Double nano-emulsions for stabilizing Vitamin C and enhancing antioxidant capacity with macadamia oil and tea tree essential oil

Vitamin C,a potent antioxidant with broad therapeutic applications,is limited by rapid degradation under environmental stressors,which compromises its stability and bioactivity.This study addresses these limitations by formulating a double nano-emulsion(W/O/W)system incorporating macadamia oil and tea tree oil,using homogenization and phase inversion temperature(PIT)techniques.Comprehensive physicochemical charac-terization,including droplet size,polydispersity index(PDI),zeta potential,turbidity,Fourier transform infrared spectroscopy(FTIR),and SEM,was conducted alongside stability assessments under varying pH,temperature,and storage conditions.The optimized nano-emulsions exhibited nanoscale droplet sizes(10-40 nm),low PDI values(indicating high uniformity),and robust stability.Interestingly,the formulation with 2%W/O loading,with a particle size of 11.57 nm and a PDI of 0.04,demonstrated an antioxidant capacity of 4622.62μg ascorbic acid equivalents(AA)/g,which was significantly higher(p<0.05)compared to both natural oils(macadamia oil:20.91μg AA/g,tea tree oil:16.86μg AA/g)and a 10%Vitamin C aqueous solution(592.94μg AA/g).FTIR analysis confirmed the molecular integrity of Vitamin C and its successful encapsulation with macadamia and tea tree oils,while SEM images revealed uniformly spherical and well-dispersed droplets.Moreover,the formulation retained its structural integrity and antioxidant functionality under diverse pH and thermal conditions.These findings underscore the potential of double nano-emulsion systems to overcome the stability challenges of Vitamin C,offering a promising approach to enhance its bioavailability and therapeutic performance in phar-maceutical and cosmetic applications. 展开更多

Background: Tea tree oil(TTO) plays an important role in antibacterial activity and alleviating the inflammatory responses. Bovine mammary epithelium and polymorphonuclear leukocytes(PMNL) can actively respond to bovine mastitis infection. However, regulatory effects of TTO extracts on the innate immune response of bovine mammary epithelial cells(BMECs) and PMNL remain not reported. Therefore, aim of the study was to evaluate the effects of TTO extracts on the m RNA levels of the genes involved in the innate immune response of BMECs and PMNL.Results: Our results demonstrated that addition of 0.025% and 0.05% TTO increased the proliferation of BMECs, and significantly enhanced(P < 0.05) the viability of BMECs exposed to Staphylococcus aureus(S. aureus). An inhibitory effect was observed against the growth of S. aureus by TTO incubation. The 0.05% TTO reduced S. aureus biofilm formation, association and invasion of S. aureus to BMECs, and changed the morphological and structural features of S. aureus. The proinflammatory cytokines IL-1β, IL-6, and TNF-α were decreased(P < 0.001) by the incubation of TTO. Interestingly, the expression of IL-8 known for PMNL chemotactic function was elevated(P < 0.05) by 0.05%TTO treatment. Consistently, 0.05% TTO increased the migration of PMNL in S. aureus-exposed BMECs when compared with S. aureus treatment alone(P < 0.05). In addition, PMNL incubated with 0.05% TTO decreased the levels of NFKB inhibitor alpha(NFKBIA) and TNF-α.Conclusions: Our results indicate that use of TTO can relieve the BMECs pro-inflammatory response caused by S.aureus and promote the migration of PMNL to mount the innate immune responses, and it may be novel strategy for the treatment of bovine mastitis caused by S. aureus. 展开更多

Eight primers selected from 40 random primers were used to amplify 45 tea samples, and a total of 95 DNA bands were amplified, of which 90(94.7%)were polymorphism. The average number of DNA bands amplified by each primer was 11.5. Based on the results of UPGMA cluster analysis of 95 DNA bands amplified by 8 primers, all the tested materials could be classified into 7 groups including 5 complex groups and 2 simple groups, which was basically identical with morphological classification. Background:The prevalence of MRSA(Methicillin-resistant Staphylococcus aureus)colonized wounds in home care residents is expected to grow continuously as a result of the substantial proportion of older people requiring institutionalized care due to chronic disease and declining functional status,which contribute to more frequent skin breakdown and wound formation.Tea tree oil has been claimed to have anti-bacterial,analgesic and anti-inflammatory effects that have been suggested in many in-vitro studies to have good efficacy against MRSA.The aims of this study were to evaluate the effectiveness of 10%topical tea tree preparation to eradicateMRSA and to ascertain its influence on wound healing forMRSA-colonized wounds.Methods:It was a randomized controlled trial,single-blind study.Those with stage II or above MRSA-colonized wounds and who had given their informed consent formed the sample.The determined sample sizewasbasedonthe effect size of our previouspilot study,whichwas 0.46.Five outcome measurements were taken for the MRSA bacterial count and wound healing condition at baseline and at 1-week intervals during the 4-week dressing intervention period.Results:Thirty-two participants were recruited from two non-government nursing homes,16 in the control group and 16 in the tea tree oil group.The control group residents received routine saline gauze dressing,while the tea tree oil group residents received the 10%topical tea tree preparation dressing.In the tea tree oil group,all chronic wounds that had previously been delayed in healing were healed within 28 days without adverse reaction.MRSA was also completely eradicated in 14(87.5%)out of 16 wounds in the group receiving the 10%topical tea tree preparation. 展开更多

Over the past few decades,complementary and alternative treatments have become increasingly popular worldwide.The purported therapeutic characteristics of natural products have come under increased scrutiny both in vitro and in vivo as part of efforts to legitimize their usage.One such product is tea tree oil(TTO),a volatile essential oil primarily obtained from the native Australian plant,Melaleuca alternifolia,which has diverse traditional and industrial applications such as topical preparations for the treatment of skin infections.Its anti-inflammatory-linked immunomodulatory actions have also been reported.This systematic review focuses on the anti-inflammatory effects of TTO and its main components that have shown strong immunomodulatory potential.An extensive literature search was performed electronically for data curation on worldwide accepted scientific databases,such as Web of Science,Google Scholar,PubMed,ScienceDirect,Scopus,and esteemed publishers such as Elsevier,Springer,Frontiers,and Taylor&Francis.Considering that the majority of pharmacological studies were conducted on crude oils only,the extracted data were critically analyzed to gain further insight into the prospects of TTO being used as a neuroprotective agent by drug formulation or dietary supplement.In addition,the active constituents contributing to the activity of TTO have not been well justified,and the core mechanisms need to be unveiled especially for anti-inflammatory and immunomodulatory effects leading to neuroprotection.Therefore,this review attempts to correlate the anti-inflammatory and immunomodulatory activity of TTO with its neuroprotective mechanisms. 展开更多

[ Result] Tea tree cultivation in Hanzhong was divided into suitable, more suitable and unsuitable planting zones by using critical value of climatic zoning. Tea is the world's oldest and most popular caffeine-containing beverage with immense economic, medicinal, and cultural importance. Here, we present the first high-quality nucleotide sequence of the repeat-rich (80.9%), 3.02-Gb genome of the cultivated tea tree Camellia sinensis. We show that an extraordinarily large genome size of tea tree is resulted from the slow, steady, and long-term amplification of a few LTR retrotransposon families. In addition to a recent whole-genome duplication event, lineage-specific expansions of genes associated with flavonoid metabolic biosynthesis were discovered, which enhance catechin production, terpene enzyme activation, and stress tolerance, important features for tea flavor and adaptation. We demonstrate an independent and rapid evolution of the tea caffeine synthesis pathway relative to cacao and coffee. A comparative study among 25 Camellia species revealed that higher expression levels of most flavonoid- and caffeinebut not theanine-related genes contribute to the increased production of catechins and caffeine and thus enhance tea-processing suitability and tea quality. These novel findings pave the way for further metabolomic and functional genomic refinement of characteristic biosynthesis pathways and will help develop a more diversified set of tea flavors that would eventually satisfy and attract more tea drinkers worldwide. 展开更多

This study aimed to prepare tea tree oil-β-cyclodextrin microcapsules using an optimized co-precipitated method.The impact of the volume fraction of ethanol in the solvent system for microencapsulation on encapsulation efficiency was investigated and analyzed sophisticatedly.Super-high encapsulation efficiency was achieved when a 40%volume fraction of ethanol was used for the microencapsulation procedure,where the recovery yield of microcapsules and the embedding fraction of tea tree oil in microcapsules were as high as 88.3%and 94.3%,respec-tively.Additionally,considering the operation cost,including time and energy consumption,an economical preparation was validated so that it would be viable for large-scale production.Based on the results of morphological and X-ray diffraction analysis,the crystal structure appeared to differ before and after microencapsulation.The results of gas chromatography-mass spectrometry and Fourier transform infrared spectroscopy confirmed the successful formation of microcapsules.Furthermore,the antibacterial activity of the fabricated microcapsules was assessed by a simple growth inhibition test using Bacillus subtilis as the study object,and the hydrophilic property was proved by a water contact angle measurement. 展开更多

To determine the age of oil-tea camellia trees, regression equations including Logistic, Mitscherlich, Gompertz, Korf, and Richards were used to calculate accumulative growth rate using basal trunk disc and investigate the relations between the age of oil-tea camellia trees and their growth rate of secondary trunk. The Gompertz equation Y=71.296 1exp (-3.874 4exp (-0.006 4t)) was the most optimal equation to simulate the accumulative growth rate of basal trunk disc. This equation could be used to estimate the age of oil-tea camellia trees that grow under similar environmental conditions. The Korf equation Y=576.900 1exp (-4.153 0x -0.314 2 ) was the best equation to describe the relation between the age and growth rate of different secondary trunks. With the adjustment coefficient and average growth of different secondary trunk discs, it is possible to predict the age of ancient oil-tea camellia trees that grow under similar environmental conditions. In addition, taking three or more discs from the same diameter group and calculating their average growth rate could lead to more accurate results. For trees that grow in different areas, environmental conditions should be carefully considered when using the above two equations to predict the age of ancient oil-tea camellia trees. 展开更多

[Method] In an ecological adult-tea garden, teas shaded by Prunus cerasoides, Prunus L., and Litsea pungens were selected and the teas without shades were taken as a control in order to explore effects of tree shading on photosynthesis, respiration and net photosynthetic intensities. [Result] In a growth cycle of one year, for teas shaded by Prunus cerasoides, Prunus L., and Litsea pungens, respiration intensity was significantly higher than that of the control; net photosynthetic intensity was extremely significant higher; photosynthesis intensity showed none rules. Both of net photosynthetic rate and intensity kept higher in winter of shaded teas. [Conclusion] It is of significance for high-yielding and high-quality teas to reduce respiration consumption and coordinate between photosynthesis and respiration given that tea grows well. 展开更多

为探究不同间作模式对茶园土壤及茶树[Camellia sinenesis(L.)O.Kuntze]木质部解剖结构的影响,本研究在云南省大理州南涧县无量山樱花谷风景区选择三种不同的茶园间作模式即马缨花(Rhododendron delavayi Franch.)-茶(MT)、冬樱花[Prunu... 为探究不同间作模式对茶园土壤及茶树[Camellia sinenesis(L.)O.Kuntze]木质部解剖结构的影响,本研究在云南省大理州南涧县无量山樱花谷风景区选择三种不同的茶园间作模式即马缨花(Rhododendron delavayi Franch.)-茶(MT)、冬樱花[Prunus cerasoides(D.Don)Sok.]-茶(DT)、核桃(Juglans mandshurica Maxim.)-茶(HT)与纯茶园(CK)进行比较,分析春、秋两季四种茶园土壤酶活性、土壤养分含量及茶树木质部解剖结构差异等。结果表明:与CK相比,春、秋季MT、DT茶园土壤酶活性总体提升效果较好,而HT茶园土壤酶活性提升效果较差;秋季,DT、MT、HT茶园总体上都能提升土壤有机质、全磷和全钾含量,改善茶园土壤理化性状;DT、HT茶园较CK显著提高茶树木质部导管长度;春季,HT茶园茶树木质部纤维长度、纤维宽度、双壁厚度和胞腔宽度等指标值均明显高于其他三种模式茶树;秋季,MT茶园茶树木质部纤维各指标值均为最大,其中纤维长度、双璧厚度与其他茶园差异达显著水平。综上表明,茶园中间作其他树种能够在一定程度上改善茶园土壤理化性状,提升茶树对水分和养分的利用效率,因此在茶园建设时可采用适宜的间作模式。 展开更多

[ Method ] Theobroma cacao caffeine syn- thase gene BCS1 was used as a probe to search the established tea EST database using BLAST; 26 tea ESTs highly homologous to BCS1 were obtained, which were assembled using CAP (contig assembly program) of BioEdit software; subsequently, two EST configs harboring ORF were obtained, which were named TCSnewl and TCSnew2, respectively. Nucleotide sequences and deduced amino acid sequences of theses two genes were compared with those of cDNA of tea caffeine synthase gene TCS in the GenBank database that was cloned with experimental biological method. A phylogenetic tree was constructed for homalogous analysis of the deduced amino acid sequences of theses three genes. [ Result] in silico cloning of functional candidate genes in tea using a homologous gene of distantly related species as a probe is a feasible technical means. 为探究蚓粪对土壤重金属Cu、Pb化学形态及茶树对土壤中Cu、Pb吸收转运的影响,在有茶树种植的Cu、Pb污染土壤系统中施入蚓粪,并以牛粪和化肥为对照,系统研究在蚓粪和茶树根系共同作用下土壤中Cu、Pb的化学形态变化以及茶树对Cu、Pb的吸... 为探究蚓粪对土壤重金属Cu、Pb化学形态及茶树对土壤中Cu、Pb吸收转运的影响,在有茶树种植的Cu、Pb污染土壤系统中施入蚓粪,并以牛粪和化肥为对照,系统研究在蚓粪和茶树根系共同作用下土壤中Cu、Pb的化学形态变化以及茶树对Cu、Pb的吸收转运情况。结果表明,在土壤中施用蚓粪可显著降低交换性氢和交换性铝的含量,增加水提和盐提pH值,有效缓解土壤酸化程度,并可显著增加有机碳、全氮以及氮、磷、钾等速效养分。在有茶树根系作用的土壤系统中,蚓粪处理组Cu、Pb的水溶-交换态含量较对照处理组分别降低66.88%和57.79%,蚓粪降低土壤水溶-交换态Cu、Pb含量的作用效果优于牛粪和化肥处理组。蚓粪处理组茶树根部和叶部Cu的含量比CK组分别下降36.61%和23.08%。可见,在种植茶树的土壤中施用蚓粪可显著降低土壤酸化程度,增加有机碳含量,这有利于降低土壤中Cu、Pb的生物有效性。本研究结果对有Cu、Pb重金属轻微污染风险的茶园土壤治理具有一定借鉴意义。 展开更多

基于物候期气候因素的茶叶产量模型构建与种植管理 被引量:1

本文聚焦于茶叶产量与符合茶树生长节律的物候期气候因素之间的复杂关系,采用茶树物候期气候指标作为因子,探索茶叶产量模型的构建,并基于模型主要气候因子制定具有区域气候适应性的茶树种植管理策略。基于1990—2022年福鼎市气象观测... 本文聚焦于茶叶产量与符合茶树生长节律的物候期气候因素之间的复杂关系,采用茶树物候期气候指标作为因子,探索茶叶产量模型的构建,并基于模型主要气候因子制定具有区域气候适应性的茶树种植管理策略。基于1990—2022年福鼎市气象观测资料和茶叶产量资料,采用线性趋势法研究气候变化趋势,运用多元线性回归法、主成分回归法及神经网络法3种方法构建福鼎市茶叶产量模型。研究结果表明:1990—2022年福鼎市年平均温度以0.436℃·(10a)^(-1)的速率快速提升,年日照时数与年降水量在不规律波动中分别以4.078 h·(10a)^(-1)和31.105 mm·(10a)^(-1)的速率提高,气候变化影响茶叶产量的稳定性;从不同茶树物候期气候指标形成的168个物候期气候要素中,筛选出14个关键气候因子,基于这些关键因子构建的神经网络产量模型的均方根误差(RMSE)为7.5956,归一化均方根误差(NRMSE)为0.0807,决定系数(R^(2))为0.9351,平均拟合准确率(P)为93.13%,该模型为最优产量模型。最后,围绕最优模型中全物候期降水日数、春茶采摘期平均气压和第2次生长期≥10℃积温3个较高重要性水平因子,新梢萌芽期蒸发量、休眠期蒸发量、休眠期平均风速、第2次生长期平均风速和第2次生长期平均日最低气温5个中等重要性水平因子,结合当地气候变暖趋势,进行分析探讨,提出优化水分、土壤保墒、通风降湿、气象灾害防治和分散气候风险等契合福鼎市气候特征的茶树种植策略。 展开更多

采用尖端菌丝纯化法,从不除草、人工耕作除草和防草膜覆盖3种不同方式的茶树根系中分离内生菌株,探讨了不同除草方式对茶树根系内生菌的种类多样性及其对病原菌的拮抗活性,旨在为茶树病害的生物防治提供理论依据。通过五点对峙培养法,... 采用尖端菌丝纯化法,从不除草、人工耕作除草和防草膜覆盖3种不同方式的茶树根系中分离内生菌株,探讨了不同除草方式对茶树根系内生菌的种类多样性及其对病原菌的拮抗活性,旨在为茶树病害的生物防治提供理论依据。通过五点对峙培养法,筛选和检测内生菌株对5种常见病原真菌的拮抗活性,并结合分子生物学方法对具有较强活性的菌株进行鉴定与分类。在分离的菌株中,共获得24株内生真菌和13株内生细菌,其中11株内生真菌和5株内生细菌对病原菌表现出较强的拮抗活性。尤其是在不除草处理中分离的Aspergillus piperis和Aspergillus brunneoviolaceus,人工耕作除草处理中分离的Achromobacter xylosoxidans,以及防草膜覆盖处理中分离的Pseudomonas tolaasii,对茶树轮斑病(Pseudopestalotiopsis camelliae-sinensis)的抑制率均超过80%,为强拮抗菌株。防草膜覆盖处理中分离的Aspergillus aculeatus对尖孢镰刀菌(Fusarium oxysporum)病原菌的抑制率也达75%以上,表现出较强的拮抗效果。此外,内生细菌如Achromobacter spanius和Pseudomonas tolaasii同时对卷枝毛霉菌(Mucor circinelloides)、茶树轮斑病、茶树炭疽病(Colletotrichum camelliae)和白地霉(Geotrichum candidum)中的3种供试病原菌菌株的抑制率均大于50%。在人工耕作除草处理中,分离的Aspergillus flavus、Purpureocillium lilacinum和Fusarium oxysporum对茶树轮斑病的抑制率在50%到66%之间,而内生细菌的拮抗活性普遍低于50%。不除草处理中的Aspergillus piperis对茶树轮斑病、尖孢镰刀菌和白地霉的抑制率均超过50%,Aspergillus brunneoviolaceus对所有5种病原菌的抑制率也均大于50%。总体来看,3种除草方式均分离出对茶树轮斑病具有强拮抗活性的内生菌株,其中防草膜覆盖方式在防治多种病害,特别是土传性病害如尖孢镰刀菌方面表现出较好的防治效果。Pseudomonas tolaasii和Aspergillus brunneoviolaceus分别在防草膜覆盖和不除草处理中表现出强拮抗活性。 展开更多

该研究探讨了茶树精油对单增李斯特菌的抑制效果及其在冷鲜猪肉保鲜中的应用。测定了茶树精油对单增李斯特菌的最小抑菌浓度(Minimal Inhibitory Concentration, MIC),观察了细菌的生长曲线以及细胞微观结构的变化,并分析了茶树精油处... 该研究探讨了茶树精油对单增李斯特菌的抑制效果及其在冷鲜猪肉保鲜中的应用。测定了茶树精油对单增李斯特菌的最小抑菌浓度(Minimal Inhibitory Concentration, MIC),观察了细菌的生长曲线以及细胞微观结构的变化,并分析了茶树精油处理的猪肉在冷藏期间其菌落总数、总挥发性盐基氮含量、硫代巴比妥酸反应物值、pH值、颜色和质构的变化情况。结果表明:茶树精油对单增李斯特菌的MIC为4.0 mg/mL,且不同茶树精油处理组均对单增李斯特菌具有明显的抑制效果,茶树精油破坏了单增李斯特菌细胞膜和细胞壁结构的完整性,引发细胞内容物外渗,从而导致菌体死亡。此外,在贮藏期间(0~12 d),MIC、2MIC和4MIC茶树精油处理组的菌落总数与对照组相比分别下降了1.18、1.37和1.56 log CFU/g,且TVB-N值和TBARS值均未超过最大阈值,茶树精油有效抑制了猪肉中微生物的生长,延缓了猪肉含氮化合物降解、脂肪氧化、色度和品质的变化。结果表明,茶树精油可抑制单增李斯特菌的生长,并延长冷鲜猪肉的货架期3 d左右,可作为冷鲜猪肉的保鲜剂。 展开更多
