目的观察腺苷A1型受体(A1AR)基因敲除小鼠的病理生理特点及A1AR在慢性低盐刺激肾素分泌中的作用。方法 C57BL/6J品系A1AR基因敲除杂合子(A1AR+/-)小鼠,经繁殖及基因鉴定后,杂合子子代小鼠用于扩大繁殖,A1AR+/+小鼠(n=13)及A1AR-/-小鼠(n... 目的观察腺苷A1型受体(A1AR)基因敲除小鼠的病理生理特点及A1AR在慢性低盐刺激肾素分泌中的作用。方法 C57BL/6J品系A1AR基因敲除杂合子(A1AR+/-)小鼠,经繁殖及基因鉴定后,杂合子子代小鼠用于扩大繁殖,A1AR+/+小鼠(n=13)及A1AR-/-小鼠(n=14)用于实验。观察两组小鼠生长发育、基本生理情况(血压、心率、肾功能和血尿电解质)、肾脏病理和免疫组化观察肾脏局部肾素表达和检测血浆醛固酮水平(A1AR+/+组n=5,A1AR-/-组n=5)。结果低盐饮食后,A1AR-/-小鼠肾皮质肾素表达明显增加(P<0.05),全身醛固酮水平和24 h尿钾排泄量显著高于A1AR+/+小鼠(P<0.05)。结论 A1AR可能参与了低盐刺激小鼠肾素分泌的过程。 展开更多

目的:探讨腺苷A1受体(A1AR)基因rs903361、rs10920573及rs3766553多态性与浙南汉族人群部分性发作癫痫耐药的相关性。方法:选取120例部分性发作癫痫患者(癫痫耐药组和非耐药组各60例)及正常对照组(正常组)60例。采用聚合酶链反应-限制... 目的:探讨腺苷A1受体(A1AR)基因rs903361、rs10920573及rs3766553多态性与浙南汉族人群部分性发作癫痫耐药的相关性。方法:选取120例部分性发作癫痫患者(癫痫耐药组和非耐药组各60例)及正常对照组(正常组)60例。采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)法检测A1AR基因rs903361、rs10920573和rs3766553的多态性,并通过病例对照研究及logistic回归分析方法分析其与癫痫耐药的相关性。结果:所有入组人群三个位点等位基因C和T分布,正常组与非耐药组三个位点各基因型分布差异均无统计学意义;耐药组rs903361 CT型分布频率低于非耐药组(P=0.017),而rs10920573 CT型分布频率高于非耐药组(P=0.010),差异均具有统计学意义;rs3766553基因型分布各组差异均无统计学意义;rs903361 CC型、TT型耐药的危险性分别为CT型的2.730倍和2.609倍,P值分别为0.043和0.047;rs10920573的CT型耐药危险性为TT型的2.572倍(P=0.036);rs903361 CC型、rs903361 TT型、rs10920573 CT型均为耐药的独立危险因素。结论:A1AR基因的rs903361 CC型、TT型和rs10920573 CT型为浙南汉族人群部分性发作癫痫耐药产生的独立危险因素。 展开更多

A three-dimensional quantitative structure-activity relationship(3 D-QSAR) study was conducted to analyze the A1 AR density(Bmax) of 56 3-aroyl-5-substituted thiophene derivatives(ASTDs) in human A1 Chinese hamster ovary(hA1 CHO) membranes by the comparative molecular field analysis(CoMFA) method. A training set of 45 compounds was used to establish the predictive model, which was verified by the test set of 17 compounds containing template molecule and 5 newly designed molecules. The cross-validation(R2 cv) and non-cross-validation(R2) coefficients of the training set were 0.655 and 0.959, respectively. The model was used to predict the activities of the compounds of the training and test sets, and the results indicated that the models had strong stability and good prediction ability. According to model analysis, the contribution of steric and electrostatic fields was 51.4% and 48.6%, respectively. Based on the 3 D contour maps, five excellent ASTDs agonists were designed, which need to be further verified by biomedical experiments. 展开更多
