菝葜HPLC指纹图谱及抗氧化活性谱效关系研究

目的:研究菝葜的指纹图谱与抗氧化活性的关联性,明确菝葜发挥抗氧化作用的药效成分群。方法:采用Agilent 5 TC-C18(2)色谱柱(250 mm×4.6 mm,5μm),柱温30℃,以甲醇-0.05%磷酸水溶液为流动相,梯度洗脱,流速0.8 mL·min^(-1),检... 目的:研究菝葜的指纹图谱与抗氧化活性的关联性,明确菝葜发挥抗氧化作用的药效成分群。方法:采用Agilent 5 TC-C18(2)色谱柱(250 mm×4.6 mm,5μm),柱温30℃,以甲醇-0.05%磷酸水溶液为流动相,梯度洗脱,流速0.8 mL·min^(-1),检测波长295 nm,进样量10μL,建立菝葜的HPLC指纹图谱,进行相似度评价并指认共有峰,同时进行聚类分析及主成分分析。采用2,2-联苯基-1-苦基肼基(DPPH)自由基清除法、2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS)自由基清除法评价29批菝葜的抗氧化活性。运用灰色关联分析和偏最小二乘回归分析2种方法进行谱效关系研究。结果:29批菝葜的HPLC指纹图谱相似度在0.784~0.996,共标定出23个共有峰,其中12个为经对照品比对确认。聚类分析、主成分分析将样品聚为3类;29批样品均有不同程度的抗氧化能力;灰色关联分析和偏最小二乘回归分析结果显示,峰6、10、11、12、13、15及18是主要的共有特征峰。通过对照品比对,其中峰6、10、11、12、13、15分别被鉴定为绿原酸、白藜芦醇苷、氧化白藜芦醇、新落新妇苷、落新妇苷和白藜芦醇。进一步的体外抗氧化活性评价及谱效相关性分析表明,这些化合物(尤其是黄酮类和二苯乙烯类成分)是菝葜发挥抗氧化作用的关键药效物质。结论:菝葜的抗氧化作用是多种成分协同作用的结果,6、10、11、12、13、15和18号色谱峰对应的化合物,可能是菝葜发挥抗氧化作用的主要活性成分。 展开更多

In the present work,the fruits of four Morus species,namely Morus alba (white mulberry),Morus nigra (black mulberry),Morus laevigata (large white fruit),and Morus laevigata (large black fruit),were analyzed for proximate composition,essential minerals,and antioxidant potentials.For this purpose,the ripe fruits were collected from the northern regions of Pakistan.The major nutritional components moisture,ash,lipids,proteins,fibres,carbohydrates,and total sugar) were found to be in the suitable range along with good computed energy.Total dry weight,pH,and titratable acidity (percent citric acid) were (17.60±1.94)–(21.97±2.34) mg/100 g,(3.20±0.07)–(4.78±0.15),and (0.84±0.40)%–(2.00±0.08)%,respectively.Low riboflavin (vitamin B2) and niacin (vitamin B3) contents were recorded in all the fruits,while ascorbic acid (vitamin C) was in the range from (15.20±1.25) to (17.03±1.71) mg/100 g fresh weight (FW).The mulberry fruits were rich with regard to the total phenol and alkaloid contents,having values of (880±7.20)–(1650±12.25) mg/100 g FW and (390±3.22)–(660±5.25) mg/100 g FW,respectively.Sufficient quantities of essential macro-(K,Ca,Mg,and Na) and micro-(Fe,Zn,and Ni) elements were found in all the fruits.K was the predominant element with concentration ranging from (1270±9.36) to (1731±11.50) mg/100 g,while Ca,Na,and Mg contents were (440±3.21)–(576±7.37),(260±3.86)–(280±3.50),and (240±3.51)–(360±4.20) mg/100 g,respectivly.The decreasing order of micro-minerals was Fe>Zn>Ni.The radical scavenging activity of methanolic extract of fruits was concentration-dependent and showed a correlation with total phenolic constituents of the respective fruits.Based on the results obtained,mulberry fruits were found to serve as a potential source of food diet and natural antioxidants. 展开更多

Objective:To search for new sources of safe and inexpensive antioxidants,the leaf,root and rhizome of Enhalus acoroides(E.acoroides) were screened for the first time for their antioxidant activities.Methods:Samples of leaf,root and rhizome from E.acoroides were tested for total phenolic content,proanthocyanidins and in vitro antioxidant activity in terms of total antioxidant assay,DPPH assay and FRAP assay.Results:The leaf sample was found to have high levels of phenolic(0.323±0.028 mg TAE/g) and proanthocyanidins(0.570 0±0.000 3 mg TAE/g) when compared to root and rhizome.The leaf samples exhibited higher total antioxidant activity (11.770±0.026 mg Ascorbic acid equivalent/g),higher percentage of DPPH radical scavenging activity(25.76±0.04) and higher reducing power(18.060±0.073) in terms of mg GAE/g.In addition, there was a significant correlation between total phenolic content and total antioxidant activity (R^2=0.923),DPPH assay(R^2=0.509).FRAP assay(R^2=0.994).Similarly,significant correlation was found between the proanthocyanidin and total antioxidant activity(R^2=0.977),DPPH assay (R^2=0.464) and the FRAP assay(R^2=0.998).Conclusions:These results suggested that E.acoroides have strong antioxidant potential.Further study is necessary for isolation and characterization of the active antioxidant agents,which can be used to treat various oxidative stress-related diseases. 展开更多

Objective: Myanmar has a long history of using medicinal plants for treatment of various diseases. To the best of our knowledge there are no previous reports on antiglycation activities of medicinal plants from Myanmar. Therefore, this study was aimed to evaluate the antioxidant, antiglycation and antimicrobial properties of 20 ethanolic extracts from 17 medicinal plants indigenous to Myanmar. Methods: In vitro scavenging assays of 2,2-diphenyl-l-picrylhydrazyl (DPPH), nitric oxide (NO), superoxide (SO) radicals were used to determine the antioxidant activities. Folin-Ciocalteu's method was performed to determine the total phenolic content. Antiglycation and antimicrobial activities were detected by bovine serum albumin-fluorescent assay and agar well diffusion method. Results: Terminalia cbebula Retz. (Fruit), containing the highest total phenolic content, showed high antioxidant activities with inhibition of 77.98% ± 0.92% 88.95% ± 2.42%, 88.56% ±1.87% and 70.74% ±2.57% for DPPH, NO, SO assays and antiglycation activity respectively. It also showed the antimicrobial activities against Staphylococcus aureus, Bacillus cereus, Escherichia coil, Pseudomonas aeruginosa and Candida albicans with inhibition zone of 19, 18, 17, 25 and 15 ram, respectively. Garcinia mangostana Linn. showed the strongest activities for SO and antiglycation assays with inhibition of 93.68% ± 2.63% and 82.37%± 1.78%. Bark of Melia sp. was the best NO radical scavenger with inhibition rate of 89.39%± 0.60%. Conclusion: The results suggest that these plants are potential sources of antioxidants with free radical-scavenging and antiglycation activities and could be useful for decreasing the oxidative stress and glycation end-product formation in glycation-related diseases. 展开更多

Objective:To ensure the safety and evaluate the anti oxidant activity of Terminalia paniculata(T.paniculata)ethanolic extract in Sprague Dawley rats.Methods:The solvent extracts(hexane,ethyl acetate and ethanol)of T.paniculata were subjected to phytochemical analysis and their DPPH radical scavenging activity was assayed.The oral acute toxicity was evaluated using ethanolic extract of T.paniculata.Results:Ethyl acetate and elhanolic extracts showed more phytochemicals,whereas highest DPPH scavenging activity was found in ethanolic extract.In an acute toxicity study,T.paniculata ethanolic extract was orally administered(1000 mg/kg body weight)to rats and observed for72 h for any toxic symptoms and the dose was continued up to 14 d.On the 15th day rats were sacrificed and blood samples were collected from control and test animals and analyzed for some biochemical parameters.We did not observe any behavioral changes in test groups in comparison with their controls.Also,there were no significant alterations in biochemical,hematological(hemoglobin content and blood cells count)and liver function parameters such as serum glutamate pyruvate transaminase,serum glutamate oxaloacetate transaminase,alkaline phosphatase,total proteins,albumin and bilirubin levels between T.paniculata ethanolic extract treated and normal control groups.Conclusions:Together our results demonstrated that T.paniculala ethanolic possessed potent antioxidant activity and it was safer and non toxic to rats even at higher doses and therefore could be well considered for further investigation for its medicinal and therapeutic efficacy. 展开更多

Background:The prime objective of the current research was to evaluate the whole plant hydroalcoholic extract of Ludwigia octovalvis(HLO)against hyperglycemia,and oxidative stress biomarkers in rats induced with diabetes comorbid depression,diabetes comorbid depression(streptozotocin-nicotinamide+electric footshocks).Methods:2,2-Diphenyl-1-picrylhydrazyl assay of HLO versus ascorbic acid was done.Effects of 200 and 400 mg/kg body weight/day HLO doses versus 25 mg/kg body weight/day metformin was studied through insulin,glucose,superoxide dismutase,lipid peroxidation,catalase,and behavioral assessment(forced swim and open field tests).Results:IC50 values of HLO and ascorbic acid were 33.52 and 27.86μg/mL respectively.Both the HLO doses showed intended results with respect to oxidative stress biomarkers in diabetes comorbid depression rats in comparison to metformin.Open field test showed better results for HLO in diabetes comorbid depression rats.However,hypoglycemic effects,and forced swim test performance of metformin was slightly higher than the 400 mg dose,followed by the 200 mg dose of HLO.Ethyl gallate,gallic acid,β-sitosterol,and quercetin in HLO might resulted in attenuating diabetic as well as depression biomarkers.Conclusion:Inhibition of glucosidase and lipase activity,and AMP-activated protein kinase phosphorylation might be the possible biochemical changes occurred in HLO treated rats. 展开更多

Gold nanostructure can be manufactured in many forms,such as spherical by using simple chemical method.X-ray powder diffraction(XRD)pattern of gold nanoparticles(AuNPs)was derived from a 400-μL trisodium citrate solution.The diffraction peaks corresponding to the diffraction planes of(111),(200),and(220)were all indexed to the gold with a face-centered cubic structure.The lattice constant calculated from the XRD pattern is 4.078Å,which matches the conventional cubic gold metal diffraction pattern well(Pattern card number(04-784)).The results of scanning electronic microscope show that the biological nanoparticles of gold have asymmetric shapes and different sizes grouped as a circular particle.It was observed that the fashioning of AuNPs increased with increase in the concentration of[6]-gingerol waste extracts.The viability of FTC-133 cells treated for 48 h with various concentrations of[6]-gingerol extract coated with AuNPs(12.5-400 g/mL)was used to determine the cytotoxicity.A dose-dependent reduction in FTC-133 cell viability caused by[6]-gingerol extract capped with Au NPs was substantial(P<0.05),reaching 75%cell mortality at 400 g/mL;the IC_(50)was 90.5%. 展开更多
