Experimental study using the dilution incubation method to assess water biostability

Water biostability is of particular concern to water supply as a major limiting factor for heterotrophic bacterial growth in water distribution systems. This study focused on bacterial growth dynamics in the series dilution of water samples with TOC(total organic carbon) values determined beforehand. The results showed that the specific growth rate of Pseudomonas fluorescens P17 varied dramatically and irregularly with TOC value when TOC concentrations were low enough during the initial periods of incubation under given conditions. According to this relationship between bacterial growth rate and TOC, a dilution incubation method was designed for the study of water biostability. With the method under a given condition, a turning-point TOC value was found at a relatively fixed point in the curve between bacterial growth rate and TOC of water sample, and the variation of growth rate had different characteristics below the turning-point TOC value relative to that over this value. A turning-point TOC value similarly existed in all experiments not only with tap water, but also with acetate and mixed solutions. And in the dilution incubation method study, the affections were analyzed by condition factors such as inoculum amount,incubation time and nature of the organic carbon source. In very low organic carbon water environments, the variation characteristics of bacterial growth rate will be useful to further understand the meaning of water biostability. 展开更多

Grazing impact of microzooplankton on phytoplankton in the Xiamen Bay using pigment-specific dilution technique 被引量:2

Phytoplankton group-specific growth and microzooplankton grazing were determined seasonally using the dilution technique with high-performance liquid chromatography (HPLC) in the Xiamen Bay, a subtropical bay in southeast China, between May 2003 and February 2004. The results showed that growth rates of phytoplankton ranged from 0.71 to 2.2 d^-1 with the highest value occurred in the inner bay in May. Mierozooplankton grazing rates ranged from 0.5 to 3.1 d^-1 with the highest value occurred in the inner bay in August. Microzooplankton grazing impact ranged from 39% to 95% on total phytoplankton Chl a biomass, and 65% to 181% on primary production. The growth and grazing rates of each phytoplankton group varied, the highest growth rate (up to 3.3 d^-1 ) was recorded for diatoms in August, while the maximum grazing rate ( up to 2.1 d ^-1 ) was recorded for chlorophytes in February in the inner bay. Among main phytoplankton groups, grazing pressure of microzooplankton ranged from 10% to 83% on Chl a biomass, and from 14% to 151% on primary production. The highest grazing pressure on biomass was observed for cryptophytes (83%) in August, while the maximum grazing pressure on primary production was observed for eyanobacteria (up to 151% ) in December in the inner bay. Net growth rates of larger phytoplanktons (diatoms and dinoflagellates) were higher than those of smaller groups ( prasinophytes, chlorophytes and cyanobacteria). Relative preference index showed that microzooplankton grazed preferentially on prasinophytes and avoided to harvest diatoms in cold seasons (December and February). 展开更多

The results show thatthe polymer mixtures are compatible in DMF solution in the absence of urea. The influence of urea addition on the degree ofcompatibility of the polymer mixtures has been studied in terms of the compatibility parameters (△b_m and △[η]_m). It wasfound that the compatibility of the polymer mixtures is decreased with increasing urea addition, passing through a minimumat 0.5 M urea. 展开更多

Optimizing the ratio of the spike to sample for isotope dilution analysis:a case study with selenium isotopes

Isotope dilution(ID)method has been widely applied to studies of elemental speciation and certification of the concentrations of geological reference materials.One of the key factors restricting the application of the ID method is the difficulty in identifying the optimal ratios of the isotope tracer(spike)to sample(S/N)and in estimating the error propagation.Here,using Se isotope as an example and employing a Monte Carlo method,we found that the optimal choice of spike and S/N are^(77)Se(spike)and 0.7,respectively.The S/N in the range from 0.1 to 4 can produce sufficiently low errors(<0.4%).Extending this method to other elements such as Cr,Fe,Ni,Sr,Cd and etc.,their optimal spike and S/N were also presented.According to the optimized parameters of Se,we determined the Se concentrations of geological reference materials(GRMs)by thiol cotton fiber(TCF)-hydride generation ID-MC-ICP-MS.The relative standard deviation is<5%,and the detection limit is reduced to0.1 ng/g.Most of the measured concentrations are consistent with those recommended by IGGE or reported previously.For some GRMs,discrepancies(e.g.6.90%for GSS-5)exist between our measurements and previous ones,which can be explained by the inhomogeneity of GRMs and/or the different digestion,purification and measurement methods among laboratories. 展开更多

[Method] The conventional method and medium dilution method were used for bacterial, mold and yeast counting in sample recovery test. [Result] The medium dilution method (1:10 test solution, 0.5 ml/plate) could effectively eliminate the inhibition effect of the Bupi Qiangli Ointment, and the recovery of Staphylococcus aureus was greater than 70% in the 3 batches of samples; and the conventional method exhibited the recoveries of E. coil, Bacillus subtilis, Candida albicans and Aspergillus greater than 70% in the 3 batches of samples. [Conclusion] Due to Bupi Qiangli Ointment has strongly antibacterial effect on Staphylococcus au- reus, the medium dilution method was used for bacterial counting, and the conventional method was used for mold and yeast counting; and the conventional method was used for controlled bacterium examination of E. coll. 展开更多

Based on the analysis of the advantages and disadvantages of some vertical coordinates applied in the calculation of the Changjiang diluted water (CDW), a new hybrid vertical coordinate is designed, which uses σ coordinate for current and σ-z coordinate for salinity. To combine the current and salinity, the Eulerian-Lagrangian method is used for the salinity calculation, and the baroclinic pressure gradient (BPG) is calculated on the salinity sited layers. The new hybrid vertical coordinate is introduced to the widely used model of POM (Princeton Ocean Model) to make a new model of POM-σ-z. The BPG calculations of an ideal case show that POM-σ-z model brings smaller error than POM model does. The simulations of CDW also show that POM-σ-z model is better than POM model on simulating the salinity and its front. 展开更多

For the prepared Zn0.9(Co1-xSmx )0.1O systems, the influences of dopant concentration of Sm on the structural and optical properties were investigated. Three additional absorption peaks around 570, 620, and 660 nm wavelengths were observed in low Sm content films, which is attributed to the d-d transitions of Co2+ ions in tetrahedral crystalline. [Objectives] This study was conducted to establish a method for microbial limit test of Compound Yu E Nose Drops.[Methods] According to the Chinese Pharmacopoei (General Rules in the fourth part of the 2015 edition),the microbial limit test method for Compound Yu E Nose Drops was verified.[Results] Compound Yu E Nose Drops has a strong inhibitory effect on Staphylococcus aureus and Bacillus subtilis,and the inhibitory activity was significantly eliminated after increasing the diluent (1∶ 20).The recoveries were all in the range of 0.5-2.0 when the total quantities of aerobic microbes were determined by the dilution method (1∶ 20).When the total quantities of mould and yeast were determined by the conventional method,the recoveries were both in the range of 0.5-2.0.When examining control bacteria,Escherichia coli,S.aureus and Pseudomonas aeruginosa can all be detected in the test groups by the test liquid dilution method.[Conclusions] For Compound Yu E Nose Drops,the total quantities of aerobic microbe can be counted by the dilution method;the quantities of mould and yeast can be examined by the conventional plate method;and the conventional method can be used for control microbe examination. 展开更多

[ Results] Tongmai Tangyanming Capsule has a strong inhibitory effect on Bacillus subtilis and Candida albicans. The antimicrobial activity was significantly eliminated after increasing the diluent ( 1 : 20) ; and the recoveries were in the range of 0.5 -2 when the total quantities of aerobic microbes were determined by dilution method (1:20). When the total quantities of mould and yeast were determined by dilution method (1:20), the recoveries were from 0.5 to 2, and Escherichia coli, cholate-tolerant Gram-negative bacteria and Salmonella can be detected by the test solution dilution method. [ Condusions] With the plate method, the total quantities of aerobic bacteria and the quantities of mould and yeast were examined by increasing diluent, and conventional method was used for examination of control bacteria including E. coli, cholatetolerant Gram-negative bacteria and Salmonella. This method has been proved effective for microbial limit test of Tongmai Tangyanming Capsule, and can effectively control the quality of the preparation. The method is accurate and reliable. 展开更多

超低排放燃煤电厂排放的总颗粒物(TPM)既包含可过滤颗粒物(FPM)也包含可凝结颗粒物(CPM)。为探究国内超低排放燃煤电厂CPM的排放特征及其水溶性离子在废气治理设施中的迁移变化规律,利用稀释冷凝法对5家超低排放燃煤电厂湿法脱硫系统(WF... 超低排放燃煤电厂排放的总颗粒物(TPM)既包含可过滤颗粒物(FPM)也包含可凝结颗粒物(CPM)。为探究国内超低排放燃煤电厂CPM的排放特征及其水溶性离子在废气治理设施中的迁移变化规律,利用稀释冷凝法对5家超低排放燃煤电厂湿法脱硫系统(WFGD)进口、出口和湿式静电除尘(WESP)出口进行CPM平行取样,并分析其质量浓度和水溶性离子浓度。结果表明,烟气中的CPM质量浓度显著高于FPM,WFGD进口、出口和WESP出口CPM质量浓度分别是FPM的5.30、4.19、9.13倍,WFGD和WESP对CPM的去除率分别为8.3%~45.3%和30.9%~45.0%。其中SO_(4)^(2-)是WFGD进口、出口和WESP出口CPM中主要的水溶性离子,占全部水溶性离子的比例分别为42.8%、60.5%和42.6%。WFGD和WESP对水溶性离子也具有协同去除作用,去除率分别为28.10%~63.50%和78.96%。因此,建议未来强化固定源CPM排放管控,考虑制定相应的检测方法和排放标准。 展开更多

示踪气体稀释法能解决电厂大直径烟囱流场复杂导致的烟气流量测量结果误差大的问题。该方法具有可溯源性,且其测量原理与常规速度面积法不同,因此具有作为流量测量现场校准方法的潜力。为此,该文基于数值模拟方法分析示踪气体稀释法在... 示踪气体稀释法能解决电厂大直径烟囱流场复杂导致的烟气流量测量结果误差大的问题。该方法具有可溯源性,且其测量原理与常规速度面积法不同,因此具有作为流量测量现场校准方法的潜力。为此,该文基于数值模拟方法分析示踪气体稀释法在火电厂烟囱烟气流量测量中的可行性与准确性,并在此基础上,研究示踪剂稀释比例、示踪剂注入截面对测量的影响,同时还设计不同示踪剂取样方案来评估测量的稳定性。结果表明:在约9D(D为烟囱直径)高度处,示踪剂与烟气实现充分混合;示踪剂稀释比例过高或过低均会影响示踪剂与烟气的混合效率;示踪剂在烟道处注入能有效减小流量测量误差。在负荷率80%下,采取烟囱注入时,各取样方案的误差波动范围较大,但3点取样具有较稳定且良好的测量效果,在3D、8D、12D截面上的测量误差分别仅为-3.59%、-0.69%、-1.05%;水平烟道注入时,各取样方案的流量测量误差均不超过±10%,且3点取样时,在3D、8D、12D截面上的测量误差分别仅为0.98%、-0.52%、0.21%,均在±1%以内。该研究证明了示踪气体稀释法在大直径烟囱烟气流量测量中的可行性与准确性。 展开更多

乳糜血对磷酸酶法检测血清锂浓度的干扰及应对处理措施

目的探讨不同程度乳糜血对磷酸酶法检测血清锂(Li)浓度的干扰影响及应对处理措施。方法收集不含锂的重度乳糜血清混合样本,制备乳糜血浓缩液。收集服用碳酸锂、血脂正常的患者混合血清样本用于甘油三酯(TG)干扰实验;同时收集未服用碳酸... 目的探讨不同程度乳糜血对磷酸酶法检测血清锂(Li)浓度的干扰影响及应对处理措施。方法收集不含锂的重度乳糜血清混合样本,制备乳糜血浓缩液。收集服用碳酸锂、血脂正常的患者混合血清样本用于甘油三酯(TG)干扰实验;同时收集未服用碳酸锂的患者血清样本28例,依据TG干扰实验结果,选择对血清锂浓度检测无干扰的最大TG浓度作为稀释的目标浓度,进而计算目标稀释倍数。向每个样本中添加锂溶液,计算真实理论值。样本均分为3组,分别采用直接检测法、生理盐水稀释法及高速离心法(13000r/min,10min)处理,测定血清锂浓度,比较不同处理方法的结果差异。结果TG浓度>4.77mmol/L的乳糜血对磷酸酶法检测血清锂产生干扰。生理盐水稀释法测定值偏差最大(t=10.87,P<0.0001),而高速离心法测定值与真实理论值最接近(t=2.97,P=0.0369),准确度较高。直接检测法虽与高速离心法结果高度相关(r=0.9765,P<0.0001),但仍存在显著均值差异(t=5.37,P<0.0001)。结论TG浓度>4.77mmol/L的乳糜血清样本应避免使用生理盐水稀释法。高速离心法可以有效去除干扰,使测定结果更接近真实理论值,推荐作为优化方法。 展开更多

提出了同位素稀释内标-超高效液相色谱-串联质谱法测定抑菌膏霜类产品中倍他米松戊酸酯、倍他米松双丙酸酯、倍氯米松、倍他米松、倍氯米松双丙酸酯、氯倍他索、倍他米松乙酸酯、氯倍他松丁酸酯、氯倍他索丙酸酯等9种常见非法添加糖皮... 提出了同位素稀释内标-超高效液相色谱-串联质谱法测定抑菌膏霜类产品中倍他米松戊酸酯、倍他米松双丙酸酯、倍氯米松、倍他米松、倍氯米松双丙酸酯、氯倍他索、倍他米松乙酸酯、氯倍他松丁酸酯、氯倍他索丙酸酯等9种常见非法添加糖皮质激素含量的方法。取0.200 g样品置于15 mL离心管中,加入100μL含2.0 mg·L^(-1)倍他米松-d5、倍他米松乙酸酯-d3、倍氯米松双丙酸酯-d10、氯倍他索丙酸酯-d5、倍氯米松-d5和氯倍他索-d5的混合同位素内标储备溶液,经5.0 mL乙腈涡旋提取,再加入1.0 mL乙腈饱和的正己烷和1.0 mL饱和氯化钠溶液,涡旋2 min,超声提取20 min,涡旋振荡15 min后静置20 min,后弃去上部正己烷层。取中间乙腈层提取液1.5 mL置于离心管中,放入-20℃冰箱冷冻2 h后弃去上层。取0.75 mL冷冻后的乙腈提取液,加0.75 mL水稀释,涡旋混匀后再次放入-20℃冰箱冷冻2 h,于4℃冷冻离心5 min。取上清液过0.22μm聚四氟乙烯滤膜,滤液中的9种糖皮质激素在Acquity UPLC?BEH C_(18)色谱柱上分离,以不同体积比的0.1%(体积分数)甲酸溶液和乙腈的混合液为流动相进行梯度洗脱。分离后的目标物经电喷雾离子源正离子模式扫描,以多反应监测模式检测,内标法定量。结果表明,9种糖皮质激素标准曲线的线性范围为0.50~80.0μg·L^(-1),检出限(3S/N)为0.9~7.5μg.·kg^(-1)。按照标准加入法进行回收试验,回收率为75.7%~112%,测定值的相对标准偏差(n=6)为1.3%~15%。方法用于分析福建省采集的110批次抑菌膏霜类产品,结果显示26批次样品检出糖皮质激素,检出率达23.64%(26/110),包括氯倍他索丙酸酯、倍他米松双丙酸酯、氯倍他松丁酸酯和倍他米松,检出量分别为0.289~332.121 mg·kg^(-1),0.308~0.312 mg·kg^(-1),0.301 mg·kg^(-1)和0.337 mg·kg^(-1)。 展开更多

【目的】探明光枝勾儿茶内生真菌QX3Y2-5次级代谢产物及其抑菌活性,为开发新型天然高效抗菌药物提供科学依据。【方法】以光枝勾儿茶内生真菌QX3Y2-5为研究对象,用硅胶柱层析法、薄层色谱法、半制备高效液相色谱法等对其代谢产物进行分... 【目的】探明光枝勾儿茶内生真菌QX3Y2-5次级代谢产物及其抑菌活性,为开发新型天然高效抗菌药物提供科学依据。【方法】以光枝勾儿茶内生真菌QX3Y2-5为研究对象,用硅胶柱层析法、薄层色谱法、半制备高效液相色谱法等对其代谢产物进行分离纯化得到单体化合物,用二倍稀释法检测次级代谢产物对大肠杆菌和沙门氏菌的抑菌活性。【结果】光枝勾儿茶内生真菌QX3Y2-5次级代谢产物粗提物对大肠杆菌和沙门氏菌具有不同程度抑制效果,最小抑菌浓度(MIC)为62.50~500.00μg/mL;粗提物经分离纯化得到的次级代谢产物中,4-羟基-4,5-二氢呋喃-2-羧酸、烟酸和阿糖尿苷表现出较好的抑菌效果,其中,4-羟基-4,5-二氢呋喃-2-羧酸抑制效果最好,对大肠杆菌和沙门氏菌的MIC分别为31.25μg/mL和15.63μg/mL,最小杀菌浓度(MBC)分别为62.50μg/mL和15.63μg/mL。【结论】光枝勾儿茶内生真菌QX3Y2-5分离所得次级代谢产物4-羟基-4,5-二氢呋喃-2-羧酸、烟酸和阿糖尿苷对大肠杆菌和沙门氏菌具有一定体外抑制效果,其中,4-羟基-4,5-二氢呋喃-2-羧酸抑制效果最好。 展开更多

为了实现高通量、高准确性和高克隆活性的单细胞分离。提出了一种基于微流控芯片和单细胞检测的新型单细胞分离方法。并对该方法在检测仓鼠卵巢细胞亚株(CHO-K1)细胞的准确性以及分离后的单细胞克隆活性进行了研究。首先,以CMOS-MEMS微... 为了实现高通量、高准确性和高克隆活性的单细胞分离。提出了一种基于微流控芯片和单细胞检测的新型单细胞分离方法。并对该方法在检测仓鼠卵巢细胞亚株(CHO-K1)细胞的准确性以及分离后的单细胞克隆活性进行了研究。首先,以CMOS-MEMS微流控芯片为核心,设计了识别640个喷嘴轮廓并定位喷嘴号的图像算法,将单细胞图像送入训练好的卷积神经网络(CNN)中,检测轮廓清晰、形状合适的单个细胞。然后,微流控芯片上的加热电阻通过解码数据通电,利用热发泡喷墨打印原理将单个细胞瞬间喷射到指定位点,将识别到的单细胞连续打印。实验结果表明:CHO-K1单细胞检测平均精度可以达到94.03%,单克隆存活率可以达到87.18%。与有限稀释法(单细胞率提高70.42%,单克隆率提高66.68%)相比,该方法的性能显著提高。 展开更多
