3-(4-Chlorophenyl)-7-[2-(piperazin-1-yl)ethoxy]-4H-chromen-4-one(CPEO-43)is a derivative of soybean isoflavone(SI),synthesized by introducing a chlorine atom and a piperazine group into the structure of natural SI.In ...3-(4-Chlorophenyl)-7-[2-(piperazin-1-yl)ethoxy]-4H-chromen-4-one(CPEO-43)is a derivative of soybean isoflavone(SI),synthesized by introducing a chlorine atom and a piperazine group into the structure of natural SI.In vitro experiments have demonstrated that CPEO-43 exhibits a notable inhibitory effect on both A549 cells and HCT116 cells.For the further development and utilization of CPEO-43,this study aims to establish and validate a liquid chromatography-tandem mass spectrometry(LC-MS/MS)quantitative analysis method for the pharmacokinetic study of CPEO-43.Normal rats were intragastrically administered different doses(2,6,and 20 mg/kg)of CPEO-43,and blood was taken from the ocular venous plexus at different time points.The blood concentration of CPEO-43 at different time points was determined using LC-MS/MS technology,and the pharmacokinetic parameters of the compound were calculated using the pharmacokinetic software DAS.The results indicated that the established LC-MS/MS method complies with the standards for bioanalytical method validation in the Chinese Pharmacopoeia(CHP)and can be applied to the pharmacokinetic study of CPEO-43.The pharmacokinetic software DAS non-compartmental model was successfully used to calculate the C_(max),T_(max),t_(1/2),AUC_(0-∞),MRTs,CL,and Vd pharmacokinetic parameters of the compound at different doses.The results were as follows:62.0±10.5,222.0±28.7,and 1384.5±376.4 ng/mL;8.5±1.2,6.0±0.0,and 11.0±6.2 h;15.6,15.0,and 18.5 h;1517.8±317.0,5328.7±864.4,and 45556.3±22735.6 ng·h/mL;17.8±1.2,17.7±0.8,and 20.0±3.2 h;1370.3±305.9,1153.5±205.6,and 505.3±179.8 mL/kg/h;30843.0±7458.0,24344.0±5237.0,and 13950.3±5996.9 mL/kg.These characteristics are of great significance for understanding the in vivo process of the drug,formulating dosing regimens,and evaluating the safety and efficacy of the drug.展开更多

The synthesized compounds were evaluated for their cytotoxic activity against five human cancer cell lines, namely, A549, MCF-7, HeLa, HT-29 and HCT-116. We found that compound 5e with two bromine atoms at the 5- and 7-positions of the indolin-2-one ring was most potent, which inhibited proliferation of five cancer cell lines in the range of 32.0%-62.3% at a concentration of 50 p,M. Our results further indicate that the connection of 5,7-dibromoindolin-2-one and 2-methylquinazolin- 4(3H)-one moiety with a carbon-carbon double bond is essential for compound 5e to exert cytotoxicity.展开更多

Their inhibitory activities in vivo were evaluated against Meloidogyne incognita. The newly prepared compounds A6, A8, A21, A28 and A38 exhibited more than 50% inhibition at the concentration of 20 mg/L. Especially compound A6 displayed 71.4% inhibition against Meloidogyne incognita at the concentration of 20 mg/L. The nematicidal activities varied significantly depending on the types and positions of the substituents, which provided guidance for further structure modification.展开更多

The catalytic activity of a-chymotrypsin was evaluated through investigating the temperature, the enzyme loading and the ratio of substrates in the enzyme-catalyzed reactions. The present method proves to be efficient and environmentally friendly in terms of short reaction time, high yield, green catalyst and the clean products obtained without further purification processes.展开更多

Effects of different parameters on extraction efficiency were evaluated. 99.96 % and 98.26 % palladium (Ⅱ) could be effectively extracted with 0.018 mol·L^-1 OIT/cyclohexane of 0.1 and 4.0 mol·L^-1 HCl medium, respectively. Nonpolar solvent and low acidity could improve the extracting efficiency, and successfully strip palladium (Ⅱ) from the loaded organic phase was achieved with 0.5 mol·L^-1 (NH2)2CS solution. It was proposed that the extraction of Pd com-plexes from HCl medium proceeded through the ion-association mechanism by slope method, NMR and FF-IR spectra.展开更多

The capitate glandular trichomes of Oenothera glazioviana(Onagraceae) were collected with laser microdissection and analyzed by gas chromatography-mass spectrometry. The volatile compound 4-hydroxy-4-methylpentan-2-one(1) was identified. We found that compound 1 displays antimicrobial, insecticidal, and phytotoxic activities. These results suggest that compound 1 might function as a defensive compound in the capitate glandular trichomes of O. glazioviana against pathogens, insect herbivores, and presumably competitive plants as well.展开更多

The crystal is of monoclinic,space group P21/n with a = 13.9854(3) ,b = 17.2678(4) ,c = 18.1828(5)A,β = 99.364(2) °,V = 4332.58(18) A^3,Z = 4,Dc = 1.338,F(000) =1824,μ = 0.185 mm^-1,MoKa radiation(λ = 0.71073) ,R = 0.0538 and wR = 0.1162 for 4728 observed reflections with I 〉 2σ(I) . X-ray diffraction analysis reveals the fused rings of benzo[4,5]furo[3,2-d]-1,2,4-triazolo[1,5-a] pyrimidin-5(1H) -one system are nearly coplanar. The crystal packing is mainly stabilized by weak intermolecular C-H···O hydrogen bond and π-π interactions.

文摘A series of some new 2-imino-5-[(Z)-1-(4-methylphenyl)methylidene]-3-[5-(2-oxo-2H-3-chromenyl)-1,3-oxazol-2-yl]-1,3- thiazolan-4-ones 5a-j has been synthesized and assayed for their antibacterial activity against Gram-positive bacteria viz.Bacillus subtilis(ATCC 6633),Staphylococcus aureus(ATCC 6538p) and Micrococcus luteus(IFC 12708),and Gram-negative bacteria viz. Proteus vulgaris(ATCC 3851).Salmonella typhimurium(ATCC 14028) and Escherichia coli(ATCC 25922).Among the screened compounds,5d,5e,5f,5g,and 5j exhibited potent inhibitory activity compared to standard drug,and emerged as potential molecules for further development.

The crystal is of monoclinic, space group P21/c with a = 13.7167(14), b = 13.113(1), c = 8.378(1) A, β = 98.992(2)^o, V = 1488.4(3) A^3, Z = 4, Dc = 1.278, F(000) = 608, μ = 0.089 mm^-1, MoKa radiation (2 = 0.71073), R = 0.0498, wR = 0.1238 for 2336 observed reflections with I 〉 2σ(I). X-ray diffraction analysis reveals that all ring atoms in the benzo[4, 5]furo [2,3-d] pyrimi- dinone moieties are almost coplanar.

文摘Bromination is used as a strategy to improve biological activity in medicinal chemistry.In order to study on the structure-activity relationships of the novel acetylcholinesterase inhibitors with 7H-thiazolo[3,2-b]-1,2,4-triazin-7-one scaffold,based on our previous work and molecular modeling,a series of novel 3-aryl-6-(bromoarylrnethyl)-7H-thiazolo[3,2-b]-1,2,4-triazin-7-one derivatives were designed by molecular docking,synthesized and characterized by mass spectra,infrared spectra,proton NMR and elemental analyses.The study of AChE inhibitory activity was carried out using the Ellman colorimetric assay with huperzine-A as the positive control.Most of all target compounds exhibited more than 45%inhibition at 10μmol/L.The preliminary structureactivity relationship was the bromine atoms and the hydroxyl group at the phenyl ring at the C6 position of the parent nucleus played significant roles in the AChE inhibitory activity of the target compounds.

文摘We discovered that a-chymotrypsin has a promiscuous ability to catalyze the cyclocondensation of aromatic and aliphatic aldehydes with 2-aminobenzamides to afford the corresponding 2,3-dihydroquinazolin-4(1H)-ones successfully in high yields(90%–98%) under alcohol solvent. The catalytic activity of a-chymotrypsin was evaluated through investigating the temperature, the enzyme loading and the ratio of substrates in the enzyme-catalyzed reactions. The present method proves to be efficient and environmentally friendly in terms of short reaction time, high yield, green catalyst and the clean products obtained without further purification processes.

文摘An environmentally benign aqueous protocol for the synthesis of 3,4-dihydropyrimidin-2-( 1 H)-ones and thiones promoted by a green catalyst, silica gel-supported L-pyrrolidine-2-carboxylic acid-4-hydrogen sulfate, has been developed. The use of nontoxic, thermally stable and inexpensive amino acid catalyst makes the process simple with minimal amount of chemical waste. Compared with classical Biginelli reaction conditions, this new method has the advantages of high yields and simple workup procedures.

文摘Vitamin B12 was applied as catalyst for the one-pot three-component synthesis of 3,4,5-trisubstituted furan-2(5H)-ones from the condensation between aldehydes, amines and dialkylacetylendicarboxylates at ambient temperature in Et OH. In addition, N-aryl-3-aminodihydropyrrol-2-one-4-carboxylates were synthesised using mentioned catalyst at ambient temperature in Et OH from condensation between formaldehyde, amines, and dialkylacetylenedicarboxylates. This methodology has number of advantages such as: use of green and nonhazardous catalyst, clean work up, short reaction times,high yields and no need to column chromatography.

文摘An efficient synthesis of symmetrical 2,2-bis(1H-indol-3-yl)-2H-acenaphthen-1-one is achieved via a reaction of acenaphthe-nequinone and indoles catalyzed by solid superacid SO4^2-/TiO2 under solvent-free conditions at room temperature by grinding, which provides an efficient route to the synthesis of symmetrical 2,2-bis(1H-indol-3-yl)-2H-acenaphthen-1-one.This procedure offers several advantages including solvent-free conditions,excellent yields of products,simple work-up as well as reuse of catalysts which makes it a useful and attractive protocol for the synthesis of these compounds.

文摘Solvent extraction of palladium (Ⅱ) from hydrochloric acid solution with 2-n-octyl-4-isothiazolin-3-one (OIT)/cyclohexane was studied. Effects of different parameters on extraction efficiency were evaluated. 99.96 % and 98.26 % palladium (Ⅱ) could be effectively extracted with 0.018 mol·L^-1 OIT/cyclohexane of 0.1 and 4.0 mol·L^-1 HCl medium, respectively. Nonpolar solvent and low acidity could improve the extracting efficiency, and successfully strip palladium (Ⅱ) from the loaded organic phase was achieved with 0.5 mol·L^-1 (NH2)2CS solution. It was proposed that the extraction of Pd com-plexes from HCl medium proceeded through the ion-association mechanism by slope method, NMR and FF-IR spectra.

文摘Glandular trichomes of plants produce a wide variety of secondary metabolites which are considered as major defensive chemicals. The capitate glandular trichomes of Oenothera glazioviana(Onagraceae) were collected with laser microdissection and analyzed by gas chromatography-mass spectrometry. The volatile compound 4-hydroxy-4-methylpentan-2-one(1) was identified. We found that compound 1 displays antimicrobial, insecticidal, and phytotoxic activities. These results suggest that compound 1 might function as a defensive compound in the capitate glandular trichomes of O. glazioviana against pathogens, insect herbivores, and presumably competitive plants as well.
