Porous carbons hold broad application prospects in the domains of electrochemical energy storage devices and sensors.In this study,porous carbon derived from sodium alginate-encapsulated ZIF-8(SA/ZIF-8-C)was suc-cessf...Porous carbons hold broad application prospects in the domains of electrochemical energy storage devices and sensors.In this study,porous carbon derived from sodium alginate-encapsulated ZIF-8(SA/ZIF-8-C)was suc-cessfully prepared by blending ZIF-8 particles with sodium alginate,forming hydrogel beads in the presence of divalent metal ions,and subsequently subjecting them to high-temperature pyrolysis.Various characterization techniques were employed to evaluate the properties of the prepared materials.The introduction of a carbon framework on ZIF-8-derived particles effectively enhanced the conductivity of the prepared materials.The SA/ZIF-8(1.0)-C sample heated at 800℃exhibited a specific capacitance of up to 208 F g^(-1)at a current density of 0.5 A g^(-1)and outstanding cyclic stability.Even after 10,000 charge and discharge cycles,its capacitance retention rate remained as high as 87.14%.The symmetric supercapacitor constructed with the composite demonstrated an excellent energy density of 14.58 Wh kg^(-1)at a power capacity of 403.85 W kg^(-1).The implementation of this study provides new ideas and inspiration for the development of high-performance supercapacitors.展开更多
Moist-electric generation,a green and environmentally friendly energy harvesting technology,is undoubt-edly one of the effective methods to alleviate energy shortages and environmental damage.However,the lack of fiber...Moist-electric generation,a green and environmentally friendly energy harvesting technology,is undoubt-edly one of the effective methods to alleviate energy shortages and environmental damage.However,the lack of fiber-like moist-electric generators(MEGs)that combine continuous power generation and high electrical output performance has constrained the development of moist-electric in the fields of flexi-ble wearable and self-power supplies.In this work,sodium alginate(SA)/multi-walled carbon nanotubes(MWCNT)fibers with axial heterogeneous(axi-he)of oxygen-containing functional groups(Ocfgs)are prepared through a mold forming method in assistance with the coagulation process.The interaction be-tween axi-he MEG and moisture is investigated by analyzing the electrical signal changes of dried MEG under moisture stimulation.The maximum output voltage and current of axi-he MEG can reach 0.35 V and 1.92μA under the stimulation of moisture.Based on the regulation of Ocfgs,axi-he MEG has a con-tinuous high moist-electric performance and environmental adaptability.The maximum output power density(Pmo)of axi-he MEG with a length of only 2 cm can reach 27.37μW cm-2 at RH=90%,which exceeds most of the MEGs reported in literature.Meanwhile,a continuous output voltage of 0.33-0.37 V for more than 15 h can be obtained from this axi-he MEG.Thus,the axi-he MEG from Ocfg distribution design and mold forming method provides a new way of clean energy generation using moisture from the ambient environment,exhibiting enormous potential in energy supply for Internet of Things(IoT)devices.展开更多

However, the mechanical strength and stiffness of alginate hydrogels are still not comparable with biological tissues such as tendons and the printability of SA solutions is often poor. Here, a novel strategy for 3D printing of alginate hydrogels with high mechanical performance is developed by using glycerol as a co-solvent for SA solutions. The addition of glycerol (GL) enables the formation of a homogenous SA/GL solution with a high solid content of 12–20 wt.% and endows crosslinked SA hydrogels with high stretchability. By applying uniaxial stretches, hydrogel filaments prepared with concentrated SA/GL solutions reveal a high tensile strength of 36.6–161.3 MPa, Young's modulus of 59.2–1964.2 MPa, and elongation at break of 8.5 %–106.2 % due to the high orientated and closely packed SA chains. SA/GL solutions become more solid-like with increasing SA concentration, and the solution with a solid content of 16 wt.% exhibits optimal 3D printability because of the appropriate rheological properties and thixotropic behavior. By designing the deforming-and-fixing process, 3D printed high-strength alginate hydrogels with complex structures are prepared, broadening the application of alginate hydrogels in load-bearing and biomedical fields.展开更多
Sodium alginate(SA)is generally considered highly hydrophilic due to two hydroxyl groups and a carboxylate group on each pyranose ring.However,SA will form a gel after dissolving in water for a certain period.The two ...Sodium alginate(SA)is generally considered highly hydrophilic due to two hydroxyl groups and a carboxylate group on each pyranose ring.However,SA will form a gel after dissolving in water for a certain period.The two properties of SA,hydrophilicity and gelation,seem to be paradoxical.In this study,to explore the mechanism behind these paradoxical properties,the single-chain behaviors of SA in various liquid environments have been investigated by using single-molecule force spectroscopy(SMFS).In nonpolar solvents such as nonane,SA exhibits its single-chain inherent elasticity consistent with the theoretical elasticity derived from quantum mechanical(QM)calculations.Notably,the experimental curve of SA obtained in water shows a long plateau in the low force region.Further research reveals that this phenomenon is driven by the hydrophobic effect.Additionally,SA shows greater rigidity than its inherent elasticity in the middle and high force regions due to electrostatic repulsion between carboxylate groups on adjacent sugar rings.Comparative single-molecule studies suggest that SA exhibits considerable hydrophobicity,offering new insights into the gelation process in water.展开更多
Alginate-based magnetic micro/millirobots have demonstrated significant potential for biomedical applications due to their flexible structures and capacity to carry various types of cargo,such as cells,enabling target...Alginate-based magnetic micro/millirobots have demonstrated significant potential for biomedical applications due to their flexible structures and capacity to carry various types of cargo,such as cells,enabling targeted therapy to specific diseased regions within the body.Their active therapy is typically achieved through magnetic actuation and magnetic heating,while monitored by medical imaging methods like CT which pose additional risks due to radiation exposure.In the last decades,a novel imaging method for superparamagnetic materials,known as magnetic particle imaging(MPI),has been under active development,offering not only positional tracking but also the ability to measure concentration and temperature.Here,we report the world's first MPI-traceable magnetic hydrogel robots,which employ a combination of iron oxide nanoflowers,NdFeB powder,and calcium alginate.Unlike previous magnetic alginate robots composed of a single magnetic material,the synergistic combination of NdFeB and nanoflowers enables these robots to exhibit triple magnetic functionalities:magnetic heating,locomotion at low magnetic fields,and tracking,all of which can be controlled using a single all-in-one electromagnetic coil system.The effects of various magnetization fields,as well as different concentrations of NdFeB and nanoflowers on the robots'magnetic properties were analyzed.This led to the development of three types of triple-function robots(spiral,droplet,and hybrid),with experimental results demonstrating biocompatibility,a magnetic heating temperature increase of over 10℃in plasma fluid under a magnetic field of 13 kA·m^(-1)at 200 kHz,locomotion speeds of up to 25 mm·s^(-1)in fields below 2 mT,and an MPI tracking error of 2.8 mm with a selection field of 0.4 mT·mm^(-1).Additionally,the robots'capacity for localized thermal therapy and selectively targeted cell delivery,as well as their locomotion within a medical phantom against a maximum flow of 50 mm·s^(-1)were demonstrated.展开更多
Alginate is an edible heteropolysaccharide that abundantly available in the brown seaweed and the capsule of bacteria such as Azotobacter sp.and Pseudomonas sp.Owing to alginate gel forming capability,it is widely use...Alginate is an edible heteropolysaccharide that abundantly available in the brown seaweed and the capsule of bacteria such as Azotobacter sp.and Pseudomonas sp.Owing to alginate gel forming capability,it is widely used in food,textile and paper industries;and to a lesser extent in biomedical applications as biomaterial to promote wound healing and tissue regeneration.This is evident from the rising use of alginate-based dressing for heavily exuding wound and their mass availability in the market nowadays.However,alginate also has limitation.When in contact with physiological environment,alginate could gelate into softer structure,consequently limits its potential in the soft tissue regeneration and becomes inappropriate for the usage related to load bearing body parts.To cater this problem,wide range of materials have been added to alginate structure,producing sturdy composite materials.For instance,the incorporation of adhesive peptide and natural polymer or synthetic polymer to alginate moieties creates an improved composite material,which not only possesses better mechanical properties compared to native alginate,but also grants additional healing capability and promote better tissue regeneration.In addition,drug release kinetic and cell viability can be further improved when alginate composite is used as encapsulating agent.In this review,preparation of alginate and alginate composite in various forms(fibre,bead,hydrogel,and 3D-printed matrices)used for biomedical application is described first,followed by the discussion of latest trend related to alginate composite utilization in wound dressing,drug delivery,and tissue engineering applications.展开更多

The N-[(2-hydroxy- 3-trimethylammonium)propyl] chitosan chloride (HTCC) was synthesized, and the quatemized chitosan nanoparticles (HTCC-T NPs) were prepared by ionic gelation of HTCC using tripolyphosphate (TPP). The alginate-coated quatemized chitosan nanoparticles (HTCC-A NPs) were prepared by coating HTCC-T NPs with alginate (ALG) solution under mild agitation. Particle size, zeta potential, surface morphology, drug loading and entrapment efficiency of HTCC-A NPs were characterized using Zeta-sizer, TEM and HPLC assays. It was found that HTCC-A NPs exhibited uniform spherical particles with the size of (322.2±8.5) nm and positive charges (14.1±0.6) mV. Our data showed that the release behavior of HTCC-A NPs was quite different from that of HTCC-T NPs (without ALG coating) when incubated with various medium at different pH values in vitro, suggesting that ALG coating over the HTCC-T NPs improved the release profile of insulin from the NPs for a successful oral delivery. The ALG coating could also improve the stability of insulin against enzymatic degradation. From circular dichroism spectrum, it was revealed that HTCC-A NPs were capable of maintaining the conformation of insulin. The relative pharmacological bioavailability of HTCC-A NPs was 8.0%±2.5% by intraduodenal administration. The HTCC-A NPs significantly increased (P〈0.05) the relative pharmacological availability (2.2 folds) compared with HTCC-T NPs after oral administration. HTCC-A NPs significantly enhanced the in vivo oral absorption of insulin and exhibited promising potentials for oral delivery.展开更多
Background Alginate oligosaccharide(AOS)holds great potential as a novel feed supplement in farm animals.However,the effects of AOS on chicken health and the underlying mechanisms are not fully understood.This study a...Background Alginate oligosaccharide(AOS)holds great potential as a novel feed supplement in farm animals.However,the effects of AOS on chicken health and the underlying mechanisms are not fully understood.This study aimed to optimize the enzymatic preparation of AOS by using bacterial alginate lyases expressed in yeast,investigate the effects of the prepared AOS on the growth performance and gut health of broiler chickens,and reveal the underlying mechanisms.Results Five alginate lyases from bacteria were cloned into Pichia pastoris GS115 and the alginate lyase PDE9 was expressed at relatively high yield,activity and stability in P.pastoris.Animal trials were carried out using 3201-day-old male Arbor Acres broilers(four groups;8 replicates/group×10 chicks/replicate)receiving either a basal diet or the same diet supplemented with 100,200 and 400 mg/kg PDE9-prepared AOS for 42 d.The results showed that dietary supplementation of 200 mg/kg AOS displayed the highest activity in promoting the birds’ADG and ADFI(P<0.05).AOS ameliorated the intestinal morphology,absorption function and barrier function,as indicated by the enhanced(P<0.05)intestinal villus height,maltase activity,and the expression of PEPT,SGLT1,ZNT1,and occludin.AOS also increased serum insulin-like growth factor-1,ghrelin(P<0.05),and growth hormone(P<0.1).Moreover,the concentrations of acetate,isobutyrate,isovalerate,valerate,and total SCFAs in cecum of birds fed AOS were significantly higher than the control birds(P<0.05).Metagenomic analysis indicated that AOS modulated the chicken gut microbiota structure,function,and microbial interactions and promoted the growth of SCFAs-producing bacteria,for example,Dorea sp.002160985;SCFAs,especially acetate,were found positively correlated with the chicken growth performance and growth-related hormone signals(P<0.05).We further verified that AOS can be utilized by Dorea sp.to grow and to produce acetate in vitro.Conclusions We demonstrated that the enzymatically produced AOS effectively promoted broiler chicken growth performance by modulating the chicken gut microbiota structure and function.For the first time,we established the connections among AOS,chicken gut microbiota/SCFAs,growth hormone signals and chicken growth performance.展开更多

Most of the current knowledge about genes involved in the alginate biosynthesis comes from bacterial systems. In this article, based on some algal and bacterial algG genes registered on GenBank and EMBL databases, we predicted 94 algG genes open reading frame (ORF) sequences of brown algae from the 1 000 Plant Transcriptome Sequencing Project (OneKP). By method of transcriptomic sequence analysis, gene structure and gene localization analysis, multiple sequence alignment and phylogenetic tree construction, we studied the algal algG gene family characteristics, the structure modeling and conserved motifs of AlgG protein, the origin of alginate biosyn-thesis and the variation incidents that might have happened during evolution in algae. Although there are different members in the algal algG gene family, almost all of them harbor the conserved epimerase region. Based on the phylogenetic analysis of algG genes, we proposed that brown algae acquired the alginate bio-synthesis pathway from an ancient bacterium by horizontal gene transfer (HGT). Afterwards, followed by duplications, chromosome disorder, mutation or recombination during evolution, brown algal algG genes were divided into different types.展开更多
The purpose of this study was to identify and compare the degradation efficiencies of free and entrapped bacterial consortia(Staphylococcus capitis CP053957.1 and Achromobacter marplatensis MT078618.1)to different pol...The purpose of this study was to identify and compare the degradation efficiencies of free and entrapped bacterial consortia(Staphylococcus capitis CP053957.1 and Achromobacter marplatensis MT078618.1)to different polymers such as Sodium Alginate(SA),Sodium Alginate/Poly(Vinyl Alcohol)(SA/PVA),and Bushnell Haas Agar(BHA).In addition to SA and SA/PVA,which are cost-effective,non-toxic and have different functional groups,BHA,which is frequently encountered in laboratory-scale studies but has not been used as an entrapment material until now.Based on these,the polymers with different surface morphologies and chemical compositions were analyzed by SEM and FT-IR.While the petroleum removal efficiency was higher with the entrapped bacterial consortia than with the free one,BHA-entrapped bacterial consortium enhanced the petroleum removal more than SA and SA/PVA.Accordingly,the degradation rate of bacterial consortia entrapped with BHA was 2.039 day^(-1),SA/PVA was 1.560,SA was 0.993,the half-life period of BHA-entrapped bacterial consortia is quite low(t_(1/2)=0.339)compared with SA(t_(1/2)=0.444)and SA/PVA(t_(1/2)=0.697).The effects of the four main factors such as:amount of BHA(0.5,1,1.5,2,2.5,3 g),disc size(4,5,6,7,8 mm),inoculum concentration(1,2.5,5,7.5,10 mL),and incubation period on petroleum removal were also investigated.The maximum petroleum removal(94.5%)was obtained at≥2.5 mL of bacterial consortium entrapped in 2 g BHA with a 7 mm disc size at 168 h and the results were also confirmed by statistical analysis.Although a decrease was observed during the reuse of bacterial consortium entrapped in BHA,the petroleum removal was still above 50%at 10th cycle.Based on GC-MS analysis,the removal capacity of BHA-entrapped consortium was over 90%for short-chain n-alkanes and 80%for medium-chain n-alkanes.Overall,the obtained data are expected to provide a potential guideline in cleaning up the large-scale oil pollution in the future.Since there has been no similar study investigating petroleum removal with the bacterial consortia entrapped with BHA,this novel entrapment material can potentially be used in the treatment of petroleum pollution in advanced remediation studies.展开更多

UV-Vis spectroscopy was employed to investigate the activity of papain to evaluate kinetics and stability.Analysis results show that the highest affinity the lowest Michaelis-Menten constant Km =11.0 mg/mL and the highest stability are obtained when using Cu2+as the crosslinker.The effect of the mass ratio of TiO2 to papain on the stability and leakage of papain is also investigated and the results show that 10∶1 TiO2∶papain is optimal because the proper use of TiO2 can reduce enzyme leakage and ensure enzyme stability.Preparing Cu/alginate/TiO2 hybrid gels via ionotropic gelation can provide a satisfactory diffusion capability and enzyme stability.展开更多

Alginate gel becomes buoyant and CO2-aerated as soon as Gaviscon comes into contact with gastric contents. This system can be characterized by several indicators, such as strength or coherence, volume, buoyancy, reflux resistance and resilience. In addition, the strength of the raft is influenced by many factors, such as molecular weight and the ratio of D-mannuronic and L-guluronic acid residues(M/G), which are intrinsic factors in this formulation. Besides, there are several extrinsic factors, such as the presence of specific cations, the amount of carbon dioxide generated and entrapped in the raft. This review focused on how to evaluate the raft, introduced some factors that impact the raft and summarized the degradation of alginate and future advance.展开更多

The traditional anion reverse flotation of hematite cannot effectively eliminate the effects of chlorite, leading to a significant decrease in the total Fe(TFe) grade of the concentrate. In this work, the effect of sodium alginate on the reverse flotation of hematite was systematically investigated. Flotation tests of artificially mixed ores were conducted, and the results showed that sodium alginate can significantly improve the removal rates of quartz and chlorite. The adsorption measurements, infrared spectroscopy, and contact angle tests demonstrated that sodium alginate adsorbs on the quartz surface by chelating with calcium ions, thereby weakening the steric hindrance of oleate ions and increasing the adsorption capacity of sodium oleate to ultimately improve the removal rate of quartz. Furthermore, owing to its lower density and fine particle size, chlorite is easily entrained into the foam layer. Sodium alginate dramatically increases the liquid-to-gas ratio of the foam layer by increasing pulp viscosity, thereby increasing the entrainment rate of chlorite and finally improving its removal rate. The core content of this thesis bears significance in improving the Fe grade in the reverse flotation of chlorite-containing hematite.展开更多

A novel environmental friendly material, calcium alginate immobilized kaolin (kaolin/CA), was prepared using a sol-gel method. The effects of contact time, pH, adsorbent dose, and temperature on Cu2+ adsorption by kaolin/CA were investigated. The Langmuir isotherm was used to describe the experimental adsorption, the maximum Cu2+ adsorption capacity of the kaolin/CA reached up to 53.63 mg/g. The thermodynamic studies showed that the adsorption reaction was a spontaneous and endothermic process.展开更多

With low viscosity and good water solubility, as well as anti-oxidant, immune regulation, anti-bacterial and antiinflammatory activities, AOS have been widely used in medical science and functional food, green agriculture and other fields. As new bio-feed additives, AOS have broad potential applications in animal husbandry. In this review, the sources of alginate, chemical structure and preparation methods of AOS, and their biological activities and application in livestock and poultry are summarized. We expect this review could contribute to lay a foundation of application and further research for AOS in livestock and poultry.展开更多

Nevertheless, the mechan...Background: Alginate oligosaccharide(AOS), produced from alginate by alginate lyase-mediated depolymerisation, is a potential substitute for antibiotics and possesses growth-enhancing effects. Nevertheless, the mechanisms by which AOS regulates porcine growth remain to be elucidated. Therefore, we investigated the AOS-mediated changes in the growth performance of weaned pigs by determining the intestinal morphology, barrier function,as well as epithelium apoptosis.Methods: Twenty-four weaned pigs were distributed into two groups(n = 12) and received either a basal diet(control group) or the same diet supplemented with 100 mg/kg AOS. On d 15, D-xylose(0.1 g/kg body weight)was orally administrated to eight randomly selected pigs per treatment, and their serum and intestinal mucosa samples were collected 1 h later.Results: Our results showed that inclusion of AOS in the diet for 2 wk increased(P < 0.05) the average daily body weight gain in weaned pigs. Notably, AOS supplementation ameliorated the intestinal morphology and barrier function, as suggested by the enhanced(P < 0.05) intestinal villus height, secretory immunoglobulin A content and goblet cell counts. Compared to the control group, AOS ingestion both decreased(P < 0.05) the total apoptotic percentage and increased(P < 0.05) the proportion of S phase in the intestinal epithelial cells. Furthermore, AOS not only up-regulated(P < 0.05) the B-cell lymphoma-2(BCL2) transcriptional level but also down-regulated(P < 0.05) the B-cell lymphoma-2-associated X protein(BAX), cysteinyl aspartate-specific proteinase-3(caspase-3) and caspase-9 transcriptional levels in the small intestine.Conclusions: In summary, this study provides evidence that supplemental AOS beneficially affects the growth performance of weaned pigs, which may result from the improved intestinal morphology and barrier function,as well as the inhibited enterocyte death, through reducing apoptosis via mitochondria-dependent apoptosis.展开更多

文摘The mini-review explores the potential use of alginates produced biotechnologically by bacteria for the development of various implantable biomaterials intended for bone and cartilage tissue regeneration in orthopedics:the recent studies on the use of algal alginate-based biomaterials in the form of hydrogels,scaffolds,and microparticles for medical applications are considered as a potential opportunity to use bacterial alginate for these applications,taking into account the advantages of biotechnological production of a polymer with desired properties.The methods of producing different alginate-based biomaterials,the manufacturing of implantable medical devices using them,and the surgical techniques for bone and cartilage tissue regeneration using these materials for orthopedic purposes are discussed.

文摘Porous carbons hold broad application prospects in the domains of electrochemical energy storage devices and sensors.In this study,porous carbon derived from sodium alginate-encapsulated ZIF-8(SA/ZIF-8-C)was suc-cessfully prepared by blending ZIF-8 particles with sodium alginate,forming hydrogel beads in the presence of divalent metal ions,and subsequently subjecting them to high-temperature pyrolysis.Various characterization techniques were employed to evaluate the properties of the prepared materials.The introduction of a carbon framework on ZIF-8-derived particles effectively enhanced the conductivity of the prepared materials.The SA/ZIF-8(1.0)-C sample heated at 800℃exhibited a specific capacitance of up to 208 F g^(-1)at a current density of 0.5 A g^(-1)and outstanding cyclic stability.Even after 10,000 charge and discharge cycles,its capacitance retention rate remained as high as 87.14%.The symmetric supercapacitor constructed with the composite demonstrated an excellent energy density of 14.58 Wh kg^(-1)at a power capacity of 403.85 W kg^(-1).The implementation of this study provides new ideas and inspiration for the development of high-performance supercapacitors.

文摘Moist-electric generation,a green and environmentally friendly energy harvesting technology,is undoubt-edly one of the effective methods to alleviate energy shortages and environmental damage.However,the lack of fiber-like moist-electric generators(MEGs)that combine continuous power generation and high electrical output performance has constrained the development of moist-electric in the fields of flexi-ble wearable and self-power supplies.In this work,sodium alginate(SA)/multi-walled carbon nanotubes(MWCNT)fibers with axial heterogeneous(axi-he)of oxygen-containing functional groups(Ocfgs)are prepared through a mold forming method in assistance with the coagulation process.The interaction be-tween axi-he MEG and moisture is investigated by analyzing the electrical signal changes of dried MEG under moisture stimulation.The maximum output voltage and current of axi-he MEG can reach 0.35 V and 1.92μA under the stimulation of moisture.Based on the regulation of Ocfgs,axi-he MEG has a con-tinuous high moist-electric performance and environmental adaptability.The maximum output power density(Pmo)of axi-he MEG with a length of only 2 cm can reach 27.37μW cm-2 at RH=90%,which exceeds most of the MEGs reported in literature.Meanwhile,a continuous output voltage of 0.33-0.37 V for more than 15 h can be obtained from this axi-he MEG.Thus,the axi-he MEG from Ocfg distribution design and mold forming method provides a new way of clean energy generation using moisture from the ambient environment,exhibiting enormous potential in energy supply for Internet of Things(IoT)devices.

文摘As a biomass material with biodegradability and biocompatibility, sodium alginate (SA) is a good candidate for constructing hydrogels for tissue-mimicking and biomedical scaffold fabricating through extrusion-based 3D printing technology. However, the mechanical strength and stiffness of alginate hydrogels are still not comparable with biological tissues such as tendons and the printability of SA solutions is often poor. Here, a novel strategy for 3D printing of alginate hydrogels with high mechanical performance is developed by using glycerol as a co-solvent for SA solutions. The addition of glycerol (GL) enables the formation of a homogenous SA/GL solution with a high solid content of 12–20 wt.% and endows crosslinked SA hydrogels with high stretchability. By applying uniaxial stretches, hydrogel filaments prepared with concentrated SA/GL solutions reveal a high tensile strength of 36.6–161.3 MPa, Young's modulus of 59.2–1964.2 MPa, and elongation at break of 8.5 %–106.2 % due to the high orientated and closely packed SA chains. SA/GL solutions become more solid-like with increasing SA concentration, and the solution with a solid content of 16 wt.% exhibits optimal 3D printability because of the appropriate rheological properties and thixotropic behavior. By designing the deforming-and-fixing process, 3D printed high-strength alginate hydrogels with complex structures are prepared, broadening the application of alginate hydrogels in load-bearing and biomedical fields.

文摘Sodium alginate(SA)is generally considered highly hydrophilic due to two hydroxyl groups and a carboxylate group on each pyranose ring.However,SA will form a gel after dissolving in water for a certain period.The two properties of SA,hydrophilicity and gelation,seem to be paradoxical.In this study,to explore the mechanism behind these paradoxical properties,the single-chain behaviors of SA in various liquid environments have been investigated by using single-molecule force spectroscopy(SMFS).In nonpolar solvents such as nonane,SA exhibits its single-chain inherent elasticity consistent with the theoretical elasticity derived from quantum mechanical(QM)calculations.Notably,the experimental curve of SA obtained in water shows a long plateau in the low force region.Further research reveals that this phenomenon is driven by the hydrophobic effect.Additionally,SA shows greater rigidity than its inherent elasticity in the middle and high force regions due to electrostatic repulsion between carboxylate groups on adjacent sugar rings.Comparative single-molecule studies suggest that SA exhibits considerable hydrophobicity,offering new insights into the gelation process in water.

文摘Alginate-based magnetic micro/millirobots have demonstrated significant potential for biomedical applications due to their flexible structures and capacity to carry various types of cargo,such as cells,enabling targeted therapy to specific diseased regions within the body.Their active therapy is typically achieved through magnetic actuation and magnetic heating,while monitored by medical imaging methods like CT which pose additional risks due to radiation exposure.In the last decades,a novel imaging method for superparamagnetic materials,known as magnetic particle imaging(MPI),has been under active development,offering not only positional tracking but also the ability to measure concentration and temperature.Here,we report the world's first MPI-traceable magnetic hydrogel robots,which employ a combination of iron oxide nanoflowers,NdFeB powder,and calcium alginate.Unlike previous magnetic alginate robots composed of a single magnetic material,the synergistic combination of NdFeB and nanoflowers enables these robots to exhibit triple magnetic functionalities:magnetic heating,locomotion at low magnetic fields,and tracking,all of which can be controlled using a single all-in-one electromagnetic coil system.The effects of various magnetization fields,as well as different concentrations of NdFeB and nanoflowers on the robots'magnetic properties were analyzed.This led to the development of three types of triple-function robots(spiral,droplet,and hybrid),with experimental results demonstrating biocompatibility,a magnetic heating temperature increase of over 10℃in plasma fluid under a magnetic field of 13 kA·m^(-1)at 200 kHz,locomotion speeds of up to 25 mm·s^(-1)in fields below 2 mT,and an MPI tracking error of 2.8 mm with a selection field of 0.4 mT·mm^(-1).Additionally,the robots'capacity for localized thermal therapy and selectively targeted cell delivery,as well as their locomotion within a medical phantom against a maximum flow of 50 mm·s^(-1)were demonstrated.

文摘Alginate is an edible heteropolysaccharide that abundantly available in the brown seaweed and the capsule of bacteria such as Azotobacter sp.and Pseudomonas sp.Owing to alginate gel forming capability,it is widely used in food,textile and paper industries;and to a lesser extent in biomedical applications as biomaterial to promote wound healing and tissue regeneration.This is evident from the rising use of alginate-based dressing for heavily exuding wound and their mass availability in the market nowadays.However,alginate also has limitation.When in contact with physiological environment,alginate could gelate into softer structure,consequently limits its potential in the soft tissue regeneration and becomes inappropriate for the usage related to load bearing body parts.To cater this problem,wide range of materials have been added to alginate structure,producing sturdy composite materials.For instance,the incorporation of adhesive peptide and natural polymer or synthetic polymer to alginate moieties creates an improved composite material,which not only possesses better mechanical properties compared to native alginate,but also grants additional healing capability and promote better tissue regeneration.In addition,drug release kinetic and cell viability can be further improved when alginate composite is used as encapsulating agent.In this review,preparation of alginate and alginate composite in various forms(fibre,bead,hydrogel,and 3D-printed matrices)used for biomedical application is described first,followed by the discussion of latest trend related to alginate composite utilization in wound dressing,drug delivery,and tissue engineering applications.

文摘In the present work, we aimed to develop alginate-coated chitosan nanoparticles for oral insulin delivery. The N-[(2-hydroxy- 3-trimethylammonium)propyl] chitosan chloride (HTCC) was synthesized, and the quatemized chitosan nanoparticles (HTCC-T NPs) were prepared by ionic gelation of HTCC using tripolyphosphate (TPP). The alginate-coated quatemized chitosan nanoparticles (HTCC-A NPs) were prepared by coating HTCC-T NPs with alginate (ALG) solution under mild agitation. Particle size, zeta potential, surface morphology, drug loading and entrapment efficiency of HTCC-A NPs were characterized using Zeta-sizer, TEM and HPLC assays. It was found that HTCC-A NPs exhibited uniform spherical particles with the size of (322.2±8.5) nm and positive charges (14.1±0.6) mV. Our data showed that the release behavior of HTCC-A NPs was quite different from that of HTCC-T NPs (without ALG coating) when incubated with various medium at different pH values in vitro, suggesting that ALG coating over the HTCC-T NPs improved the release profile of insulin from the NPs for a successful oral delivery. The ALG coating could also improve the stability of insulin against enzymatic degradation. From circular dichroism spectrum, it was revealed that HTCC-A NPs were capable of maintaining the conformation of insulin. The relative pharmacological bioavailability of HTCC-A NPs was 8.0%±2.5% by intraduodenal administration. The HTCC-A NPs significantly increased (P〈0.05) the relative pharmacological availability (2.2 folds) compared with HTCC-T NPs after oral administration. HTCC-A NPs significantly enhanced the in vivo oral absorption of insulin and exhibited promising potentials for oral delivery.

文摘Background Alginate oligosaccharide(AOS)holds great potential as a novel feed supplement in farm animals.However,the effects of AOS on chicken health and the underlying mechanisms are not fully understood.This study aimed to optimize the enzymatic preparation of AOS by using bacterial alginate lyases expressed in yeast,investigate the effects of the prepared AOS on the growth performance and gut health of broiler chickens,and reveal the underlying mechanisms.Results Five alginate lyases from bacteria were cloned into Pichia pastoris GS115 and the alginate lyase PDE9 was expressed at relatively high yield,activity and stability in P.pastoris.Animal trials were carried out using 3201-day-old male Arbor Acres broilers(four groups;8 replicates/group×10 chicks/replicate)receiving either a basal diet or the same diet supplemented with 100,200 and 400 mg/kg PDE9-prepared AOS for 42 d.The results showed that dietary supplementation of 200 mg/kg AOS displayed the highest activity in promoting the birds’ADG and ADFI(P<0.05).AOS ameliorated the intestinal morphology,absorption function and barrier function,as indicated by the enhanced(P<0.05)intestinal villus height,maltase activity,and the expression of PEPT,SGLT1,ZNT1,and occludin.AOS also increased serum insulin-like growth factor-1,ghrelin(P<0.05),and growth hormone(P<0.1).Moreover,the concentrations of acetate,isobutyrate,isovalerate,valerate,and total SCFAs in cecum of birds fed AOS were significantly higher than the control birds(P<0.05).Metagenomic analysis indicated that AOS modulated the chicken gut microbiota structure,function,and microbial interactions and promoted the growth of SCFAs-producing bacteria,for example,Dorea sp.002160985;SCFAs,especially acetate,were found positively correlated with the chicken growth performance and growth-related hormone signals(P<0.05).We further verified that AOS can be utilized by Dorea sp.to grow and to produce acetate in vitro.Conclusions We demonstrated that the enzymatically produced AOS effectively promoted broiler chicken growth performance by modulating the chicken gut microbiota structure and function.For the first time,we established the connections among AOS,chicken gut microbiota/SCFAs,growth hormone signals and chicken growth performance.

文摘The coding product of alginate-c5-mannuronan-epimerase gene (algG gene) can catalyze the conversion of mannuronate to guluronate and determine the M/G ratio of alginate. Most of the current knowledge about genes involved in the alginate biosynthesis comes from bacterial systems. In this article, based on some algal and bacterial algG genes registered on GenBank and EMBL databases, we predicted 94 algG genes open reading frame (ORF) sequences of brown algae from the 1 000 Plant Transcriptome Sequencing Project (OneKP). By method of transcriptomic sequence analysis, gene structure and gene localization analysis, multiple sequence alignment and phylogenetic tree construction, we studied the algal algG gene family characteristics, the structure modeling and conserved motifs of AlgG protein, the origin of alginate biosyn-thesis and the variation incidents that might have happened during evolution in algae. Although there are different members in the algal algG gene family, almost all of them harbor the conserved epimerase region. Based on the phylogenetic analysis of algG genes, we proposed that brown algae acquired the alginate bio-synthesis pathway from an ancient bacterium by horizontal gene transfer (HGT). Afterwards, followed by duplications, chromosome disorder, mutation or recombination during evolution, brown algal algG genes were divided into different types.
文摘The purpose of this study was to identify and compare the degradation efficiencies of free and entrapped bacterial consortia(Staphylococcus capitis CP053957.1 and Achromobacter marplatensis MT078618.1)to different polymers such as Sodium Alginate(SA),Sodium Alginate/Poly(Vinyl Alcohol)(SA/PVA),and Bushnell Haas Agar(BHA).In addition to SA and SA/PVA,which are cost-effective,non-toxic and have different functional groups,BHA,which is frequently encountered in laboratory-scale studies but has not been used as an entrapment material until now.Based on these,the polymers with different surface morphologies and chemical compositions were analyzed by SEM and FT-IR.While the petroleum removal efficiency was higher with the entrapped bacterial consortia than with the free one,BHA-entrapped bacterial consortium enhanced the petroleum removal more than SA and SA/PVA.Accordingly,the degradation rate of bacterial consortia entrapped with BHA was 2.039 day^(-1),SA/PVA was 1.560,SA was 0.993,the half-life period of BHA-entrapped bacterial consortia is quite low(t_(1/2)=0.339)compared with SA(t_(1/2)=0.444)and SA/PVA(t_(1/2)=0.697).The effects of the four main factors such as:amount of BHA(0.5,1,1.5,2,2.5,3 g),disc size(4,5,6,7,8 mm),inoculum concentration(1,2.5,5,7.5,10 mL),and incubation period on petroleum removal were also investigated.The maximum petroleum removal(94.5%)was obtained at≥2.5 mL of bacterial consortium entrapped in 2 g BHA with a 7 mm disc size at 168 h and the results were also confirmed by statistical analysis.Although a decrease was observed during the reuse of bacterial consortium entrapped in BHA,the petroleum removal was still above 50%at 10th cycle.Based on GC-MS analysis,the removal capacity of BHA-entrapped consortium was over 90%for short-chain n-alkanes and 80%for medium-chain n-alkanes.Overall,the obtained data are expected to provide a potential guideline in cleaning up the large-scale oil pollution in the future.Since there has been no similar study investigating petroleum removal with the bacterial consortia entrapped with BHA,this novel entrapment material can potentially be used in the treatment of petroleum pollution in advanced remediation studies.

文摘In order to improve the substrate diffusion properties and stability of an immobilized enzyme alginate microgels modified with TiO2 nanoparticles were employed as the enzyme immobilizing support.Ionotropic gelation was applied for the preparation of hybrid gels while Ca2+ Ce3+ Ni2+Cu2+and Fe3+were employed as the crosslinkers.Papain was selected as the model enzyme. UV-Vis spectroscopy was employed to investigate the activity of papain to evaluate kinetics and stability.Analysis results show that the highest affinity the lowest Michaelis-Menten constant Km =11.0 mg/mL and the highest stability are obtained when using Cu2+as the crosslinker.The effect of the mass ratio of TiO2 to papain on the stability and leakage of papain is also investigated and the results show that 10∶1 TiO2∶papain is optimal because the proper use of TiO2 can reduce enzyme leakage and ensure enzyme stability.Preparing Cu/alginate/TiO2 hybrid gels via ionotropic gelation can provide a satisfactory diffusion capability and enzyme stability.
文摘Formulations of Gaviscon based on novel alginate raft forming system have been developed to prevent gastroesophageal reflux disease(GERD) symptoms. Alginate gel becomes buoyant and CO2-aerated as soon as Gaviscon comes into contact with gastric contents. This system can be characterized by several indicators, such as strength or coherence, volume, buoyancy, reflux resistance and resilience. In addition, the strength of the raft is influenced by many factors, such as molecular weight and the ratio of D-mannuronic and L-guluronic acid residues(M/G), which are intrinsic factors in this formulation. Besides, there are several extrinsic factors, such as the presence of specific cations, the amount of carbon dioxide generated and entrapped in the raft. This review focused on how to evaluate the raft, introduced some factors that impact the raft and summarized the degradation of alginate and future advance.

文摘Polymer binder plays a pivotal role in electrochemical performance of high-capacity silicon(Si)anode that usually suffers from severe capacity fading due to enormous substantial volume change of Si during cycling.In an effort to find efficient polymer binder that could mitigate such capacity fading,alginate-carboxymethyl chitosan(Alg-C-chitosan)composite polymer was investigated as a low-cost watersoluble binder for silicon anodes in lithium-ion batteries.The electrostatic interaction between carboxylate(-COO-)of Alg and protonated amines(-NH3+)of C-chitosan forms a selfhealing porous scaffold structure.Synergistic effect on the enhanced porous scaffold structure and self-healing electrostatic interaction of Alg-C-chitosan binder effectively can tolerate the tremendous volume change of Si and maintain an integrated electrode structure during cycling process.The Si nanopowder electrodes with Alg-C-chitosan composite binder exhibit an excellent cycling stability,with a capacity of750 mAh·g-1 remaining after 100 th cycling.In addition,an extraordinary areal capacity of 3.76 mAh·cm-2 is achieved for Si-based anodes with Alg-C-chitosan binder.

文摘Given the gradual increase in the chlorite content of hematite ores, pulp properties seriously deteriorate during flotation. The traditional anion reverse flotation of hematite cannot effectively eliminate the effects of chlorite, leading to a significant decrease in the total Fe(TFe) grade of the concentrate. In this work, the effect of sodium alginate on the reverse flotation of hematite was systematically investigated. Flotation tests of artificially mixed ores were conducted, and the results showed that sodium alginate can significantly improve the removal rates of quartz and chlorite. The adsorption measurements, infrared spectroscopy, and contact angle tests demonstrated that sodium alginate adsorbs on the quartz surface by chelating with calcium ions, thereby weakening the steric hindrance of oleate ions and increasing the adsorption capacity of sodium oleate to ultimately improve the removal rate of quartz. Furthermore, owing to its lower density and fine particle size, chlorite is easily entrained into the foam layer. Sodium alginate dramatically increases the liquid-to-gas ratio of the foam layer by increasing pulp viscosity, thereby increasing the entrainment rate of chlorite and finally improving its removal rate. The core content of this thesis bears significance in improving the Fe grade in the reverse flotation of chlorite-containing hematite.

文摘Gelatin/Alginate hydrogels were engineered for bioplotting in tissue engineering.One major drawback of hydrogel scaffolds is the lack of adequate mechanical properties.In this study,using a bioplotter,we constructed the scaffolds with different pore architectures by deposition of gelatin/alginate hydrogels layerby-layer.The scaffolds with different crosslinking degree were obtained by post-crosslinking methods.Their physicochemical properties,as well as cell viability,were assessed.Different crosslinking methods had little influence on scaffold architecture,porosity,pore size and distribution.By contrast,the water absorption ability,degradation rate and mechanical properties of the scaffolds were dramatically affected by treatment with various concentrations of crosslinking agent(glutaraldehyde).The crosslinking process using glutaraldehyde markedly improved the stability and mechanical strength of the hydrogel scaf-folds.Besides the post-processing methods,the pore architecture can also evidently affect the mechanical properties of the scaffolds.The crosslinked gelatin/alginate scaffolds showed a good potential to encap-sulate cells or drugs.

文摘Kaolin has been widely used as an adsorbent to remove heavy metal ions from aqueous solutions. However, the lower heavy metal adsorption capacity of kaolin limits its practical application. A novel environmental friendly material, calcium alginate immobilized kaolin (kaolin/CA), was prepared using a sol-gel method. The effects of contact time, pH, adsorbent dose, and temperature on Cu2+ adsorption by kaolin/CA were investigated. The Langmuir isotherm was used to describe the experimental adsorption, the maximum Cu2+ adsorption capacity of the kaolin/CA reached up to 53.63 mg/g. The thermodynamic studies showed that the adsorption reaction was a spontaneous and endothermic process.

文摘Alginate oligosaccharides(AOS), belonging to the class of functional marine oligosaccharides, are low-molecular polymers linked by β-1,4-mannuronic acid(M) and α-1,4-guluronic acid(G), which could be classically obtained by enzymatic hydrolysis of alginate. With low viscosity and good water solubility, as well as anti-oxidant, immune regulation, anti-bacterial and antiinflammatory activities, AOS have been widely used in medical science and functional food, green agriculture and other fields. As new bio-feed additives, AOS have broad potential applications in animal husbandry. In this review, the sources of alginate, chemical structure and preparation methods of AOS, and their biological activities and application in livestock and poultry are summarized. We expect this review could contribute to lay a foundation of application and further research for AOS in livestock and poultry.

文摘Background: Alginate oligosaccharide(AOS), produced from alginate by alginate lyase-mediated depolymerisation, is a potential substitute for antibiotics and possesses growth-enhancing effects. Nevertheless, the mechanisms by which AOS regulates porcine growth remain to be elucidated. Therefore, we investigated the AOS-mediated changes in the growth performance of weaned pigs by determining the intestinal morphology, barrier function,as well as epithelium apoptosis.Methods: Twenty-four weaned pigs were distributed into two groups(n = 12) and received either a basal diet(control group) or the same diet supplemented with 100 mg/kg AOS. On d 15, D-xylose(0.1 g/kg body weight)was orally administrated to eight randomly selected pigs per treatment, and their serum and intestinal mucosa samples were collected 1 h later.Results: Our results showed that inclusion of AOS in the diet for 2 wk increased(P < 0.05) the average daily body weight gain in weaned pigs. Notably, AOS supplementation ameliorated the intestinal morphology and barrier function, as suggested by the enhanced(P < 0.05) intestinal villus height, secretory immunoglobulin A content and goblet cell counts. Compared to the control group, AOS ingestion both decreased(P < 0.05) the total apoptotic percentage and increased(P < 0.05) the proportion of S phase in the intestinal epithelial cells. Furthermore, AOS not only up-regulated(P < 0.05) the B-cell lymphoma-2(BCL2) transcriptional level but also down-regulated(P < 0.05) the B-cell lymphoma-2-associated X protein(BAX), cysteinyl aspartate-specific proteinase-3(caspase-3) and caspase-9 transcriptional levels in the small intestine.Conclusions: In summary, this study provides evidence that supplemental AOS beneficially affects the growth performance of weaned pigs, which may result from the improved intestinal morphology and barrier function,as well as the inhibited enterocyte death, through reducing apoptosis via mitochondria-dependent apoptosis.
