A simple and accurate high-performance liquid chromatography(HPLC)method to generate the fingerprints of crude Pu-erh tea(CPT)and ripened Pu-erh tea(RPT)is described.A suitable chromatographic system was establi...A simple and accurate high-performance liquid chromatography(HPLC)method to generate the fingerprints of crude Pu-erh tea(CPT)and ripened Pu-erh tea(RPT)is described.A suitable chromatographic system was established using a linear gradient elution with acetonitrile and water containing 0.6% formic acid as the mobile phase and a detection wavelength of 300 nm.HPLC analysis identified 24 common peaks among RPT and 21 common peaks among CPT.This study revealed that crude Pu-erh tea and ripened Pu-erh tea contained some similar major constituents,but with distinctive peaks,which may be caused by the difference in the fermentation process.This HPLC fingerprint method could be used to evaluate and authenticate crude Pu-erh tea and ripened Pu-erh tea.展开更多

The rhizome of Alpinia officinarum Hance is a well-known traditional Chinese medicine(TCM)and has been widely used for the remedy of gastrointestinal diseases.In the present study,a simple and rapid HPLC-DAD was devel...The rhizome of Alpinia officinarum Hance is a well-known traditional Chinese medicine(TCM)and has been widely used for the remedy of gastrointestinal diseases.In the present study,a simple and rapid HPLC-DAD was developed for the quality control of the rhizomes of A.officinarum.Its chemical fingerprint was established using raw material of 15 different origins in China.Similarity analysis(SA)and hierarchical clusting analysis(HCA)were applied to select the qualitative markers.Principal components analysis(PCA)was conducted to select the quantitative markers of the rhizomes of A.officinarum samples from different origins.The constituents were confirmed by(+)electrospray ionization LC-MS.12 constituents were selected as common peaks and 10 of them were confirmed by(+)electrospray ionization LC-MS.Six bioactive constituents including DPHA,galangin flavanone,galangin,galangin 3-methylether,DPHB and DPHC were simultaneous determination by using the HPLC-DAD analysis.The developed method was able to determine the bioactive components with excellent resolution,precision and recovery.The results indicated that chromatographic fingerprint combination with multi-components determination method is suitable for quality assessment of the rhizomes of A.officinarum.展开更多
To evaluate the quality of Polyporus umbellatus,we established a simple,repeatable and reliable method based on high performance liquid chromatography(HPLC)for specific chromatogram and fingerprint analysis,which was ...To evaluate the quality of Polyporus umbellatus,we established a simple,repeatable and reliable method based on high performance liquid chromatography(HPLC)for specific chromatogram and fingerprint analysis,which was applied to analyze samples of medicinal materials and decoction pieces collected from different regions.Finally,ten characteristic peaks were designated in the specific chromatograms and applied to the authenticate identification of P.umbellatus samples.Nine common peaks were designated in the fingerprints,and then the similarities between 32 batches of samples were calculated.Among them,eight compounds were identified by HPLC-APCI-IT-TOF-MS^(n),four of which were identified in specific chromatograms and four in fingerprints.In the present study,we,for the first time,combined HPLC specific chromatograms and fingerprints for the species identification and quality evaluation of P.umbellatus.Collectively,our findings provided a new method for establishing a comprehensive quality standard of P.umbellatus.展开更多
