This paper undertakes a systematic combing of the development of research on integrating Chinese culture into foreign language education in China from the 1980s to 2025,dividing it into three stages:cultural attachmen...This paper undertakes a systematic combing of the development of research on integrating Chinese culture into foreign language education in China from the 1980s to 2025,dividing it into three stages:cultural attachment,cultural compensation,and cultural symbiosis,and reveals the logical shift of the research from the dominance of target language culture to the construction of the subjectivity of Chinese culture.Through quantitative and qualitative analysis of 435 CSSCI papers,three core themes are extracted:what to integrate,why to integrate,and how to integrate.This paper critically analyzes three pairs of contradictions:the imbalance between instrumentality and humanism,the separation of national narrative and individual expression,and the disconnection between traditional inheritance and modern transformation.It is proposed that future research should reconstruct the educational logic based on the Chinese context,integrate the national and individual dimensions,and build a dialogue mechanism between tradition and modernity,so as to provide theoretical and practical reference for the construction of a foreign language education system with Chinese characteristics.展开更多

Probiotics can regulate gut microbes to maintain human health.However,the sensitivity of probiotics to environmental conditions reduces their bioavailability.In contrast,the formation of probiotic biofilm provides a n...Probiotics can regulate gut microbes to maintain human health.However,the sensitivity of probiotics to environmental conditions reduces their bioavailability.In contrast,the formation of probiotic biofilm provides a natural physical barrier against external interference.Our previous study established a dynamic culture system of the biofilm-state Bifidobacterium adolescentis Gr19(B-DC-B.adolescentis Gr19),forming higher density and more structurally stable biofilms,which enhanced its potential probiotic properties in vivo.Thus,the protective effect and mechanism of B-DC-B.adolescentis Gr19 on lipopolysaccharide(LPS)-induced intestinal barrier dysfunction were investigated in this study.The results showed that B-DC-B.adolescentis Gr19 not only had high resistance and adhesion activity,but also improved the intestinal barrier by increasing goblet cells and promoting the expression of tight junction(TJ)-related proteins.Moreover,B-DC-B.adolescentis Gr19 effectively attenuated intestinal barrier injury in Caco-2 cells by improving intestinal permeability and integrity.Remarkably,B-DC-B.adolescentis Gr19 enhanced expression of TJ proteins,restored localization of cytoskeleton and reduced intestinal inflammation by suppressing the Ras homolog family member A/Rho-associated coiled-coil-forming kinases/nuclear factor kappa B/myosin light chain kinase/myosin light chain(RhoA/ROCK/NF-κB/MLCK/MLC)pathway.Therefore,B-DC-B.adolescentis Gr19 plays a key role in mitigating LPS-induced intestinal barrier dysfunction.Overall,the present study provides a theoretical basis for ameliorating intestinal barrier dysfunction and developing novel functional foods by using biofilm-state probiotics under dynamic culture.展开更多

Background:Therapeutic responses of breast cancer vary among patients and lead to drug resistance and recurrence due to the heterogeneity.Current preclinical models,however,are inadequate for predicting individual pat...Background:Therapeutic responses of breast cancer vary among patients and lead to drug resistance and recurrence due to the heterogeneity.Current preclinical models,however,are inadequate for predicting individual patient responses towards different drugs.This study aimed to investigate the patient-derived breast cancer culture models for drug sensitivity evaluations.Methods:Tumor and adjacent tissues from female breast cancer patients were collected during surgery.Patient-derived breast cancer cells were cultured using the conditional reprogramming technique to establish 2D models.The obtained patient-derived conditional reprogramming breast cancer(CRBC)cells were subsequently embedded in alginate-gelatin methacryloyl hydrogel microspheres to form 3D culture models.Comparisons between 2D and 3D models were made using immunohistochemistry(tumor markers),MTS assays(cell viability),flow cytometry(apoptosis),transwell assays(migration),and Western blotting(protein expression).Drug sensitivity tests were conducted to evaluate patient-specific responses to anti-cancer agents.Results:2D and 3D culture models were successfully established using samples from eight patients.The 3D models retained histological and marker characteristics of the original tumors.Compared to 2D cultures,3D models exhibited increased apoptosis,enhanced drug resistance,elevated stem cell marker expression,and greater migration ability—features more reflective of in vivo tumor behavior.Conclusion:Patient-derived 3D CRBC models effectively mimic the in vivo tumor microenvironment and demonstrate stronger resistance to anti-cancer drugs than 2D models.These hydrogel-based models offer a cost-effective and clinically relevant platform for drug screening and personalized breast cancer treatment.展开更多

Transcription factor-mediated cell conversion has been reported in the central nervous system of both rodents and nonhuman primates.In particular,glia-to-neuron conversion has been achieved in the brain and spinal cor...Transcription factor-mediated cell conversion has been reported in the central nervous system of both rodents and nonhuman primates.In particular,glia-to-neuron conversion has been achieved in the brain and spinal cord of animal models for neural regeneration and repair.However,whether glia-to-neuron conversion can be used for brain repair in humans needs to be explored.To investigate the use of glia-to-neuron conversion technology in the human brain,we established a long-term ex vivo culture system using human brain tissue that was surgically removed from epileptic patients to test glia-to-neuron conversion directly.We found that neural transcription factors NeuroD1 and Ascl1 both converted human glial cells into neurons.Immunostaining and electrophysiological recordings showed that the glia-converted neurons demonstrated immature properties during the initial 7-14 days of conversion,and then acquired more mature neuronal properties after 21-27 days of conversion.These ex vivo conversion studies in human brain tissue pave the way toward future clinical trials using a transcription factor-based glia-to-neuron conversion approach to treat neurological disorders.展开更多
Cell cultured meat has been extensively studied as an environmentally friendly,energy-saving and more effective technology.However,there are many technical bottlenecks,especially the regulatory mechanism and manufactu...Cell cultured meat has been extensively studied as an environmentally friendly,energy-saving and more effective technology.However,there are many technical bottlenecks,especially the regulatory mechanism and manufacturing method of in vitro myogenesis.Based on an edible modified silk protein scaffold,with 3D culturing,in situ differentiated and transcriptome analysis,this study describes novel scaffolds and fabrication methods for cell cultured meat.The results showed that the effective space and utilization efficiency for cell culture of the scaffold is 26–1000 that of the traditional culture dish;it could form a tissue-like structure.Transcriptomics revealed the regulatory pathways and key factors of different cycles.It clarifies that the multi-cycle process of myoblast myogenesis in vitro is different from the single feedback regulation in vivo.More importantly,a novel scaffold-based cell cultured meat manufacturing method was developed,further develop a new tissue culture solution that is different from existing cell culture meat production.For manufacturing processes,it provides a new cell culture meat technology system,provides a theoretical basis for the regulation of cell proliferation and muscle growth,and lays the technical foundation for in situ tissue culture of cell cultured meat in vitro.展开更多

Liposarcoma is one of the most common soft tissue sarcomas,however,its occurrence rate is still rare compared to other cancers.Due to its rarity,in vitro experiments are an essential approach to elucidate liposarcoma ...Liposarcoma is one of the most common soft tissue sarcomas,however,its occurrence rate is still rare compared to other cancers.Due to its rarity,in vitro experiments are an essential approach to elucidate liposarcoma pathobiology.Conventional cell culture-based research(2D cell culture)is still playing a pivotal role,while several shortcomings have been recently under discussion.In vivo,mouse models are usually adopted for pre-clinical analyses with expectations to overcome the issues of 2D cell culture.However,they do not fully recapitulate human dedifferentiated liposarcoma(DDLPS)characteristics.Therefore,three-dimensional(3D)culture systems have been the recent research focus in the cell biology field with the expectation to overcome at the same time the disadvantages of 2D cell culture and in vivo animal models and fill in the gap between them.Given the liposarcoma rarity,we believe that 3D cell culture techniques,including 3D cell cultures/co-cultures,and Patient-Derived tumor Organoids(PDOs),represent a promising approach to facilitate liposarcoma investigation and elucidate its molecular mechanisms and effective therapy development.In this review,we first provide a general overview of 3D cell cultures compared to 2D cell cultures.We then focus on one of the recent 3D cell culture applications,Patient-Derived Organoids(PDOs),summarizing and discussing several PDO methodologies.Finally,we discuss the current and future applications of PDOs to sarcoma,particularly in the field of liposarcoma.展开更多

文摘Probiotics can regulate gut microbes to maintain human health.However,the sensitivity of probiotics to environmental conditions reduces their bioavailability.In contrast,the formation of probiotic biofilm provides a natural physical barrier against external interference.Our previous study established a dynamic culture system of the biofilm-state Bifidobacterium adolescentis Gr19(B-DC-B.adolescentis Gr19),forming higher density and more structurally stable biofilms,which enhanced its potential probiotic properties in vivo.Thus,the protective effect and mechanism of B-DC-B.adolescentis Gr19 on lipopolysaccharide(LPS)-induced intestinal barrier dysfunction were investigated in this study.The results showed that B-DC-B.adolescentis Gr19 not only had high resistance and adhesion activity,but also improved the intestinal barrier by increasing goblet cells and promoting the expression of tight junction(TJ)-related proteins.Moreover,B-DC-B.adolescentis Gr19 effectively attenuated intestinal barrier injury in Caco-2 cells by improving intestinal permeability and integrity.Remarkably,B-DC-B.adolescentis Gr19 enhanced expression of TJ proteins,restored localization of cytoskeleton and reduced intestinal inflammation by suppressing the Ras homolog family member A/Rho-associated coiled-coil-forming kinases/nuclear factor kappa B/myosin light chain kinase/myosin light chain(RhoA/ROCK/NF-κB/MLCK/MLC)pathway.Therefore,B-DC-B.adolescentis Gr19 plays a key role in mitigating LPS-induced intestinal barrier dysfunction.Overall,the present study provides a theoretical basis for ameliorating intestinal barrier dysfunction and developing novel functional foods by using biofilm-state probiotics under dynamic culture.

文摘Background:Therapeutic responses of breast cancer vary among patients and lead to drug resistance and recurrence due to the heterogeneity.Current preclinical models,however,are inadequate for predicting individual patient responses towards different drugs.This study aimed to investigate the patient-derived breast cancer culture models for drug sensitivity evaluations.Methods:Tumor and adjacent tissues from female breast cancer patients were collected during surgery.Patient-derived breast cancer cells were cultured using the conditional reprogramming technique to establish 2D models.The obtained patient-derived conditional reprogramming breast cancer(CRBC)cells were subsequently embedded in alginate-gelatin methacryloyl hydrogel microspheres to form 3D culture models.Comparisons between 2D and 3D models were made using immunohistochemistry(tumor markers),MTS assays(cell viability),flow cytometry(apoptosis),transwell assays(migration),and Western blotting(protein expression).Drug sensitivity tests were conducted to evaluate patient-specific responses to anti-cancer agents.Results:2D and 3D culture models were successfully established using samples from eight patients.The 3D models retained histological and marker characteristics of the original tumors.Compared to 2D cultures,3D models exhibited increased apoptosis,enhanced drug resistance,elevated stem cell marker expression,and greater migration ability—features more reflective of in vivo tumor behavior.Conclusion:Patient-derived 3D CRBC models effectively mimic the in vivo tumor microenvironment and demonstrate stronger resistance to anti-cancer drugs than 2D models.These hydrogel-based models offer a cost-effective and clinically relevant platform for drug screening and personalized breast cancer treatment.

文摘Transcription factor-mediated cell conversion has been reported in the central nervous system of both rodents and nonhuman primates.In particular,glia-to-neuron conversion has been achieved in the brain and spinal cord of animal models for neural regeneration and repair.However,whether glia-to-neuron conversion can be used for brain repair in humans needs to be explored.To investigate the use of glia-to-neuron conversion technology in the human brain,we established a long-term ex vivo culture system using human brain tissue that was surgically removed from epileptic patients to test glia-to-neuron conversion directly.We found that neural transcription factors NeuroD1 and Ascl1 both converted human glial cells into neurons.Immunostaining and electrophysiological recordings showed that the glia-converted neurons demonstrated immature properties during the initial 7-14 days of conversion,and then acquired more mature neuronal properties after 21-27 days of conversion.These ex vivo conversion studies in human brain tissue pave the way toward future clinical trials using a transcription factor-based glia-to-neuron conversion approach to treat neurological disorders.

文摘Cell cultured meat has been extensively studied as an environmentally friendly,energy-saving and more effective technology.However,there are many technical bottlenecks,especially the regulatory mechanism and manufacturing method of in vitro myogenesis.Based on an edible modified silk protein scaffold,with 3D culturing,in situ differentiated and transcriptome analysis,this study describes novel scaffolds and fabrication methods for cell cultured meat.The results showed that the effective space and utilization efficiency for cell culture of the scaffold is 26–1000 that of the traditional culture dish;it could form a tissue-like structure.Transcriptomics revealed the regulatory pathways and key factors of different cycles.It clarifies that the multi-cycle process of myoblast myogenesis in vitro is different from the single feedback regulation in vivo.More importantly,a novel scaffold-based cell cultured meat manufacturing method was developed,further develop a new tissue culture solution that is different from existing cell culture meat production.For manufacturing processes,it provides a new cell culture meat technology system,provides a theoretical basis for the regulation of cell proliferation and muscle growth,and lays the technical foundation for in situ tissue culture of cell cultured meat in vitro.

文摘The Yangtze River Economic Belt is the birthplace of the world s rice farming culture,and is the origin of common rice(Oryza sativa L.),wild rice(Oryza Rufipogon Griff.),medicinal rice(Oryza officinalis Wall.ex G.Watt),and wart-grain rice(Oryza granate Nees et Arn.ex Hook.f.).Rice culture is the main theme of agricultural culture in this economic belt and an outstanding representative of excellent traditional Chinese culture.This study provides an overview of the history of rice cultivation in the belt from the prehistory,post-history and modern progress.It studies the agricultural cultural heritage resources of the whole rice industry chain from the biological genetic resources,world cultural heritage,key cultural relics protection units,national archaeological site parks,movable cultural relics,important agricultural cultural heritage,intangible cultural heritage,China time-honored brands,historical names,traditional markers,tribute culture,archival and documentary heritage.It also analyzes the six main problems in the inheritance and development of rice farming culture,and proposes 8 strategies,including promoting the inheritance and innovative development of excellent traditional Chinese culture through the inheritance of rice farming culture,promoting the Exploration of the Origins of Chinese Civilization Project through rice farming culture,and creating the Yangtze River National Cultural Park with rice farming culture as the main theme.
文摘Liposarcoma is one of the most common soft tissue sarcomas,however,its occurrence rate is still rare compared to other cancers.Due to its rarity,in vitro experiments are an essential approach to elucidate liposarcoma pathobiology.Conventional cell culture-based research(2D cell culture)is still playing a pivotal role,while several shortcomings have been recently under discussion.In vivo,mouse models are usually adopted for pre-clinical analyses with expectations to overcome the issues of 2D cell culture.However,they do not fully recapitulate human dedifferentiated liposarcoma(DDLPS)characteristics.Therefore,three-dimensional(3D)culture systems have been the recent research focus in the cell biology field with the expectation to overcome at the same time the disadvantages of 2D cell culture and in vivo animal models and fill in the gap between them.Given the liposarcoma rarity,we believe that 3D cell culture techniques,including 3D cell cultures/co-cultures,and Patient-Derived tumor Organoids(PDOs),represent a promising approach to facilitate liposarcoma investigation and elucidate its molecular mechanisms and effective therapy development.In this review,we first provide a general overview of 3D cell cultures compared to 2D cell cultures.We then focus on one of the recent 3D cell culture applications,Patient-Derived Organoids(PDOs),summarizing and discussing several PDO methodologies.Finally,we discuss the current and future applications of PDOs to sarcoma,particularly in the field of liposarcoma.
文摘In the context of rural revitalization,the landscape design of public spaces in Enshi Xiaomaopoying Village presents both new opportunities and challenges.This study explores the significance of integrating regional culture into rural public spaces.Field research has been conducted to assess the current utilization of regional culture in the public spaces of Xiaomaopoying Village.Subsequently,design optimization strategies are analyzed and proposed.Additionally,the practical application of regional cultural elements is examined,in order to offer guidance for the application of cultural creation of traditional characteristic villages in rural public spaces.
