BACKGROUND To date,there has been no effective treatment for intervertebral disc degeneration(IDD).Nucleus pulposus-derived mesenchymal stem cells(NPMSCs)showed encouraging results in IDD treatment,but the overexpression of reactive oxygen species(ROS)impaired the endogenous repair abilities of NPMSCs.6-gingerol(6-GIN)is an antioxidant and anti-inflammatory reagent that might protect NPMSCs from injury.AIM To investigate the effect of 6-GIN on NPMSCs under oxidative conditions and the potential mechanism.METHODS The cholecystokinin-8 assay was used to evaluate the cytotoxicity of hydrogen peroxide and the protective effects of 6-GIN.ROS levels were measured by 2´7´-dichlorofluorescin diacetate analysis.Matrix metalloproteinase(MMP)was detected by the tetraethylbenzimidazolylcarbocyanine iodide assay.TUNEL assay and Annexin V/PI double-staining were used to determine the apoptosis rate.Additionally,autophagy-related proteins(Beclin-1,LC-3,and p62),apoptosisassociated proteins(Bcl-2,Bax,and caspase-3),and PI3K/Akt signaling pathwayrelated proteins(PI3K and Akt)were evaluated by Western blot analysis.Autophagosomes were detected by transmission electron microscopy in NPMSCs.LC-3 was also detected by immunofluorescence.The mRNA expression of collagen II and aggrecan was evaluated by real-time polymerase chain reaction(RT-PCR),and the changes in collagen II and MMP-13 expression were verified through an immunofluorescence assay.RESULTS 6-GIN exhibited protective effects against hydrogen peroxide-induced injury in NPMSCs,decreased hydrogen peroxide-induced intracellular ROS levels,and inhibited cell apoptosis.6-GIN could increase Bcl-2 expression and decrease Bax and caspase-3 expression.The MMP,Annexin V-FITC/PI flow cytometry and TUNEL assay results further confirmed that 6-GIN treatment significantly inhibited NPMSC apoptosis induced by hydrogen peroxide.6-GIN treatment promoted extracellular matrix(ECM)expression by reducing the oxidative stress injury-induced increase in MMP-13 expression.6-GIN activated autophagy by increasing the expression of autophagy-related markers(Beclin-1 and LC-3)and decreasing the expression of p62.Autophagosomes were visualized by transmission electron microscopy.Pretreatment with 3-MA and BAF further confirmed that 6-GIN-mediated stimulation of autophagy did not reduce autophagosome turnover but increased autophagic flux.The PI3K/Akt pathway was also found to be activated by 6-GIN.6-GIN inhibited NPMSC apoptosis and ECM degeneration,in which autophagy and the PI3K/Akt pathway were involved.CONCLUSION 6-GIN efficiently decreases ROS levels,attenuates hydrogen peroxide-induced NPMSCs apoptosis,and protects the ECM from degeneration.6-GIN is a promising candidate for treating IDD. 展开更多

Objective:To develop and validate a simple,accurate HPTLC method for the analysis of 8-gingerol and to determine the quantity of 8-gingerol inZingiber officinaleextract and gingercontaining dietary supplements,teas and commercial creams.Methods:The analysis was performed on 10×20 cm aluminium-backed plates coated with 0.2 mm layers of silica gel 60 F254(E-Merck,Germany)with n-hexane:ethyl acetate 60:40(v/v)as mobile phase.Camag TLC Scanner III was used for the UV densitometric scanning at 569.Results:This system was found to give a compact spot of 8-gingerol at retention factor(Rf) value of(0.39依0.04)and linearity was found in the ranges 50-500 ng/spot(r2=0.9987).Limit of detection(12.76 ng/spot),limit of quantification(26.32 ng/spot),accuracy(less than 2%)and recovery(ranging from 98.22-99.20)were found satisfactory.Conclusions:The HPTLC method developed for quantification of 8-gingerol was found to be simple,accurate,reproducible,sensitive and is applicable to the analysis of 8-gingerol in Zingiber officinaleextract and ginger-containing dietary supplements,teas and commercial creams. 展开更多

Objective: To study in vitro anti-hydatic and immunomodulatory effects of ginger and [6]-gingerol as an alternative therapy for Cystic echinococcosis. Methods: Effect of a commonly used herbal product and ginger(Zingiber officinale) towards protoscoleces(PSC) and cyst wall in vitro was studied. The effect of [6]-gingerol, and the pungent constituent of ginger, was also evaluated on PSC culture. Furthermore, the activity of both extracts in association with interferon-gamma(IFN-γ) on PSC co-cultured with mononuclear cells of hydatic patients was evaluated. The nitric oxide(NO) production was measured in each co-culture. Results: Ginger exhibited a concentration- and time-dependent cytotoxic effect against PSC and cyst wall. Interestingly, ginger was more effective than the [6]-gingerol. Moreover, additional parasitic effect between extracts and IFN-γ are also observed in co-cultures. Furthermore, both extracts attenuated the NO production elicited by this infection or by the IFN-γ. Conclusions: Ginger has an important anti-hydatic effect in vitro. This effect is amplified in the presence of IFN-γ. Moreover, this herbal product may protect against host's cell death by reducing the high levels of NO. Ginger may act, at least, through the [6]-gingerol. All our data suggest the promising use of ginger in the treatment of Echinococcus granulosus infection. 展开更多

Objective:To explore the effects of 6-gingerol,asarinin,and deoxyschizandrindthe main components of Zingiber officinale(Willd.)Rosc.(Gan Jiang),Asarum heterotropoides f.var.mandshuricum(Maxim.)(Xi Xin),and Schisandra chinensis(Turcz.)Baill.(Wu Wei Zi),respectivelydon an interleukin(IL)-13einduced BEAS-2B cell model in vitro.Methods:The BEAS-2B cell model was established using 25 ng/mL IL-13 combined with 1%fetal bovine serum(FBS)in vitro.Mitoquinone mesylate(Mito-Q)treatment was used as a positive control group,and different concentrations of 6-gingerol,asarinin,and deoxyschizandrin were used to treat the models.The level of reactive oxygen species(ROS)production was detected by flow cytometry.The expression levels of LC3B,Beclin1,adenosine 50-monophosphate(AMP)eactivated protein kinase(AMPK),phosphory-lated-AMPeactivated protein kinase(P-AMPK),dynamin-related protein 1(DRP1),and mitochondrial fusion protein 2(MFN2)were detected by Western blot.Mitochondrial membrane potential(MMP)assay kit with JC-1 was utilized to detect the level of MMP.Results:The BEAS-2B cells exposed to 25 ng/mL IL-13 with 1%FBS showed an increased ROS level and a decreased MMP.6-Gingerol,asarinin,and deoxyschizandrin were able to downregulate ROS level and upregulate the MMP in the BEAS-2B model.Asarinin and deoxyschizandrin reduced the expression of autophagy protein LC3B,while deoxyschizandrin significantly increased the expression of DRP1 in the BEAS-2B model.Conclusion:6-Gingerol,asarinin,and deoxyschizandrin can reduce ROS generation and increase MMP,but have different regulatory effects on the expression of autophagy protein and mitochondrial mitotic protein.The three components have both synergistic and complementary effects in classic medicine compatibility.This study may provide an innovative strategy to reduce the lung inflammation related to IL-13. 展开更多

The morbidity and mortality of cardiovascular diseases are very high,which has attracted more and more attention all over the world.Common treatment methods for clinical treatment of acute myocardial infarction include direct percutaneous coronary intervention and coronary artery bypass grafting,which can quickly restore blocked coronary blood flow and reduce the infarct size.However,the inevitable ischemia/reperfusion injury will occur during the recovery of coronary blood flow,its pathological mechanism is complicated,and the Western medicine countermeasures are very limited.Among the current drugs for the treatment of cardiovascular diseases,traditional Chinese medicine has become a research hotspot due to its multiple targets,safety,and low side effects.Ginger is the fresh rhizome of Zingiber officinale Rosc.,a perennial herbaceous plant in the ginger family.It is a dual-purpose resource of medicine and food.Ginger has the functions of relieving the appearance and dispelling cold,warming up and relieving vomiting,resolving phlegm and relieving cough,and relieving fish and crab poison.The chemical components of ginger mainly include volatile oil,gingerol,diphenylheptane,etc..Among them,6-gingerol,as the main active component of gingerols,has obvious pharmacological effects in myocardial protection,anti-oxidation,anti-inflammatory,etc..Studies have shown that 6-gingerol protects myocardium mainly through anti-oxidative stress,anti-inflammatory,inhibiting cell apoptosis,and preventing calcium influx.①Anti-oxidative stress:oxidative stress is a state where oxidation and anti-oxidation in the body are out of balance,and it is also an important factor leading to myocardial damage.Many studies have confirmed that 6-gingerol has an antioxidant effect,and it is considered a natural antioxidant.6-gingerol can significantly reduce the degree of oxidative stress and the level of reactive oxygen species caused by cardiomyocyte damage,and has a significant cardioprotective effect.②Anti-inflammatory:inflammation can cause substantial cell damage and organ dysfunction,which is another important cause of myocardial damage.6-gingerol can reduce the levels of inflammatory factors such as interleukin-6,interleukin-1β,and tumor necrosis factor-αin cardiomyocytes,and at the same time inhibit the TLR4/NF-κB signaling pathway,an important regulatory pathway of inflammation,showing that it may improve myocardial damage through anti-inflammatory effects.③Inhibition of apoptosis:apoptosis is a complex and orderly process in the autonomous biochemical process of cells,and one of the main mechanisms of myocardial injury.This process can be roughly divided into three pathways:mitochondria,endoplasmic reticulum,and death receptors.Among them,the mitochondrial pathway plays an important role,and Bcl-2 and Bax located upstream of this pathway can regulate the entire process of cell apoptosis by regulating the permeability of the mitochondrial membrane.Studies have found that the preventive application of 6-gingerol can reduce cell damage,reduce the number of apoptotic cells,reduce the activity of Bax and caspase-3,and increase the expression of Bcl-2.Therefore,6-gingerol pretreatment can reduce the damage of cardiomyocytes,and its mechanism may be related to the inhibition of apoptosis.④Prevent calcium influx:calcium overload is involved in the pathogenesis of myocardial ischemic injury,which may be related to excessive contracture,arrhythmia,and mitochondrial Ca2+accumulation that impairs myocardial function.6-gingerol inhibits the increase of intracellular Ca2+concentration by inhibiting L-type calcium current,thereby reducing extracellular Ca2+influx,thereby avoiding calcium overload and playing a cardioprotective effect.In summary,6-gingerol can effectively treat and improve myocardial ischemia/reperfusion injury,and it has great development potential in the fields of medicine and health products. 展开更多

Ginger(Zingiber officinale Roscoe)has high economic value as medicinal and food resources.6-gingerol is the core medicinal constituents of ginger.In the present study,a local ginger cultivar of Chongqing was taken as the research material.The ZoWRKY1 gene was cloned to determine its expression level in different ginger developmental phases and to analyze its correlation with 6-gingerol content.The expression level of ZoWRKY1 under different concentrations of NaCl stress was tested,and so was the correlation between ZoWRKY1’s expression level and the contents of 6-gingerol synthase genes,i.e.ZoPAL,ZoC4H and Zo4CL.The results showed that the cDNA of the cloned ZoWRKY1 gene is 1026 bp in total length,and ZoWRKY1 belongs to the second type member of the WRKY family;the expression level of ZoWRKY1 rose sharply in the second developmental phase of the ginger which was about one month after sowing,and there was a significant correlation between the expression level of ZoWRKY1 and the increase of 6-gingerol content;the expressions of ZoWRKY1 and 6-gingerol synthase genes ZoPAL,ZoC4H and Zo4CL had sharp rises under 25 g/L NaCl stress,and the expression level of ZoWRKY1 was closely related to that of ZoC4H or Zo4CL.Therefore,it was speculated that there was a regulatory correlation between ZoWRKY1 and ZoC4H or Zo4CL that can further affect the biosynthesis of 6-gingerol. 展开更多

目的:探讨6-姜酚(6-gingerol,6-G)调控酰基辅酶A合成酶4(ACSL4)/溶质载体7家族成员11,SLC7A11(xCT)/谷胱甘肽过氧化物酶4(GPX4)抑制ApoE^((-/-))动脉粥样硬化(atherosclerosis,AS)小鼠铁死亡的作用及机制。方法:6只普通饲料喂养C57BL/6... 目的:探讨6-姜酚(6-gingerol,6-G)调控酰基辅酶A合成酶4(ACSL4)/溶质载体7家族成员11,SLC7A11(xCT)/谷胱甘肽过氧化物酶4(GPX4)抑制ApoE^((-/-))动脉粥样硬化(atherosclerosis,AS)小鼠铁死亡的作用及机制。方法:6只普通饲料喂养C57BL/6J雄性小鼠为对照(Control,Con)组,按随机数字表法将18只SPF级别的ApoE^((-/-))小鼠分成AS模型(atherosclerosis Model,Mod)组,6-G治疗(6-Gingerol,6-G)组,铁死亡抑制剂(Ferrostatin-1,Fer-1)组,每组6只。Con组喂养普通饲料,除Con组,其余组别均喂养高脂饲料,持续喂养10周,诱导构建AS动物模型。造模成功后,6-G组给予20 mg/kg的6-G灌胃处理,Fer-1组给予1 mg/kg的Fer-1腹腔注射,除Con组仍饲喂普通饲料外,其余组仍继续饲喂高脂饲料,继续喂养6周。全自动生化分析仪检测血脂水平,油红O染色法检测小鼠动脉组织粥样硬化水平,透射电子显微镜观察各组主动脉线粒体超微形态结构改变,试剂盒检测Fe^(2+)水平,ELISA法测定血清还原型谷胱甘肽(GSH)和丙二醛(MDA)的水平。Western blot评估小鼠主动脉ACSL4、xCT和GPX4蛋白表达情况。结果:与Con相比,Mod小鼠主动脉见明显脂质斑块附着,血清TC、TG、LDL-C、Fe^(2+)、MDA水平升高,HDL-C、GSH水平降低(P<0.01),主动脉组织ACSL4蛋白表达明显上升,GPX4、xCT表达明显下降(P<0.01),线粒体结构形态损伤严重。与Mod组相比,6-G组以及Fer-1组小鼠治疗后主动脉脂质斑块沉积减少,血清TC、TG、LDL-C、Fe^(2+)、MDA水平降低,HDL-C、GSH水平升高,差异具有统计学意义(P<0.05),主动脉组织ACSL4蛋白表达降低,GPX4、xCT的蛋白表达量明显升高,差异具有统计学意义(P<0.05),线粒体结构趋近正常。结论:6-姜酚可以减轻动脉粥样硬化水平,其作用机制可能是通过调节ACSL4/xCT/GPX4途径抑制铁死亡和脂质过氧化发挥对主动脉的保护作用。 展开更多

该文建立了生姜药材特征图谱及以单一内标物计算多种成分相对含量的“质-量”双标的质量评价方法。具体方法为:采用超高效液相色谱(UPLC)和四极杆-飞行时间质谱(Q-TOF MS)技术建立了生姜药材特征图谱,标定了其共有特征峰并明确其中的化... 该文建立了生姜药材特征图谱及以单一内标物计算多种成分相对含量的“质-量”双标的质量评价方法。具体方法为:采用超高效液相色谱(UPLC)和四极杆-飞行时间质谱(Q-TOF MS)技术建立了生姜药材特征图谱,标定了其共有特征峰并明确其中的化学成分,通过比对供试品药材和对照药材的相似度,即可明确药材的真伪,即为“质”;对内标成分“6-姜辣素”进行准确的定量,同时通过内标成分计算各特征峰的相对含量,再根据各特征峰的相对含量下限评价生姜药材的优劣,即为“量”。结果表明,通过该方法建立的特征图谱和内标成分含量测定方法均符合方法学要求;经过分析,特征图谱中发现了10个共同的特征峰,并且确定了其中的4种化学成分;此外,不同产地生姜药材与对照药材的相似度均超过了0.900,同时也设定了各特征峰的相对含量下限。该方法不依赖多个对照品且能同时在质和量两方面做到全面、快速、准确地对药材的真伪优劣进行评价,为全面控制生姜药材的质量提供了参考依据。 展开更多

Gold nanostructure can be manufactured in many forms,such as spherical by using simple chemical method.X-ray powder diffraction(XRD)pattern of gold nanoparticles(AuNPs)was derived from a 400-μL trisodium citrate solution.The diffraction peaks corresponding to the diffraction planes of(111),(200),and(220)were all indexed to the gold with a face-centered cubic structure.The lattice constant calculated from the XRD pattern is 4.078Å,which matches the conventional cubic gold metal diffraction pattern well(Pattern card number(04-784)).The results of scanning electronic microscope show that the biological nanoparticles of gold have asymmetric shapes and different sizes grouped as a circular particle.It was observed that the fashioning of AuNPs increased with increase in the concentration of[6]-gingerol waste extracts.The viability of FTC-133 cells treated for 48 h with various concentrations of[6]-gingerol extract coated with AuNPs(12.5-400 g/mL)was used to determine the cytotoxicity.A dose-dependent reduction in FTC-133 cell viability caused by[6]-gingerol extract capped with Au NPs was substantial(P<0.05),reaching 75%cell mortality at 400 g/mL;the IC_(50)was 90.5%. 展开更多

以霍山小黄姜、水口生姜、马厂黄姜、洪里生姜、临泉狮子头、临泉小黄姜、岳西小黄姜、舒城小黄姜、广西红芽姜、金寨小黄姜、铜陵白姜等16份地方生姜资源为试验材料进行品质与产量比较试验。结果表明,岳西小黄姜的姜辣素总和最高(22548... 以霍山小黄姜、水口生姜、马厂黄姜、洪里生姜、临泉狮子头、临泉小黄姜、岳西小黄姜、舒城小黄姜、广西红芽姜、金寨小黄姜、铜陵白姜等16份地方生姜资源为试验材料进行品质与产量比较试验。结果表明,岳西小黄姜的姜辣素总和最高(22548.290μg/g),最低为临泉小黄姜(13694.800μg/g);粗纤维含量最高为西林火姜(46.707 mg/g),最低为玉林园肉生姜(3.437 mg/g);粗蛋白含量最高为玉林园肉生姜(311.863 mg/g),最低为水口生姜(200.917 mg/g);可溶性糖含量最高为临泉狮子头(91.687 mg/g),最低为水口生姜(31.000 mg/g);含水量最高为玉林园肉生姜(932.53 g/kg),最低为铜陵白姜(90.420 mg/g);产量最高是广西红芽姜(108075.0 kg/hm^(2)),最低是水口生姜(74475.0 kg/hm^(2))。岳西小黄姜、马厂黄姜、平顶4号、洪里生姜和临泉狮子头可以作为生产生姜调味品的育种材料。马厂黄姜、平顶4号、水口生姜、金寨小黄姜、石塘姜、玉林园肉生姜和舒城小黄姜适合以嫩姜进行加工。广西红芽姜、临泉狮子头、岳西小黄姜、洪里生姜、平顶4号、马厂黄姜、铜陵白姜和西林火姜更符合产量育种需求。 展开更多

目的:评价生姜、干姜、炮姜姜辣素部位对慢性阻塞性肺疾病(COPD)寒饮蕴肺证的温肺作用,并探讨其作用机制。方法:采用烟熏结合鼻腔滴注脂多糖,饮用冰水加寒冷刺激的方法建立COPD寒饮蕴肺证病证结合大鼠模型。SD大鼠分为正常对照组,模型组... 目的:评价生姜、干姜、炮姜姜辣素部位对慢性阻塞性肺疾病(COPD)寒饮蕴肺证的温肺作用,并探讨其作用机制。方法:采用烟熏结合鼻腔滴注脂多糖,饮用冰水加寒冷刺激的方法建立COPD寒饮蕴肺证病证结合大鼠模型。SD大鼠分为正常对照组,模型组,阳性对照组,生姜高剂量组,生姜低剂量组,干姜高剂量组,干姜低剂量组,炮姜高剂量组,炮姜低剂量组。分别给予生姜、干姜、炮姜姜辣素高、低剂量,阳性药为小青龙颗粒,进行大鼠一般形态观察、肺功能检测,并检测各项指标、观察肺组织和气管病理改变。结果:与模型组相比较,各给药组均可不同程度地改善大鼠上述一般形态;不同显著程度增加大鼠的体质量(P<0.05,P<0.01)、饮水量(P<0.05,P<0.01)、0.2秒用力呼气容积/用力肺活量(FEV_(0.1)/FVC)(P<0.05)、0.3秒用力呼气容积/用力肺活量(FEV_(0.2)/FVC)(P<0.05,P<0.01)和最大呼气流速(PEF)值(P<0.05,P<0.01);各给药组的肺组织炎症细胞浸润面积不同程度缩小,肺泡腔变宽,肺气肿情况得到改善;气管上皮细胞脱落情况、炎症细胞浸润及血管充血情况得到不同程度的改善;显著降低血清中炎症因子的表达,上调肺组织中水通道蛋白-1(AQP1)的表达(P<0.05,P<0.01)、下调黏蛋白5AC(MUC5AC)的表达(P<0.05,P<0.01),不同程度地下调基质蛋白酶-2(MMP-2)(P<0.01)、基质蛋白酶-9(MMP-9)(P<0.05,P<0.01)、基质金属蛋白酶抑制剂-1(TIMP-1)(P<0.05,P<0.01)和基质蛋白酶-12(MMP-12)(P<0.01)的表达,增加气管中超氧化物歧化酶(SOD)(P<0.05)和过氧化氢酶(CAT)(P<0.05,P<0.01)的含量、降低丙二醛(MDA)(P<0.05,P<0.01)的含量。结论:生姜、干姜、炮姜的姜辣素部位是其温肺的功效部位,其作用机制与降低血清中炎性因子的表达、降低肺组织中基质金属蛋白酶的过度表达、阻止气管中氧化应激的发生,从而改善肺组织和气管病理形态,并增加肺组织AQP1的表达、降低MUC5AC的表达密切相关。 展开更多

目的:基于液质联用技术探讨益心饮在正常和房颤大鼠血浆及心脏移行成分的时量特征。方法:连续30 d用酒精液体饲料饲喂大鼠,检测心超确定左心房扩张后,采用连续7 d尾静脉注射药物乙酰胆碱-氯化钙促使大鼠心房电活动紊乱,造成房颤模型。... 目的:基于液质联用技术探讨益心饮在正常和房颤大鼠血浆及心脏移行成分的时量特征。方法:连续30 d用酒精液体饲料饲喂大鼠,检测心超确定左心房扩张后,采用连续7 d尾静脉注射药物乙酰胆碱-氯化钙促使大鼠心房电活动紊乱,造成房颤模型。大鼠随机分为正常组、模型组、正常+益心饮组、模型+益心饮组。益心饮给药剂量为31 g·kg^(-1)·d^(-1),不给药组给予同体积0.9%NaCl溶液。连续灌胃14 d后采集多个时间点的全血,以及终点的心脏组织,采用超高效液相色谱-四极杆-飞行时间质谱(UHPLC-Q-TOF/MS)技术检测各组大鼠血浆、心脏组织的移行成分。结果:与正常组比较,模型组房颤诱发率、房颤易感性、房颤持续时间显著升高(P<0.05),提示造模成功。益心饮体外鉴定得111种化学成分,正常及房颤大鼠血浆、心脏分别均可检测到27种、8种化学成分。时量特征结果表明4-羟基肉桂酸、香兰素、丹酚酸B、芒柄花苷、大豆黄素、毛蕊异黄酮、异甘草素、肉豆蔻醚、洋川芎内酯A、甘草次酸在房颤大鼠血浆中含量较高;甘草苷、毛蕊异黄酮苷、姜辣素等8种成分还可在心脏组织中被检测到。结论:靶向入心成分甘草苷、毛蕊异黄酮苷、姜辣素可能是益心饮治疗房颤的关键药效成分。 展开更多

该研究采用核磁共振氢谱(proton nuclear magnetic resonance,^(1)H NMR)技术对冻干生姜与烘干生姜中的姜辣素和姜黄素类化合物的变化进行定性定量分析,发现姜中姜辣素提取物主要包括6-姜酚、8-姜酚、10-姜酚等姜酚类物质以及少量姜烯... 该研究采用核磁共振氢谱(proton nuclear magnetic resonance,^(1)H NMR)技术对冻干生姜与烘干生姜中的姜辣素和姜黄素类化合物的变化进行定性定量分析,发现姜中姜辣素提取物主要包括6-姜酚、8-姜酚、10-姜酚等姜酚类物质以及少量姜烯酚。经过烘干处理后,总姜酚的含量由(1.5±0.3)mg/g降低至(0.89±0.06)mg/g(鲜重),总姜辣素的含量由(2.3±0.4)mg/g降低至(1.4±0.1)mg/g,6-姜烯酚的含量由(4.8×10^(-3)±2.7×10^(-3))mg/g升高至(0.0209±0.0004)mg/g。烘干后姜黄素类化合物的含量由(4.4±0.2)mg/100 g降低至(1.5±0.6)mg/100 g,香草醛的含量由(1.20±0.07)mg/100 g升高至(1.87±0.01)mg/100 g,说明热处理会导致生姜中的姜辣素与姜黄素类化合物降解,姜酚类物质向姜烯酚转化。该试验结果为研究姜中姜辣素与姜黄素类化合物的变化机理以及姜制品的生产与工艺优化提供了参考。 展开更多

该研究建立了快速测定生姜中6-姜酚、8-姜酚、10-姜酚、6-姜醇的超临界流体色谱-质谱(SFC-MS)方法。使用HSS C_(18)SB(100 mm×2.1 mm,1.8μm)色谱柱,以0.2%(体积分数)甲酸甲醇为改性剂,与超临界流体二氧化碳混合进行梯度洗脱,柱温... 该研究建立了快速测定生姜中6-姜酚、8-姜酚、10-姜酚、6-姜醇的超临界流体色谱-质谱(SFC-MS)方法。使用HSS C_(18)SB(100 mm×2.1 mm,1.8μm)色谱柱,以0.2%(体积分数)甲酸甲醇为改性剂,与超临界流体二氧化碳混合进行梯度洗脱,柱温为40℃,背压为13.79 MPa(2000 psi),进样量为1μL,流速为1 mL/min,采用电喷雾电离源,正离子选择离子监测模式对目标物定性,外标法定量。结果表明,4种化合物在17 min内完成分析,标准曲线的相关系数(r^(2))均大于0.995,检出限为0.05~0.08μg/mL,定量下限为0.5~0.8μg/mL,平均加标回收率为84.1%~105%,相对标准偏差为0.47%~3.5%,方法具有良好的精密度和准确度。采用SFC-MS法对生姜样品进行分析并与高效液相色谱法进行比较,两者定量结果在误差范围内具有可比性。所建方法的显著优点为分析时间缩短一半,可作为生姜中姜酚类物质的高效快速分析方法。 展开更多

道地麻柳姜活性成分与金属含量分析

目的基于温胆汤指纹图谱和指标成分定量测定,结合化学计量学方法分析其共煎传统汤剂(traditional decoction,TD)与单煎混合TD化学成分的差异,为温胆汤等方药科学制备和质量控制提供参考。方法采用HPLC法建立温胆汤10批共煎TD与10批单煎... 目的基于温胆汤指纹图谱和指标成分定量测定,结合化学计量学方法分析其共煎传统汤剂(traditional decoction,TD)与单煎混合TD化学成分的差异,为温胆汤等方药科学制备和质量控制提供参考。方法采用HPLC法建立温胆汤10批共煎TD与10批单煎混合TD的指纹图谱,并评价其相似度;测定温胆汤中腺苷、辛弗林、甘草苷、柚皮苷、橙皮苷、6-姜酚和环磷酸腺苷7种指标成分的含量;采用主成分分析(principal component analysis,PCA)、多维标度分析(multidimensional scaling analysis,MDSA)和正交偏最小二乘法-判别分析(orthogonal partial least squares discriminant analysis,OPLS-DA)对指标成分进行数据分析。结果以10批温胆汤共煎TD的指纹图谱生成共有模式图谱,对照共有模式图谱,共煎TD与单煎混合TD的指纹图谱相似度分别为相似度1:0.897±0.043、0.845±0.069(n=10,P<0.01),相似度2:0.842±0.083、0.759±0.274(n=10,P>0.05);通过指纹图谱及指标成分定量测定发现,温胆汤共煎TD与单煎混合TD在化学成分种类上未见明显差异,含量上温胆汤共煎TD中腺苷、辛弗林、甘草苷、橙皮苷、6-姜酚的含量高于单煎混合TD(P<0.01),分别为单煎混合TD的2.037、1.926、2.505、4.265、1.964倍,柚皮苷、环磷酸腺苷含量与单煎混合TD无显著性差异(P>0.05);PCA、MDSA和OPLS-DA结果一致,可将温胆汤共煎TD与单煎混合TD分为2类。结论所建立的HPLC指纹图谱结合PCA、MDSA和OPLS-DA及多成分定量测定能客观、有效地显示温胆汤共煎TD与单煎混合TD中主要成分的差异,可为温胆汤质量控制和临床合理用药提供参考。 展开更多

目的建立黄芪桂枝五物汤(Huangqi Guizhi Wuwu Decoction,HGWD)基准样品的指纹图谱方法,并建立HGWD基准样品中4个关键指标性成分定量测定的HPLC方法,为HGWD基准样品质量控制提供参考。方法采用HPLC法建立HGWD指纹图谱,通过基准样品与各... 目的建立黄芪桂枝五物汤(Huangqi Guizhi Wuwu Decoction,HGWD)基准样品的指纹图谱方法,并建立HGWD基准样品中4个关键指标性成分定量测定的HPLC方法,为HGWD基准样品质量控制提供参考。方法采用HPLC法建立HGWD指纹图谱,通过基准样品与各单味药材的相关性分析进行共有峰归属,以对照品进行共有峰指认,通过各批次间相似度评价结合化学模式识别分析15批HGWD基准样品的质量差异。同时建立指标性成分毛蕊异黄酮葡萄糖苷、芍药苷、肉桂酸、桂皮醛定量测定方法,结合干膏率及定量测定结果进行量值传递研究。结果建立了15批HGWD基准样品的HPLC指纹图谱,共标定14个共有峰,经对照品比对,确定其中3号峰为芍药内酯苷、4号峰为芍药苷,5号峰为毛蕊异黄酮葡萄糖苷、8号峰为香豆素、11号峰为肉桂酸、12号峰为桂皮醛、14号峰为6-姜辣素;各批次间相似度均≥0.904,可聚为2类,5个主成分累积贡献率达84.205%;毛蕊异黄酮葡萄糖苷、芍药苷、肉桂酸和桂皮醛的质量分数分别为0.503~0.920、16.452~28.520、0.344~0.866、0.842~1.735 mg/g,HGWD饮片-基准样品的转移率分别为23.480%~38.717%、36.468%~53.193%、20.054%~33.694%、2.782%~4.570%。结论采用指纹图谱及多指标性成分定量测定方法为经典名方HGWD的基准样品量值传递进行分析,为HGWD基准样品质量控制和复方制剂的开发提供依据。 展开更多

目的 探究不同煎煮方法与纯化工艺对吴茱萸汤相态以及成分的影响,力争在经典名方开发中还原“一碗汤”的药性。方法 通过文献考证与实验确定吴茱萸汤传统煎煮方法,以《医疗机构中药煎药室管理规范》中的中药汤剂煎煮方法作为现代煎煮方... 目的 探究不同煎煮方法与纯化工艺对吴茱萸汤相态以及成分的影响,力争在经典名方开发中还原“一碗汤”的药性。方法 通过文献考证与实验确定吴茱萸汤传统煎煮方法,以《医疗机构中药煎药室管理规范》中的中药汤剂煎煮方法作为现代煎煮方法,煎煮吴茱萸汤;采用醇沉法、澄清剂法精制2种煎煮方法的吴茱萸汤;采用差速离心结合透析法分离吴茱萸汤中的纳米相态和沉淀相态,对汤剂中纳米相态的形貌、粒径、ζ电位、颗粒浓度及纯度、多糖含量、蛋白浓度等进行测定和比较,测定原汤及2种相态的冻干粉得率,并利用UPLC-UV法测定6-O-反式咖啡酰葡萄糖酸、绿原酸、金丝桃苷、去氢吴茱萸碱、人参皂苷Rg1、人参皂苷Rb1、柠檬苦素、6-姜辣素、吴茱萸碱、吴茱萸次碱10种指标成分含量。结果 2种煎煮方法所得吴茱萸汤中均存在200 nm左右、表面呈现介孔状的球形颗粒(纳米相态);2种纯化工艺减少了纳米相态中的颗粒数量,破坏颗粒结构并降低其蛋白质及多糖含量;现代煎煮吴茱萸汤及其纳米相态冻干粉得率均显著高于传统煎法;现代煎煮吴茱萸汤中除吴茱萸碱、吴茱萸次碱外的其他8种成分均显著高于传统煎法;澄清剂处理后纳米相态中多数成分含量降低或有降低的趋势,醇沉处理后多数成分含量升高或有升高的趋势,各成分分布比例也异于原汤纳米相态。结论 不同的煎煮与纯化方法均会显著改变经典名方吴茱萸汤成分与相态等理化性质,影响药性传递,在经典名方及其他中药复方制剂的研发过程中应予以重视。 展开更多
