Alpiniae oxyphyllae Fructus ameliorates renal lipid accumulation in diabetic kidney disease via activating PPARα

Objective:To investigate the effects of Alpiniae oxyphyllae Fructus(AOF)on renal lipid deposition in diabetic kidney disease(DKD)and elucidate its molecular mechanisms.Methods:The mechanism of AOF in treating DKD was explored by network pharmacological enrichment analysis,molecular docking,and molecular dynamics simulation.The effects of AOF on renal function and lipid deposition were assessed in a mouse model of DKD and high glucose-stressed HK-2 cells.Cell viability and lipid accumulation were detected by CCK8 and oil red O staining.The expressions of PPARαand fatty acid oxidation-related genes(ACOX1 and CPT1A)were detected by quantitative RT-PCR,Western blot,and immunofluorescence.Furthermore,PPARαknockdown was performed to examine the molecular mechanism of AOF in treating DKD.Results:Network pharmacological enrichment analysis,molecular docking,and molecular dynamics simulation showed that the active compounds in AOF targeted PPARαand thus transcriptionally regulated ACOX1 and CPT1A.AOF lowered blood glucose,improved dyslipidemia,and attenuated renal injury in DKD mice.AOF-containing serum accentuated high glucose-induced decrease in cell viability and ameliorated lipid accumulation.Additionally,it significantly upregulated the expression of PPARα,ACOX1,and CPT1A in both in vivo and in vitro experiments,which was reversed by PPARαknockdown.Conclusions:AOF may promote fatty acid oxidation via PPARαto ameliorate renal lipid deposition in DKD. 展开更多

The analysis of chloroplast gene characteristics in Alpinia japonica(Thunb.)Miq.is of great significance for developing relevant genetic resources.The high-throughput sequencing and bioinformatic research were performed to analyze the chloroplast genome characteristics of A.japonica.The total chloroplast genome length of A.japonica was 161,906 bp,with a typical circular tetrameric structure.And 133 genes were annotated,comprising 87 protein-coding,38 tRNA,and 8 rRNA genes.Furthermore,22 genes contained two copies,and 18 genes owned introns.Repeat sequence analysis showed that it contains 321 simple sequence repeats(SSRs)and 37 long segment repeats.Compared with the chloroplast genomes of eight representative plants in the genus Alpinia,the gene structure,type,and quantity were relatively conservative.Rps12 was the highest variation site in the entire chloroplast gene.A phylogenetic tree showed that the genus Alpinia was the most closely related to the genus Amomum.Meanwhile,A.japonica is the most closely related to Alpinia chinensis belonging to the genus Alpinia.Overall,the chloroplast genome of a new species was reported in the genus Alpinia,and a basis was provided for the utilization of Alpinia plants as a medical resource. 展开更多

理论和实验研究表明片断化是导致生物多样性丧失的主要因素之一,但有关研究基本上集中在人类活动引发的片断化生境体系中。由于长期气候变化以及地形等因素的影响,一些物种的种群之间天然情况下就存在隔离,对这些种群进行研究可以弥... 理论和实验研究表明片断化是导致生物多样性丧失的主要因素之一,但有关研究基本上集中在人类活动引发的片断化生境体系中。由于长期气候变化以及地形等因素的影响,一些物种的种群之间天然情况下就存在隔离,对这些种群进行研究可以弥补人类活动引起的片断化体系经历时间较短的不足。山姜是一个喜湿耐荫的多年生植物,分布在热带亚热带地区,由于气候和地形原因,该物种在浙江东部呈现天然片断化状态,如在浙江天童森林公园及其周边地区,仅分布在少数几个沟谷中。以这些种群为对象,采用RAPD标记分析经历长期天然片断化山姜种群遗传多样性程度和遗传分化格局。7条RAPD引物获得了69个位点,其中多态位点68个。种群水平遗传多样性较高,多态位点百分比、期望杂合度和Shannon多样性指数分别为78.81%-85.51%、0.3170-0.3430、0.4560-0.4914,这与其异交的繁殖方式有关,同时与山姜克隆生长,每个世代维持的时间长,种群更新降低也有关系。遗传变异大多分配在种群内,但种群问遗传分化达到显著程度,φST为0.297,计算的基因流中等,为0.592,不足以克服长期遗传漂变导致的种群分化,Mantel检验表明遗传距离与空间距离不存在相关关系,显示了长期片断化状态下遗传漂变的作用。 展开更多

These two compounds are the first example of naturally occurring sesquiterpenoids with a 4,5-secoeudesmane skeleton in the family of Zingiberaceae and oxyphyllone A (1) is the first 4,5-secoeudesmane type of 13-norsesquiterpenoid. Objective:To study the effect of Alpinia officinarum Hance(A.officinarum) 80% alcohol extract on the primary dysmenorrhea.Methods:A.officinarum 80% alcohol extract were enriched by macroporous adsorption resins.Female mice of primary dysmenorrhea model were established by oxytocin induction; the effects of A.officinarum 80% alcohol extract on primary dysmenorrhea were observed by body twist method; and the homogenate level of prostaglandin F_(2α)(PGF_(2α)),prostaglandin E_2(PGE_2) and Ca^(2+) in the uterus were observed in oxytocin-induced female mice.Results:The writhing frequency of primary dysmenorrhea mice was significantly decreased after treatment of A.officinarum 80% alcohol extract and the level of PGF_(2α),PGE_2 and Ca^(2+) in mice uterus was significantly decreased(P<0.05,P<0.01) in groups of mice treated with middle and high dosage of A.officinarum 80% alcohol extract compared with that of model group.Conclusions:These findings suggest that A.Officinarum 80% alcohol extract can significantly relieve primary dysmenorrhea. 展开更多

Alpinia oxyphylla,a traditional herb,is widely used for its neuroprotective,antioxidant and memory-improving effects.However,the neuroprotective mechanisms of action of its active ingredients are unclear.In this study,we investigated the neuroprotective effects of various organic extracts of Alpinia oxyphylla on PC12 cells exposed to hydrogen peroxide-induced oxidative injury in vitro.Alpinia oxyphylla was extracted three times with 95%ethanol(representing extracts 1–3).The third 95%ethanol extract was dried and resuspended in water,and then extracted successively with petroleum ether,ethyl acetate and n-butanol(representing extracts 4–6).The cell counting kit-8 assay and microscopy were used to evaluate cell viability and observe the morphology of PC12 cells.The protective effect of the three ethanol extracts(at tested concentrations of 50,100 and 200μg/mL)against cytotoxicity to PC12 cells increased in a concentration-dependent manner.The ethyl acetate,petroleum ether and n-butanol extracts(each tested at 100,150 and 200μg/mL)had neuroprotective effects as well.The optimum effective concentration ranged from 50–200μg/mL,and the protective effect of the ethyl acetate extract was comparatively robust.These results demonstrate that organic extracts of Alpinia oxyphylla protect PC12 cells against apoptosis induced by hydrogen peroxide.Our findings should help identify the bioactive neuroprotective components in Alpinia oxyphylla. 展开更多

AIM: To study the chemical constituents of the rhizomes of Alpinia officinarum Hance. METHOD: Compounds were isolated by repeated column chromatography, and their structures were elucidated on the basis of spectral analysis. The cytotoxic activities of these compounds were evaluated with the T98G and B16F10 cell lines by the MTT assay. RESULTS: A dimeric diarylheptanoid, named alpinin B(1), along with three known diarylheptanoids were obtained, and their structures were identified as alpinin B(1), 1, 7-diphenyl-3,5-heptanedione(2),(4E)-1, 7-diphenylhept-4-en-3-one(3) and(4E)-7-(4-hydroxyphenyl)-1-phenylhept-4-en-3-one(4). CONCLUSION: Compound 1 is a new dimeric diarylheptanoid. The biosynthetic pathway of 1 was speculated to originate from a Michael reaction between compounds 2 and 3. Compound 3 showed cytotoxicity against the human glioblastoma T98G cell line with IC50 of 27 μmol L 1. 展开更多

Objective:To investigate antioxidant potential of methanol extract of Alpinia nigra leaves.Methods:The study was done by using various in vitro methods such as 1,1-diphenyl-2-picrylhydrazyl(DPPH),2,2-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid)(ABTS),nitric oxide and hydrogen peroxide radical scavenging assays.Phytochemical constituents,total phenolic content and total flavonoid content of the extract at different concentrations(10-500μg/mL)were determined.Results:Alpinia nigra leaves showed high free radical scavenging activity as evidenced by the low IG_(50)values in DPPH(64.51μg/mL),in ABTS(28.32μg/mL),in nitric oxide(80.02μg/mL)and in H_2O_2(77.45μg/mL)scavenging assays.Furthermore the TPC and TFC of the extract were found to be 69.25 mg gallic acid equivalent per gram of extract and 78.84 mg quercetin equivalent per gram of extract respectively.Conclusions:The results of present comprehensive analysis demonstrated that Alpinia nigra leaves possess high phenolic,flavonoid contents and potential antioxidant activity,and could be used as a viable source of natural antioxidants and might be exploited for functional foods and neutraceutical applications. 展开更多

In this review, the botany and uses of Alpinia zerumbet(yan shan jiang) are described, and the current knowledge of its phytochemistry, pharmacological properties, and clinical trials is summarized. An important ginger crop in East Asia, A. zerumbet has many uses, both medicinal and non-medicinal. Leaves are used to produce essential oils and herbal teas. Rhizomes are consumed as spices, and stem fibers are made into paper, fabrics, and handicrafts. In Brazil, tea from A. zerumbet leaves is believed to have hypotensive, diuretic, and anti-ulcerogenic properties. This species possesses many medicinal properties due to its chemical constituents, including flavonoids, phenolic acids, phenylpropanoids, kava pyrones, sterols, and terpenoids. Extracts of A. zerumbet display antioxidant, antimicrobial, insecticidal, anthelmintic, tyrosinase and melanogenesis inhibitory, anti-atherogenic, anti-aging, anti-glycation, integrase and neuraminidase inhibitory, lifespan prolongation, hair growth promotion, anticancer, antidepressant, anxiolytic, anti-obesity, analgesic, anti-inflammatory, hypolipidemic, anti-ulcerogenic, anti-platelet, osteoblastic, osteogenic, thrombolytic, and cardiac arrhythmogenic activities. Essential oils of A. zerumbet leaves have antimicrobial, larvicidal, antinociceptive, hypotensive, vasorelaxant, myorelaxant, antispasmodic, antidepressant, anxiolytic, anti-neuraminidase, anti-atherogenic, anti-aging, anti-melanogenic, anti-tyrosinase, cytoprotective, cardiodepressive, antipsychotic, analgesic, anti-inflammatory, and tissue healing activities. Clinical trials conducted in Brazil showed that extracts of A. zerumbet have hypotensive and diuretic effects whereas topical application of the essential oil has positive therapeutic effects on patients with fibromyalgia. Spanning two continents of Asia and South America, A. zerumbet is truly a multi-purpose ginger plant with promising medicinal properties. 展开更多

The rhizome of Alpinia officinarum Hance is a well-known traditional Chinese medicine(TCM)and has been widely used for the remedy of gastrointestinal diseases.In the present study,a simple and rapid HPLC-DAD was developed for the quality control of the rhizomes of A.officinarum.Its chemical fingerprint was established using raw material of 15 different origins in China.Similarity analysis(SA)and hierarchical clusting analysis(HCA)were applied to select the qualitative markers.Principal components analysis(PCA)was conducted to select the quantitative markers of the rhizomes of A.officinarum samples from different origins.The constituents were confirmed by(+)electrospray ionization LC-MS.12 constituents were selected as common peaks and 10 of them were confirmed by(+)electrospray ionization LC-MS.Six bioactive constituents including DPHA,galangin flavanone,galangin,galangin 3-methylether,DPHB and DPHC were simultaneous determination by using the HPLC-DAD analysis.The developed method was able to determine the bioactive components with excellent resolution,precision and recovery.The results indicated that chromatographic fingerprint combination with multi-components determination method is suitable for quality assessment of the rhizomes of A.officinarum. 展开更多

The purpose of this study was to investigate the possible prophylactic effects of Alpinia officinarum hance on experimentally induced acute and chronic colitis models,in-vivo and in-vitro.Acute and chronic colitis were induced in Male Wistar rats by administration of Dextran Sulfate Sodium(DSS)in drinking water.DSS induction exhibited colon shrinkage,increased the Disease Activity Index(DAI)score,increased the levels of inflammatory markers and caused severe anemia.DSS induced animals,co-treated with the hexane extract of Alpinia officinarum(HEAO)(200 mg/kg body wt),effectively suppressed colonic injury that was evidenced by the reduced DAI score,colon weight/length ratio,histological damage,proinflammatory markers and MPO activity.Further,it restored the colonic antioxidants near to normal levels by regulating the oxidative stress via attenuation of lipid peroxidation.Our results revealed that the degree of colitis caused by the administration of DSS was significantly attenuated by HEAO.In addition,the in-vitro study showed that HEAO treatment inhibited the proliferation of HT-29 cells and down regulated the mRNA expression of NF-B and COX-2.Taken together,these results suggest that HEAO is a promising anti-oxidant and anti-inflammatory agent that support its possible therapeutic role in the treatment of colitis. 展开更多

Objective: To evaluate antibacterial activity and the bioactive compounds of 50% hydroethanolic extract of Alpinia zerumbet(A. zerumbet) rhizomes.Methods: Eight reference microbial strains including two Gram-positive bacteria [Staphylococcus aureus(ATCC 29213) and Enterococcus faecalis(ATCC 29212)] and six Gram-negative bacteria [Escherichia coli(ATCC 25922), Klebsiella pneumoniae(ATTC 700603), Proteus mirabilis(DMST 8212), Salmonella enterica subsp. enterica serovar Vellore.(ATCC 15611), Shigella flexneri(ATCC 12022) and Pseudomonas aeruginosa(ATCC 27853)], were used to test antimicrobial susceptibility by the broth microdilution method. Bioactive compounds were analyzed by using HPLC.Results: The minimum inhibitory concentration values of A. zerumbet extract were 8 mg/mL for Staphylococcus aureus, Escherichia coli and Shigella flexneri and 16 mg/mL for Enterococcus faecalis and the other four Gram-negative bacilli. HPLC chromatograms revealed that the A. zerumbet extract contained hydroxybenzoic acids, hydroxycinnamic acids and flavonoids. Conclusions: The constituents of A. zerumbet rhizomes could be a potential source of antibacterial compounds, warranting further study of A. zerumbet extract. 展开更多

Objective:To explore cytotoxic activity of ethanol extract of Alpinia calcarata Rose(EEAC) rhizome against Ehiiich ascites carcinoma(EAC) tumor bearing Swiss Albino mice.Methods: In the present study,its anti-neoplastic activity has been studied by monitoring parameters like tumor weight measurement,survival time,tumor cell growth inhibition,haematological characteristics etc.Results:It was found that EEAC at dose 8 mg/kg/day(i.p.) significantly decreased tumor weight(62.0%;P【0.01),increased life span(70.25%;P 【0.01) and reduced tumor cell growth rate(85.7%;P【0.01) in comparison to those of EAC bearing mice.The plant extract also improved the depleted haematological parameters like RBC WBC,Hb%,differential counts(e.g.lymphocytes,neutrophils,monocytes etc) of EAC bearing mice towards normal.The host toxic effects were not very high and recovered gradually towards normal within a few days after treatment.ConchiskHis:EEAC exhibits potent in vivo cytotoxic activity against EAC tumor bearing Swiss Albino mice.So,the plant can be considered as a probable new source of antitumor agents. 展开更多

Four previously unreported diarylheptanoids(1a/1b-2a/2b),one undescribed sesquiterpenoid(8),one new diterpenoid(12),and twelve known analogs were isolated from the fruits of Alpinia oxyphylla.The structural elucidation of these compounds was achieved through a comprehensive analysis of spectroscopic data,single-crystal X-ray diffraction,electronic circular dichroism(ECD),and modified Mosher's method.Enantiomeric mixtures(1a/1b,2a/2b,3a/3b,4a/4b,and 5a/5b)were separated on a chiral column using acetonitrile-water mixtures as eluents.Among them,compounds 3a/3b and 4a/4b were isolated as optically pure enantiomers in the initial chiral separation.Furthermore,most of the isolates were evaluated for their inhibitory effects against the production of nitric oxide(NO)and interleukin-6(IL-6)in lipopolysaccharide(LPS)-induced RAW264.7 macrophages.Interestingly,2 and 4 showed significant inhibitory activities against NO production with IC50 values of 33.65 and 9.88μmol·L^(-1)(hydrocortisone:IC_(50)34.26μmol·L^(-1)),respectively.Additionally,they also partially reduced the secretion of IL-6. 展开更多

[Objectives]To explore the protective effect of Alpinia zerumbet(Pers.)Burttet Smith ethanol extraction(YSJ)on gastric ulcers.[Methods]The ethanol-induced gastric ulcer model was generated by male Kunming mice.The mice were divided into six groups,including control,model(GU),positive omeprazole enteric-coated capsules(18 mg/kg),YSJ low(1.17 g/kg),medium(2.34 g/kg)and high(4.68 g/kg)dose groups.To observe the state of gastric tissues,hematoxylin and eosin(H&E)staining was applied and immunohistochemistry(IHC)was used to evaluate the expression of nuclear factor kappa-B p65(NF-κB p65),interleukin 6(IL-6),tumor necrosis factor-α(TNF-α),and Cyclooxygenase-2(COX-2)in gastric tissues.The ulcer index,biochemical parameters,and inflammatory proteins were evaluated.In vitro,GES-1 cells were induced by anhydrous ethanol to found gastric ulcer model.The groups were the same as in vivo experiment.Cytotoxicity was tested by MTT and IL-1β,TNF-α,and IL-6 were detected by Elisa assays.[Results]The injuries of gastric tissue in the model group were unambiguously observed and improved after YSJ treatment.The levels of interleukin 1 beta(IL-1β),interleukin 6(IL-6),interleukin 8(IL-8),and TNF-αin serum and tissues were decreased(P<0.05 or 0.01),whereas interleukin 4(IL-4)and prostaglandin E 2(PGE 2)were increased after treatment with YSJ(P<0.05 or 0.01).The levels of IL-1β,IL-6,TNF-α,and IL-8 were decreased in YSJ groups,while IL-4 and PGE 2 presented the opposite trend.The protein expression of p-NF-κB,NF-κB,p-IκBα,IκBα,and TNF-αwas inhibited after treatment with YSJ(P<0.05 or 0.01).[Conclusions]These results demonstrate that YSJ alleviates the occurrence and development of gastric ulcers via inhibiting inflammation. 展开更多

OBJECTIVE:To detect the effects of active compounds of Caodoukou(Semen Alpinia Katsumadai)(ACAK)on the proliferation,migration and invasion of pancreatic cancer,and explain the possible molecular mechanism of ACAK interacting with these processes.Methods:Cell counting kit-8 method,cell scratch repair experiment,Transwell migration and invasion experiment,immunohistochemistry,western blot assay and real-time polymerase chain reaction experiment were used to evaluate the effect of ACAK on the proliferation,migration and invasion of pancreatic cancer cells.The levels of active molecules involved in the phosphoinosmde-3-kinase(PI3K)/Akt/the mammalian target of rapamycin(m TOR)signal transduction were detected by Western blot assay.In addition,the function of ACAK in vivo was evaluated by xenotransplantation tumor model in nude mice.Results:The inhibitory effect of ACAK on the proliferation of pancreatic cancer cells showed certain time-dose dependence.The results of scratch repair test,Transwell test,Western blotting and real time polymerase chain reaction assay showed that ACAK could inhibit the migration and invasion of pancreatic cancer cells in vitro.In addition,the regulatory effect of ACAK on epithelialmesenchymal transition(EMT)is partly attributed to PI3K/Akt/mT OR signaling pathway.The experimental results in vivo showed that ACAK regulated the development of pancreatic cancer.Conclusions:ACAK can partly inhibit the activity of EMT and matrix metallopeptidases by down-regulating the downstream proteins of PI3K/Akt/mTOR signal pathway,thus inhibiting the ability of migration and invasion of pancreatic cancer. 展开更多

Alpinia galanga, Eurycoma longifolia Jack and Syzygium aromaticum have been widely used in traditional medicine for decades. Antimicrobial activities for individual crude extract were well established. Crude methanolic extracts of Alpinia galanga, Eurycoma longifolia Jack and Syzygium aromaticum and the combination for all extracts were tested using well diffusion techniques against Saccharomyces cerevisiae (ATCC 9763) and Escherichia coli (ATCC 25922). The mixed extracts were prepared based on the concentration ratio of 50 μg/μL which are 1:1, 1:1:1 and 1:2:2. Single Syzygium aromaticum extract showed higher inhibition zone on Saccharomyces cerevisiae compared to Escherichia coli. There is reduction in diameter of inhibition zone for single extract and mixture extracts either in combination of two or three extracts tested on Saccharomyces cerevisiae and Escherichia coli growth but they are not significant. In conclusion, Syzygium aromaticum showed highest activity against Saccharomyces cerevisiae and Escherichia coli. Reduction in diameter of inhibition zone indicated that Alpinia galanga and Eurycoma longifolia Jack extracts had antagonistic effect with Syzygium aromaticum. 展开更多

Objective:To explore whether Yizhi(Alpinia oxyphylla,AO)could treat diabetic nephropathy(DN)through the inhibition of p47phox expression which is related to oxidative stress.Methods:Model of DN was established in SD rats with high glucose and fat diet for 4 weeks after STZ intraperitoneal injection.The rats were randomly divided into blank group,model group,AO group and irbesartan(IRB)group.Drugs were administered for 4 weeks.Blood glucose and 24-hour urinary protein were measured regularly.The index of ROS,SOD,MDA and Nox2 were tested by ELISA and the expression of p47phox protein in renal tissues were tested by techniques of RT-PCR and Western blot.Results:The decrease of blood sugar in AO group and IRB group were not obvious(P>0.05)while the drugs could significantly reduce the amount of protein in urine(P<0.05).AO could decrease ROS,MDA and Nox2 and increase SOD(P<0.05).It also could inhibit the expression of p47phox which was increased in model group(P<0.05).The IRB group has the similar effects compared with AO group(P>0.05).Conclusion:AO could reduce the amount of 24-hour urinary protein in DN rats and the mechanism may be related to the inhibition of p47phox expression thus alleviates excessive oxidative stress response. 展开更多
