克柔念珠菌诱导奶牛乳腺上皮细胞microRNA差异表达分析

非编码小RNA(microRNA)可参与奶牛乳腺固有免疫应答,可作为病原菌感染宿主细胞和奶牛乳腺炎的潜在生物标志物之一,但克柔念珠菌(Candida krusei)感染奶牛乳腺上皮细胞(MAC-T)后microRNA的表达模式尚不清晰。本研究旨在分析Candida kruse... 非编码小RNA(microRNA)可参与奶牛乳腺固有免疫应答,可作为病原菌感染宿主细胞和奶牛乳腺炎的潜在生物标志物之一,但克柔念珠菌(Candida krusei)感染奶牛乳腺上皮细胞(MAC-T)后microRNA的表达模式尚不清晰。本研究旨在分析Candida krusei诱导MAC-T中的差异表达microRNA(DEmicroRNA)及其功能,为揭示Candida krusei感染MAC-T的标志microRNA和后续研究microRNA调控宿主细胞免疫应答的调控机制提供基础。利用转录组(RNA-Seq)测序技术和生物信息学方法对克柔念珠菌感染(感染复数=1)后的MAC-T进行microRNA测序、DEmicroRNA分析及GO和KEGG功能富集分析。结果表明,在正常组和感染组MAC-T中共检测出1465个microRNA,相比于正常组,感染组筛选出16个表达显著上调和7个表达量显著下调的microRNA。应用TargetScan和miRanda软件对11个极显著性差异表达microRNA进行靶基因预测,共预测到6739个靶基因。GO和KEGG功能富集分析结果表明,上述11个极显著性差异表达microRNA可通过免疫相关信号通路而调节奶牛乳腺炎症,进一步分析发现,显著下调的bta-miR-2377,显著上调的bta-miR-2285i可能通过潜在靶基因参与MAPK、NF-κB、Toll样受体信号通路进而调控宿主细胞炎症的发生与发展过程。在Candida krusei诱导的奶牛乳腺上皮细胞中获得了23个DEmicroRNA,可能通过潜在靶基因调控奶牛乳腺上皮细胞炎症反应的发生与发展过程,为揭示microRNA调控克柔念珠菌诱导奶牛乳腺炎的致病机制提供了科学基础。 展开更多

单颗粒-电感耦合等离子体质谱(SP-ICP-MS)法超灵敏检测外泌体microRNA

外泌体microRNA(miRNA)作为极具潜力的液体活检生物标志物,其准确定量对疾病的早期诊断与预后评估至关重要。然而,由于外泌体中miRNA含量极低且样品基质复杂,现有检测方法常面临灵敏度不足的挑战。为解决此问题,本研究构建了一种结合滚... 外泌体microRNA(miRNA)作为极具潜力的液体活检生物标志物,其准确定量对疾病的早期诊断与预后评估至关重要。然而,由于外泌体中miRNA含量极低且样品基质复杂,现有检测方法常面临灵敏度不足的挑战。为解决此问题,本研究构建了一种结合滚环扩增(RCA)与超小型上转换纳米颗粒(UCNPs)标记的超灵敏免洗检测方法。该方法利用RCA固有的高特异性实现靶标的精准识别,并借助单颗粒-电感耦合等离子体质谱(SP-ICP-MS)的尺寸分辨能力彻底避免了繁琐的分离和清洗步骤。利用目标miRNA-21触发的RCA反应产生长的重复DNA序列作为“放大骨架”,并通过互补序列将超小型UCNPs高效标记于RCA产物上;随后,基于SP-ICP-MS技术,仅当纳米颗粒标记到足够大的RCA产物上时才能被计数,而未结合的UCNPs因尺寸低于检测限而被有效排除,从而实现免洗检测。结果表明,该方法对miRNA-21的检测限低至1.86 fmol/L,并在10~5000 fmol/L三个数量级的范围内呈现出良好的线性(R2=0.97)。本研究成功实现了对生物样本中外泌体miRNA的高灵敏检测。该策略为超低丰度生物分子的检测提供了新思路,在临床诊断领域具有广阔的应用前景。 展开更多

MicroRNA-98对糖氧剥夺诱导的SH-SY5Y细胞损伤的影响

目的 探讨microRNA-98(miR-98)在人神经母细胞瘤细胞SH-SY5Y中的表达及与缺血性脑卒中细胞凋亡和炎症损伤的关系。方法 采用氧糖剥夺/再灌注(OGD/R)诱导SH-SY5Y细胞构建体外缺血性脑卒中细胞模型。细胞分为对照组(未经任何处理)和OGD/R... 目的 探讨microRNA-98(miR-98)在人神经母细胞瘤细胞SH-SY5Y中的表达及与缺血性脑卒中细胞凋亡和炎症损伤的关系。方法 采用氧糖剥夺/再灌注(OGD/R)诱导SH-SY5Y细胞构建体外缺血性脑卒中细胞模型。细胞分为对照组(未经任何处理)和OGD/R组,采用MTT法检测细胞活力,采用实时荧光定量聚合酶链反应(qRT-PCR)检测miR-98、Rho相关卷曲螺旋激酶2(ROCK2)表达;通过生物信息学的方法,寻找与ROCK2结合特异性好、稳定性强的miRNAs,采用双萤光素酶报告基因检测萤光素酶活性。Western blotting检测ROCK2蛋白相对表达量。将miR-98模拟物、miR-98抑制剂、pcDNA3.1-ROCK2和相应的阴性对照组转染到SH-SY5Y细胞中,并进行OGD/R处理。采用流式细胞术、TUNEL法检测细胞凋亡情况;采用吸光光度法检测凋亡相关蛋白胱天蛋白酶3(Caspase-3)和胱天蛋白酶9(Caspase-9)水平;qRT-PCR检测TNF-α和IL-6表达。结果 与对照组相比,OGD/R组细胞活力降低,miR-98相对表达量降低(P<0.05),ROCK2 mRNA相对表达量升高(P<0.05)。联合生物信息学和双萤光素酶实验分析发现ROCK2是miR-98的靶点之一(P<0.05)。与对照组相比,miR-98模拟物转染组miR-98 mRNA相对表达量升高(P<0.05),而ROCK2蛋白相对表达量降低(P<0.05);miR-98抑制剂转染组miR-98 mRNA相对表达量降低,而ROCK2蛋白相对表达量升高(P<0.05)。与对照组相比,OGD/R组细胞凋亡率、Caspase-3和Caspase-9水平升高(P<0.05);与模拟物对照组相比,miR-98模拟物组细胞凋亡率、Caspase-3和Caspase-9水平降低(P<0.05);与miR-98模拟物组相比,miR-98模拟物组+pcDNA3.1-ROCK2组的细胞凋亡率、Caspase-3和Caspase-9水平升高(P<0.05)。与对照组相比,OGD/R组TNF-α和IL-6 mRNA相对表达量升高(P<0.05);与模拟物对照组相比,miR-98模拟物组TNF-α和IL-6 mRNA相对表达量降低(P<0.05);与miR-98模拟物组相比,miR-98模拟物组+pcDNA3.1-ROCK2组的TNF-α和IL-6 mRNA相对表达量升高(P<0.05)。结论 miR-98通过靶向抑制ROCK2的表达,抑制神经母细胞瘤SH-SY5Y细胞的凋亡和炎症损伤,在缺血性脑卒中神经元损伤中发挥保护作用。 展开更多

目的:采用双样本孟德尔随机化(MR)方法探索microRNAs(miRNAs)与子宫内膜异位症(EMs)发病危险之间的潜在因果关系。方法:整合miRNA表达数量性状位点(miR-eQTL)数据和EMs全基因组关联研究(GWAS)数据集。采用逆方差加权法(IVW)以及加权中... 目的:采用双样本孟德尔随机化(MR)方法探索microRNAs(miRNAs)与子宫内膜异位症(EMs)发病危险之间的潜在因果关系。方法:整合miRNA表达数量性状位点(miR-eQTL)数据和EMs全基因组关联研究(GWAS)数据集。采用逆方差加权法(IVW)以及加权中位数法进行MR分析,系统评估miRNAs与EMs之间的因果关系。利用MiRNet 2.0和miRTarBase数据库预测显著关联miRNAs的靶基因并进行功能富集分析。结果:miR-130a-3p的表达与EMs发病风险呈正相关(加权中位数:OR=1.263,95%CI:1.142~1.398,FDR校正后P=0.010;IVW:OR=1.202,95%CI:1.125~1.285,FDR校正后P=8.83×10^(-5)),而miR-30b-5p和miR-30d-5p则表现出保护性作用,尤其与直肠阴道间隔深部浸润型EMs风险降低相关。GO功能分析显示上述3个miRNA的靶基因显著富集在miRNAs代谢过程和转录调控相关通路。结论:miR-130a-3p高表达是EMs潜在危险因素,而miR-30b-5p和miR-30d-5p高表达则可能具有保护作用,它们与EMs发病之间存在潜在的因果关系。这些发现为EMs的临床诊治提供了潜在的循环生物标志物。 展开更多

The hot compression deformation behavior of Mg-6Zn-1Mn-0.5Ca(ZM61-0.5Ca)and Mg-6Zn-1Mn-2Sn-0.5Ca(ZMT612-0.5Ca)alloys was investigated at deformation temperatures ranging from 250℃to 400℃and strain rates varying from 0.001 s^(-1) to 1 s^(-1).The results show that the addition of Sn promotes dynamic recrystallization(DRX),and CaMgSn phases can act as nucleation sites during the compression deformation.Flow stress increases with increasing the strain rate and decreasing the temperature.Both the ZM61-0.5Ca and ZMT612-0.5Ca alloys exhibit obvious DRX characteristics.CaMgSn phases can effectively inhibit dislocation motion with the addition of Sn,thus increasing the peak fl ow stress of the alloy.The addition of Sn increases the hot deformation activation energy of the ZM61-0.5Ca alloy from 199.654 kJ/mol to 276.649 kJ/mol,thus improving the thermal stability of the alloy.For the ZMT612-0.5Ca alloy,the optimal hot deformation parameters are determined to be a deformation temperature range of 350–400℃and a strain rate range of 0.001–0.01 s^(-1). 展开更多

Colorectal cancer remains one of the leading causes of morbidity and mortality worldwide.Despite notable advances in early detection and therapeutic strategies,the molecular mechanisms underlying tumor survival,chemotherapy resistance,and metastasis are not yet fully understood.MicroRNAs(miRNAs)have emerged as pivotal regulators of cancer development,as they modulate gene expression and orchestrate key signaling pathways.However,the epigenetic mechanisms that control miRNA expression and their downstream gene targets remain largely unclear.In this review,we highlight the critical role of the colorectal cancer microenvironment in influencing miRNA expression and discuss how this regulation contributes to tumorigenesis.A better understanding of these processes may lead to the identification of novel therapeutic targets and strategies to prevent recurrence. 展开更多

Alzheimer s disease is a progressive neurodegenerative disease chara cterized by memory decline and the accumulation of abnormal protein aggregates in the brain.While the precise cause of Alzheimer s disease remains under investigation,recent research suggests that dys regulation of brainspecific microRNAs(miRs)plays a significant role in Alzheimer s disease pathogenesis.Brain-specific miRs are predominantly expressed within the central nervous system and are crucial for neuronal development,and function,potentially in brain disorders.This review identifies some key brainspecific miRs in Alzheimer's disease,including miR-9,miR-26b,miR-34a,miR-107,miR-124,miR-125b,miR-128,miR-132,miR-146a,miR-155,miR-219,miR-501-3p,and miR-502-3p.The review also shed light on the brain-specific location of these miRs,their dysregulation in Alzheimer s disease,and how they are involved in disease progression.Apparently,these brain-specific miRs modulate specific genes and are therefo re crucial for various cellular processes,including autophagy,cell cycle,tau phosphorylation,amyloid-beta production,and neuroinflammation.Moreover,these miRs are potent disease-modifying factors and their expression levels co uld serve as potential biomarkers for diagnosing or monitoring Alzheimer s disease progression. 展开更多

This literature review explores the complex interaction between p53 and microRNAs(miRNAs)in the occurrence and progression of breast cancer(BC),the most common and lethal tumor type among women.BC is a multifactorial disease resulting from a combination of genetic and epigenetic alterations in cell DNA,influencing proliferation,differentiation,and migration.TP53 gene,which codifies p53 protein,is a known tumor suppressor,and it plays an important role in cell maintenance as DNA repair,cell proliferation control,and apoptosis activation.TP53 expression can be modulated by several miRNAs,as miR-30c,miR-34a,and the miR-200 family,inhibiting p53 production and silencing its tumor suppressor effects.On the other hand,p53 protein can modulate several miRNAs expression,as miR-146a,miR-192,and the miR-200 family,by acting as a transcription factor or by modulating miRNA processing,interfering with BC aggressiveness and progression.Understanding the role of p53 and miRNAs in BC may aid in identifying new biomarkers and developing new targeted therapies for patient treatment. 展开更多

This study investigated the effects of xu-argument-based continuation writing on learners’processing of source texts.Seventy-five participants were randomly assigned to three conditions:(1)continuation writing,(2)summary writing,or(3)reading comprehension.Eye-tracking data were collected during reading,measuring early(first fixation duration,first pass duration)and late(go-past time,total fixation duration)eye movements.During writing,source-text rereading was tracked via fixation counts and durations.Results showed that task type did not affect initial lexical access,as first fixation duration showed no group differences.However,both production groups exhibited significantly longer first pass durations than the reading comprehension group.Late measures revealed a gradient pattern:the continuation writing group spent significantly longer gopast time and total fixation duration than the summary writing group,which exceeded the reading comprehension group.This indicates that continuation tasks promoted deeper cognitive engagement during reading.During writing,the continuation writing group spent more time rereading the source text with higher fixation counts than the summary writing group.These findings suggest that continuation writing triggers more intensive reader-text interaction during pre-writing and enhances comprehension-production coupling through sustained attention to input during writing.This study sheds light on the cognitive mechanisms underlying the theoretical and pedagogical value of xu-argument. 展开更多

Laser micro-nano processing technologies have been developed to address challenges that are otherwise difficult to solve in industrial applications and diverse scientific fields.These technologies offer designable patterning,arraying capabilities,three-dimensional(3D)processing,and high precision.Recent advancements in laser technologies have demonstrated their effectiveness as powerful tools for micro-nano processing of optoelectronic materials.By utilizing various laser techniques—such as laser-induced polymerization,laser ablation,laser-induced transfer,laser-directed assembly,and laser-assisted crystallization—broad applications in image sensors,displays,solar cells,lasers,anti-counterfeiting,and information encryption have been enabled.This review comprehensively summarizes recent progress in the laser micro-nano processing of optoelectronic materials,including the technologies used for preparation,patterning,arraying,and modification.These laser fabrication methods uniquely provide capabilities such as annealing,phase transitions,and ion exchange in optoelectronic materials.We also discuss the perspectives and challenges for future developments,including the advantages,disadvantages,and potential applications of different laser micro-nano processing technologies.With the rapid advancements in laser micro-nanofabrication,we foresee significant growth in advanced,high-performance optoelectronic applications.This review aims to provide researchers with insights into the current state and future prospects of laser-based micro-nano processing,encouraging further exploration and innovation in this promising field. 展开更多

Microseismic signal processing and rockburst disaster identification:A multi-task deep learning and machine learning approach

Underground engineering projects such as deep tunnel excavation often encounter rockburst disasters accompanied by numerous microseismic events.Rapid interpretation of microseismic signals is crucial for the timely identification of rockbursts.However,conventional processing encompasses multi-step workflows,including classification,denoising,picking,locating,and computational analysis,coupled with manual intervention,which collectively compromise the reliability of early warnings.To address these challenges,this study innovatively proposes the“microseismic stethoscope"-a multi-task machine learning and deep learning model designed for the automated processing of massive microseismic signals.This model efficiently extracts three key parameters that are necessary for recognizing rockburst disasters:rupture location,microseismic energy,and moment magnitude.Specifically,the model extracts raw waveform features from three dedicated sub-networks:a classifier for source zone classification,and two regressors for microseismic energy and moment magnitude estimation.This model demonstrates superior efficiency compared to traditional processing and semi-automated processing,reducing per-event processing time from 0.71 s to 0.49 s to merely 0.036 s.It concurrently achieves 98%accuracy in source zone classification,with microseismic energy and moment magnitude estimation errors of 0.13 and 0.05,respectively.This model has been well applied and validated in the Daxiagu Tunnel case in Sichuan,China.The application results indicate that the model is as accurate as traditional methods in determining source parameters,and thus can be used to identify potential geomechanical processes of rockburst disasters.By enhancing the signal processing reliability of microseismic events,the proposed model in this study presents a significant advancement in the identification of rockburst disasters. 展开更多
