Despite demonstrating significant anti-tumor potential as an artemisinin derivative,artesunate faces delivery efficiency challenges due to low water solubility and insufficient targeting specificity.To improve the del...Despite demonstrating significant anti-tumor potential as an artemisinin derivative,artesunate faces delivery efficiency challenges due to low water solubility and insufficient targeting specificity.To improve the delivery efficiency,we engineered three artesunate(ART) derivatives,AC_(15)-L(linear),AC_(15)-B(branched),and AC_(15)-C(cyclic) with distinct aliphatic chain architectures.Unexpectedly,we observed that AC_(15)-C exhibited superior cytotoxicity against 4T1 breast cancer cells,and had the highest binding affinity for Lon protease 1(LONP1)(-72.6 kcal/mol).Subsequently,disulfide bond-containing lipid-PEG(DSPESS-PEG2K) modified chain architecture-engineered ART derivatives nanoassemblies(NAs) were developed to mitigate solubility-related limitations while enhancing targeting precision.Molecular docking and experimental validation demonstrated that ART derivatives inhibited LONP1 through hydrophobic interactions while preserved Fe^(2+)-mediated Fenton-like reaction activity.In vitro and in vivo evaluations demonstrated that AC_(15)-C NAs outperformed free ART and other NAs,suppressing 4T1 tumor growth via dual action:LONP1-directed mitochondrial proteostasis collapse and reactive oxygen species(ROS) amplification through Fe^(2+)-ART interactions.This study elucidated a novel anti-tumor mechanism of ART through the rational design of derivatives with spatially configured aliphatic chains,and developed reductionresponsive NAs to provide an advanced delivery strategy.展开更多
Rabies is caused by infection of rabies virus(RABV)and remains a serious threat to the global public health.Except for the requirement for cold chain and high cost of human rabies immune globulin,no small molecule dru...Rabies is caused by infection of rabies virus(RABV)and remains a serious threat to the global public health.Except for the requirement for cold chain and high cost of human rabies immune globulin,no small molecule drugs are currently available for clinical treatment of rabies.So,it is of great importance to identify novel compounds that can effectively inhibit RABV infection.Artesunate(ART)and dihydroartemisinin(DHA),two derivatives of artemisinin,are widely used for treatment of malaria in adults and children,showing high safety.In this study,we found that both ART and DHA were able to inhibit RABV replication in host cells at a low concentration(0.1μmol/L).The antiviral effects of ART and DHA were independent of viral strains and cell lines.Pre-treatment with ART or DHA for 2 h in vitro did not affect the viral replication in host cells,implying that ART and DHA neither reduced the viability of RABV directly nor inhibited the binding and entrance of the virus to host cells.Further studies revealed that ART and DHA inhibited RABV genomic RNA synthesis and viral gene transcription.Treatment with ART or DHA(5 mg/kg)by intramuscular injection improved,to some extent,the survival rate of RABV-challenged mice.Combination treatment with derivatives of artemisinin and mannitol significantly improved the survival rate of RABV-challenged mice.The results suggest that ART and DHA have a great potential to be explored as new anti-rabies agents for treatment of rabies.展开更多

Methods: The human breast tumor cell line MCF-7 was transplanted into nude mice, and the animals were treated with various doses of ART alone or in combination with cyclophosphamide (CTX) or normal saline (NS). The tumor inhibitory effects were observed and compared, and the ultrastructural morphology of the transplanted tumor cells was observed by electron microscopy. The apoptosis rates and cell cycle status were detected by flow cytometry (FCM). The expression of apoptosis-related proteins p53, Bcl-2, Bax and Caspase-3 were detected by immunohistochemistry and IGF-IR was detected by western blot. The expression correlation for these proteins was also analyzed. Results: The tumor inhibition rates in the low dose ART group, high dose ART group, CTX group and combined drug therapy group were (24.39±10.20)%, (40.24±7.02)%, (57.01±5.84)% and (68.29±5.1)%, respectively. The cell cycle was arrested in phase G0/Gt after treatment with ART. The expression of Bcl-2 was significantly reduced, and the expression levels of Bax and Caspase-3 were significantly increased in the ART group compared to the negative control saline group. There was no significant difference detected in p53 expression. The Bcl-2 level was negatively related to Bax and Caspase-3. The western blotting results showed IGF-IR downregulation. Conclusions: ART inhibits the growth of MCF-7 breast tumor cell xenografts in nude mice. The anti-tumor mechanism of ART for human breast carcinoma in nude mice might be correlated with the alteration of apoptosis related protein expression, which may further induce apoptosis and inhibit cell proliferation.展开更多

This study aims to compare the therapeutic responses of two generic formulations of this antimalarial agent, Artéfan® and Coartem®, which have been available in Côte d’Ivoire’s healthcare facilities since 2008. Method: We conducted a comparative analysis of a bi-centric clinical trial in Côte d’Ivoire, evaluating the therapeutic efficacy of Artéfan® in Daloa and Coartem® in Ayamé between 2018 and 2019, following the WHO’s 42-day protocol. The primary evaluation metric was the Adequate Clinical and Parasitological Response (ACPR) at day 42 (D42) post-PCR correction. Results: A total of sixty-two patients were included at each site. The fever clearance time was 24 hours in both Daloa and Ayamé. Parasitic clearance occurred in 48 hours in Daloa and 72 hours in Ayamé. In the per-protocol analysis at D42, the ACPR was 92% in Daloa and 98% in Ayamé, with no significant difference noted (p = 0.1). Conclusion: Artéfan® and Coartem® exhibited statistically equivalent therapeutic efficacy. Nonetheless, the Artemether-Lumefantrine combination warrants further surveillance due to notable re-infestation rates.展开更多

目的探讨影响男男性行为者(men who have sex with men,MSM)人类免疫缺陷病毒(human immunodeficiency virus,HIV)感染者确诊后快速启动抗病毒治疗(antiretroviral therapy,ART)的相关因素,为临床制定针对性干预策略提供依据。方法选取2...目的探讨影响男男性行为者(men who have sex with men,MSM)人类免疫缺陷病毒(human immunodeficiency virus,HIV)感染者确诊后快速启动抗病毒治疗(antiretroviral therapy,ART)的相关因素,为临床制定针对性干预策略提供依据。方法选取2021年3月至2022年9月在北京佑安医院性病艾滋病门诊所有新诊断的成人MSM HIV感染者作为研究对象。通过问卷调查收集社会学及人口学信息,并从医院电子病历系统提取临床资料。根据确诊后14 d内是否启动ART,将研究对象分为快速治疗组(Rapid组)和非快速治疗组(非Rapid组)。采用t检验或χ^(2)检验进行单因素分析,并使用二元Logistic回归模型分析快速启动ART的独立影响因素。结果共纳入研究对象353例,其中Rapid组256例(72.5%),非Rapid组97例(27.5%)。单因素分析显示,独居状态、主动寻求HIV检测、认为快速启动ART有必要及身边有HIV感染的朋友/同伴,在两组间差异均具有统计学意义(χ^(2)分别为5.902、4.573、4.709、7.037,均P<0.05)。多因素Logistic回归分析显示,主动寻求HIV检测、和家人朋友或者同伴居住及身边有HIV感染者是愿意快速启动ART的独立促进因素(OR值分别为2.062、1.970、2.128,均P<0.05)。结论主动寻求HIV检测、身边有感染者同伴以及和家人朋友或者同伴居住的MSM HIV感染者,在确诊后更易快速启动ART,临床应重点关注和引导不具备相关条件的感染者,加强关爱和支持,以提升其快速启动治疗率。展开更多
