We find that the oxygen abundance gradient in Arp 86 is flatter than that in normal disk galaxies, which confirms that gas inflows caused by tidal forces during encounters can flatten the metallicity distributions in galaxies. The companion galaxy NGC 7752 is currently experiencing a galaxy-wide starburst with a higher star for- marion rate surface density than the main galaxy NGC 7753, which can be explained in that the companion galaxy is more susceptible to the effects of interaction than the primary. We also find that the galaxy 2MASX J23470758+2926531 has similar abun- dance and star formation properties to NGC 7753, and may be a part of the Arp 86 system.展开更多
We have examined an isotropic and homogeneous cosmological model in f(R,T^(φ))gravity,where R represents the Ricci scalar and T^(φ)exhibits the energy momentum tensor's trace.We examine the stability criteria by...We have examined an isotropic and homogeneous cosmological model in f(R,T^(φ))gravity,where R represents the Ricci scalar and T^(φ)exhibits the energy momentum tensor's trace.We examine the stability criteria by performing the dynamical system analysis for our model f(R,T^(φ))=R+2(a T^(φ)+b),where a and b are the constants.We derive a set of autonomous equations and find their solutions by assuming a flat potential V0.We assess the equilibrium points from these equations and find the eigenvalues.We analyze the physical interpretation of the phase space for this system.We obtain three stable equilibrium points.We also examine the interaction between the scalar field and dark energy,represented by Q=ψH_(ρde)and determine the equilibrium points for this interaction.We identify four stable equilibrium points for this interaction.We calculate the values of the physical parameters for both scenarios at each equilibrium point,indicating the Universe's accelerated expansion.展开更多
Background: The human gut microbiome is an important target for disease treatment and prevention. Various microbial species within the complex ecosystem of the microbiome have been shown to play important roles in dis...Background: The human gut microbiome is an important target for disease treatment and prevention. Various microbial species within the complex ecosystem of the microbiome have been shown to play important roles in disease. Identification of bioactive materials capable of altering the abundances of these species both safely and effectively is a major goal in microbiome research. Many traditional Chinese medicines (TCMs) have been reported to affect the composition of the gut microbiome. Here, we summarize studies that have used TCMs to alter the gut microbiome and discuss the response relationship between TCMs and gut microbial species. Methods: We searched the PubMed, Web of Science, and Knowledge Network databases using the terms “traditional Chinese medicine,” “gut microbiome,” and specific system disease names (endocrine, immune, nervous, cardiovascular, and digestive). Studies were excluded if irrelevant or if the experimental procedures were unclear. Results: TCMs have been reported to affect a wide range of gut microbial taxa spanning major phyla, including Firmicutes, Bacteroidetes, Proteobacteria, Verrucomicrobiota, Actinobacteria, and Fusobacteria. In all, 54 TCMs including compounds and extracts have been tested in rodents and 30 have been examined in human trials. Almost all studies have reported positive results in regulating the gut microbiome as well as modulating corresponding phenotypes, spanning diseases of the endocrine, immune, nervous, cardiovascular, and digestive systems. Gut species, including Akkermansia, Bacteroides, Fusobacterium, Faecalibacterium, and E. coli, were found to be regulated by 19 TCMs. A network was constructed to visualize the interactions between TCMs and these taxa. Conclusion: There exists a complex and close relationship between intestinal microflora and diseases. Sufficient experimental data and studies have proved that the imbalance of intestinal microflora affects health by mediating metabolism, immune regulation, inflammation and signal transduction. Many characteristic alterations of intestinal microflora are positively correlated with diseases, so intestinal microflora has become a potential risk index and treatment target for many diseases. Many TCMs affect the relative abundances of microbial species in the gut, and therefore may be useful for modulating the gut microbiome. This review provides a reference for prioritizing candidate TCMs from the enormous repertoire of such medicines to test which specific gut microbes are targeted.展开更多
Background:The Colorectal Cancer(CRC)pathogenesis and therapeutic efficacy are influenced by the gut microbiome,making it a promising biomarker for predicting treatment responses and adverse effects.This systematic re...Background:The Colorectal Cancer(CRC)pathogenesis and therapeutic efficacy are influenced by the gut microbiome,making it a promising biomarker for predicting treatment responses and adverse effects.This systematic review aims to outline the gut microbiome composition in individuals with CRC undergoing the same therapeutic regimen and evaluate interindividual microbiome profile variations to better understand how these differences may influence therapeutic outcomes.Methods:Key studies investigating the microbiome’s role in therapeutic approaches for CRC were searched in both PubMed and Cochrane databases on 12 and 22 March 2025,respectively.Eligible studies included free full-text English-language randomized clinical trials and human observational studies reporting on gut microbiome composition and treatment outcomes.RoB 2 and ROBINS-I were employed in the evaluation of bias for randomized trials and observational studies,respectively.Data extracted was narratively analyzed.Results:Six studies involving a total of 361 individuals were included.Therapeutic interventions,either standard treatments and/or those targeting the gut microbiome,generally increased probiotic taxa and reduced pro-carcinogenic bacteria.However,no consistent pattern of improved clinical outcomes was observed,suggesting that treatment mechanisms,the tumor’s nature,and individual characteristics play critical roles in microbiome modulation.Conclusion:The gut microbiome holds significant potential in clinical settings.Nonetheless,further research is needed to better understand its functional aspects and to consider the influence of treatment mechanisms,the tumor’s nature,and individual characteristics as modulators,in order to optimize clinical outcomes.展开更多
Magnetorheological(MR)bearings,with their field-controllable rheological properties,offer new possibilities for control of rotor instabilities.However,their nonlinear dynamic behaviors and the underlying physical mech...Magnetorheological(MR)bearings,with their field-controllable rheological properties,offer new possibilities for control of rotor instabilities.However,their nonlinear dynamic behaviors and the underlying physical mechanisms governing these instabilities remain insufficiently understood.This work develops a coupled MR bearingrotor system model,where the oil film force is derived from a novel bilinear constitutive equation to capture the field-sensitive shear behaviors of MR fluids.Complex nonlinear dynamic behaviors including period doubling,quasi-period,and chaos are revealed,which emerge from the interaction between oil film vortex dynamics and magnetic excitation.The critical instability mechanism is identified from the evolution of intrinsic dynamic characteristics of MR bearings.When the whirl speed within the oil film reaches approximately half of the rotor speed,the damping force balances the destabilizing force,thereby defining a critical threshold beyond which the system transitions to instability.This threshold can be effectively tuned by adjusting the excitation current,which modifies the yield stress of MR fluids and consequently regulates the damping force.As a result,the nonlinear vibrations of oil whirl and whip can be suppressed,and the system stability can be significantly enhanced.These findings provide both theoretical insight and practical guidance for the design and control of MR bearing supported rotor systems.展开更多

The algorithm is mainly based on Kalman filter and interacting multiple model(IMM). It processes crosscorrelated sensor noises by using augmented fusion before model interacting. And eigenvalue decomposition is utilized to reduce calculation complexity and implement parallel computing. In simulation part, the feasibility of the algorithm was tested and verified, and the relationship between sensor number and the estimation precision was studied. Results show that simply increasing the number of sensor cannot always improve the performance of the estimation. Type and number of sensors should be optimized in practical applications.展开更多

METHODS: We constructed p7 protein bait plasmid by cloning the gene of p7 protein into pGBKTT, then transformed it into yeast AH109 (a type). The transformed yeast was mated with yeast Y187 (α type) containing liver cDNA library plasmid, pACT2 in 2×YPDA medium. Diploid yeast was plated on synthetic dropout nutrient medium (SD/- Trp-Leu-His-Ade) containing x-α-gal for selection and screening. After extracting and sequencing of plasmids from blue colonies, we performed sequence analysis by bioinformatics. RESULTS: Fifty colonies were selected and sequenced. Among them, one colony was Homo sapiens signal sequence receptor, seven colonies were Homo sapiens H19, seven colonies were immunoglobulin superfamily containing leucine-rich repeat, three colonies were spermatid peri-nuclear RNA binding proteins, two colonies were membrane-spanning 4-domains, 24 colonies were cancer-associated antigens, four colonies were nudeoporin 214 ku and two colonies were CLL-associated antigens. CONCLUSION: The successful cloning of gene of protein interacting with p7 protein paves a way for the study of the physiological function of p7 protein and its assodated protein.展开更多

HPV 18 E6 mRNA was expressed in yeast and there was no self-activation and toxicity in AH109. Seven proteins that interacted with HPV18 E6, including transmembrane protein 87B, phosphonoformate immuno-associated protein 5, vimentin, KM-HN-1 protein, dedicator of cytokinesis 7, vaccinia related kinase 2 and a hypothetical protein, were identified. It was suggested that yeast two-hybrid system is an efficient for screening interacting proteins. The high-risk HPV 18 E6 oncogene may interact with the proteins, which may be associated with signal transduction and transcriptional control, epithelial cell invasion and migration, as well as humoral and cellular immune etc. This investigation provides functional clues for further exploration of potential oncogenesis targets for cancer biotherapy.展开更多
Clubroot disease is a severe threat to Brassica crops globally,particularly in western Canada.Genetic resistance,achieved through pyramiding clubroot resistance(CR)genes with different modes of action,is the most impo...Clubroot disease is a severe threat to Brassica crops globally,particularly in western Canada.Genetic resistance,achieved through pyramiding clubroot resistance(CR)genes with different modes of action,is the most important strategy for managing the disease.However,studies on the CR gene functions are quite limited.In this study,we have conducted investigations into the temporal,structural,and interacting features of a newly cloned CR gene,Rcr1,using CRISPR/Cas9 technology.For temporal functionality,we developed a novel CRISPR/Cas9-based binary vector,pHHIGR-Hsp18.2,to deliver Rcr1 into a susceptible canola line(DH12075)and observed that early expression of Rcr1 is critical for conferring resistance.For structural functionality,several independent mutations in specific domains of Rcr1 resulted in loss-offunction,highlighting their importance for CR phenotype.In the study of the interacting features of Rcr1,a cysteine protease gene and its homologous allele in canola were successfully disrupted via CRISPR/Cas9 as an interacting component with Rcr1 protein,resulting in the conversion from clubroot resistant to susceptible in plants carrying intact Rcr1.These results indicated an indispensable role of these two cysteine proteases in Rcr1-mediated resistance response.This study,the first of its kind,provides valuable insights into the functionality of Rcr1.Further,the new vector p HHIGR-Hsp18.2 demonstrated an inducible feature on the removal of add-on traits,which should be useful for functional genomics and other similar research in brassica crops.展开更多
The E3 ubiquitin ligase,carboxyl terminus of heat shock protein 70(Hsp70)interacting protein(CHIP),also functions as a co-chaperone and plays a crucial role in the protein quality control system.In this study,we aimed...The E3 ubiquitin ligase,carboxyl terminus of heat shock protein 70(Hsp70)interacting protein(CHIP),also functions as a co-chaperone and plays a crucial role in the protein quality control system.In this study,we aimed to investigate the neuroprotective effect of overexpressed CHIP on Alzheimer’s disease.We used an adeno-associated virus vector that can cross the blood-brain barrier to mediate CHIP overexpression in APP/PS1 mouse brain.CHIP overexpression significantly ameliorated the performance of APP/PS1 mice in the Morris water maze and nest building tests,reduced amyloid-βplaques,and decreased the expression of both amyloid-βand phosphorylated tau.CHIP also alleviated the concentration of microglia and astrocytes around plaques.In APP/PS1 mice of a younger age,CHIP overexpression promoted an increase in ADAM10 expression and inhibitedβ-site APP cleaving enzyme 1,insulin degrading enzyme,and neprilysin expression.Levels of HSP70 and HSP40,which have functional relevance to CHIP,were also increased.Single nuclei transcriptome sequencing in the hippocampus of CHIP overexpressed mice showed that the lysosomal pathway and oligodendrocyte-related biological processes were up-regulated,which may also reflect a potential mechanism for the neuroprotective effect of CHIP.Our research shows that CHIP effectively reduces the behavior and pathological manifestations of APP/PS1 mice.Indeed,overexpression of CHIP could be a beneficial approach for the treatment of Alzheimer’s disease.展开更多

The algorithm describes the motion mode of vehicle by using three state spacemode]s. At first, the parallel filter of each model is realized by using multiple nonlinear filters. Then the weight integration of filtering result is carried out by using the model matching likelihood function so as to get the system positioning information. The method has advantages of nonlinear system filter and overcomes disadvantages of single model of filtering algorithm that has poor effects on positioning the maneuvering target. At last, the paper uses IMM and EKF methods to simulate the global positioning system (OPS)/inertial navigation system (INS)/dead reckoning (DR) integrated positioning system, respectively. The results indicate that the IMM algorithm is obviously superior to EKF filter used in the integrated positioning system at present. Moreover, it can greatly enhance the stability and positioning precision of integrated positioning system.展开更多

Using the dimensionality-reduction technique of Ott and Antonsen, we explore different types of synchronization dynamics when the incoherent state becomes unstable. We find that the inter-population coupling is crucial to the synchronization. When the intra-population interaction is repulsive, the local synchronization can still be maintained through the inter-population coupling. For attractive inter-population coupling, the local order parameters in different populations are of in-phase while the local synchronization are of anti-phase for repulsive inter-population coupling.展开更多

[Result] There was no toxicity and self activationof bait protein in yeast hybridization system, and six proteins (FTH1, LGALS3, CORO1C, SNRPG, KRTAP5-3, ZNF598) interacting with N proteinwere indentified. It was confirmed that LGALS3 and SNRPG had specific interaction with N protein by return experiment and co-immunoprecipitation(CoIP) test. This work demonstrates experimentally the close relation between return currents from relativistic laser-driven target polarization and the quality of the relativistic laser–plasma interaction for laser-driven second...This work demonstrates experimentally the close relation between return currents from relativistic laser-driven target polarization and the quality of the relativistic laser–plasma interaction for laser-driven secondary sources,taking as an example ion acceleration by target normal sheath acceleration.The Pearson linear correlation of maximum return current amplitude and proton spectrum cutoff energy is found to be in the range from~0.70 to 0.94.kA-scale return currents rise in all interaction schemes where targets of any kind are charged by escaping laser-accelerated relativistic electrons.Their precise measurement is demonstrated using an inductive scheme that allows operation at high repetition rates.Thus,return currents can be used as a metrological online tool for the optimization of many laser-driven secondary sources and for diagnosing their stability.In particular,in two parametric studies of laser-driven ion acceleration,we carry out a noninvasive online measurement of return currents in a tape target system irradiated by the 1 PW VEGA-3 laser at Centro de Láseres Pulsados:first the size of the irradiated area is varied at best compression of the laser pulse;second,the pulse duration is varied by means of induced group delay dispersion at best focus.This work paves the way to the development of feedback systems that operate at the high repetition rates of PW-class lasers.展开更多
FCS-like zinc finger(FLZ)gene family members are C2-C2 zinc finger proteins that take part in seed dormancy,resistance to Myzus persicae 1,sucrose signaling and abiotic stresse tolerance.However,their functions,especi...FCS-like zinc finger(FLZ)gene family members are C2-C2 zinc finger proteins that take part in seed dormancy,resistance to Myzus persicae 1,sucrose signaling and abiotic stresse tolerance.However,their functions,especially the molecular mechanism through which FLZs function,are not well understood.In this study,we characterized 120FLZs in wheat and revealed the function and mechanism of TaFLZ54D increasing salt stress tolerance in transgenic wheat.Expression analysis demonstrated that TaFLZ54D can be induced by NaCl treatment and it had the highest expression level under NaCl treatment among the 120 FLZs.Over-expression of TaFLZ54D increased wheat salt stress tolerance and the transgenic plants had higher levels of superoxide dismutase(SOD)and peroxidase(POD)activities and soluble sugar content,but a lower Na^(+)/K^(+)ratio and malondialdehyde(MDA)content than the wild type(WT)plants.Potassium ion transmembrane transporters and serine/threonine kinase inhibitor proteins showed differential expression between Ta FLZ54D transgenic wheat and the WT.Yeast two hybrid and luciferase complementation assays revealed that TaSGT1 and TaPP2C are the proteins that interact directly with TaFLZ54D.In summary,TaFLZ54D enhances salt stress tolerance through interaction with TaSGT1 and TaPP2C to reduce Na^(+)absorption and mitigate oxidative stress.The interaction between TaFLZ54D and TaSGT1,as well as TaPP2C indicated a link between salt stress tolerance of TaFLZ54D and the ubiquitin-mediated degradation of negative regulatory proteins.展开更多
Food systems are deeply affected by climate change and air pollution,while being key contributors to these environmental challenges.Understanding the complex interactions among food systems,climate change,and air poll...Food systems are deeply affected by climate change and air pollution,while being key contributors to these environmental challenges.Understanding the complex interactions among food systems,climate change,and air pollution is crucial for mitigating climate change,improving air quality,and promoting the sustainable development of food systems.However,the literature lacks a comprehensive review of these interactions,particularly in the current phase of rapid development in the field.To address this gap,this study systematically reviews recent research on the impacts of climate change and air pollution on food systems,as well as the greenhouse gas and air pollutant emissions from agri-food systems and their contribution to global climate change and air pollution.In addition,this study summarizes various strategies for mitigation and adaptation,including adjustments in agricultural practices and food supply chains.Profound changes in food systems are urgently needed to enhance adaptability and reduce emissions.This review offers a critical overview of current research on the interactions among food systems,climate change,and air pollution and highlights future research directions to support the transition to sustainable food systems.展开更多

文摘We have examined an isotropic and homogeneous cosmological model in f(R,T^(φ))gravity,where R represents the Ricci scalar and T^(φ)exhibits the energy momentum tensor's trace.We examine the stability criteria by performing the dynamical system analysis for our model f(R,T^(φ))=R+2(a T^(φ)+b),where a and b are the constants.We derive a set of autonomous equations and find their solutions by assuming a flat potential V0.We assess the equilibrium points from these equations and find the eigenvalues.We analyze the physical interpretation of the phase space for this system.We obtain three stable equilibrium points.We also examine the interaction between the scalar field and dark energy,represented by Q=ψH_(ρde)and determine the equilibrium points for this interaction.We identify four stable equilibrium points for this interaction.We calculate the values of the physical parameters for both scenarios at each equilibrium point,indicating the Universe's accelerated expansion.

文摘Background: The human gut microbiome is an important target for disease treatment and prevention. Various microbial species within the complex ecosystem of the microbiome have been shown to play important roles in disease. Identification of bioactive materials capable of altering the abundances of these species both safely and effectively is a major goal in microbiome research. Many traditional Chinese medicines (TCMs) have been reported to affect the composition of the gut microbiome. Here, we summarize studies that have used TCMs to alter the gut microbiome and discuss the response relationship between TCMs and gut microbial species. Methods: We searched the PubMed, Web of Science, and Knowledge Network databases using the terms “traditional Chinese medicine,” “gut microbiome,” and specific system disease names (endocrine, immune, nervous, cardiovascular, and digestive). Studies were excluded if irrelevant or if the experimental procedures were unclear. Results: TCMs have been reported to affect a wide range of gut microbial taxa spanning major phyla, including Firmicutes, Bacteroidetes, Proteobacteria, Verrucomicrobiota, Actinobacteria, and Fusobacteria. In all, 54 TCMs including compounds and extracts have been tested in rodents and 30 have been examined in human trials. Almost all studies have reported positive results in regulating the gut microbiome as well as modulating corresponding phenotypes, spanning diseases of the endocrine, immune, nervous, cardiovascular, and digestive systems. Gut species, including Akkermansia, Bacteroides, Fusobacterium, Faecalibacterium, and E. coli, were found to be regulated by 19 TCMs. A network was constructed to visualize the interactions between TCMs and these taxa. Conclusion: There exists a complex and close relationship between intestinal microflora and diseases. Sufficient experimental data and studies have proved that the imbalance of intestinal microflora affects health by mediating metabolism, immune regulation, inflammation and signal transduction. Many characteristic alterations of intestinal microflora are positively correlated with diseases, so intestinal microflora has become a potential risk index and treatment target for many diseases. Many TCMs affect the relative abundances of microbial species in the gut, and therefore may be useful for modulating the gut microbiome. This review provides a reference for prioritizing candidate TCMs from the enormous repertoire of such medicines to test which specific gut microbes are targeted.

文摘Background:The Colorectal Cancer(CRC)pathogenesis and therapeutic efficacy are influenced by the gut microbiome,making it a promising biomarker for predicting treatment responses and adverse effects.This systematic review aims to outline the gut microbiome composition in individuals with CRC undergoing the same therapeutic regimen and evaluate interindividual microbiome profile variations to better understand how these differences may influence therapeutic outcomes.Methods:Key studies investigating the microbiome’s role in therapeutic approaches for CRC were searched in both PubMed and Cochrane databases on 12 and 22 March 2025,respectively.Eligible studies included free full-text English-language randomized clinical trials and human observational studies reporting on gut microbiome composition and treatment outcomes.RoB 2 and ROBINS-I were employed in the evaluation of bias for randomized trials and observational studies,respectively.Data extracted was narratively analyzed.Results:Six studies involving a total of 361 individuals were included.Therapeutic interventions,either standard treatments and/or those targeting the gut microbiome,generally increased probiotic taxa and reduced pro-carcinogenic bacteria.However,no consistent pattern of improved clinical outcomes was observed,suggesting that treatment mechanisms,the tumor’s nature,and individual characteristics play critical roles in microbiome modulation.Conclusion:The gut microbiome holds significant potential in clinical settings.Nonetheless,further research is needed to better understand its functional aspects and to consider the influence of treatment mechanisms,the tumor’s nature,and individual characteristics as modulators,in order to optimize clinical outcomes.

文摘Magnetorheological(MR)bearings,with their field-controllable rheological properties,offer new possibilities for control of rotor instabilities.However,their nonlinear dynamic behaviors and the underlying physical mechanisms governing these instabilities remain insufficiently understood.This work develops a coupled MR bearingrotor system model,where the oil film force is derived from a novel bilinear constitutive equation to capture the field-sensitive shear behaviors of MR fluids.Complex nonlinear dynamic behaviors including period doubling,quasi-period,and chaos are revealed,which emerge from the interaction between oil film vortex dynamics and magnetic excitation.The critical instability mechanism is identified from the evolution of intrinsic dynamic characteristics of MR bearings.When the whirl speed within the oil film reaches approximately half of the rotor speed,the damping force balances the destabilizing force,thereby defining a critical threshold beyond which the system transitions to instability.This threshold can be effectively tuned by adjusting the excitation current,which modifies the yield stress of MR fluids and consequently regulates the damping force.As a result,the nonlinear vibrations of oil whirl and whip can be suppressed,and the system stability can be significantly enhanced.These findings provide both theoretical insight and practical guidance for the design and control of MR bearing supported rotor systems.

文摘We investigate the alpha helical protein structure characterized by fourth-order interspine coupling,focusing on a three-coupled fourth-order nonlinear Schr??dinger system.We introduce a generalized Darboux transformation,departing from the classical Darboux transformation.Based on this,we construct the two-and three-degenerate soliton solutions and four-degenerate asymptotic soliton solutions.Based on the asymptotic analysis,we find that the amplitudes of interacting solitons are retained upon the interactions.Elastic interactions between two degenerate solitons exhibiting four curve-type asymptotic solitons are depicted.When the lattice parameterβchanges,the velocities of the two degenerate solitons also change.Elastic interaction among three degenerate solitons comprising four curve-type asymptotic solitons and two line-type solitons is presented.Interaction among one soliton and two degenerate solitons with different velocities is shown.Elastic interaction among four degenerate solitons comprising eight curve-type asymptotic solitons is also presented.Interaction among two two-degenerate solitons with two spectral parameters is shown.The relative distance between two asymptotic solitons exhibits logarithmic growth with|t|,where t represents the retarded time.Acceleration of soliton separation decays exponentially with relative distance,and eventually approaches zero.Phase shifts depend on t.

文摘This paper presents a data fusion algorithm for dynamic system with multi-sensor and uncertain system models. The algorithm is mainly based on Kalman filter and interacting multiple model(IMM). It processes crosscorrelated sensor noises by using augmented fusion before model interacting. And eigenvalue decomposition is utilized to reduce calculation complexity and implement parallel computing. In simulation part, the feasibility of the algorithm was tested and verified, and the relationship between sensor number and the estimation precision was studied. Results show that simply increasing the number of sensor cannot always improve the performance of the estimation. Type and number of sensors should be optimized in practical applications.

文摘AIM: To investigate the biological function of p7 protein and to look for proteins interacting with p7 protein in hepatocytes. METHODS: We constructed p7 protein bait plasmid by cloning the gene of p7 protein into pGBKTT, then transformed it into yeast AH109 (a type). The transformed yeast was mated with yeast Y187 (α type) containing liver cDNA library plasmid, pACT2 in 2×YPDA medium. Diploid yeast was plated on synthetic dropout nutrient medium (SD/- Trp-Leu-His-Ade) containing x-α-gal for selection and screening. After extracting and sequencing of plasmids from blue colonies, we performed sequence analysis by bioinformatics. RESULTS: Fifty colonies were selected and sequenced. Among them, one colony was Homo sapiens signal sequence receptor, seven colonies were Homo sapiens H19, seven colonies were immunoglobulin superfamily containing leucine-rich repeat, three colonies were spermatid peri-nuclear RNA binding proteins, two colonies were membrane-spanning 4-domains, 24 colonies were cancer-associated antigens, four colonies were nudeoporin 214 ku and two colonies were CLL-associated antigens. CONCLUSION: The successful cloning of gene of protein interacting with p7 protein paves a way for the study of the physiological function of p7 protein and its assodated protein.

文摘To screen for novel binding proteins interacting with high-risk HPV 18 E6 oncogene, the strain AH 109 was transformed with pGBKT7-HPV 18 E6 plasmid, and subsequent transference was utilized to screen for interacting proteins with HPV 18 E6 in human Hela cDNA library. HPV 18 E6 mRNA was expressed in yeast and there was no self-activation and toxicity in AH109. Seven proteins that interacted with HPV18 E6, including transmembrane protein 87B, phosphonoformate immuno-associated protein 5, vimentin, KM-HN-1 protein, dedicator of cytokinesis 7, vaccinia related kinase 2 and a hypothetical protein, were identified. It was suggested that yeast two-hybrid system is an efficient for screening interacting proteins. The high-risk HPV 18 E6 oncogene may interact with the proteins, which may be associated with signal transduction and transcriptional control, epithelial cell invasion and migration, as well as humoral and cellular immune etc. This investigation provides functional clues for further exploration of potential oncogenesis targets for cancer biotherapy.

文摘Clubroot disease is a severe threat to Brassica crops globally,particularly in western Canada.Genetic resistance,achieved through pyramiding clubroot resistance(CR)genes with different modes of action,is the most important strategy for managing the disease.However,studies on the CR gene functions are quite limited.In this study,we have conducted investigations into the temporal,structural,and interacting features of a newly cloned CR gene,Rcr1,using CRISPR/Cas9 technology.For temporal functionality,we developed a novel CRISPR/Cas9-based binary vector,pHHIGR-Hsp18.2,to deliver Rcr1 into a susceptible canola line(DH12075)and observed that early expression of Rcr1 is critical for conferring resistance.For structural functionality,several independent mutations in specific domains of Rcr1 resulted in loss-offunction,highlighting their importance for CR phenotype.In the study of the interacting features of Rcr1,a cysteine protease gene and its homologous allele in canola were successfully disrupted via CRISPR/Cas9 as an interacting component with Rcr1 protein,resulting in the conversion from clubroot resistant to susceptible in plants carrying intact Rcr1.These results indicated an indispensable role of these two cysteine proteases in Rcr1-mediated resistance response.This study,the first of its kind,provides valuable insights into the functionality of Rcr1.Further,the new vector p HHIGR-Hsp18.2 demonstrated an inducible feature on the removal of add-on traits,which should be useful for functional genomics and other similar research in brassica crops.

文摘The E3 ubiquitin ligase,carboxyl terminus of heat shock protein 70(Hsp70)interacting protein(CHIP),also functions as a co-chaperone and plays a crucial role in the protein quality control system.In this study,we aimed to investigate the neuroprotective effect of overexpressed CHIP on Alzheimer’s disease.We used an adeno-associated virus vector that can cross the blood-brain barrier to mediate CHIP overexpression in APP/PS1 mouse brain.CHIP overexpression significantly ameliorated the performance of APP/PS1 mice in the Morris water maze and nest building tests,reduced amyloid-βplaques,and decreased the expression of both amyloid-βand phosphorylated tau.CHIP also alleviated the concentration of microglia and astrocytes around plaques.In APP/PS1 mice of a younger age,CHIP overexpression promoted an increase in ADAM10 expression and inhibitedβ-site APP cleaving enzyme 1,insulin degrading enzyme,and neprilysin expression.Levels of HSP70 and HSP40,which have functional relevance to CHIP,were also increased.Single nuclei transcriptome sequencing in the hippocampus of CHIP overexpressed mice showed that the lysosomal pathway and oligodendrocyte-related biological processes were up-regulated,which may also reflect a potential mechanism for the neuroprotective effect of CHIP.Our research shows that CHIP effectively reduces the behavior and pathological manifestations of APP/PS1 mice.Indeed,overexpression of CHIP could be a beneficial approach for the treatment of Alzheimer’s disease.

文摘To solve low precision and poor stability of the extended Kalman filter (EKF) in the vehicle integrated positioning system owing to acceleration, deceleration and turning (hereinafter referred to as maneuvering) , the paper presents an adaptive filter algorithm that combines interacting multiple model (IMM) and non linear Kalman filter. The algorithm describes the motion mode of vehicle by using three state spacemode]s. At first, the parallel filter of each model is realized by using multiple nonlinear filters. Then the weight integration of filtering result is carried out by using the model matching likelihood function so as to get the system positioning information. The method has advantages of nonlinear system filter and overcomes disadvantages of single model of filtering algorithm that has poor effects on positioning the maneuvering target. At last, the paper uses IMM and EKF methods to simulate the global positioning system (OPS)/inertial navigation system (INS)/dead reckoning (DR) integrated positioning system, respectively. The results indicate that the IMM algorithm is obviously superior to EKF filter used in the integrated positioning system at present. Moreover, it can greatly enhance the stability and positioning precision of integrated positioning system.
文摘We study the synchronization dynamics in a system of multiple interacting populations of phase oscillators. Using the dimensionality-reduction technique of Ott and Antonsen, we explore different types of synchronization dynamics when the incoherent state becomes unstable. We find that the inter-population coupling is crucial to the synchronization. When the intra-population interaction is repulsive, the local synchronization can still be maintained through the inter-population coupling. For attractive inter-population coupling, the local order parameters in different populations are of in-phase while the local synchronization are of anti-phase for repulsive inter-population coupling.

文摘[Objective] The paper was to obtain host proteins interacting with porcine epidemic diarrhea virus (PEDV) N protein. [Method] The re-combinant vector pGBKT7-N of PEDV N gene was constructed and used as the bait plasmid to screen the proteins interacting with N protein ofPEDV from the cDNA library of porcine alveolar macrophage (PAM) by yeast two-hybrid method. [Result] There was no toxicity and self activationof bait protein in yeast hybridization system, and six proteins (FTH1, LGALS3, CORO1C, SNRPG, KRTAP5-3, ZNF598) interacting with N proteinwere indentified. It was confirmed that LGALS3 and SNRPG had specific interaction with N protein by return experiment and co-immunoprecipitation(CoIP) test. [Conclusion] The study lays a foundation for further studying the function of PEDV N protein and the pathogenic mechanism of PEDV.
文摘The behavior of the quantum correlations, information scrambling and the non-Markovianity of three entangling qubits systems via Rashba is discussed. The results showed that, the three physical quantities oscillate between their upper and lower bounds, where the number of oscillations increases as the Rashba interaction strength increases. The exchanging rate of these three quantities depends on the Rashba strength, and whether the entangled state is generated via direct/indirect interaction. Moreover, the coherence parameter can be used as a control parameter to maximize or minimize the three physical quantities.

文摘This work demonstrates experimentally the close relation between return currents from relativistic laser-driven target polarization and the quality of the relativistic laser–plasma interaction for laser-driven secondary sources,taking as an example ion acceleration by target normal sheath acceleration.The Pearson linear correlation of maximum return current amplitude and proton spectrum cutoff energy is found to be in the range from~0.70 to 0.94.kA-scale return currents rise in all interaction schemes where targets of any kind are charged by escaping laser-accelerated relativistic electrons.Their precise measurement is demonstrated using an inductive scheme that allows operation at high repetition rates.Thus,return currents can be used as a metrological online tool for the optimization of many laser-driven secondary sources and for diagnosing their stability.In particular,in two parametric studies of laser-driven ion acceleration,we carry out a noninvasive online measurement of return currents in a tape target system irradiated by the 1 PW VEGA-3 laser at Centro de Láseres Pulsados:first the size of the irradiated area is varied at best compression of the laser pulse;second,the pulse duration is varied by means of induced group delay dispersion at best focus.This work paves the way to the development of feedback systems that operate at the high repetition rates of PW-class lasers.

文摘FCS-like zinc finger(FLZ)gene family members are C2-C2 zinc finger proteins that take part in seed dormancy,resistance to Myzus persicae 1,sucrose signaling and abiotic stresse tolerance.However,their functions,especially the molecular mechanism through which FLZs function,are not well understood.In this study,we characterized 120FLZs in wheat and revealed the function and mechanism of TaFLZ54D increasing salt stress tolerance in transgenic wheat.Expression analysis demonstrated that TaFLZ54D can be induced by NaCl treatment and it had the highest expression level under NaCl treatment among the 120 FLZs.Over-expression of TaFLZ54D increased wheat salt stress tolerance and the transgenic plants had higher levels of superoxide dismutase(SOD)and peroxidase(POD)activities and soluble sugar content,but a lower Na^(+)/K^(+)ratio and malondialdehyde(MDA)content than the wild type(WT)plants.Potassium ion transmembrane transporters and serine/threonine kinase inhibitor proteins showed differential expression between Ta FLZ54D transgenic wheat and the WT.Yeast two hybrid and luciferase complementation assays revealed that TaSGT1 and TaPP2C are the proteins that interact directly with TaFLZ54D.In summary,TaFLZ54D enhances salt stress tolerance through interaction with TaSGT1 and TaPP2C to reduce Na^(+)absorption and mitigate oxidative stress.The interaction between TaFLZ54D and TaSGT1,as well as TaPP2C indicated a link between salt stress tolerance of TaFLZ54D and the ubiquitin-mediated degradation of negative regulatory proteins.

文摘Food systems are deeply affected by climate change and air pollution,while being key contributors to these environmental challenges.Understanding the complex interactions among food systems,climate change,and air pollution is crucial for mitigating climate change,improving air quality,and promoting the sustainable development of food systems.However,the literature lacks a comprehensive review of these interactions,particularly in the current phase of rapid development in the field.To address this gap,this study systematically reviews recent research on the impacts of climate change and air pollution on food systems,as well as the greenhouse gas and air pollutant emissions from agri-food systems and their contribution to global climate change and air pollution.In addition,this study summarizes various strategies for mitigation and adaptation,including adjustments in agricultural practices and food supply chains.Profound changes in food systems are urgently needed to enhance adaptability and reduce emissions.This review offers a critical overview of current research on the interactions among food systems,climate change,and air pollution and highlights future research directions to support the transition to sustainable food systems.
