[Objectives]This study was conducted to detect and analyze tomato leaf curl New Delhi virus(ToLCNDV).[Methods]Through PCR detection,sequence analysis,and pathogenicity verification,tomato leaf curl New Delhi virus(ToL...[Objectives]This study was conducted to detect and analyze tomato leaf curl New Delhi virus(ToLCNDV).[Methods]Through PCR detection,sequence analysis,and pathogenicity verification,tomato leaf curl New Delhi virus(ToLCNDV)was identified in zucchini exhibiting systemic disease symptoms during a 2024 outbreak in Qingzhou City,Shandong Province,and was designated as ToLCNDV-SD.[Results]Specific primer amplification showed that all eight diseased samples produced bands of 504 bp(DNA-A)and 892 bp(DNA-B).Sequencing analysis revealed that ToLCNDV-SD DNA-A shared 96.10%homology with an Indonesian melon isolate(LC421834.1),while DNA-B showed 88.31%homology with a Malaysian bitter gourd isolate(MW248678.1).Phylogenetic analysis indicated its closest relationship with Southeast Asian cucurbit-infecting isolates.Friction transmission tests confirmed that the virus could spread mechanically,inducing typical symptoms 14 d after inoculation with positive PCR detection.[Conclusions]This study provides important insights for understanding the epidemic mechanisms and control strategies of ToLCNDV in China.展开更多

However, this action faces immense challenges due to the lack of early postearthquake data and the unreliability of forecasts. To obtain foundational data for sequence parameters of the land-sea adjacent zone and establish a reliable and operational aftershock forecasting framework, we combined the initial sequence parameters extracted from envelope functions and incorporated small-earthquake information into our model to construct a Bayesian algorithm for the early postearthquake stage. We performed parameter fitting and early postearthquake aftershock occurrence rate forecasting and effectiveness evaluation for 36 earthquake sequences with M ≥ 4.0 in the Bohai Rim region since 2010. According to the results, during the early stage after the mainshock, earthquake sequence parameters exhibited relatively drastic fl uctuations with signifi cant errors. The integration of prior information can mitigate the intensity of these changes and reduce errors. The initial and stable sequence parameters generally display advantageous distribution characteristics, with each parameter’s distribution being relatively concentrated and showing good symmetry and remarkable consistency. The sequence parameter p-values were relatively small, which indicates the comparatively slow attenuation of signifi cant earthquake events in the Bohai Rim region. A certain positive correlation was observed between earthquake sequence parameters b and p. However, sequence parameters are unrelated to the mainshock magnitude, which implies that their statistical characteristics and trends are universal. The Bayesian algorithm revealed a good forecasting capability for aftershocks in the early postearthquake period (2 h) in the Bohai Rim region, with an overall forecasting effi cacy rate of 76.39%. The proportion of “too low” failures exceeded that of “too high” failures, and the number of forecasting failures for the next three days was greater than that for the next day.展开更多
Pyrola atropurpurea Franch is an important annual herbaceous plant.Few genomic analyses have been conducted on this plant,and chloroplast genome research will enrich its genomics basis.This study is based on high-thro...Pyrola atropurpurea Franch is an important annual herbaceous plant.Few genomic analyses have been conducted on this plant,and chloroplast genome research will enrich its genomics basis.This study is based on high-throughput sequencing technology and Bioinformatics methods to obtain the sequence,structure,and other characteristics of the P.atropurpurea chloroplast genome.The result showed that the chloroplast genome of P.atropurpurea has a double-stranded circular structure with a total length of 172,535 bp and a typical four-segment structure.The genome has annotated a total of 132 functional genes,including 43 tRNAs,8 rRNAs,76 protein-coding genes,and 5 pseudo-genes.In total,358 SSR loci were checked out,mainly composed of mononucleotide and trinucleotide repeat.There are three types of scattered repetitive sequences,totaling 4223,including 2452 forward repeats,1763 palindrome repeats,and eight reverse repeats.The optimal codon usage frequency is relatively high with AT usage preference in this genome.Chloroplast genome comparative analysis in the family Ericaceae shows that the overall sequence is more complex,and there are more variations in the gene interval region.The collinearity analysis indicated that there is a complex rearrangement of species between different genera in Ericaceae.The selection pressure analysis showed that the protein-encoding genes rpl33 and rps16 were positively selected among the seven medicinal plants in Ericaceae.The maximum likelihood tree shows that the genetic relationship among P.atropurpurea,Pyrola rotundifolia,and Chimaphila japonica is relatively close.Therefore,an important data basis was provided for species identification,genetic diversity,and phylogenetic studies of P.atropurpurea and even this genus of plants.展开更多

The parasite is transmitted cyclically between canines and numerous intermediate herbivorous livestock animals. Also, other Taeniid tapeworms could infect domestic dogs and they pose significant veterinary and public health concerns worldwide. This study aimed to develop a sensitive molecular method for detecting Echinococcus spp. DNA in dog fecal samples using next-generation sequencing (NGS). A set of PCR primers targeting conserved regions of Taeniid tapeworms’ 18s rRNA genes was designed and tested for amplifying genomic DNA from various tapeworm species. The PCR system demonstrated high sensitivity, amplifying DNA from all tested tapeworm species, with differences observed in amplified band sizes. The primers were adapted for NGS analysis by adding forward and reverse adapters, enabling the sequencing of amplified DNA fragments. Application of the developed PCR system to dog fecal samples collected from Yatta town, Palestine, revealed the presence of E. granulosus DNA in five out of 50 samples. NGS analysis confirmed the specificity of the amplified DNA fragments, showing 98% - 99% similarity with the 18s rDNA gene of E. granulosus. This study demonstrates the utility of NGS-based molecular methods for accurate and sensitive detection of Echinococcus spp. in dog fecal samples, providing valuable insights for epidemiological surveillance and control programs of echinococcosis in endemic regions.展开更多
Sweet potatoes are significant cash crops,however,their yield and quality are greatly compromised by viral diseases.In this study,the complete genomic sequences of two Sweet Potato Virus 2(SPV2)isolates from infected ...Sweet potatoes are significant cash crops,however,their yield and quality are greatly compromised by viral diseases.In this study,the complete genomic sequences of two Sweet Potato Virus 2(SPV2)isolates from infected sweet potato leaves in the Shandong(designated as SPV2-SDYT,GenBank No.PQ855660.1)and Jiangsu(designated as SPV2-JSXZ,GenBank No.PQ855661.1)provinces in China were obtained using 5′RACE and RT-PCR amplification.Consistency,phylogeny,codon usage bias,recombination,and selection pressure analyses were conducted using the SPV2-SDYT and SPV2-JSXZ genome sequences.The complete genome sequences of SPV2-SDYT and SPV2-JSXZ were 10561 nucleotides(nt)in length,with respective nucleotide and amino acid identities of 99.25%and 99.12%,respectively.Both isolates were closely related to the SPV2 isolate from China(SPV2-LN).In both SPV2-SDYT and SPV2-JSXZ,the identity of the P1 protein was the highest,whereas that of the P3 protein was the lowest.There were 26 codons with relatively synonymous codon usage(RSCU)values greater than 1 in SPV2-SDYT and 27 codons with RSCU values greater than 1 in SPV2-JSXZ.High-frequency codons in their genomes were predominantly found to end with A/U.Recombination analysis revealed no major recombination sites in either SPV2-SDYT or SPV2-JSXZ.Further selection pressure analysis showed that the non-synonymous substitution rate/synonymous substitution rate(dN/dS)value of all 10 SPV2 proteins was less than 1.This is the first report on the evolutionary relationships of the 17 known SPV2 isolates.Our findings lay the molecular groundwork for preventing and controlling SPV2 infection in root-tuber crops.These findings also contribute to our understanding of the spread and evolution of SPV2,its pathogenic mechanisms,and the development of antiviral strategies against it.展开更多
Machine learning(ML)efficiently and accurately processes dense seismic array data,improving earthquake catalog creation,which is crucial for understanding earthquake sequences and fault systems;analyzing its reliabili...Machine learning(ML)efficiently and accurately processes dense seismic array data,improving earthquake catalog creation,which is crucial for understanding earthquake sequences and fault systems;analyzing its reliability is also essential.An M5.8 earthquake struck Alxa Left Banner,Inner Mongolia,China on April 15,2015,a region with limited CENC monitoring capabilities,making analysis challenging.However,abundant data from ChinArray provided valuable observations for assessing the event.This study leveraged ChinArray data from the 2015 Alxa Left Banner earthquake sequence,employing machine learning(specifically PhaseNet,a deep learning method,and GaMMA,a Bayesian approach)for automated seismic phase picking,association,and location analysis.Our generated catalog,comprising 10,432 phases from 708 events,is roughly ten times larger than the CENC catalog,encompassing all CENC events with strong consistency.A slight magnitude overestimation is observed only at lower magnitudes.Furthermore,the catalog adheres to the Gutenberg-Richter and Omori laws spatially,temporally,and in magnitude distribution,demonstrating its high reliability.Double-difference tomography refined locations for 366 events,yielding a more compact spatial distribution with horizontal errors within 100m,vertical errors within 300m,and travel-time residuals within 0.05s.Depths predominantly range from 10-30km.Aftershocks align primarily NEE,with the mainshock east of the aftershock zone.The near-vertical main fault plane dips northwestward,exhibiting a Y-shaped branching structure,converging at depth and expanding towards the surface.FOCMEC analysis,using first motion and amplitude ratios,yielded focal mechanism solutions for 10 events,including the mainshock.These solutions consistently indicate a strike-slip mechanism with a minor extensional component.Integrating the earthquake sequence's spatial distribution and focal mechanisms suggests the seismogenic structure is a negative flower structure,consistent with the Dengkou-Benjing fault.Comparing the CENC and ML-generated catalogs using the maximum curvature(MAXC)method reveals a 0.6 decrease in completeness magnitude(M_(C)).However,magnitude-frequency distribution discrepancies above the MAXC-estimated M_(C)suggest MAXC may underestimate both M_(C)and the b-value.This study analyzes the 2015 Alxa Left Banner M5.8 earthquake using a reliable,MLgenerated earthquake catalog,revealing detailed information about the sequence,faulting structure,aftershock distribution,and stress characteristics.展开更多
By integrating core observations,logging data and seismic interpretation,this study takes the massive Cretaceous carbonates in the M block of the Santos Basin,Brazil,as an example to establish the sequence filling pat...By integrating core observations,logging data and seismic interpretation,this study takes the massive Cretaceous carbonates in the M block of the Santos Basin,Brazil,as an example to establish the sequence filling pattern of fault-bounded isolated platforms in rift lake basins,reveal the control mechanisms of shoal-body development and reservoir formation,and reconstruct the evolutionary history of lithofacies paleogeography.The following results are obtained.(1)Three tertiary sequences(SQ1-SQ3)are identified in the Lower Cretaceous Itapema-Barra Velha of the M block.During the depositional period of SQ1,the rift basement faults controlled the stratigraphic distribution pattern of thick on both sides and thin in the middle.The strata overlapped to uplift in the early stage.During the depositional period of SQ2-SQ3,the synsedimentary faults controlled the paleogeomorphic reworking process with subsidence in the northwest and uplifting in the northeast,accompanied with the relative fall of lake level.(2)The Lower Cretaceous in the M block was deposited in a littoral-shallow lake,with the lithofacies paleogeographic pattern transiting from the inner clastic shoals and outer shelly shoals in SQ1 to the alternation of mounds and shoals in SQ2-SQ3.(3)Under the joint control of relative lake-level fluctuation,synsedimentary faults and volcanic activity,the shelly shoals in SQ1 tend to accumulated vertically in the raised area,and the mound-shoal complex in SQ2-SQ3 tends to migrate laterally towards the slope-break belt due to the reduction of accommodation space.(4)The evolution pattern of high-energy mounds and shoals,which were vertically accumulated in the early stage and laterally migrated in the later stage,controlled the transformation of high-quality reservoirs from“centralized”to“ring shaped”distribution.The research findings clarify the sedimentary patterns of mounds and shoals and the distribution of favorable reservoirs in the fault-controlled lacustrine isolated platform,providing support for the deepwater hydrocarbon exploration in the subsalt carbonate rocks in the Santos Basin.展开更多

To ad...Wavelet transformation is a widely used method in high-frequency sequence stratigraphic analysis.However, the application is problematic since different wavelets always return the same sequence analysis results. To address this issue, we applied five commonly used wavelets to theoretical sequence models to document some application criteria. Five gradual scale-change sequence models were simplified from the glutenite succession deposition by gravity flows to form the fining-upwards cycle sequences(FUCS) and coarsening-upwards cycle sequences(CUCS). After conducting theoretical sequence model tests, the optimal wavelet(sym4) was selected and successfully used with actual data to identify the sequence boundaries. We also proposed a new method to optimize the scale of continuous wavelet transformation(CWT) for sequence boundary determination. We found that the balloon-like marks in scalograms of db4, sym4, and coif4 wavelet determine, respectively, the fourth-order sequence boundary, the thick succession sequence boundaries in FUCS, and the thick succession sequence in FUCS and CUCS. Comparing the sequence identification results shows that the asymmetric wavelets had an advantage in high-frequency sequence boundary determination and sedimentary cycle discrimination through the amplitude trend of the coefficient, in which the sym4 wavelet is the most effective. In conclusion, the asymmetry of wavelets is the first selection principle, of which asymmetric wavelets are more sensitive to sediment deposition by flood flows. The match of the wavelet between the sequence is the second selection principle, in which the correlation of time-frequency impacts the accuracy of sequence surface localization. However, the waveform of the wavelet is a visual and abstract parameter for sequence boundary detection. The appropriate wavelet for lacustrine sequence analysis is the asymmetric wavelet with a weak number of side lobes. The depositional flows, depositional process,and autogenic are three sedimentary factors that influence the sequence analysis results.展开更多
Matrix-assisted laser desorption/ionization(MALDI)mass spectrometry(MS)plays an indispensable role in analyzing protein covalent structures.The reliable identification of amino acid residues and modifications relies o...Matrix-assisted laser desorption/ionization(MALDI)mass spectrometry(MS)plays an indispensable role in analyzing protein covalent structures.The reliable identification of amino acid residues and modifications relies on the mass accuracy,which is highly dependent on calibration.However,the accuracy provided by the currently available calibrants still needs further improvement in terms of compatibility with multiple tandem MS modes or ion polarity modes,calibratable range,and minimizing suppression of and interference with analyte signals.Here aiming at developing a versatile calibrant to solve these problem,we designed a synthetic peptide format of calibrant R_x(GDP_n)_m(referred to as“Gly-Asp-Pro,GDP”)according to the chemical natures of amino acids and polypeptide fragmentation rules in tandem MS.With four types of amino acid residues selected and arranged through rational designs,a GDP peptide produces highly regulated fragments that give rise to evenly spaced signals in each tandem MS mode and is compatible with both positive and negative ion modes.In internal calibration,its regulated fragmentation pattern minimizes interference with analyte signals,and using a single peptide as the input minimizes suppression of the analyte signals.As demonstrated by analyses of proteins including monoclonal antibody and Aβ-42,these features allowed significant increase of the mass accuracy and precision,which improved sequence coverage and sequence resolution in sequence analyses(including de novo sequencing).This rational design strategy may also inspire further development of synthetic calibrants that benefit structural analysis of biomolecules.展开更多
Objective:To address the phylogenetic and phylogeographic relationship between different lineages of Anopheles(An.)subpictus species complex in most parts of the Asian continent by maximum utilization of Internal Tran...Objective:To address the phylogenetic and phylogeographic relationship between different lineages of Anopheles(An.)subpictus species complex in most parts of the Asian continent by maximum utilization of Internal Transcriber Spacer 2(ITS2)and cytochrome C oxidase I(COI)sequences deposited at the GenBank.Methods:Seventy-five ITS2,210 COI and 26 concatenated sequences available in the NCBI database were used.Phylogenetic analysis was performed using Bayesian likelihood trees,whereas median-joining haplotype networks and time-scale divergence trees were generated for phylogeographic analysis.Genetic diversity indices and genetic differentiation were also calculated.Results:Two genetically divergent molecular forms of An.subpictus species complex corresponding to sibling species A and B are established.Species A evolved around 37-82 million years ago in Sri Lanka,India,and the Netherlands,and species B evolved around 22-79 million years ago in Sri Lanka,India,and Myanmar.Vietnam,Thailand,and Cambodia have two molecular forms:one is phylogenetically similar to species B.Other forms differ from species A and B and evolved recently in the above mentioned countries,Indonesia and the Philippines.Genetic subdivision among Sri Lanka,India,and the Netherlands is almost absent.A substantial genetic differentiation was obtained for some populations due to isolation by large geographical distances.Genetic diversity indices reveal the presence of a long-established stable mosquito population,at mutation-drift equilibrium,regardless of population fluctuations.Conclusions:An.subpictus species complex consists of more than two genetically divergent molecular forms.Species A is highly divergent from the rest.Sri Lanka and India contain only species A and B.展开更多
The present study is devoted to understanding the evolution of the Upper Jurassic Sab'atayn Formation in the Marib-Shabwa Basin,Yemen,through a sequence stratigraphic analysis based on integrating datasets of sedi...The present study is devoted to understanding the evolution of the Upper Jurassic Sab'atayn Formation in the Marib-Shabwa Basin,Yemen,through a sequence stratigraphic analysis based on integrating datasets of sedimentology,seismic sections,and well logs.The Sab'atayn Formation(Tithonian age)is represented by a series of clastic and evaporites that were deposited under fluvio-deltaic to prodeltaic settings.It is divided into four members including Yah(at the base),upwards to Seen,Alif,and Safir at the top.Two third-order depositional sequences were determined for the Tithonian succession which were separated by three sequence boundaries.These sequences were classified into their systems tracts signifying several sedimentation patterns of progradational,aggradational,and retrogradational parasequence sets.The first depositional sequence corresponds to the early-middle Tithonian Yah and Seen units that can be classified into lowstand,transgressive,and highstand systems tracts.The second sequence comprises the late Tithonian Alif unit that can be subdivided into transgressive and highstand systems tracts.The sandy deposits of the Alif Member(highstand deposits)represent the most productive hydrocarbon reservoir in the basin.The Upper Jurassic sediments in the study area were resulted from a combination of eustatic and tectonic effects.展开更多

Until recently, the functions of border-associated macrophages have been poorly understood and largely overlooked. However, a recent study reported that border-associated macrophages participate in stroke-induced inflammation, although many details and the underlying mechanisms remain unclear. In this study, we performed a comprehensive single-cell analysis of mouse border-associated macrophages using sequencing data obtained from the Gene Expression Omnibus(GEO) database(GSE174574 and GSE225948). Differentially expressed genes were identified, and enrichment analysis was performed to identify the transcription profile of border-associated macrophages. CellChat analysis was conducted to determine the cell communication network of border-associated macrophages. Transcription factors were predicted using the ‘pySCENIC' tool. We found that, in response to hypoxia, borderassociated macrophages underwent dynamic transcriptional changes and participated in the regulation of inflammatory-related pathways. Notably, the tumor necrosis factor pathway was activated by border-associated macrophages following ischemic stroke. The pySCENIC analysis indicated that the activity of signal transducer and activator of transcription 3(Stat3) was obviously upregulated in stroke, suggesting that Stat3 inhibition may be a promising strategy for treating border-associated macrophages-induced neuroinflammation. Finally, we constructed an animal model to investigate the effects of border-associated macrophages depletion following a stroke. Treatment with liposomes containing clodronate significantly reduced infarct volume in the animals and improved neurological scores compared with untreated animals. Taken together, our results demonstrate comprehensive changes in border-associated macrophages following a stroke, providing a theoretical basis for targeting border-associated macrophages-induced neuroinflammation in stroke treatment.展开更多
BACKGROUND Gallbladder neuroendocrine carcinoma(NEC)represents a subtype of gallbladder malignancies characterized by a low incidence,aggressive nature,and poor prognosis.Despite its clinical severity,the genetic alte...BACKGROUND Gallbladder neuroendocrine carcinoma(NEC)represents a subtype of gallbladder malignancies characterized by a low incidence,aggressive nature,and poor prognosis.Despite its clinical severity,the genetic alterations,mechanisms,and signaling pathways underlying gallbladder NEC remain unclear.CASE SUMMARY This case study presents a rare instance of primary gallbladder NEC in a 73-year-old female patient,who underwent a radical cholecystectomy with hepatic hilar lymphadenectomy and resection of liver segments IV-B and V.Targeted gene sequencing and bioinformatics analysis tools,including STRING,GeneMANIA,Metascape,TRRUST,Sangerbox,cBioPortal and GSCA,were used to analyze the biological functions and features of mutated genes in gallbladder NEC.Twelve mutations(APC,ARID2,IFNA6,KEAP1,RB1,SMAD4,TP53,BTK,GATA1,GNAS,and PRDM3)were identified,and the tumor mutation burden was determined to be 9.52 muts/Mb via targeted gene sequencing.A protein-protein interaction network showed significant interactions among the twelve mutated genes.Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analyses were used to assess mutation functions and pathways.The results revealed 40 tumor-related pathways.A key regulatory factor for gallbladder NEC-related genes was identified,and its biological functions and features were compared with those of gallbladder carcinoma.CONCLUSION Gallbladder NEC requires standardized treatment.Comparisons with other gallbladder carcinomas revealed clinical phenotypes,molecular alterations,functional characteristics,and enriched pathways.展开更多
The SKP1 gene is an important component of the SCF(SKP1-Cullin1-F-box)complex and serves as a bridge connecting the F-box and Cullin1genes(F-box-SKP1-Cullin1).The pattern of S-RNase being ubiquitously labelled by the ...The SKP1 gene is an important component of the SCF(SKP1-Cullin1-F-box)complex and serves as a bridge connecting the F-box and Cullin1genes(F-box-SKP1-Cullin1).The pattern of S-RNase being ubiquitously labelled by the SCF complex and degraded by the 26S protease accounts for the bulk of the available self-incompatibility studies.In this study,15 ClSKP1s from the‘Xiangshui'lemon genome and ubiquitome exist in the same SKP1 conserved domain(CD)as SKP1s in other species.The q PCR results showed that SKP1-6 and SKP1-14 have tissue expression patterns specific for expression in pollen.In addition,SKP1-6 and SKP1-14 in the stigma,style and ovary were significantly upregulated after self-pollination compared to those after cross-pollination.A subcellular location showed that SKP1-6 and SKP1-14 were located in the nucleus.In addition,yeast two-hybrid(Y2H)assays,bimolecular fluorescence complementation(BiFC)and luciferase complementation imaging(LCI)assays showed that SKP1-6 interacted with F-box1,F-box33,F-box34,F-box17,F-box19,Cullin1-2 and 26S proteasome subunit 4 homolog A(26S PS4HA).SKP1-14 interacted with F-box17,F-box19,F-box35,Cullin1-2 and 26S PS4HA.The interaction of Cullin1-2 and the F-box with SKP1 as a bridge was verified by a yeast three-hybrid experiment.The ability of S3-RNase to inhibit pollen and pollen tube growth and development was assessed using in vitro pollen co-culture experiments with recombinant S3-RNase proteins.Overall,this study provides important experimental evidence and theoretical basis for understanding the mechanism of self-incompatibility in plants by revealing the key role of the SCF complex in‘Xiangshui'lemon,which is bridged by ClSKP1-6,in self-incompatibility.The results of this study are of great significance for the future indepth exploration of the molecular mechanism of the SCF complex and its wide application in the self-incompatibility of plants.展开更多
Peripheral nerve injury is a common neurological condition that often leads to severe functional limitations and disabilities.Research on the pathogenesis of peripheral nerve injury has focused on pathological changes...Peripheral nerve injury is a common neurological condition that often leads to severe functional limitations and disabilities.Research on the pathogenesis of peripheral nerve injury has focused on pathological changes at individual injury sites,neglecting multilevel pathological analysis of the overall nervous system and target organs.This has led to restrictions on current therapeutic approaches.In this paper,we first summarize the potential mechanisms of peripheral nerve injury from a holistic perspective,covering the central nervous system,peripheral nervous system,and target organs.After peripheral nerve injury,the cortical plasticity of the brain is altered due to damage to and regeneration of peripheral nerves;changes such as neuronal apoptosis and axonal demyelination occur in the spinal cord.The nerve will undergo axonal regeneration,activation of Schwann cells,inflammatory response,and vascular system regeneration at the injury site.Corresponding damage to target organs can occur,including skeletal muscle atrophy and sensory receptor disruption.We then provide a brief review of the research advances in therapeutic approaches to peripheral nerve injury.The main current treatments are conducted passively and include physical factor rehabilitation,pharmacological treatments,cell-based therapies,and physical exercise.However,most treatments only partially address the problem and cannot complete the systematic recovery of the entire central nervous system-peripheral nervous system-target organ pathway.Therefore,we should further explore multilevel treatment options that produce effective,long-lasting results,perhaps requiring a combination of passive(traditional)and active(novel)treatment methods to stimulate rehabilitation at the central-peripheral-target organ levels to achieve better functional recovery.展开更多
As a means of quantitative interpretation,forward calculations of the global lithospheric magnetic field in the Spherical Harmonic(SH)domain have been widely used to reveal geophysical,lithological,and geothermal vari...As a means of quantitative interpretation,forward calculations of the global lithospheric magnetic field in the Spherical Harmonic(SH)domain have been widely used to reveal geophysical,lithological,and geothermal variations in the lithosphere.Traditional approaches either do not consider the non-axial dipolar terms of the inducing field and its radial variation or do so by means of complicated formulae.Moreover,existing methods treat the magnetic lithosphere either as an infinitesimally thin layer or as a radially uniform spherical shell of constant thickness.Here,we present alternative forward formulae that account for an arbitrarily high maximum degree of the inducing field and for a magnetic lithosphere of variable thickness.Our simulations based on these formulae suggest that the satellite magnetic anomaly field is sensitive to the non-axial dipolar terms of the inducing field but not to its radial variation.Therefore,in forward and inverse calculations of satellite magnetic anomaly data,the non-axial dipolar terms of the inducing field should not be ignored.Furthermore,our results show that the satellite magnetic anomaly field is sensitive to variability in the lateral thickness of the magnetized shell.In particular,we show that for a given vertically integrated susceptibility distribution,underestimating the thickness of the magnetic layer overestimates the induced magnetic field.This discovery bridges the greatest part of the alleged gap between the susceptibility values measured from rock samples and the susceptibility values required to match the observed magnetic field signal.We expect the formulae and conclusions of this study to be a valuable tool for the quantitative interpretation of the Earth's global lithospheric magnetic field,through an inverse or forward modelling approach.展开更多
The Fushan Depression is one of the petroliferous depressions in the Beibuwan Basin,South China Sea.Previous studies have preliminarily explored the origin and source of crude oils in some areas of this depression.Nev...The Fushan Depression is one of the petroliferous depressions in the Beibuwan Basin,South China Sea.Previous studies have preliminarily explored the origin and source of crude oils in some areas of this depression.Nevertheless,no systematic investigations on the classification and origin of oils and hy-drocarbon migration processes have been made for the entire petroleum system in this depression,which has significantly hindered the hydrocarbon exploration in the region.A total of 32 mudstone and 58 oil samples from the Fushan Depression were analyzed to definite the detailed oil-source correlation within the sequence and sedimentary framework.The organic matter of third member of Paleogene Liushagang Formation(Els(3))source rocks,both deltaic and lacustrine mudstone,are algal-dominated with high abundance of C_(23)tricyclic terpane and C_(30)4-methylsteranes.The deltaic source rocks occur-ring in the first member(Els_(1))and second member(Els_(2))of the Paleogene Liushagang Formation are characterized by high abundance of C_(19+20)tricyclic terpane and oleanane,reflecting a more terrestrial plants contribution.While lacustrine source rocks of Els_(1)and Els_(2)display the reduced input of terrige-nous organic matter with relatively low abundance of C 19+20 tricyclic terpane and oleanane.Three types of oils were identified by their biomarker compositions in this study.Most of the oils discovered in the Huachang and Bailian Els_(1)reservoir belong to group A and were derived from lacustrine source rocks of Els_(1)and Els_(2).Group B oils are found within the Els_(1)and Els_(2)reservoirs,showing a close relation to the deltaic source rocks of Els_(1)and Els_(2),respectively.Group C oils,occurring in the Els3 reservoirs,have a good affinity with the Els3 source rocks.The spatial distribution and accumulation of different groups of oils are mainly controlled by the sedimentary facies and specific structural conditions.The Els_(2)reservoir in the Yong'an area belonging to Group B oil,are adjacent to the source kitchen and could be considered as the favorable exploration area in the future.展开更多
Because of the challenge of compounding lightweight,high-strength Ti/Al alloys due to their considerable disparity in properties,Al 6063 as intermediate layer was proposed to fabricate TC4/Al 6063/Al 7075 three-layer ...Because of the challenge of compounding lightweight,high-strength Ti/Al alloys due to their considerable disparity in properties,Al 6063 as intermediate layer was proposed to fabricate TC4/Al 6063/Al 7075 three-layer composite plate by explosive welding.The microscopic properties of each bonding interface were elucidated through field emission scanning electron microscope and electron backscattered diffraction(EBSD).A methodology combining finite element method-smoothed particle hydrodynamics(FEM-SPH)and molecular dynamics(MD)was proposed for the analysis of the forming and evolution characteristics of explosive welding interfaces at multi-scale.The results demonstrate that the bonding interface morphologies of TC4/Al 6063 and Al 6063/Al 7075 exhibit a flat and wavy configuration,without discernible defects or cracks.The phenomenon of grain refinement is observed in the vicinity of the two bonding interfaces.Furthermore,the degree of plastic deformation of TC4 and Al 7075 is more pronounced than that of Al 6063 in the intermediate layer.The interface morphology characteristics obtained by FEM-SPH simulation exhibit a high degree of similarity to the experimental results.MD simulations reveal that the diffusion of interfacial elements predominantly occurs during the unloading phase,and the simulated thickness of interfacial diffusion aligns well with experimental outcomes.The introduction of intermediate layer in the explosive welding process can effectively produce high-quality titanium/aluminum alloy composite plates.Furthermore,this approach offers a multi-scale simulation strategy for the study of explosive welding bonding interfaces.展开更多
Joint Multimodal Aspect-based Sentiment Analysis(JMASA)is a significant task in the research of multimodal fine-grained sentiment analysis,which combines two subtasks:Multimodal Aspect Term Extraction(MATE)and Multimo...Joint Multimodal Aspect-based Sentiment Analysis(JMASA)is a significant task in the research of multimodal fine-grained sentiment analysis,which combines two subtasks:Multimodal Aspect Term Extraction(MATE)and Multimodal Aspect-oriented Sentiment Classification(MASC).Currently,most existing models for JMASA only perform text and image feature encoding from a basic level,but often neglect the in-depth analysis of unimodal intrinsic features,which may lead to the low accuracy of aspect term extraction and the poor ability of sentiment prediction due to the insufficient learning of intra-modal features.Given this problem,we propose a Text-Image Feature Fine-grained Learning(TIFFL)model for JMASA.First,we construct an enhanced adjacency matrix of word dependencies and adopt graph convolutional network to learn the syntactic structure features for text,which addresses the context interference problem of identifying different aspect terms.Then,the adjective-noun pairs extracted from image are introduced to enable the semantic representation of visual features more intuitive,which addresses the ambiguous semantic extraction problem during image feature learning.Thereby,the model performance of aspect term extraction and sentiment polarity prediction can be further optimized and enhanced.Experiments on two Twitter benchmark datasets demonstrate that TIFFL achieves competitive results for JMASA,MATE and MASC,thus validating the effectiveness of our proposed methods.展开更多
The growth trajectory of hailstones within clouds has remained elusive due to the inability to trace them directly,impeding the comprehension of their underlying growth mechanisms.This study investigated hailstone ver...The growth trajectory of hailstones within clouds has remained elusive due to the inability to trace them directly,impeding the comprehension of their underlying growth mechanisms.This study investigated hailstone vertical growth trajectories by detecting the stable isotope signatures(2H and 18O compositions)of different shells in 27 hailstones from 9hailstorms,which allowed us to capture the ambient temperature during hailstone growth.The vertical growth trajectories were obtained by comparing the isotopic compositions of water condensate in clouds,derived from the Adiabatic Model,with those measured in hailstones.Although hailstone growth was primarily observed in the–10°C to–30°C temperature layer,the embryo formation height and subsequent growth trajectories significantly varied among hailstones.Embryos formed over a wide range of temperatures(–8.7°C to–33.4°C);four originated at temperatures above–15°C and 16originated at temperatures below–20°C,suggesting ice nuclei composed of bioproteins and mineral dust,respectively.Among the 27 measured hailstones,3 exhibited minimal vertical movement,16 exhibited a monotonic rise or fall,and the remaining 8 exhibited alternating up-down trajectories;only one experienced“recycling”during up-down drifting.Trajectory analysis revealed that similar-sized hailstones from a single storm tended to form at similar heights,whereas those larger than 25 mm in diameter exhibited at least one period of upward growth.Vertical trajectories derived from isotopic analysis were corroborated by radar hydrometeor observations.展开更多

文摘[Objectives]This study was conducted to detect and analyze tomato leaf curl New Delhi virus(ToLCNDV).[Methods]Through PCR detection,sequence analysis,and pathogenicity verification,tomato leaf curl New Delhi virus(ToLCNDV)was identified in zucchini exhibiting systemic disease symptoms during a 2024 outbreak in Qingzhou City,Shandong Province,and was designated as ToLCNDV-SD.[Results]Specific primer amplification showed that all eight diseased samples produced bands of 504 bp(DNA-A)and 892 bp(DNA-B).Sequencing analysis revealed that ToLCNDV-SD DNA-A shared 96.10%homology with an Indonesian melon isolate(LC421834.1),while DNA-B showed 88.31%homology with a Malaysian bitter gourd isolate(MW248678.1).Phylogenetic analysis indicated its closest relationship with Southeast Asian cucurbit-infecting isolates.Friction transmission tests confirmed that the virus could spread mechanically,inducing typical symptoms 14 d after inoculation with positive PCR detection.[Conclusions]This study provides important insights for understanding the epidemic mechanisms and control strategies of ToLCNDV in China.

文摘Disaster mitigation necessitates scientifi c and accurate aftershock forecasting during the critical 2 h after an earthquake. However, this action faces immense challenges due to the lack of early postearthquake data and the unreliability of forecasts. To obtain foundational data for sequence parameters of the land-sea adjacent zone and establish a reliable and operational aftershock forecasting framework, we combined the initial sequence parameters extracted from envelope functions and incorporated small-earthquake information into our model to construct a Bayesian algorithm for the early postearthquake stage. We performed parameter fitting and early postearthquake aftershock occurrence rate forecasting and effectiveness evaluation for 36 earthquake sequences with M ≥ 4.0 in the Bohai Rim region since 2010. According to the results, during the early stage after the mainshock, earthquake sequence parameters exhibited relatively drastic fl uctuations with signifi cant errors. The integration of prior information can mitigate the intensity of these changes and reduce errors. The initial and stable sequence parameters generally display advantageous distribution characteristics, with each parameter’s distribution being relatively concentrated and showing good symmetry and remarkable consistency. The sequence parameter p-values were relatively small, which indicates the comparatively slow attenuation of signifi cant earthquake events in the Bohai Rim region. A certain positive correlation was observed between earthquake sequence parameters b and p. However, sequence parameters are unrelated to the mainshock magnitude, which implies that their statistical characteristics and trends are universal. The Bayesian algorithm revealed a good forecasting capability for aftershocks in the early postearthquake period (2 h) in the Bohai Rim region, with an overall forecasting effi cacy rate of 76.39%. The proportion of “too low” failures exceeded that of “too high” failures, and the number of forecasting failures for the next three days was greater than that for the next day.

文摘Pyrola atropurpurea Franch is an important annual herbaceous plant.Few genomic analyses have been conducted on this plant,and chloroplast genome research will enrich its genomics basis.This study is based on high-throughput sequencing technology and Bioinformatics methods to obtain the sequence,structure,and other characteristics of the P.atropurpurea chloroplast genome.The result showed that the chloroplast genome of P.atropurpurea has a double-stranded circular structure with a total length of 172,535 bp and a typical four-segment structure.The genome has annotated a total of 132 functional genes,including 43 tRNAs,8 rRNAs,76 protein-coding genes,and 5 pseudo-genes.In total,358 SSR loci were checked out,mainly composed of mononucleotide and trinucleotide repeat.There are three types of scattered repetitive sequences,totaling 4223,including 2452 forward repeats,1763 palindrome repeats,and eight reverse repeats.The optimal codon usage frequency is relatively high with AT usage preference in this genome.Chloroplast genome comparative analysis in the family Ericaceae shows that the overall sequence is more complex,and there are more variations in the gene interval region.The collinearity analysis indicated that there is a complex rearrangement of species between different genera in Ericaceae.The selection pressure analysis showed that the protein-encoding genes rpl33 and rps16 were positively selected among the seven medicinal plants in Ericaceae.The maximum likelihood tree shows that the genetic relationship among P.atropurpurea,Pyrola rotundifolia,and Chimaphila japonica is relatively close.Therefore,an important data basis was provided for species identification,genetic diversity,and phylogenetic studies of P.atropurpurea and even this genus of plants.
文摘Cystic echinococcosis (CE) is a prevalent zoonotic disease caused by Echinococcus granulosus, with a cosmopolitan distribution. The parasite is transmitted cyclically between canines and numerous intermediate herbivorous livestock animals. Also, other Taeniid tapeworms could infect domestic dogs and they pose significant veterinary and public health concerns worldwide. This study aimed to develop a sensitive molecular method for detecting Echinococcus spp. DNA in dog fecal samples using next-generation sequencing (NGS). A set of PCR primers targeting conserved regions of Taeniid tapeworms’ 18s rRNA genes was designed and tested for amplifying genomic DNA from various tapeworm species. The PCR system demonstrated high sensitivity, amplifying DNA from all tested tapeworm species, with differences observed in amplified band sizes. The primers were adapted for NGS analysis by adding forward and reverse adapters, enabling the sequencing of amplified DNA fragments. Application of the developed PCR system to dog fecal samples collected from Yatta town, Palestine, revealed the presence of E. granulosus DNA in five out of 50 samples. NGS analysis confirmed the specificity of the amplified DNA fragments, showing 98% - 99% similarity with the 18s rDNA gene of E. granulosus. This study demonstrates the utility of NGS-based molecular methods for accurate and sensitive detection of Echinococcus spp. in dog fecal samples, providing valuable insights for epidemiological surveillance and control programs of echinococcosis in endemic regions.

文摘Sweet potatoes are significant cash crops,however,their yield and quality are greatly compromised by viral diseases.In this study,the complete genomic sequences of two Sweet Potato Virus 2(SPV2)isolates from infected sweet potato leaves in the Shandong(designated as SPV2-SDYT,GenBank No.PQ855660.1)and Jiangsu(designated as SPV2-JSXZ,GenBank No.PQ855661.1)provinces in China were obtained using 5′RACE and RT-PCR amplification.Consistency,phylogeny,codon usage bias,recombination,and selection pressure analyses were conducted using the SPV2-SDYT and SPV2-JSXZ genome sequences.The complete genome sequences of SPV2-SDYT and SPV2-JSXZ were 10561 nucleotides(nt)in length,with respective nucleotide and amino acid identities of 99.25%and 99.12%,respectively.Both isolates were closely related to the SPV2 isolate from China(SPV2-LN).In both SPV2-SDYT and SPV2-JSXZ,the identity of the P1 protein was the highest,whereas that of the P3 protein was the lowest.There were 26 codons with relatively synonymous codon usage(RSCU)values greater than 1 in SPV2-SDYT and 27 codons with RSCU values greater than 1 in SPV2-JSXZ.High-frequency codons in their genomes were predominantly found to end with A/U.Recombination analysis revealed no major recombination sites in either SPV2-SDYT or SPV2-JSXZ.Further selection pressure analysis showed that the non-synonymous substitution rate/synonymous substitution rate(dN/dS)value of all 10 SPV2 proteins was less than 1.This is the first report on the evolutionary relationships of the 17 known SPV2 isolates.Our findings lay the molecular groundwork for preventing and controlling SPV2 infection in root-tuber crops.These findings also contribute to our understanding of the spread and evolution of SPV2,its pathogenic mechanisms,and the development of antiviral strategies against it.

文摘Machine learning(ML)efficiently and accurately processes dense seismic array data,improving earthquake catalog creation,which is crucial for understanding earthquake sequences and fault systems;analyzing its reliability is also essential.An M5.8 earthquake struck Alxa Left Banner,Inner Mongolia,China on April 15,2015,a region with limited CENC monitoring capabilities,making analysis challenging.However,abundant data from ChinArray provided valuable observations for assessing the event.This study leveraged ChinArray data from the 2015 Alxa Left Banner earthquake sequence,employing machine learning(specifically PhaseNet,a deep learning method,and GaMMA,a Bayesian approach)for automated seismic phase picking,association,and location analysis.Our generated catalog,comprising 10,432 phases from 708 events,is roughly ten times larger than the CENC catalog,encompassing all CENC events with strong consistency.A slight magnitude overestimation is observed only at lower magnitudes.Furthermore,the catalog adheres to the Gutenberg-Richter and Omori laws spatially,temporally,and in magnitude distribution,demonstrating its high reliability.Double-difference tomography refined locations for 366 events,yielding a more compact spatial distribution with horizontal errors within 100m,vertical errors within 300m,and travel-time residuals within 0.05s.Depths predominantly range from 10-30km.Aftershocks align primarily NEE,with the mainshock east of the aftershock zone.The near-vertical main fault plane dips northwestward,exhibiting a Y-shaped branching structure,converging at depth and expanding towards the surface.FOCMEC analysis,using first motion and amplitude ratios,yielded focal mechanism solutions for 10 events,including the mainshock.These solutions consistently indicate a strike-slip mechanism with a minor extensional component.Integrating the earthquake sequence's spatial distribution and focal mechanisms suggests the seismogenic structure is a negative flower structure,consistent with the Dengkou-Benjing fault.Comparing the CENC and ML-generated catalogs using the maximum curvature(MAXC)method reveals a 0.6 decrease in completeness magnitude(M_(C)).However,magnitude-frequency distribution discrepancies above the MAXC-estimated M_(C)suggest MAXC may underestimate both M_(C)and the b-value.This study analyzes the 2015 Alxa Left Banner M5.8 earthquake using a reliable,MLgenerated earthquake catalog,revealing detailed information about the sequence,faulting structure,aftershock distribution,and stress characteristics.

文摘By integrating core observations,logging data and seismic interpretation,this study takes the massive Cretaceous carbonates in the M block of the Santos Basin,Brazil,as an example to establish the sequence filling pattern of fault-bounded isolated platforms in rift lake basins,reveal the control mechanisms of shoal-body development and reservoir formation,and reconstruct the evolutionary history of lithofacies paleogeography.The following results are obtained.(1)Three tertiary sequences(SQ1-SQ3)are identified in the Lower Cretaceous Itapema-Barra Velha of the M block.During the depositional period of SQ1,the rift basement faults controlled the stratigraphic distribution pattern of thick on both sides and thin in the middle.The strata overlapped to uplift in the early stage.During the depositional period of SQ2-SQ3,the synsedimentary faults controlled the paleogeomorphic reworking process with subsidence in the northwest and uplifting in the northeast,accompanied with the relative fall of lake level.(2)The Lower Cretaceous in the M block was deposited in a littoral-shallow lake,with the lithofacies paleogeographic pattern transiting from the inner clastic shoals and outer shelly shoals in SQ1 to the alternation of mounds and shoals in SQ2-SQ3.(3)Under the joint control of relative lake-level fluctuation,synsedimentary faults and volcanic activity,the shelly shoals in SQ1 tend to accumulated vertically in the raised area,and the mound-shoal complex in SQ2-SQ3 tends to migrate laterally towards the slope-break belt due to the reduction of accommodation space.(4)The evolution pattern of high-energy mounds and shoals,which were vertically accumulated in the early stage and laterally migrated in the later stage,controlled the transformation of high-quality reservoirs from“centralized”to“ring shaped”distribution.The research findings clarify the sedimentary patterns of mounds and shoals and the distribution of favorable reservoirs in the fault-controlled lacustrine isolated platform,providing support for the deepwater hydrocarbon exploration in the subsalt carbonate rocks in the Santos Basin.
文摘Wavelet transformation is a widely used method in high-frequency sequence stratigraphic analysis.However, the application is problematic since different wavelets always return the same sequence analysis results. To address this issue, we applied five commonly used wavelets to theoretical sequence models to document some application criteria. Five gradual scale-change sequence models were simplified from the glutenite succession deposition by gravity flows to form the fining-upwards cycle sequences(FUCS) and coarsening-upwards cycle sequences(CUCS). After conducting theoretical sequence model tests, the optimal wavelet(sym4) was selected and successfully used with actual data to identify the sequence boundaries. We also proposed a new method to optimize the scale of continuous wavelet transformation(CWT) for sequence boundary determination. We found that the balloon-like marks in scalograms of db4, sym4, and coif4 wavelet determine, respectively, the fourth-order sequence boundary, the thick succession sequence boundaries in FUCS, and the thick succession sequence in FUCS and CUCS. Comparing the sequence identification results shows that the asymmetric wavelets had an advantage in high-frequency sequence boundary determination and sedimentary cycle discrimination through the amplitude trend of the coefficient, in which the sym4 wavelet is the most effective. In conclusion, the asymmetry of wavelets is the first selection principle, of which asymmetric wavelets are more sensitive to sediment deposition by flood flows. The match of the wavelet between the sequence is the second selection principle, in which the correlation of time-frequency impacts the accuracy of sequence surface localization. However, the waveform of the wavelet is a visual and abstract parameter for sequence boundary detection. The appropriate wavelet for lacustrine sequence analysis is the asymmetric wavelet with a weak number of side lobes. The depositional flows, depositional process,and autogenic are three sedimentary factors that influence the sequence analysis results.

文摘Matrix-assisted laser desorption/ionization(MALDI)mass spectrometry(MS)plays an indispensable role in analyzing protein covalent structures.The reliable identification of amino acid residues and modifications relies on the mass accuracy,which is highly dependent on calibration.However,the accuracy provided by the currently available calibrants still needs further improvement in terms of compatibility with multiple tandem MS modes or ion polarity modes,calibratable range,and minimizing suppression of and interference with analyte signals.Here aiming at developing a versatile calibrant to solve these problem,we designed a synthetic peptide format of calibrant R_x(GDP_n)_m(referred to as“Gly-Asp-Pro,GDP”)according to the chemical natures of amino acids and polypeptide fragmentation rules in tandem MS.With four types of amino acid residues selected and arranged through rational designs,a GDP peptide produces highly regulated fragments that give rise to evenly spaced signals in each tandem MS mode and is compatible with both positive and negative ion modes.In internal calibration,its regulated fragmentation pattern minimizes interference with analyte signals,and using a single peptide as the input minimizes suppression of the analyte signals.As demonstrated by analyses of proteins including monoclonal antibody and Aβ-42,these features allowed significant increase of the mass accuracy and precision,which improved sequence coverage and sequence resolution in sequence analyses(including de novo sequencing).This rational design strategy may also inspire further development of synthetic calibrants that benefit structural analysis of biomolecules.
文摘Objective:To address the phylogenetic and phylogeographic relationship between different lineages of Anopheles(An.)subpictus species complex in most parts of the Asian continent by maximum utilization of Internal Transcriber Spacer 2(ITS2)and cytochrome C oxidase I(COI)sequences deposited at the GenBank.Methods:Seventy-five ITS2,210 COI and 26 concatenated sequences available in the NCBI database were used.Phylogenetic analysis was performed using Bayesian likelihood trees,whereas median-joining haplotype networks and time-scale divergence trees were generated for phylogeographic analysis.Genetic diversity indices and genetic differentiation were also calculated.Results:Two genetically divergent molecular forms of An.subpictus species complex corresponding to sibling species A and B are established.Species A evolved around 37-82 million years ago in Sri Lanka,India,and the Netherlands,and species B evolved around 22-79 million years ago in Sri Lanka,India,and Myanmar.Vietnam,Thailand,and Cambodia have two molecular forms:one is phylogenetically similar to species B.Other forms differ from species A and B and evolved recently in the above mentioned countries,Indonesia and the Philippines.Genetic subdivision among Sri Lanka,India,and the Netherlands is almost absent.A substantial genetic differentiation was obtained for some populations due to isolation by large geographical distances.Genetic diversity indices reveal the presence of a long-established stable mosquito population,at mutation-drift equilibrium,regardless of population fluctuations.Conclusions:An.subpictus species complex consists of more than two genetically divergent molecular forms.Species A is highly divergent from the rest.Sri Lanka and India contain only species A and B.
文摘The present study is devoted to understanding the evolution of the Upper Jurassic Sab'atayn Formation in the Marib-Shabwa Basin,Yemen,through a sequence stratigraphic analysis based on integrating datasets of sedimentology,seismic sections,and well logs.The Sab'atayn Formation(Tithonian age)is represented by a series of clastic and evaporites that were deposited under fluvio-deltaic to prodeltaic settings.It is divided into four members including Yah(at the base),upwards to Seen,Alif,and Safir at the top.Two third-order depositional sequences were determined for the Tithonian succession which were separated by three sequence boundaries.These sequences were classified into their systems tracts signifying several sedimentation patterns of progradational,aggradational,and retrogradational parasequence sets.The first depositional sequence corresponds to the early-middle Tithonian Yah and Seen units that can be classified into lowstand,transgressive,and highstand systems tracts.The second sequence comprises the late Tithonian Alif unit that can be subdivided into transgressive and highstand systems tracts.The sandy deposits of the Alif Member(highstand deposits)represent the most productive hydrocarbon reservoir in the basin.The Upper Jurassic sediments in the study area were resulted from a combination of eustatic and tectonic effects.

文摘Border-associated macrophages are located at the interface between the brain and the periphery, including the perivascular spaces, choroid plexus, and meninges. Until recently, the functions of border-associated macrophages have been poorly understood and largely overlooked. However, a recent study reported that border-associated macrophages participate in stroke-induced inflammation, although many details and the underlying mechanisms remain unclear. In this study, we performed a comprehensive single-cell analysis of mouse border-associated macrophages using sequencing data obtained from the Gene Expression Omnibus(GEO) database(GSE174574 and GSE225948). Differentially expressed genes were identified, and enrichment analysis was performed to identify the transcription profile of border-associated macrophages. CellChat analysis was conducted to determine the cell communication network of border-associated macrophages. Transcription factors were predicted using the ‘pySCENIC' tool. We found that, in response to hypoxia, borderassociated macrophages underwent dynamic transcriptional changes and participated in the regulation of inflammatory-related pathways. Notably, the tumor necrosis factor pathway was activated by border-associated macrophages following ischemic stroke. The pySCENIC analysis indicated that the activity of signal transducer and activator of transcription 3(Stat3) was obviously upregulated in stroke, suggesting that Stat3 inhibition may be a promising strategy for treating border-associated macrophages-induced neuroinflammation. Finally, we constructed an animal model to investigate the effects of border-associated macrophages depletion following a stroke. Treatment with liposomes containing clodronate significantly reduced infarct volume in the animals and improved neurological scores compared with untreated animals. Taken together, our results demonstrate comprehensive changes in border-associated macrophages following a stroke, providing a theoretical basis for targeting border-associated macrophages-induced neuroinflammation in stroke treatment.

文摘BACKGROUND Gallbladder neuroendocrine carcinoma(NEC)represents a subtype of gallbladder malignancies characterized by a low incidence,aggressive nature,and poor prognosis.Despite its clinical severity,the genetic alterations,mechanisms,and signaling pathways underlying gallbladder NEC remain unclear.CASE SUMMARY This case study presents a rare instance of primary gallbladder NEC in a 73-year-old female patient,who underwent a radical cholecystectomy with hepatic hilar lymphadenectomy and resection of liver segments IV-B and V.Targeted gene sequencing and bioinformatics analysis tools,including STRING,GeneMANIA,Metascape,TRRUST,Sangerbox,cBioPortal and GSCA,were used to analyze the biological functions and features of mutated genes in gallbladder NEC.Twelve mutations(APC,ARID2,IFNA6,KEAP1,RB1,SMAD4,TP53,BTK,GATA1,GNAS,and PRDM3)were identified,and the tumor mutation burden was determined to be 9.52 muts/Mb via targeted gene sequencing.A protein-protein interaction network showed significant interactions among the twelve mutated genes.Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analyses were used to assess mutation functions and pathways.The results revealed 40 tumor-related pathways.A key regulatory factor for gallbladder NEC-related genes was identified,and its biological functions and features were compared with those of gallbladder carcinoma.CONCLUSION Gallbladder NEC requires standardized treatment.Comparisons with other gallbladder carcinomas revealed clinical phenotypes,molecular alterations,functional characteristics,and enriched pathways.

文摘The SKP1 gene is an important component of the SCF(SKP1-Cullin1-F-box)complex and serves as a bridge connecting the F-box and Cullin1genes(F-box-SKP1-Cullin1).The pattern of S-RNase being ubiquitously labelled by the SCF complex and degraded by the 26S protease accounts for the bulk of the available self-incompatibility studies.In this study,15 ClSKP1s from the‘Xiangshui'lemon genome and ubiquitome exist in the same SKP1 conserved domain(CD)as SKP1s in other species.The q PCR results showed that SKP1-6 and SKP1-14 have tissue expression patterns specific for expression in pollen.In addition,SKP1-6 and SKP1-14 in the stigma,style and ovary were significantly upregulated after self-pollination compared to those after cross-pollination.A subcellular location showed that SKP1-6 and SKP1-14 were located in the nucleus.In addition,yeast two-hybrid(Y2H)assays,bimolecular fluorescence complementation(BiFC)and luciferase complementation imaging(LCI)assays showed that SKP1-6 interacted with F-box1,F-box33,F-box34,F-box17,F-box19,Cullin1-2 and 26S proteasome subunit 4 homolog A(26S PS4HA).SKP1-14 interacted with F-box17,F-box19,F-box35,Cullin1-2 and 26S PS4HA.The interaction of Cullin1-2 and the F-box with SKP1 as a bridge was verified by a yeast three-hybrid experiment.The ability of S3-RNase to inhibit pollen and pollen tube growth and development was assessed using in vitro pollen co-culture experiments with recombinant S3-RNase proteins.Overall,this study provides important experimental evidence and theoretical basis for understanding the mechanism of self-incompatibility in plants by revealing the key role of the SCF complex in‘Xiangshui'lemon,which is bridged by ClSKP1-6,in self-incompatibility.The results of this study are of great significance for the future indepth exploration of the molecular mechanism of the SCF complex and its wide application in the self-incompatibility of plants.

文摘Peripheral nerve injury is a common neurological condition that often leads to severe functional limitations and disabilities.Research on the pathogenesis of peripheral nerve injury has focused on pathological changes at individual injury sites,neglecting multilevel pathological analysis of the overall nervous system and target organs.This has led to restrictions on current therapeutic approaches.In this paper,we first summarize the potential mechanisms of peripheral nerve injury from a holistic perspective,covering the central nervous system,peripheral nervous system,and target organs.After peripheral nerve injury,the cortical plasticity of the brain is altered due to damage to and regeneration of peripheral nerves;changes such as neuronal apoptosis and axonal demyelination occur in the spinal cord.The nerve will undergo axonal regeneration,activation of Schwann cells,inflammatory response,and vascular system regeneration at the injury site.Corresponding damage to target organs can occur,including skeletal muscle atrophy and sensory receptor disruption.We then provide a brief review of the research advances in therapeutic approaches to peripheral nerve injury.The main current treatments are conducted passively and include physical factor rehabilitation,pharmacological treatments,cell-based therapies,and physical exercise.However,most treatments only partially address the problem and cannot complete the systematic recovery of the entire central nervous system-peripheral nervous system-target organ pathway.Therefore,we should further explore multilevel treatment options that produce effective,long-lasting results,perhaps requiring a combination of passive(traditional)and active(novel)treatment methods to stimulate rehabilitation at the central-peripheral-target organ levels to achieve better functional recovery.

文摘As a means of quantitative interpretation,forward calculations of the global lithospheric magnetic field in the Spherical Harmonic(SH)domain have been widely used to reveal geophysical,lithological,and geothermal variations in the lithosphere.Traditional approaches either do not consider the non-axial dipolar terms of the inducing field and its radial variation or do so by means of complicated formulae.Moreover,existing methods treat the magnetic lithosphere either as an infinitesimally thin layer or as a radially uniform spherical shell of constant thickness.Here,we present alternative forward formulae that account for an arbitrarily high maximum degree of the inducing field and for a magnetic lithosphere of variable thickness.Our simulations based on these formulae suggest that the satellite magnetic anomaly field is sensitive to the non-axial dipolar terms of the inducing field but not to its radial variation.Therefore,in forward and inverse calculations of satellite magnetic anomaly data,the non-axial dipolar terms of the inducing field should not be ignored.Furthermore,our results show that the satellite magnetic anomaly field is sensitive to variability in the lateral thickness of the magnetized shell.In particular,we show that for a given vertically integrated susceptibility distribution,underestimating the thickness of the magnetic layer overestimates the induced magnetic field.This discovery bridges the greatest part of the alleged gap between the susceptibility values measured from rock samples and the susceptibility values required to match the observed magnetic field signal.We expect the formulae and conclusions of this study to be a valuable tool for the quantitative interpretation of the Earth's global lithospheric magnetic field,through an inverse or forward modelling approach.

文摘The Fushan Depression is one of the petroliferous depressions in the Beibuwan Basin,South China Sea.Previous studies have preliminarily explored the origin and source of crude oils in some areas of this depression.Nevertheless,no systematic investigations on the classification and origin of oils and hy-drocarbon migration processes have been made for the entire petroleum system in this depression,which has significantly hindered the hydrocarbon exploration in the region.A total of 32 mudstone and 58 oil samples from the Fushan Depression were analyzed to definite the detailed oil-source correlation within the sequence and sedimentary framework.The organic matter of third member of Paleogene Liushagang Formation(Els(3))source rocks,both deltaic and lacustrine mudstone,are algal-dominated with high abundance of C_(23)tricyclic terpane and C_(30)4-methylsteranes.The deltaic source rocks occur-ring in the first member(Els_(1))and second member(Els_(2))of the Paleogene Liushagang Formation are characterized by high abundance of C_(19+20)tricyclic terpane and oleanane,reflecting a more terrestrial plants contribution.While lacustrine source rocks of Els_(1)and Els_(2)display the reduced input of terrige-nous organic matter with relatively low abundance of C 19+20 tricyclic terpane and oleanane.Three types of oils were identified by their biomarker compositions in this study.Most of the oils discovered in the Huachang and Bailian Els_(1)reservoir belong to group A and were derived from lacustrine source rocks of Els_(1)and Els_(2).Group B oils are found within the Els_(1)and Els_(2)reservoirs,showing a close relation to the deltaic source rocks of Els_(1)and Els_(2),respectively.Group C oils,occurring in the Els3 reservoirs,have a good affinity with the Els3 source rocks.The spatial distribution and accumulation of different groups of oils are mainly controlled by the sedimentary facies and specific structural conditions.The Els_(2)reservoir in the Yong'an area belonging to Group B oil,are adjacent to the source kitchen and could be considered as the favorable exploration area in the future.

文摘Because of the challenge of compounding lightweight,high-strength Ti/Al alloys due to their considerable disparity in properties,Al 6063 as intermediate layer was proposed to fabricate TC4/Al 6063/Al 7075 three-layer composite plate by explosive welding.The microscopic properties of each bonding interface were elucidated through field emission scanning electron microscope and electron backscattered diffraction(EBSD).A methodology combining finite element method-smoothed particle hydrodynamics(FEM-SPH)and molecular dynamics(MD)was proposed for the analysis of the forming and evolution characteristics of explosive welding interfaces at multi-scale.The results demonstrate that the bonding interface morphologies of TC4/Al 6063 and Al 6063/Al 7075 exhibit a flat and wavy configuration,without discernible defects or cracks.The phenomenon of grain refinement is observed in the vicinity of the two bonding interfaces.Furthermore,the degree of plastic deformation of TC4 and Al 7075 is more pronounced than that of Al 6063 in the intermediate layer.The interface morphology characteristics obtained by FEM-SPH simulation exhibit a high degree of similarity to the experimental results.MD simulations reveal that the diffusion of interfacial elements predominantly occurs during the unloading phase,and the simulated thickness of interfacial diffusion aligns well with experimental outcomes.The introduction of intermediate layer in the explosive welding process can effectively produce high-quality titanium/aluminum alloy composite plates.Furthermore,this approach offers a multi-scale simulation strategy for the study of explosive welding bonding interfaces.

文摘Joint Multimodal Aspect-based Sentiment Analysis(JMASA)is a significant task in the research of multimodal fine-grained sentiment analysis,which combines two subtasks:Multimodal Aspect Term Extraction(MATE)and Multimodal Aspect-oriented Sentiment Classification(MASC).Currently,most existing models for JMASA only perform text and image feature encoding from a basic level,but often neglect the in-depth analysis of unimodal intrinsic features,which may lead to the low accuracy of aspect term extraction and the poor ability of sentiment prediction due to the insufficient learning of intra-modal features.Given this problem,we propose a Text-Image Feature Fine-grained Learning(TIFFL)model for JMASA.First,we construct an enhanced adjacency matrix of word dependencies and adopt graph convolutional network to learn the syntactic structure features for text,which addresses the context interference problem of identifying different aspect terms.Then,the adjective-noun pairs extracted from image are introduced to enable the semantic representation of visual features more intuitive,which addresses the ambiguous semantic extraction problem during image feature learning.Thereby,the model performance of aspect term extraction and sentiment polarity prediction can be further optimized and enhanced.Experiments on two Twitter benchmark datasets demonstrate that TIFFL achieves competitive results for JMASA,MATE and MASC,thus validating the effectiveness of our proposed methods.

文摘The growth trajectory of hailstones within clouds has remained elusive due to the inability to trace them directly,impeding the comprehension of their underlying growth mechanisms.This study investigated hailstone vertical growth trajectories by detecting the stable isotope signatures(2H and 18O compositions)of different shells in 27 hailstones from 9hailstorms,which allowed us to capture the ambient temperature during hailstone growth.The vertical growth trajectories were obtained by comparing the isotopic compositions of water condensate in clouds,derived from the Adiabatic Model,with those measured in hailstones.Although hailstone growth was primarily observed in the–10°C to–30°C temperature layer,the embryo formation height and subsequent growth trajectories significantly varied among hailstones.Embryos formed over a wide range of temperatures(–8.7°C to–33.4°C);four originated at temperatures above–15°C and 16originated at temperatures below–20°C,suggesting ice nuclei composed of bioproteins and mineral dust,respectively.Among the 27 measured hailstones,3 exhibited minimal vertical movement,16 exhibited a monotonic rise or fall,and the remaining 8 exhibited alternating up-down trajectories;only one experienced“recycling”during up-down drifting.Trajectory analysis revealed that similar-sized hailstones from a single storm tended to form at similar heights,whereas those larger than 25 mm in diameter exhibited at least one period of upward growth.Vertical trajectories derived from isotopic analysis were corroborated by radar hydrometeor observations.
