大型强子对撞机上发现的希格斯玻色子完成了标准模型(SM)的最后一块拼图,但仍有许多问题无法在SM的框架下得到完美的解释,如SM预言的电荷-宇称(CP)破坏效应不足以产生足够的重子数,因此无法合理解释宇宙中观测到的物质-反物质不对称性... 大型强子对撞机上发现的希格斯玻色子完成了标准模型(SM)的最后一块拼图,但仍有许多问题无法在SM的框架下得到完美的解释,如SM预言的电荷-宇称(CP)破坏效应不足以产生足够的重子数,因此无法合理解释宇宙中观测到的物质-反物质不对称性问题。在CP守恒的双希格斯二重态模型(2HDM)的TypeⅠ/X类型下,选取CP为偶的重希格斯粒子为类SM的希格斯玻色子进行研究,系统分析模型中可能生成强一阶电弱相变的参数区域。在计算中,对希格斯势能进行圈图修正并考虑有限温度场论的效应来计算相变温度,同时还考虑所有相关的理论和最新的实验约束。研究证实,在2HDM类型TypeⅠ/X中,仍存在着大量满足强一阶电弱相变且可被对撞机观测的参数空间。 展开更多

Cosmological imprints of Dirac neutrinos in a keV-vacuum 2HDM

The Dirac neutrino masses could be simply generated by a neutrinophilic scalar doublet with a vacuum being dramatically different from the electroweak one.While the case with an eV-scale vacuum has been widely explored previously,we exploit in this work the desert where the scalar vacuum is of O(keV)scale.In this regime,there would be rare hope to probe the keV-vacuum neutrinophilic scalar model via the lepton-flavor-violating processes,which makes it distinguishable from the widely considered eV-scale vacuum.Although such a keV-vacuum scenario is inert in the low-energy flavor physics,we show that the baryogenesis realized via the lightest Dirac neutrino can be a natural candidate in explaining the baryon asymmetry of the Universe.Furthermore,the Dirac neutrinos with a keV-vacuum scalar can generate a shift of the effective neutrino number within the range 0.097≤ΔN_(eff)≤0.112,which can be probed by the future Simons Observatory experiments.In particular,the model with a minimal valueΔN_(eff)=0.097 can already be falsified by the future CMB Stage-Ⅳand Large Scale Structure surveys,providing consequently striking exploratory avenues in the cosmological regime for such a keV-vacuum scenario. 展开更多

Background:KIT proto-oncogene,receptor tyrosine kinase(KIT,CD117)and platelet-derived growth factor-alpha(PDGFRA)are key drivers of gastrointestinal stromal tumors(GIST),but resistance to targeted therapy often arises from tumor protein p53(p53)alterations and loss of cell cycle control.However,the role of p53 status in GIST therapeutic potential has rarely been studied,so this study aimed to employ both wild-type andmutant p53 GIST models to investigate how p53 dysfunction influences the efficacy of p53 pathway-targeted therapies.Methods:The efficacy of the mouse double minute 2 homolog(MDM2)inhibitor(HDM201)and the Wee1 G2 checkpoint kinase(Wee1)inhibitor(adavosertib)was confirmed in both p53 wild-type(p53 WT)and p53 mutant(p53 MT)GIST cells.The anti-proliferative effects were assessed using the Cell Counting Kit-8(CCK-8)assay.Flow cytometry(FACS)and immunoblotting were employed to evaluate apoptosis and the expression of proteins related to drug efficacy.These findings were further validated in a xenograft model.Results:HDM201 selectively inhibited growth and triggered apoptosis in p53WT GIST cells,while adavosertib was effective mainly in p53 MT cells.Western blot analysis revealed thatHDM201 increased p53 and p21 levels in p53WT cells,and adavosertib affectedWee1 and phospho-cdc2 expression in both p53WT and p53 MT cells.In a xenograft mouse model,HDM201 significantly reduced the tumor volume and weight in p53WTGIST cells,whereas p53MT tumors showed only a moderate size reduction with adavosertib,without significant changes.Conclusions:Our results highlight the importance of p53 status in guiding GIST treatment.p53 WT tumors respond toMDM2 inhibitors,while p53 MTtumors show greater sensitivity toWee1 inhibitors,supporting p53 pathway targeting as a promising strategy for GIST patients. 展开更多

Charged Higgs Pair Production in a General Two Higgs Doublet Model at e^+e^- and μ^+μ^- Linear Colliders

In this paper, charged Higgs pair production through l^+l^- → H^+ H^-, where l = e or μ, is studied within the framework of a general Two Higgs Doublet Model (2HDM). The analysis is relevant to a future e^+e^- or μ^+ μ^- collider operating at center of mass energy of √s = 500 GeV. Two different scenarios of small and large a values are studied. Here a is the parameter, which diagonMizes the neutral CP-even Higgs boson mass matrix. Within the Minimal Supersymmetric Standard Model (MSSM), cross section of this process is almost the same at e+ e- and #+#- colliders. It is shown that at e^+e^- eolliders within a general 2HDM, cross section is not sensitive to the mass of neutral Higgs bosons, however, it can acquire large values up to several picobarn at μ^+μ^- colliders with the presence of heavy neutral Higgs bosons. A scan over Higgs boson mass parameter space is performed to analyze the effect of large masses of neutral Higgs bosons involved in the s-channel propagator and thus in the total cross section of this process. 展开更多

A Two-Higgs-Doublet Model without Flavor-Changing Neutral Currents at Tree-Level

The flavor-changing neutral current (FCNC) problem at tree-level is a very critical defect of the two Higgs doublet extension of standard model (SM). In this article, a two-Higgs-doublet model (2HDM) in which such defects do not exist at all is to be demonstrated. The general pattern of matrix pairs which can be diagonalized simul-taneously by a same unitary transformation is proposed without extra constraints like symmetries or zeros in M matrices. Only an assumption of the hermiticity of mass matrices is employed in the derivation. With this assumption, number of parameters in the mass matrix of a specific fermion type is reduced from eighteen down to five. Eigenvalues and eigenvectors are analytically derived and it is surprising that unitary transformation matrix thus derived depends on only two parameters. It is a very general and elegant way to solve the tree-level FCNC problem radically and it includes previous similar models as special cases with specific parameter values. 展开更多

Using the form factors calculated both in lattice QCD and HQET, we analyze the branching ratio, doublelepton polarization asymmetries and averaged double-lepton polarization asymmetries of Λ_b →Λμ^+μ^- decay in the2 HDM Ⅲ, respectively. Combining the experimental constrains on the 2HDM Ⅲ parameters, we take two set of representative parameter spaces in our calculations. For the branching ratio of Λ+b →Λμμ-decay, considering the uncertainties of the form factors and input parameters, we find the results of both SM and 2HDM Ⅲ can satisfy the current experimental data in the framework of lattice QCD, but not in HQET. Then we calculate the double-lepton polarization asymmetries Pij and their averaged values Pij, we find the double-lepton polarization asymmetries PLT,PNN, and PTT are sensitive to the 2HDM Ⅲ. However, PLN, PNL, PNT, and PTN show the opposite results. The contribution of the 2HDM Ⅲ to their averaged values have similar results. 展开更多

Neural Networks Search for Charged Higgs Boson in Two Doublet Higgs Model at the Hadrons Colliders

In this work we present an analysis of a search for charged Higgs boson in the context of Two Doublet Higgs Model (2HDM) which is an extension of the Standard Model of particles physics where the 2HDM predicts by existence scalar sector with new five Higgs bosons;two of them are electrically charged and the other three Higgs bosons are neutral charged. Our analysis based on the Monte Carlo data produced from the simulation of 2HDM with proton antiproton collisions at the Tevatron = 1.96 TeV (Fermi Lab) and proton proton collisions at the LHC = 14 TeV (CERN) with final state includes electron, muon, multiple jets and missing transverse energy via the production and decay of the new Higgs in the hard process where the dominant background (electrons and muons) for this process comes from the Standard Model processes via the production and decay of top quark pair. We assumed that the branching ratio of charged Higgs boson to tau lepton and neutrino is 100%. We used the Artificial Neural Networks (ANNs) which are an efficient technique to discriminate the signal of charged Higgs boson from the SM background for charged Higgs boson masses between 80 GeV and 160 GeV. Also we calculated the production cross section at different energies, decay width, branching ration and different kinematics distribution for charged Higgs boson and for the final state particles. 展开更多

目的:探究Numb蛋白在三阴乳腺癌患者中的表达降低情况,及Numb蛋白在三阴乳腺癌中对抑癌因子p53蛋白水平的影响及调控机制,进一步研究Numb蛋白的降低与三阴乳腺癌发生发展的相关性,从而为缺乏有效治疗方法的三阴乳腺癌提供一个潜在的治... 目的:探究Numb蛋白在三阴乳腺癌患者中的表达降低情况,及Numb蛋白在三阴乳腺癌中对抑癌因子p53蛋白水平的影响及调控机制,进一步研究Numb蛋白的降低与三阴乳腺癌发生发展的相关性,从而为缺乏有效治疗方法的三阴乳腺癌提供一个潜在的治疗新靶点。方法:40例三阴乳腺癌患者病理组织切片取自重庆医科大学临床病理诊断中心,采用免疫组化法检测Numb蛋白在三阴乳腺癌患者中的表达情况。MCF-10A细胞株和MDA-MB-231细胞株均为ATCC来源,采用qPCR和Western blot法检测对比Numb、HDM2、p53三者的转录水平和蛋白质水平在以上两个细胞株中差异。采用增强型绿色荧光蛋白(enhance green fluorescent protein,EGFP)质粒转染的方法在MDA-MB-231细胞中重表达Numb,采用q PCR和Western blot法验证Numb、HDM2、p53三者表达的变化。结果:转染NUMB-EGFP后MDA-MB-231细胞中Numb的mRNA和蛋白质水平均明显上调,HDM2无显著改变,p53在转录水平无明显变化,但在蛋白质水平显著升高。在231细胞中上调Numb蛋白可以在转录后水平调节p53水平,使p53蛋白随之显著升高。结论:Numb蛋白在三阴乳腺癌患者中表达降低的比列很高,为55%,且Numb蛋白在三阴乳腺癌细胞MDA-MB-231中可以调控抑癌因子p53蛋白水平,Numb蛋白水平与p53蛋白水平呈正相关。 展开更多

AIM: To investigate if non-peptidic small molecular inhibitors of the p53-HDM2 interaction could restore p53 function and kill tumor cells.METHODS: A series of non-peptidic small HDM2 inhibitors were designed by computer-aided model and synthesized by chemical method. Syl-155 was one of these inhibitors. Cytotoxic effect of syl-155 on three tumor cell lines with various states of p53, HT1080 (wild-type p53), KYSE510 (mutant p53), MG63 (p53 deficiency) was evaluated by MTT assay, Western blot and flow cytometry.RESULTS: Syl-155 stimulated the accumulation of p53 and p21 protein in HT1080 cells expressing wild-type p53, but not in KYSE510 and MG63 cells. Consequently, syl-155 induced cell cycle arrest and apoptosis in HT1080 cells.CONCLUSION: Non-peptidic small molecular inhibitors of the p53-HDM2 interaction show promise in treatment of tumors expressing wild-type p53. 展开更多

Differential effects of p63 mutants on transactivation of p53 and/or p63 responsive genes 被引量:1

Differential splicin... p63, known to play a role in development, has more recently also been implicated in cancer progression. Mutations in p63 have been shown to be responsible for several human developmental diseases. Differential splicing of the p63 gene gives rise to p63 isoforms, which can act either as tumor suppressors or as oncogene. In this report, we studied the effects of naturally occurring TAp637 mutants on the regulation of p53/p63 and p63 specific target genes. We observed significant differences among p63 mutants to regulate the p53/p63 and p63 specific target genes. Additionally, we observed a differential effect of p63 mutants on wildtype-p63-mediated induction ofp53/p63 and p63 specific target genes. We also demonstrated that these mutants differentially regulate the binding of wildtype p63 to the promoter of target genes. Furthermore, the effects of these mutants on cell death and survival were consistent with their ability to regulate the downstream targets when compared to wildtype TAp63T. In summary, our data demonstrate that p63 mutants exhibit differential effects on p63 and p53/p63 specific target genes and on the induction of apoptosis, and provide further insight into the function of p63. 展开更多

This study explores the diphoton and bb excesses at 95.4 GeV,as well as nano-Hertz gravitational waves originating from domain walls,within the framework of the next-to-two-Higgs-doublet model(N2HDM),which extends the two-Higgs-doublet model by introducing a real singlet scalar subject to a discrete Z_(2) symmetry.The Z_(2) symmetry is spontaneously broken by the non-zero vacuum expectation value of the singlet scalar,v_(s),which leads to the formation of domain walls.Two different scenarios are discussed:in scenario A,the 95.4 GeV Higgs boson predominantly originates from the singlet field,while in scenario B,it arises mainly from the CP-even components of the Higgs doublets.Accounting for relevant theoretical and experimental constraints,scenario A can fully account for both the diphoton and bb excesses at 95.4 GeV within the 1σrange.In the parameter space accommodating both excesses,scenario A fails to provide a valid explanation for the NANOGrav data up to v_(s)=1000 TeV,and the predicted gravitational wave spectrum can exceed the SKA sensitivity curve in the low frequency region.Scenario B only marginally accounts for the diphoton and bb excesses at the 1σlevel,but can simultaneously explain the NANOGrav data well. 展开更多

Probing heavy charged Higgs bosons at gamma-gamma colliders using a multivariate technique

This study explores the production of charged Higgs particles through photon-photon collisions within the context of the Two Higgs Doublet Model,including one-loop-level scattering amplitudes of electroweak and QED radiation.The cross-section has been scanned for the plane(m_(φ^(0)),√s)to investigate the process ofγγ→H^(+)H^(-).Three particular numerical scenarios,i.e.,low-,non-alignment,and short-cascade are employed.The decay channels for charged Higgs particles are examined using h^(0)for low-m_(H^(0))and H^(0)for non-alignment and short-cascade scenarios incorporating the new experimental and theoretical constraints along with the analysis for cross-sections.We find that,at a low energy,the cross-section is consistently higher for all scenarios.However,as√s increases,it reaches a peak value at 1 TeV for all benchmark scenarios.The branching ratio of the decay channels indicates that for non-alignment,the mode of decay W^(±)h^(0)takes control,and for a short cascade,the prominent decay mode remains tb,whereas in the low-scenario,the dominant decay channel is of W^(±)h^(0).In our research,we employ contemporary machine-learning methodologies to investigate the production of high-energy Higgs bosons within a 3.0 TeVγγcollider.We have used multivariate approaches such as Boosted Decision Trees(BDT),LikelihoodD,and Multilayer Perceptron(MLP)to show the observability of heavy-charged Higgs Bosons versus the most significant Standard Model backgrounds.The purity of the signal efficiency and background rejection are measured for each cut value. 展开更多
