目的研究spd1672基因在肺炎链球菌感染过程中的作用。方法通过腹腔攻毒实验和细菌入血载量分析、细菌的粘附侵袭实验、全血杀菌实验以及相关细胞因子检测等观察spd1672基因敲除(D39△1672菌株)后对肺炎链球菌毒力的影响。结果D39△1672... 目的研究spd1672基因在肺炎链球菌感染过程中的作用。方法通过腹腔攻毒实验和细菌入血载量分析、细菌的粘附侵袭实验、全血杀菌实验以及相关细胞因子检测等观察spd1672基因敲除(D39△1672菌株)后对肺炎链球菌毒力的影响。结果D39△1672菌株感染组小鼠中位生存时间和生存率显著高于野生菌株组(P<0.05),而血中细菌载量则显著低于野生菌株感染组(P<0.05)。与野生菌株比较,D39△1672菌株对A549细胞的粘附能力无明显差异,但其侵袭能力显著受损(P<0.05)。此外,D39△1672菌株对全血杀菌作用的抵抗能力亦较野生菌显著减弱(P<0.05),小鼠体内炎性细胞因子水平显著降低。结论spd1672基因与肺炎链球菌的侵袭、抗宿主能力和致炎反应等过程相关。 展开更多

目的原核表达肺炎链球菌SPD1672蛋白膜外EL4功能环,制备并鉴定其多克隆抗血清。方法生物信息学分析SPD1672蛋白结构功能区,用p Cold TF质粒构建含EL4功能环的原核表达载体,IPTG诱导蛋白质表达后行镍柱纯化。经HRV 3C蛋白酶酶切、鉴定后... 目的原核表达肺炎链球菌SPD1672蛋白膜外EL4功能环,制备并鉴定其多克隆抗血清。方法生物信息学分析SPD1672蛋白结构功能区,用p Cold TF质粒构建含EL4功能环的原核表达载体,IPTG诱导蛋白质表达后行镍柱纯化。经HRV 3C蛋白酶酶切、鉴定后,免疫新西兰大白兔以制备多克隆抗血清,间接ELISA鉴定效价,western blot鉴定抗血清与肺炎链球菌D39菌株天然蛋白质的亲合力。结果 EL4环可能为SPD1672蛋白酶活性中心。成功构建p Cold TF-SPD1672EL4原核表达载体,诱导表达后获得纯度大于95%的重组融合目的蛋白质。该蛋白质免疫新西兰大白兔后获得效价达到5.12×106的多克隆抗血清,此抗血清与重组的SPD1672EL4蛋白及肺炎链球菌中天然SPD1672蛋白有较好的亲合力。结论成功获得高纯度的肺炎链球菌SPD1672EL4蛋白及其特异的多克隆抗血清,为进一步对SPD1672蛋白的结构与功能研究奠定了基础。 展开更多

目的查找桐庐县某单位发生的1起食源性疾病事件病因,提出控制措施,防止类似事件再次发生。方法开展病例搜索与个案调查,调查描述病例的临床表现和疾病分布特征,开展病例对照研究和现场卫生学勘查,采集可疑样品进行实验室检测,分析推断... 目的查找桐庐县某单位发生的1起食源性疾病事件病因,提出控制措施,防止类似事件再次发生。方法开展病例搜索与个案调查,调查描述病例的临床表现和疾病分布特征,开展病例对照研究和现场卫生学勘查,采集可疑样品进行实验室检测,分析推断引发事件的可疑食物及有关危险因素。结果事件发生于2018年1月6日傍晚,共发现19例患者均为某单位员工,在发病前均有单位食堂共同就餐史,临床主要表现为恶心(占68.42%)、呕吐(89.47%)、腹痛(52.63%)、腹泻(36.84%)等症状,疑为群体性食源性疾病事件,推测事件发生当天(1月6日)的午餐为暴露餐次,发病潜伏期中位数为3.5h;该午餐菜谱分析结果显示,病例组(100.00%)和对照组(14.29%)间四季豆炒肉暴露率差异有统计学意义(OR=95.00,P<0.01),对午餐食用的四季豆炒肉样品作皂素定性检测结果阳性;现埸勘查和危害分析发现食堂在制作四季豆炒肉时,由于加工量大,烹饪时间过短,导致四季豆中的皂苷成分未被加热过程完全破坏。结论这是1起因共同食用加热不透的四季豆菜肴所致的皂苷中毒事件。 展开更多

In order to decrease the deformation and stress and increase the natural frequency of the fixed table,a method of optimization driven by the sensitivity and topology analyses is proposed.The finite element model of the fixed table is constructed and analyzed by using ANSYS software.Based on the results of static analysis and modal analysis,the maximum deformation,the maximum stress,and natural frequencies are obtained.Then,the sensitivity analysis and topology optimization are carried out to find out the parameters to be optimized.The fixed table is reconstructed according to optimal design scheme.In the comparison of the results between original model and the optimized one,the maximum deformation and stress are decreased by 71.73%and 60.27%respectively.At the same time,the natural frequencies from the first mode to the sixth mode are increased by 30.28%,29.57%,29.51%,31.52%,22.19%,and 21.80%,respectively.The method can provide technology guide for the design and optimization of machining structure. 展开更多

Inspired by the idea that bionic non-smooth surfaces(BNSS)can reduce water flow resistance,the application of BNSS resistance reduction method in grooves surface of antiskid tire tread pattern has been investigated for increasing hydroplaning velocity of tire by using computational fluid dynamics(CFD)simulation.Three kinds of BNSS(riblet,convex dome,and dimple concave)are arranged in tire tread grooves to study the water flow resistance effects in grooves with non-smooth characteristics.A tire-water coupled model is established and CFD technique is applied to simulating hydroplaning.The simulation results show that BNSS grooves can reduce water flow resistance and increase mean flow rate by disturbing the eddy movement in boundary layers.The drag forces of riblet and dimple surface are lower and drainage capacity is higher than those of smooth surface under the same void space on tread pattern,but it is not in dome.BNSS is a good way to promote antiskid performance without increasing additional groove space;extra tire-road noise production is therefore avoided due to groove space enlargement. 展开更多

In order to realize the parametric design of the conjugate cam weft insertion mechanism,according to the necessary parameters of weft insertion process,ideal kinematic curves of the weft insertion mechanism were given,and the mathematical model of reverse solution for this mechanism was established.The parameters of this mechanism were obtained by reverse solution on the basis of the given ideal kinematic curves and kinematic requirements.The parametric design platform which was integrated with the functions of parametric reverse solution,motion simulation,three-dimensional modeling and virtual assembly was developed based on VB.NET and Unigraphics(UG) NX.After entering the technological parameters of weft insertion process and essential structural parameters by users,three-dimensional drawing of the main parts such as the conjugate cam and shaft,can be obtained by this platform,also the processing data of the cam can be calculated.This platform provides a rapid approach for parametric design of weft insertion mechanism. 展开更多

Jet pipe electro-hydraulic servo valve is the heart of feedback control systems,and it is one of the mechatronic components used for precision flow control application.It consists of several precision and ddicate components.The performance of the jet pipe servo valve depends on many parameters.During the developmental stage,it is very difficult to ascertain the function parameters.The steady-state analysis of jet pipe electro-hydraulic servo valve has been made to simulate its fluid characteristics (flowin,flow-out,leakage flow,recovery or load pressure,etc.) by mathematical modeling.Theoretical model was conducted on various affecting parameters on the pressure,the main flow rate of fluid,or leakage flow through the receiver holes.The major parameters studied are jet pipe nozzle diameters,receiver hole diameters,angle between the two centre-lines of receiver hole,nozzle offset,and nozzle stand-of distance.In this paper the research is important to determine and optimize the structural parameters of jet pipe servo valve.Thus,equations of the pressure and flow characteristics are set up and the optimal structural parameters of jet pipe are established. 展开更多

A self-tuning reaching law based sliding mode control(SMC)theory is proposed to stabilize the nonlinear continuous stirred tank reactor(CSTR).T-S fuzzy logic is used to build a global fuzzy state-space linear model.Combing the traits of SMC and CSTR,three fuzzy rules can meet the requirements of controlled system.The self-tuning switch control law which can drive the state variables to the sliding surface as soon as possible is designed to ensure the robustness of uncertain fuzzy system.Lyapunov equation is applied to proving the stability of the sliding surface.The simulations show that the proposed approach can achieve desired performance with less chattering problem. 展开更多

Increasing complexity of distributed hydrological model (DHM) has lowered the efficiency of convergence.In this study,global sensitivity analysis (SA) was introduced by combining multiobjective (MO) optimization for DHM calibration.Latin Hypercube-once at a time (LH-OAT) was adopted in global parameter SA to obtain relative sensitivity of model parameter,which can be categorized into different sensitivity levels.Two comparative study cases were conducted to present the efficiency and feasibility by combining SA with MO(SA-MO).WetSpa model with non-dominated sorting genetic algorithm-Ⅱ (NSGA-Ⅱ) algorithm and EasyDHM model with multi-objective sequential complex evolutionary metropolis-uncertainty analysis (MOSCEM-UA)algorithm were adopted to demonstrate the general feasibility of combining SA in optimization.Results showed that the LH-OAT was globally effective in selecting high sensitivity parameters.It proves that using parameter from high sensitivity groups results in higher convergence efficiency.Study case Ⅰ showed a better Pareto front distribution and convergence compared with model calibration without SA.Study case Ⅱ indicated a more efficient convergence of parameters in sequential evolution of MOSCEM-UA under the same iteration.It indicates that SA-MO is feasible and efficient for high dimensional DHM calibration. 展开更多

To extract features of fabric defects effectively and reduce dimension of feature space,a feature extraction method of fabric defects based on complex contourlet transform (CCT) and principal component analysis (PCA) is proposed.Firstly,training samples of fabric defect images are decomposed by CCT.Secondly,PCA is applied in the obtained low-frequency component and part of highfrequency components to get a lower dimensional feature space.Finally,components of testing samples obtained by CCT are projected onto the feature space where different types of fabric defects are distinguished by the minimum Euclidean distance method.A large number of experimental results show that,compared with PCA,the method combining wavdet low-frequency component with PCA (WLPCA),the method combining contourlet transform with PCA (CPCA),and the method combining wavelet low-frequency and highfrequency components with PCA (WPCA),the proposed method can extract features of common fabric defect types effectively.The recognition rate is greatly improved while the dimension is reduced. 展开更多
