OBJECTIVE:To elucidate the regulatory effects of salvianolic acid B(Sal B)on trophoblast cells in preeclampsia(PE).METHODS:3-(4,5-dimethylthiazol-2-yl)-2,5 diphenyltetrazolium bromide(MTT)assays were used to detect th...OBJECTIVE:To elucidate the regulatory effects of salvianolic acid B(Sal B)on trophoblast cells in preeclampsia(PE).METHODS:3-(4,5-dimethylthiazol-2-yl)-2,5 diphenyltetrazolium bromide(MTT)assays were used to detect the viability of human extravillous trophoblast HTR-8/Svneo cells induced by H_(2)O_(2)following treatment with different concentrations of Sal B.The levels of oxidative stressrelated molecules,including superoxide dismutase,glutathione-Px and malondialdehyde were detected using corresponding kits.Cell apoptosis was detected using a Terminal deoxynucleotidyl transferase(Td T)d UTP NickEnd Labeling(TUNEL)assay,and the expression of apoptosis-related proteins was detected using western blot analysis.In the present study,wound healing and Transwell assays were performed to measure the levels of cell invasion and migration.Western blot analysis was also used to detect the expression levels of epithelialmesenchymal transition-related proteins.The mechanisms underlying Sal B were further investigated using reverse transcription-quantitative real-time polymerase chain reaction(RT-q PCR)and western blot analysis,to determine the expression levels of matrix metallopeptidase 9(MMP-9)and phosphatidylinositol-4,5-bisphosphate 3-kinase(PI3K)/protein kinase B(Akt).RESULTS:Sal B increased the activity of HTR-8/Svneo cells,inhibited oxidative damage and promoted the invasion and migration of trophoblast cells induced by H_(2)O_(2).Furthermore,the expression levels of MMP-9 and members of the PI3K/Akt signaling pathway were significantly decreased.The pathway agonist,LY294002,and MMP-9 inhibitor,GM6001,reversed the effects of Sal B on H_(2)O_(2)-induced cells.CONCLUSIONS:Sal B promoted the invasion and migration of H_(2)O_(2)-induced HTR-8/Svneo trophoblast cells by upregulating MMP-9 via the PI3K/Akt signaling pathway.展开更多

Methods ELISA method was adopted to detect serum MMP-8 co...Objective To study the clinical role of the variation of serum matrix metalloproteinase-8 (MMP-8) concentration in patients with acute coronary syndrome (ACS). Methods ELISA method was adopted to detect serum MMP-8 concentration and to observe concentration’s differences and features among 80 selected ACS cases (43 acute myocardial infarction and 37 unstable angina pectoris), 43 stable angina pectoris (SAP) cases and 37 control cases. And meanwhile the atherosclerosis risk factors of each case, such as age, sex, hypertension, body mass index, smoking, family history, diabetes, and hyperlipidemia were collected and analyzed as a whole. Results First, serum MMP-8 concentration reached the highest point in ACS, and there was significant difference between SAP and control groups (P<0.01). Second, serum MMP-8 in AMI was much higher than that in UAP with significant difference (P<0.01). There was no difference between UAP and SAP groups (P>0.05). Third, Logistic regression analysis revealed that serum MMP-8 concentration might be the indicator of ACS (B=4.493, P=0.000), particularly, that of AMI (B=9.961, P=0.000). Fourth, linear correlation and linear regression analysis found that only neutrophil was likely to influence serum MMP-8 concentration (r=0.274, P=0.001). Fifth, in the diagnosis of ACS, the area under ROC curve of MMP-8 was 0.785, the sensitivity and specificity were 68.6% and 76.5%, respectively. Conclusion ① Serum MMP-8 concentration has close relationship with the occurrence of ACS, particularly with AMI; ② Serum MMP-8 concentration may be one of the predicting indicators of ACS and particularly of AMI; ③ Neutrophil may be correlated with serum MMP-8 concentration; ④ MMP-8 is of somewhat valuable in diagnosing ACS.展开更多
Zeolitic imidazolate framework-8(ZIF-8)is a typical filler used to fabricate mixed matrix membranes(MMMs)on account of its attractive advantage of high selective permeability for gas separation.However,the performance...Zeolitic imidazolate framework-8(ZIF-8)is a typical filler used to fabricate mixed matrix membranes(MMMs)on account of its attractive advantage of high selective permeability for gas separation.However,the performance is usually affected by filler aggregation due to strong interactions among fillers and weak interactions between the polymer and fillers,However,the performance is usually affected by filler aggregation due to strong interactions among fillers and weak interactions between the polymer and fillers,which will lead to a decrease of selectivity and the performance of gas separation will be strongly influenced.Herein,we modified ZIF-8 with 3-amino-1,2,4-triazole to obtain ZIF-8-NH_(2),Kapton polyamide acid was selected as the polymer matrix.Results showed that the ZIF-8-NH_(2)/Kapton MMMs has a good compatibility interface between ZIF-8 and Kapton because of the covalent bridging,even the filler loading up to 45%(mass).The 45%(mass)of ZIF-8-NH_(2)/Kapton membrane showed 297 barrer(1 barrer=10^(-10)10 cm^(3)·cm·cm^(-2)·s^(-1)·cmHg^(-1),1 cmHg=1333.22 Pa,standard temperature and pressure)of the permeability of H_(2)and 43.9 and 62.2 of selectivities for H_(2)/N_(2)and for H_(2)/CH_(4),respectively,which are beyond the upper limit of Robeson 2008.展开更多
