Colorectal cancer remains one of the leading causes of morbidity and mortality worldwide.Despite notable advances in early detection and therapeutic strategies,the molecular mechanisms underlying tumor survival,chemot...Colorectal cancer remains one of the leading causes of morbidity and mortality worldwide.Despite notable advances in early detection and therapeutic strategies,the molecular mechanisms underlying tumor survival,chemotherapy resistance,and metastasis are not yet fully understood.MicroRNAs(miRNAs)have emerged as pivotal regulators of cancer development,as they modulate gene expression and orchestrate key signaling pathways.However,the epigenetic mechanisms that control miRNA expression and their downstream gene targets remain largely unclear.In this review,we highlight the critical role of the colorectal cancer microenvironment in influencing miRNA expression and discuss how this regulation contributes to tumorigenesis.A better understanding of these processes may lead to the identification of novel therapeutic targets and strategies to prevent recurrence.展开更多

Oxide dispersion strengthened(ODS)alloys are extensively used owing to high thermostability and creep strength contributed from uniformly dispersed fine oxides particles.However,the existence of these strengthening pa...Oxide dispersion strengthened(ODS)alloys are extensively used owing to high thermostability and creep strength contributed from uniformly dispersed fine oxides particles.However,the existence of these strengthening particles also deteriorates the processability and it is of great importance to establish accurate processing maps to guide the thermomechanical processes to enhance the formability.In this study,we performed particle swarm optimization-based back propagation artificial neural network model to predict the high temperature flow behavior of 0.25wt%Al2O3 particle-reinforced Cu alloys,and compared the accuracy with that of derived by Arrhenius-type constitutive model and back propagation artificial neural network model.To train these models,we obtained the raw data by fabricating ODS Cu alloys using the internal oxidation and reduction method,and conducting systematic hot compression tests between 400 and800℃with strain rates of 10^(-2)-10 S^(-1).At last,processing maps for ODS Cu alloys were proposed by combining processing parameters,mechanical behavior,microstructure characterization,and the modeling results achieved a coefficient of determination higher than>99%.展开更多
In this study,an automated multimodal system for detecting,classifying,and dating fruit was developed using a two-stage YOLOv11 pipeline.In the first stage,the YOLOv11 detection model locates individual date fruits in...In this study,an automated multimodal system for detecting,classifying,and dating fruit was developed using a two-stage YOLOv11 pipeline.In the first stage,the YOLOv11 detection model locates individual date fruits in real time by drawing bounding boxes around them.These bounding boxes are subsequently passed to a YOLOv11 classification model,which analyzes cropped images and assigns class labels.An additional counting module automatically tallies the detected fruits,offering a near-instantaneous estimation of quantity.The experimental results suggest high precision and recall for detection,high classification accuracy(across 15 classes),and near-perfect counting in real time.This paper presents a multi-stage pipeline for date fruit detection,classification,and automated counting,employing YOLOv11-based models to achieve high accuracy while maintaining real-time throughput.The results demonstrated that the detection precision exceeded 90%,the classification accuracy approached 92%,and the counting module correlated closely with the manual tallies.These findings confirm the potential of reducing manual labour and enhancing operational efficiency in post-harvesting processes.Future studies will include dataset expansion,user-centric interfaces,and integration with harvesting robotics.展开更多

Recently, noncoding RNAs such as micro RNAs(mi RNAs) and circular RNAs(circ R...Synaptic dysfunction and abnormal processing of amyloid precursor protein(APP) are early pathological features in Alzheimer’s disease(AD). Recently, noncoding RNAs such as micro RNAs(mi RNAs) and circular RNAs(circ RNAs) have been reported to contribute to the pathogenesis of AD. We found an age-dependent elevation of mi R-138 in APP/PS1(presenilin-1) mice. Mi R-138 inhibited the expression of ADAM10 [a disintegrin and metalloproteinase domain-containing protein 10], promoted amyloid beta(Ab) production, and induced synaptic and learning/memory deficits in APP/PS1 mice, while its suppression alleviated the AD-like phenotype in these mice. Overexpression of sirtuin 1(Sirt1), a target of mi R-138, ameliorated the mi R-138-induced inhibition of ADAM10 and elevation of Ab in vitro. The circ RNA HDAC9(circ HDAC9) was predicted to contain a mi R-138 binding site in several databases. Its expression was inversely correlated with mi R-138 in both Ab-oligomertreated N2 a cells and APP/PS1 mice, and it co-localized with mi R-138 in the cytoplasm of N2 a cells. Circ HDAC9 acted as a mi R-138 sponge, decreasing mi R-138 expression, and reversing the Sirt1 suppression and excessive Ab production induced by mi R-138 in vitro. Moreover,circ HDAC9 was decreased in the serum of both AD patients and individuals with mild cognitive impairment.These results suggest that the circ HDAC9/mi R-138/Sirt1 pathway mediates synaptic function and APP processing in AD, providing a potential therapeutic target for its treatment.展开更多
Circulating microRNAs are robustly present in plasma or serum and have become a research focus as biomarkers for tumor diagnosis and prognosis.Centrifugation is a necessary procedure for obtaining highquality blood su...Circulating microRNAs are robustly present in plasma or serum and have become a research focus as biomarkers for tumor diagnosis and prognosis.Centrifugation is a necessary procedure for obtaining highquality blood supernatant.Herein,we investigated one-step and two-step centrifugations,two centrifugal methods routinely used in microRNA study,to explore their effects on plasma microRNA quantification.The microRNAs obtained from one-step and two-step centrifugations were quantified by microarray and TaqMan-based real-time quantitative polymerase chain reaction(Q-PCR).Dynamic light scattering was performed to explore the difference underlying the two centrifugal methods.The results from the microarray containing 1,347 microRNAs showed that the signal detection rate was greatly decreased in the plasma sample prepared by two-step centrifugation.More importantly,the microRNAs missing in this plasma sample could be recovered and detected in the precipitate generated from the second centrifugation.Consistent with the results from microarray,a marked decrease of three representative microRNAs in twostep centrifugal plasma was validated by Q-PCR.According to the size distribution of all nanoparticles in plasma,there were fewer nanoparticles with size>1,000 nm in two-step centrifugal plasma.Our experiments directly demonstrated that different centrifugation methods produced distinct quantities of plasma microRNAs.Thus,exosomes or protein complexes containing microRNAs may be involved in large nanoparticle formation and may be precipitated after two-step centrifugation.Our results remind us that sample processing methods should be first considered in conducting research.展开更多
