OBJECTIVE:To determine direct targeting of localized adiposity through Morus alba Linne bark injection based on pharmacology network analysis.METHODS:Male C57BL/6J mice were fed a high-fat diet(HFD)to induce obesity.A...OBJECTIVE:To determine direct targeting of localized adiposity through Morus alba Linne bark injection based on pharmacology network analysis.METHODS:Male C57BL/6J mice were fed a high-fat diet(HFD)to induce obesity.After 6 weeks on HFD,the water extract of Morus alba L.bark(MAB,2 mg/mL)was locally injected into one inguinal fat pad,while saline was injected into the other side,3 times/week for 6 weeks(n=6/group).The water extract of MAB was freeze-dried and then diluted in saline before use.RESULTS:HFD-fed mice treated with local MAB topical injection showed reduced adipocyte weight and size in inguinal fat pads by dual-energy X-ray absorptiometry.No toxicity changes seen in liver,spleen,kidney tissue,or alanine aminotransferase/aspartate aminotransferase levels in serum by MAB injection.Protein levels of phosphorylated insulin receptor substrate-1 and glucose transporter type 4,and mRNA expression of adiponectin,were increased in inguinal adipose tissue injected with MAB locally.Locally MAB injection led to a decrease in glucose-6-phosphatase and phosphoenolpyruvate carboxykinase,linked to gluconeogenesis,while forkhead box protein O1,which regulates these factors,was increased.Moreover,there was an increase in adenosine 5‘-monophosphate-activated protein kinase,related to lipogenesis,as well as elevated levels of hormone-sensitive lipase and fatty acid synthase,both associated with lipolysis.These results support the'insulin signaling pathway'and'regulation of lipolysis in adipocytes'identified in the Kyoto Encyclopedia of Genes and Genomes pathway through network analysis.CONCLUSION:This study suggests that MAB topical injection exhibits localized fat reduction by inhibiting insulin resistance,gluconeogenesis and lipogenesis mediator,while activating lipolysis enzymes within targeted adipose site.展开更多
OBJECTIVE:To elucidate the potential molecular mechanisms of Baishao(Radix Paeoniae Alba)(APR)and Gancao(Radix Glycyrrhizae)(GR)in the treatment of major depressive disorder(MDD).METHODS:Based on the network pharmacol...OBJECTIVE:To elucidate the potential molecular mechanisms of Baishao(Radix Paeoniae Alba)(APR)and Gancao(Radix Glycyrrhizae)(GR)in the treatment of major depressive disorder(MDD).METHODS:Based on the network pharmacology strategy,the therapeutic targets of APR-GR for MDD are predicted,differentially expressed genes from the Integrated Gene Expression database for MDD patients.Topological networks are constructed,Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathways are enriched,their pharmacological potential molecular mechanisms are discussed,and molecular docking analysis is performed to further motivate compositional and target interactions.Finally,the CUMS mouse model is used for validation.RESULTS:Based on the pharmacological network analysis,17 candidate genes were identified,including muscarinic acetylcholine receptor M1(CHRM1),muscarinic acetylcholine receptor M2(CHRM2),β2-adrenergic receptor(ADRB2),adrenergicα1A receptor(ADRA1A)and 5-hydroxytryptamine transfer protein(SLC6A4),etc.which are primarily involved in reactive oxygen species metabolism,neural response,oxidative stress response and other biological processes.Further analysis revealed that these targets are closely related to Ca^(2+),cyclic adenosine monophosphate,etc.,and exhibit optimal binding sites after molecular docking.Finally,in vivo experiments were performed and it was found that APR-GR significantly improved depression-like behavior and hippocampal impairment in mouse models,increasing brain levels of 5-hydroxytryptamine,dopamine and norepinephrine and decreasing serum levels of corticotropin releasing hormone,corticosterone and adreno cortico tropic hormone,while upregulating the expression of CHRM1,CHRM2 and ADRA1A in the hippocampus and downregulating the expression of SLC6A4 and ADRB2.CNCLUSION:This research sheds light on the potential molecular mechanism of APR-GR to improve MDD.展开更多

In order to evaluate the crude and pr...The crude and processed Paeoniae Radix Alba-Atractylodis Macrocephalae Rhizoma herbal pairs, originated from Bai-zhu-shao-yao-san, are used to treat different diseases clinically. In order to evaluate the crude and processed Paeoniae Radix-Atractylodis Macrocephalae Rhizoma herbal pairs, a simple, easy, and sensitive high-performance liquid chromatography coupled with diode array detectors was developed for simultaneous determination of nine bioactive components in the herbal pairs. The calibration curve exhibited good linearity(r2≥0.9992). The LODs and LOQs were ≤7.30 and 11.53 μg/m L, respectively. The intra-, inter-day and repeatability RSD values of the nine compounds were less than 3.86%, 2.71%, and 4.29%, respectively. The RSD stability values were less than 3.64%. The recovery of the method was in the range of 96.70%–102.10%, with RSD values less than 3.52%. The developed method can be applied to the intrinsic quality control of crude and processed Paeoniae Radix-Atractylodis Macrocephalae Rhizoma herbal pairs.展开更多
[Objective]The aim was to explore the active compound of Morus alba leaves with the analysis of active components of hexane extractives of M.alba leaves.[Method]Antibacterial assays of hexane extractives of M.alba lea...[Objective]The aim was to explore the active compound of Morus alba leaves with the analysis of active components of hexane extractives of M.alba leaves.[Method]Antibacterial assays of hexane extractives of M.alba leaves were done and the volatile components of hexane extractives of M.alba leaves with the strongest antibacterial activity were analyzed by gas chromatography-mass spectrometry(GC-MS).[Result]The results of inhibitory effect of hexane extractives of M.alba leaves on Staphylococcus aureus and Escherichia coli showed that hexane extractives of M.alba leaves collected in June,July and August had antibacterial activity against S.aureus and had no antibacterial effect against E.coli.Furthermore,hexane extractives of M.alba leaves collected in June had the strongest antibacterial activity against S.aureus(inhibition diameter 10.95 mm).The hexane extractives of M.alba leaves collected in June was analyzed by GC-MS,the main volatile components were tetradecane(16.76%),dodecane(13.20%),diisobutyl phthalate(10.26%),decane(9.10%),hexadecane(8.71%),linolenyl alcohol(7.25%),octadecane(5.88%),eicosane(3.26%),dibutyl phthalate(2.59%).[Conclusion]Linolenyl alcohol was the potential antibacterial compound.展开更多

[Result] Alcohols [phytol (59.47%,extracted 3 h for M. alba leaves),α-cadinol (38.77%,extracted 6 h for C. pisifera outer heartwood)] were their first principal volatile components. Phytol was usually used as pharmaceutical raw material and α-cadinol was a kind of active organic compound with antibacterial function. The present review is aimed at providing a comprehensive summary on the botany,utility,phytochemistry,pharmacology,and clinical trials of Morus alba(mulberry or sang shu).The mulberry foliage has remained the primary ...The present review is aimed at providing a comprehensive summary on the botany,utility,phytochemistry,pharmacology,and clinical trials of Morus alba(mulberry or sang shu).The mulberry foliage has remained the primary food for silkworms for centuries.Its leaves have also been used as animal feed for livestock and its fruits have been made into a variety of food products.With flavonoids as major constituents,mulberry leaves possess various biological activities,including antioxidant,antimicrobial,skin-whitening,cytotoxic,anti-diabetic,glucosidase inhibition,anti-hyperlipidemic,anti-atherosclerotic,anti-obesity,cardioprotective,and cognitive enhancement activities.Rich in anthocyanins and alkaloids,mulberry fruits have pharmacological properties,such as antioxidant,anti-diabetic,anti-atherosclerotic,anti-obesity,and hepatoprotective activities.The root bark of mulberry,containing flavonoids,alkaloids and stilbenoids,has antimicrobial,skin-whitening,cytotoxic,anti-inflammatory,and anti-hyperlipidemic properties.Other pharmacological properties of M.alba include anti-platelet,anxiolytic,anti-asthmatic,anthelmintic,antidepressant,cardioprotective,and immunomodulatory activities.Clinical trials on the efficiency of M.alba extracts in reducing blood glucose and cholesterol levels and enhancing cognitive ability have been conducted.The phytochemistry and pharmacology of the different parts of the mulberry tree confer its traditional and current uses as fodder,food,cosmetics,and medicine.Overall,M.alba is a multi-functional plant with promising medicinal properties.展开更多

Objective:The present study deals with the investigation of larvicidal and ovicidal activities of benzene,hexane,ethyl acetate,methanol and chloroform leaf extract of Eclipta alba(E.alba) against dengue vector,Aedes...Objective:The present study deals with the investigation of larvicidal and ovicidal activities of benzene,hexane,ethyl acetate,methanol and chloroform leaf extract of Eclipta alba(E.alba) against dengue vector,Aedes aegypti(Ae.Aegypti).Methods:Twenty five earlyⅢinstar larvae of Ae.aegypti was exposed to various concentrations(50-300 ppm) and was assayed in the laboratory by using the protocol of WHO 2005;the 24 h LC<sub>50</sub> values of the E.alba leaf extract was determined by Probit analysis.For ovicidal activity,slightly modified method of Su and Mulla was performed.The ovicidal activity was determined against Ae.aegypti to various concentrations ranging from 100-350 ppm under the laboratory conditions.The egg hatch rates were assessed 48 h post treatment.Results:The LC<sub>50</sub> values of benzene,hexane,ethyl acetate,methanol and chloroform extract of E.alba against early third instar larvae of Ae.aegypti were 151.38,165.10, 154.88,127.64 and 146.28 ppm,respectively.Maximum larvicidal activity was observed in the methanol extract followed by chloroform,benzene,ethyl acetate and hexane extract.No mortality was observed in control.Among five solvent tested the methanol extract was found to be most effective for ovicidal activity against Ae.aegypti.The methanol extracts exerted 100%mortality (zero hatchability) at 300 ppm.Conclusions:From the results it can be concluded the crude extract of E.alba was an excellent potential for controlling Ae.aegypti mosquito.展开更多

Correspondence analysis (a commonly used multivariate statistical approach) and method of effective number of codons (ENc)-plot were conducted to analyze synonymous codon usage. The results of correspondence analysis showed that the distribution of genes on the major axis was significantly correlated with the frequency of use of G+C in synonymously variable third position of sense codon (GC3S), (r=0.349), and the positions of genes on the axis 2 and axis 3 were significantly correlated with CAI (r=-0.348, p〈0.01 and r=0.602, p〈0.01). The ENc for most genes was similar to that for the expected ENc based on the GC3s, but several genes with low ENc values were lying below the expected curve. All of these data indicated that codon usage was dominated by a mutational bias in chloroplast gcnome ofP. alba. The selection in nature for translational efficiency only played a minor role in shaping codon usage in the chloroplast genome ofP alba.展开更多

In this study, radial variation of sap flow velocity(Js) was investigated at five depths of the xylem(1, 2, 3, 5 and 8 cm under the cambium) in three mature Xinjiang poplar(Populus alba L. var. pyramidalis) trees grown at the Gansu Minqin National Studies Station for Desert Steppe Ecosystem from May to October 2011. Thermal dissipation probes of various lengths manufactured according to the Granier's design were installed into each tree for simultaneous observation of the radial patterns of Js through the xylem. The radial patterns were found to fit the four-parameter GaussAmp equation. The peak Js was about 27.02±0.95 kg/(dm2?d) at approximately 3 to 5 cm deep from the cambium of the three trees,and the lowest Js appeared at 1 cm deep in most of the time. Approximately 50% of the total sap flow in Xinjiang poplar occurred within one-third of the xylem from its outer radius, whereas 90% of the total sap flow occurred within two-fifth of the xylem. In addition, the innermost point of the xylem(at 8-cm depth), which appeared as the penultimate sap flow in most cases during the study period, was hydro-active with Js,8 of 7.55±3.83 kg/(dm2?d). The radial pattern of Js was found to be steeper in midday than in other time of the day, and steeper diurnal fluctuations were recorded in June, July and August(the mid-growing season). Maximum differences between the lowest Js(Js,1 or Js,8) and the highest Js(Js,3 or Js,5) from May through October were 12.41, 17.35, 16.30, 18.52, 12.60 and 16.04 g/(cm2?h), respectively. The time-dependent changes of Js along the radial profile(except at 1-cm depth) were strongly related to the reference evapotranspiration(ET0). Due to significant radial variability of Js, the mean daily sap flow at the whole-tree level could be over-estimated by up to 29.69% when only a single probe at depth of 2 cm was used. However, the accuracy of the estimation of sap flow in Xinjiang poplar could be significantly improved using a correction coefficient of 0.885.展开更多
Ultra-performance liquid chromatography coupled with quadrupole time-of-flight tandem mass spectrometry (UPLC-Q-TOF/MS) was developed to identify the absorbed parent components and metabolites in rat bile, plasma an...Ultra-performance liquid chromatography coupled with quadrupole time-of-flight tandem mass spectrometry (UPLC-Q-TOF/MS) was developed to identify the absorbed parent components and metabolites in rat bile, plasma and urine after oral administration of Radix Paeoniae Alba extract (RPAE). A total of 65 compounds were detected in rat bile, plasma and urine samples, including 11 parent compounds and 54 metabolites. The results indicated that glucuronidation, hydroxylation and methylation were the major metabolic pathways of the components of RPAE. Furthermore, the results of this work demonstrated that UPLC-Q-TOF/MS combined with MetaboLynx? software and mass defect filtering (MDF) could provide unique high throughput capabilities for drug metabolism study, with excellent MS mass accuracy and enhanced MSE data acquisition. With the MSE technique, both precursor and fragment mass spectra can be simultaneously acquired by alternating between high and low collision energy during a single chromatographic run.展开更多
Radix Bupleuri(RB)is commonly used to treat depression,but it can also lead to hepatotoxicity after longterm use.In many anti-depression prescriptions,RB is often used in combination with Radix Paeoniae Alba(RPA)as an...Radix Bupleuri(RB)is commonly used to treat depression,but it can also lead to hepatotoxicity after longterm use.In many anti-depression prescriptions,RB is often used in combination with Radix Paeoniae Alba(RPA)as an herb pair.However,whether RPA can alleviate RB-induced hepatotoxicity remain unclear.In this work,the results confirmed that RB had a dose-dependent antidepressant effect,but the optimal antidepressant dose caused hepatotoxicity.Notably,RPA effectively reversed RB-induced hepatotoxicity.Afterward,the mechanism of RB-induced hepatotoxicity was confirmed.The results showed that saikosaponin A and saikosaponin D could inhibit GSH synthase(GSS)activity in the liver,and further cause liver injury through oxidative stress and nuclear factor kappa B(NF-kB)/NOD-like receptor thermal protein domain associated protein 3(NLRP3)pathway.Furthermore,the mechanisms by which RPA attenuates RBinduced hepatotoxicity were investigated.The results demonstrated that RPA increased the abundance of intestinal bacteria with glycosidase activity,thereby promoting the conversion of saikosaponins to saikogenins in vivo.Different from saikosaponin A and saikosaponin D,which are directly combined with GSS as an inhibitor,their deglycosylation conversion products saikogenin F and saikogenin G exhibited no GSS binding activity.Based on this,RPA can alleviate the inhibitory effect of saikosaponins on GSS activity to reshape the liver redox balance and further reverse the RB-induced liver inflammatory response by the NFkB/NLRP3 pathway.In conclusion,the present study suggests that promoting the conversion of saikosaponins by modulating gut microbiota to attenuate the inhibition of GSS is the potential mechanism by which RPA prevents RB-induced hepatotoxicity.展开更多

文摘The crude and processed Paeoniae Radix Alba-Atractylodis Macrocephalae Rhizoma herbal pairs, originated from Bai-zhu-shao-yao-san, are used to treat different diseases clinically. In order to evaluate the crude and processed Paeoniae Radix-Atractylodis Macrocephalae Rhizoma herbal pairs, a simple, easy, and sensitive high-performance liquid chromatography coupled with diode array detectors was developed for simultaneous determination of nine bioactive components in the herbal pairs. The calibration curve exhibited good linearity(r2≥0.9992). The LODs and LOQs were ≤7.30 and 11.53 μg/m L, respectively. The intra-, inter-day and repeatability RSD values of the nine compounds were less than 3.86%, 2.71%, and 4.29%, respectively. The RSD stability values were less than 3.64%. The recovery of the method was in the range of 96.70%–102.10%, with RSD values less than 3.52%. The developed method can be applied to the intrinsic quality control of crude and processed Paeoniae Radix-Atractylodis Macrocephalae Rhizoma herbal pairs.

文摘[Objective]The aim was to explore the active compound of Morus alba leaves with the analysis of active components of hexane extractives of M.alba leaves.[Method]Antibacterial assays of hexane extractives of M.alba leaves were done and the volatile components of hexane extractives of M.alba leaves with the strongest antibacterial activity were analyzed by gas chromatography-mass spectrometry(GC-MS).[Result]The results of inhibitory effect of hexane extractives of M.alba leaves on Staphylococcus aureus and Escherichia coli showed that hexane extractives of M.alba leaves collected in June,July and August had antibacterial activity against S.aureus and had no antibacterial effect against E.coli.Furthermore,hexane extractives of M.alba leaves collected in June had the strongest antibacterial activity against S.aureus(inhibition diameter 10.95 mm).The hexane extractives of M.alba leaves collected in June was analyzed by GC-MS,the main volatile components were tetradecane(16.76%),dodecane(13.20%),diisobutyl phthalate(10.26%),decane(9.10%),hexadecane(8.71%),linolenyl alcohol(7.25%),octadecane(5.88%),eicosane(3.26%),dibutyl phthalate(2.59%).[Conclusion]Linolenyl alcohol was the potential antibacterial compound.

文摘[Objective] The aim was to explore the composition of essential oils of Morus alba leaves and Chamaecyparis pisifera outer heartwood. [Method] The essential oils of M. alba leaves and C. pisifera outer heartwood were analyzed by gas chromatography-mass spectrometry,respectively. [Result] Alcohols [phytol (59.47%,extracted 3 h for M. alba leaves),α-cadinol (38.77%,extracted 6 h for C. pisifera outer heartwood)] were their first principal volatile components. Phytol was usually used as pharmaceutical raw material and α-cadinol was a kind of active organic compound with antibacterial function. [Conclusion] The essential oils of M. alba leaves and C. pisifera outer heartwood with hydrodistillation have medicinal value.

Hassk.Their structures were deduced as 3β,16a-dihydroxy olean-12-ene-28-oic acid-3β-O-β-D-glu-copyranoside(3)and 3β-O-[β-D-glucopyranosyl(1-4)]-β-D-glucopyranosyl-16a-hydroxy olean-12-ene-28-oic acid 28-O-β-D-glucopyranoside(4),based on spectral analysis and chemical evidences as well as results ofhydrolysis.
文摘Two new triterpenoid saponins, monepaloside K (1) and monepaloside L (2), together with a known saponin, mazusaponin I (3), were isolated from the water-soluble part of the whole plant of Marina nepalensis var. alba Hand.-Mazz. The structures of monepalosides K and L were determined to be 3-O-α-L-arabinopyranosyl-(1-3)-α-D-xylopyranosyl siaresinolic acid (1) and 3-O-β-D-glucopyranosyl-( 1-3)-β-L-arabinopyranosy1 siaresinolic acid 28-O-β-D-glucopyranosyl-(1-6)-β-D-glucopyranoside (2) respectively, on the basis of chemical and spectroscopic evidence.
文摘The present review is aimed at providing a comprehensive summary on the botany,utility,phytochemistry,pharmacology,and clinical trials of Morus alba(mulberry or sang shu).The mulberry foliage has remained the primary food for silkworms for centuries.Its leaves have also been used as animal feed for livestock and its fruits have been made into a variety of food products.With flavonoids as major constituents,mulberry leaves possess various biological activities,including antioxidant,antimicrobial,skin-whitening,cytotoxic,anti-diabetic,glucosidase inhibition,anti-hyperlipidemic,anti-atherosclerotic,anti-obesity,cardioprotective,and cognitive enhancement activities.Rich in anthocyanins and alkaloids,mulberry fruits have pharmacological properties,such as antioxidant,anti-diabetic,anti-atherosclerotic,anti-obesity,and hepatoprotective activities.The root bark of mulberry,containing flavonoids,alkaloids and stilbenoids,has antimicrobial,skin-whitening,cytotoxic,anti-inflammatory,and anti-hyperlipidemic properties.Other pharmacological properties of M.alba include anti-platelet,anxiolytic,anti-asthmatic,anthelmintic,antidepressant,cardioprotective,and immunomodulatory activities.Clinical trials on the efficiency of M.alba extracts in reducing blood glucose and cholesterol levels and enhancing cognitive ability have been conducted.The phytochemistry and pharmacology of the different parts of the mulberry tree confer its traditional and current uses as fodder,food,cosmetics,and medicine.Overall,M.alba is a multi-functional plant with promising medicinal properties.

文摘Objective:The present study deals with the investigation of larvicidal and ovicidal activities of benzene,hexane,ethyl acetate,methanol and chloroform leaf extract of Eclipta alba(E.alba) against dengue vector,Aedes aegypti(Ae.Aegypti).Methods:Twenty five earlyⅢinstar larvae of Ae.aegypti was exposed to various concentrations(50-300 ppm) and was assayed in the laboratory by using the protocol of WHO 2005;the 24 h LC<sub>50</sub> values of the E.alba leaf extract was determined by Probit analysis.For ovicidal activity,slightly modified method of Su and Mulla was performed.The ovicidal activity was determined against Ae.aegypti to various concentrations ranging from 100-350 ppm under the laboratory conditions.The egg hatch rates were assessed 48 h post treatment.Results:The LC<sub>50</sub> values of benzene,hexane,ethyl acetate,methanol and chloroform extract of E.alba against early third instar larvae of Ae.aegypti were 151.38,165.10, 154.88,127.64 and 146.28 ppm,respectively.Maximum larvicidal activity was observed in the methanol extract followed by chloroform,benzene,ethyl acetate and hexane extract.No mortality was observed in control.Among five solvent tested the methanol extract was found to be most effective for ovicidal activity against Ae.aegypti.The methanol extracts exerted 100%mortality (zero hatchability) at 300 ppm.Conclusions:From the results it can be concluded the crude extract of E.alba was an excellent potential for controlling Ae.aegypti mosquito.

文摘The pattern of codon usage in the chloroplast genome of Populus alba was investigated. Correspondence analysis (a commonly used multivariate statistical approach) and method of effective number of codons (ENc)-plot were conducted to analyze synonymous codon usage. The results of correspondence analysis showed that the distribution of genes on the major axis was significantly correlated with the frequency of use of G+C in synonymously variable third position of sense codon (GC3S), (r=0.349), and the positions of genes on the axis 2 and axis 3 were significantly correlated with CAI (r=-0.348, p〈0.01 and r=0.602, p〈0.01). The ENc for most genes was similar to that for the expected ENc based on the GC3s, but several genes with low ENc values were lying below the expected curve. All of these data indicated that codon usage was dominated by a mutational bias in chloroplast gcnome ofP. alba. The selection in nature for translational efficiency only played a minor role in shaping codon usage in the chloroplast genome ofP alba.

文摘Estimation of the transpiration rate for a tree is generally based on sap flow measurements within the hydro-active stem xylem. In this study, radial variation of sap flow velocity(Js) was investigated at five depths of the xylem(1, 2, 3, 5 and 8 cm under the cambium) in three mature Xinjiang poplar(Populus alba L. var. pyramidalis) trees grown at the Gansu Minqin National Studies Station for Desert Steppe Ecosystem from May to October 2011. Thermal dissipation probes of various lengths manufactured according to the Granier's design were installed into each tree for simultaneous observation of the radial patterns of Js through the xylem. The radial patterns were found to fit the four-parameter GaussAmp equation. The peak Js was about 27.02±0.95 kg/(dm2?d) at approximately 3 to 5 cm deep from the cambium of the three trees,and the lowest Js appeared at 1 cm deep in most of the time. Approximately 50% of the total sap flow in Xinjiang poplar occurred within one-third of the xylem from its outer radius, whereas 90% of the total sap flow occurred within two-fifth of the xylem. In addition, the innermost point of the xylem(at 8-cm depth), which appeared as the penultimate sap flow in most cases during the study period, was hydro-active with Js,8 of 7.55±3.83 kg/(dm2?d). The radial pattern of Js was found to be steeper in midday than in other time of the day, and steeper diurnal fluctuations were recorded in June, July and August(the mid-growing season). Maximum differences between the lowest Js(Js,1 or Js,8) and the highest Js(Js,3 or Js,5) from May through October were 12.41, 17.35, 16.30, 18.52, 12.60 and 16.04 g/(cm2?h), respectively. The time-dependent changes of Js along the radial profile(except at 1-cm depth) were strongly related to the reference evapotranspiration(ET0). Due to significant radial variability of Js, the mean daily sap flow at the whole-tree level could be over-estimated by up to 29.69% when only a single probe at depth of 2 cm was used. However, the accuracy of the estimation of sap flow in Xinjiang poplar could be significantly improved using a correction coefficient of 0.885.
文摘Ultra-performance liquid chromatography coupled with quadrupole time-of-flight tandem mass spectrometry (UPLC-Q-TOF/MS) was developed to identify the absorbed parent components and metabolites in rat bile, plasma and urine after oral administration of Radix Paeoniae Alba extract (RPAE). A total of 65 compounds were detected in rat bile, plasma and urine samples, including 11 parent compounds and 54 metabolites. The results indicated that glucuronidation, hydroxylation and methylation were the major metabolic pathways of the components of RPAE. Furthermore, the results of this work demonstrated that UPLC-Q-TOF/MS combined with MetaboLynx? software and mass defect filtering (MDF) could provide unique high throughput capabilities for drug metabolism study, with excellent MS mass accuracy and enhanced MSE data acquisition. With the MSE technique, both precursor and fragment mass spectra can be simultaneously acquired by alternating between high and low collision energy during a single chromatographic run.

文摘Radix Bupleuri(RB)is commonly used to treat depression,but it can also lead to hepatotoxicity after longterm use.In many anti-depression prescriptions,RB is often used in combination with Radix Paeoniae Alba(RPA)as an herb pair.However,whether RPA can alleviate RB-induced hepatotoxicity remain unclear.In this work,the results confirmed that RB had a dose-dependent antidepressant effect,but the optimal antidepressant dose caused hepatotoxicity.Notably,RPA effectively reversed RB-induced hepatotoxicity.Afterward,the mechanism of RB-induced hepatotoxicity was confirmed.The results showed that saikosaponin A and saikosaponin D could inhibit GSH synthase(GSS)activity in the liver,and further cause liver injury through oxidative stress and nuclear factor kappa B(NF-kB)/NOD-like receptor thermal protein domain associated protein 3(NLRP3)pathway.Furthermore,the mechanisms by which RPA attenuates RBinduced hepatotoxicity were investigated.The results demonstrated that RPA increased the abundance of intestinal bacteria with glycosidase activity,thereby promoting the conversion of saikosaponins to saikogenins in vivo.Different from saikosaponin A and saikosaponin D,which are directly combined with GSS as an inhibitor,their deglycosylation conversion products saikogenin F and saikogenin G exhibited no GSS binding activity.Based on this,RPA can alleviate the inhibitory effect of saikosaponins on GSS activity to reshape the liver redox balance and further reverse the RB-induced liver inflammatory response by the NFkB/NLRP3 pathway.In conclusion,the present study suggests that promoting the conversion of saikosaponins by modulating gut microbiota to attenuate the inhibition of GSS is the potential mechanism by which RPA prevents RB-induced hepatotoxicity.
