Methods HCMV UL144 open reading frame (ORF) was amplified by PCR assay in 72 lowpassage isolates [65 con-genitally infective children and 7 healthy children who were HCMV-DNA positive by quantitative PCR (qPCR)]. All positive PCR products were analyzed by heteroduplex mobility assay and single-stranded conformation polymorphism (HMA-SSCP) and 32 of them were sequenced. Results Fifty-five patient isolates and five healthy children isolates were HCMV-UL144 positive by PCR. Sequencing and HMA-SSCP analysis showed that significant strain-specific variability was present in the UL144 ORF. Phylogenetic analysis indicated that the nucleotide sequences could be separated into 3 major genotypes. Comparing between UL144 se-quences and the corresponding symptoms showed that genotype 2 did not exist in megacolon isolates. And genotype 1 and 3 were the major types among microcephaly and jaundice isolates respectively. Conclusions HCMV-UL144 existed in most of low passage isolates and sequences were hypervariable. The UL144 ORF and its predicted product with the high level of sequence variability in different kinds of isolates suggest that UL144 ORF might play a role in HCMV infectivity and subsequent diseases.展开更多

They may cause a significant increase in both stiffness and strength, reducing the deformation demand and increasing the energy dissipation capacity of the system. On the other hand, irregular arrangements of the masonry panels may lead to the concentration of damage in some regions, with negative effects; for example soft story mechanisms and shear failures in short columns. Therefore, the presence ofinfill walls should not be neglected, especially in regions of moderate and high seismicity. To this aim, simple models are available for solid infills walls, such as the diagonal no-tension strut model, while infilled frames with openings have not been adequately investigated. In this study, the effect of openings on the strength and stiffness of infilled frames is investigated by means of about 150 experimental and numerical tests. The main parameters involved are identified and a simple model to take into account the openings in the infills is developed and compared with other models proposed by different researchers. The model, which is based on the use of strength and stiffness reduction factors, takes into account the opening dimensions and presence of reinforcing elements around the opening. To address the issue of incorrect fusion results caused by conflicting evidence due to inaccurate evidence and incomplete recognition frameworks in radar airborne target tactical intention recognition,a spatiotemporal...To address the issue of incorrect fusion results caused by conflicting evidence due to inaccurate evidence and incomplete recognition frameworks in radar airborne target tactical intention recognition,a spatiotemporal evidence fusion algorithm is proposed.To resolve the conflict evidence fusion problem caused by inaccurate evidence,the algorithm performs discounting of evidence from both spatial and temporal dimensions.Spatial discounting is influenced by both inter-evidence inconsistency and intra-evidence inconsistency,while temporal discounting is determined by time intervals and information entropy.For the problem of conflicting evidence fusion due to an incomplete recognition framework,an open recognition architecture based on dynamic composite focal elements is proposed.This approach allocates some conflicting information to temporary composite focal elements,avoiding excessive basic probability assignment(BPA)of the empty set after fusion,which can lead to deviations from the actual fusion results.Simulation experiments comparing various methods indicate that the proposed method can effectively improve target intention recognition accuracy and demonstrates good stability.展开更多
The accuracy of genomic annotation is crucial for subsequent functional investigations;however,computational protocols used in high-throughput annotation of open reading frames(ORFs)can introduce inconsistencies.These...The accuracy of genomic annotation is crucial for subsequent functional investigations;however,computational protocols used in high-throughput annotation of open reading frames(ORFs)can introduce inconsistencies.These inconsistencies,which lead to non-uniform extension or truncation of sequence ends,pose challenges for downstream analyses.Existing strategies to rectify these inconsistencies are time-consuming and labor-intensive,lacking specific approaches.To address this gap,we developed to GC,a tool that integrates genomic annotation with RNA-seq datasets to rectify annotation inconsistencies.Using to GC,we achieved an accuracy of nearly 100%accuracy in correcting inconsistencies in published Phytophthora sojae ORFs.We applied this innovative pipeline to the GPCR-bigrams gene family,which was predicted to have 42 members in the P.sojae genome but lacked experimental validation.By employing to GC,we identified 32 GPCR-bigram ORFs with inconsistencies between previous annotations and to GC-corrected sequences.Notably,among these were 5 genes(GPCR-TKL9,GPCR-TKL15,GPCR-PDE3,GPCR-AC3,and GPCR-AC4)showed substantial inconsistencies.Experimental gene annotation confirmed the effectiveness of to GC,as sequences obtained through cloning matched those annotated by to GC.Importantly,we discovered two novel GPCRs(GPCR-AC3 and GPCR-AC4),which were previously mispredicted as a single gene.CRISPR/Cas9-mediated knockout experiments revealed the involvement of GPCR-AC4 but not GPCR-AC3 in oospore production,further confirming their status as two separate genes.In addition to P.sojae,the reliability of the to GC pipeline in Phytophthora capsici and Pythium ultimum further emphasizes the robustness of this pipeline.Our findings highlight the utility of to GC for reliable gene model correction,facilitating investigations into biological functions and offering potential applications in diverse species analyses.展开更多
