更多

Objective: A method for separating mRNAs by means of the polymerase chain reaction (differential display mRNA), and identifying the genes related to radiation-induced lung cancer was introduced. Methods: The RNAs were isolated from two pairs of samples, SV40-immortalized human fetal tracheal fibroblast cell (SHTF) versus αSHTF cell (transformed SHTF cell induced by α particles) and lung cancer tissue versus normal lung tissue obtained from one miner, and amplified by RTPCR. The differential expressed gene fragments were displayed by autoradiograph or silver nitrate stain. Results: The differential display mRNA method was established using both cell and tissue samples. The bands stained by silver nitrate were clearer than those on X-ray film. The rate of reamplification of differentially expressed gene fragments stained by silver nitrate is 80%, higher than that by autoradiograph, 50%. Conclusion: Differential display mRNA method was established successfully on both cell and tissue samples. The modified method for staining band increased the rate of reamplification and established the basis for confirming relative genes. 展开更多
