Piezo2,a mechanosensitive ion channel,serves as a crucial mechanotransducer in dental primary afferent(DPA)neurons and is potentially involved in hypersensitivity to mild mechanical irritations observed in dental pati...Piezo2,a mechanosensitive ion channel,serves as a crucial mechanotransducer in dental primary afferent(DPA)neurons and is potentially involved in hypersensitivity to mild mechanical irritations observed in dental patients.Given Piezo2’s widespread expression across diverse subpopulations of DPA neurons,this study aimed to characterize the mechanosensory properties of Piezo2-expressing DPA neurons with a focus on distinct features of voltage-gated sodium channels(VGSCs)and neuropeptide profiles.Using whole-cell patch-clamp recordings,we observed mechanically activated action potentials(APs)and classified AP waveforms based on the presence or absence of a hump during the repolarization phase.Single-cell reverse transcription polymerase chain reaction combined with patch-clamp recordings revealed specific associations between AP waveforms and molecular properties,including tetrodotoxin-resistant VGSCs(NaV1.8 and NaV1.9)and TRPV1 expression.Reanalysis of the transcriptomic dataset of DPA neurons identified correlations between neuropeptides—including two CGRP isoforms(α-CGRP andβ-CGRP),Substance P,and Galanin—and the expression of NaV1.8 and NaV1.9,which were linked to defined AP subtypes.These molecular associations were further validated in Piezo2+DPA neurons using fluorescence in situ hybridization.Together,these findings highlight the electrophysiological and neurochemical heterogeneity of Piezo2-expressing DPA neurons and their specialized roles in distinct mechanosensory signal transmission.展开更多
The strategic design and synthesis of photothermal/photocatalytic materials are pivotal to realizing photothermal conversion water evaporation coupled with photocatalytic sewage purification functions.In this work,bas...The strategic design and synthesis of photothermal/photocatalytic materials are pivotal to realizing photothermal conversion water evaporation coupled with photocatalytic sewage purification functions.In this work,based on the principle of three primary colors,brick-red g-C_(3)N_(4)/Ag_(2)CrO_(4)composite was loaded onto a green polyurethane(PU)sponge using polyvinyl alcohol(PVA)as the linking agent.The resultant PU/PVA/g-C_(3)N_(4)/Ag_(2)CrO_(4)composite exhibits outstanding performance in simultaneous photothermal/photocatalytic water evaporation,pollutant degradation,sterilization,and thermoelectric generation.Under 1.0 kW m^(-2)irradiation,the water evaporation rate reaches 3.19 kg m^(-2)h-1,while a single thermoelectric module generates a maximum thermoelectric output power of 0.25 W m^(-2).Concurrently,rhodamine B(RhB)at a concentration of 4.0×10^(-4)mol L^(-1)undergoes complete photocatalytic degradation within 40 min.When the light intensity is 2.0 kW m^(-2),the evaporation rate soars to 8.52 kg m^(-2)h^(-1),and the thermoelectric power output increases to 1.1 W m^(-2).Furthermore,this photothermal/photocatalytic material based on the principle of three primary colors has excellent photothermal/photocatalytic antibacterial activity against Escherichia coli.By abandoning black light-absorbing materials,more active sites of the photocatalyst can be exposed.The g-C_(3)N_(4)/Ag_(2)CrO_(4)heterojunction accelerates the separation of photogenerated carriers,while the hydrophilic groups in the photothermal/photocatalytic materials reduce the water evaporation enthalpy.This research provides a novel approach for fabricating multi-function photothermal/photocatalytic materials,which could quicken the development of solution to freshwater and electricity energy shortages as well as environmental pollution issues.展开更多
Schwann cell transplantation is considered one of the most promising cell-based therapy to repair injured spinal cord due to its unique growth-promoting and myelin-forming properties.A the Food and Drug Administration...Schwann cell transplantation is considered one of the most promising cell-based therapy to repair injured spinal cord due to its unique growth-promoting and myelin-forming properties.A the Food and Drug Administration-approved Phase I clinical trial has been conducted to evaluate the safety of transplanted human autologous Schwann cells to treat patients with spinal cord injury.A major challenge for Schwann cell transplantation is that grafted Schwann cells are confined within the lesion cavity,and they do not migrate into the host environment due to the inhibitory barrier formed by injury-induced glial scar,thus limiting axonal reentry into the host spinal cord.Here we introduce a combinatorial strategy by suppressing the inhibitory extracellular environment with injection of lentivirus-mediated transfection of chondroitinase ABC gene at the rostral and caudal borders of the lesion site and simultaneously leveraging the repair capacity of transplanted Schwann cells in adult rats following a mid-thoracic contusive spinal cord injury.We report that when the glial scar was degraded by chondroitinase ABC at the rostral and caudal lesion borders,Schwann cells migrated for considerable distances in both rostral and caudal directions.Such Schwann cell migration led to enhanced axonal regrowth,including the serotonergic and dopaminergic axons originating from supraspinal regions,and promoted recovery of locomotor and urinary bladder functions.Importantly,the Schwann cell survival and axonal regrowth persisted up to 6 months after the injury,even when treatment was delayed for 3 months to mimic chronic spinal cord injury.These findings collectively show promising evidence for a combinatorial strategy with chondroitinase ABC and Schwann cells in promoting remodeling and recovery of function following spinal cord injury.展开更多

This study investigated the impact of rehabilitation exercise on functional recovery and morphological changes following thoracic contusive spinal cord injury. After a 7-day recovery period after spinal cord injury, mice were assigned to either a trained group(10 weeks of voluntary running wheel or forced treadmill exercise) or an untrained group. Bi-weekly assessments revealed that the exercise-trained group, particularly the voluntary wheel exercise subgroup, displayed significantly improved locomotor recovery, more plasticity of dopaminergic and serotonin modulation compared with the untrained group. Additionally, exercise interventions led to gait pattern restoration and enhanced transcranial magnetic motor-evoked potentials. Despite consistent injury areas across groups, exercise training promoted terminal innervation of descending axons. In summary, voluntary wheel exercise shows promise for enhancing outcomes after thoracic contusive spinal cord injury, emphasizing the role of exercise modality in promoting recovery and morphological changes in spinal cord injuries. Our findings will influence future strategies for rehabilitation exercises, restoring functional movement after spinal cord injury.展开更多
The M1/M2 phenotypic shift of microglia after spinal cord injury plays an important role in the regulation of neuroinflammation during the secondary injury phase of spinal cord injury.Regulation of shifting microglia ...The M1/M2 phenotypic shift of microglia after spinal cord injury plays an important role in the regulation of neuroinflammation during the secondary injury phase of spinal cord injury.Regulation of shifting microglia polarization from M1(neurotoxic and proinflammatory type)to M2(neuroprotective and anti-inflammatory type)after spinal cord injury appears to be crucial.Tryptanthrin possesses an anti-inflammatory biological function.However,its roles and the underlying molecular mechanisms in spinal cord injury remain unknown.In this study,we found that tryptanthrin inhibited microglia-derived inflammation by promoting polarization to the M2 phenotype in vitro.Tryptanthrin promoted M2 polarization through inactivating the cGAS/STING/NF-κB pathway.Additionally,we found that targeting the cGAS/STING/NF-κB pathway with tryptanthrin shifted microglia from the M1 to M2 phenotype after spinal cord injury,inhibited neuronal loss,and promoted tissue repair and functional recovery in a mouse model of spinal cord injury.Finally,using a conditional co-culture system,we found that microglia treated with tryptanthrin suppressed endoplasmic reticulum stress-related neuronal apoptosis.Taken together,these results suggest that by targeting the cGAS/STING/NF-κB axis,tryptanthrin attenuates microglia-derived neuroinflammation and promotes functional recovery after spinal cord injury through shifting microglia polarization to the M2 phenotype.展开更多

BACKGROUND Primary biliary cholangitis(PBC)is a chronic autoimmune-mediated cholestatic liver disease.Nanoparticles encapsulating rapamycin(ImmTOR)suppress adaptive immune responses and induce the hepatic tolerogenic ...BACKGROUND Primary biliary cholangitis(PBC)is a chronic autoimmune-mediated cholestatic liver disease.Nanoparticles encapsulating rapamycin(ImmTOR)suppress adaptive immune responses and induce the hepatic tolerogenic immune response.AIM To investigate the effects of ImmTOR in PBC mouse models.METHODS PBC models were induced in C57BL/6 mice by two immunizations of 2-octynoic acid-coupled bovine serum albumin at two-week intervals,and polycytidylic acid every three days.The PBC mouse models were separated into the treatment group and the control group.The levels of alkaline phosphatase(ALP)and alanine aminotransferase in the mice were detected using an automatic biochemical analyzer.Liver and spleen mononuclear cells were analyzed by flow cytometry,and serum anti-mitochondrial antibodies(AMA)and the related cytokines were analyzed by enzyme-linked immunosorbent assay.Liver histopathology was examined by hematoxylin and eosin staining and scored.RESULTS After treatment with ImmTOR,the ALP level was significantly decreased(189.60 U/L±27.25 U/L vs 156.00 U/L±17.21 U/L,P<0.05),the level of AMA was reduced(1.28 ng/mL±0.27 ng/mL vs 0.56 ng/mL±0.07 ng/mL,P<0.001)and the expression levels of interferon gamma and tumor necrosis factorαwere significantly decreased(48.29 pg/mL±10.84 pg/mL vs 25.01 pg/mL±1.49 pg/mL,P<0.0001)and(84.24 pg/mL±23.47 pg/mL vs 40.66 pg/mL±14.65 pg/mL,P<0.001).The CD4+T lymphocytes,CD8+T lymphocytes and B lymphocytes in the liver were significantly reduced,with statistically significant differences(24.21%±6.55%vs 15.98%±3.03%,P<0.05;9.09%±1.91%vs 5.49%±1.00%,P<0.001;80.51%±2.96%vs 75.31%±4.34%,P<0.05).The expression of CD8+T lymphocytes and B lymphocytes in the ImmTOR treatment group also decreased(9.09%±1.91%vs 5.49%±1.00%,P<0.001;80.51%±2.96%vs 75.31%±4.34%,P<0.05).The liver pathology of PBC mice in the treatment group showed reduced inflammation and a decreased total pathology score,and the difference in the scores was statistically significant(4.50±2.88 vs 1.75±1.28,P<0.05).CONCLUSION ImmTOR can improve biochemistry and pathology of liver obvious by inhibiting the expression of CD8+T cells and B cells,and reducing the titer of AMA.展开更多

In clinical practice, effective prognostic tools for early identifying recipien...Background: Primary non-function(PNF) and early allograft failure(EAF) after liver transplantation(LT) seriously affect patient outcomes. In clinical practice, effective prognostic tools for early identifying recipients at high risk of PNF and EAF were urgently needed. Recently, the Model for Early Allograft Function(MEAF), PNF score by King's College(King-PNF) and Balance-and-Risk-Lactate(BAR-Lac) score were developed to assess the risks of PNF and EAF. This study aimed to externally validate and compare the prognostic performance of these three scores for predicting PNF and EAF. Methods: A retrospective study included 720 patients with primary LT between January 2015 and December 2020. MEAF, King-PNF and BAR-Lac scores were compared using receiver operating characteristic(ROC) and the net reclassification improvement(NRI) and integrated discrimination improvement(IDI) analyses. Results: Of all 720 patients, 28(3.9%) developed PNF and 67(9.3%) developed EAF in 3 months. The overall early allograft dysfunction(EAD) rate was 39.0%. The 3-month patient mortality was 8.6% while 1-year graft-failure-free survival was 89.2%. The median MEAF, King-PNF and BAR-Lac scores were 5.0(3.5–6.3),-2.1(-2.6 to-1.2), and 5.0(2.0–11.0), respectively. For predicting PNF, MEAF and King-PNF scores had excellent area under curves(AUCs) of 0.872 and 0.891, superior to BAR-Lac(AUC = 0.830). The NRI and IDI analyses confirmed that King-PNF score had the best performance in predicting PNF while MEAF served as a better predictor of EAD. The EAF risk curve and 1-year graft-failure-free survival curve showed that King-PNF was superior to MEAF and BAR-Lac scores for stratifying the risk of EAF. Conclusions: MEAF, King-PNF and BAR-Lac were validated as practical and effective risk assessment tools of PNF. King-PNF score outperformed MEAF and BAR-Lac in predicting PNF and EAF within 6 months. BAR-Lac score had a huge advantage in the prediction for PNF without post-transplant variables. Proper use of these scores will help early identify PNF, standardize grading of EAF and reasonably select clinical endpoints in relative studies.展开更多

The Red Sea-Gulf of Suez-Cairo-Alexandria Clysmic-Trend in northern Egypt is the main earthquake zone in the country,with a moderate-to-high seismic hazard and a history of significant earthquakes caused by rifting an...The Red Sea-Gulf of Suez-Cairo-Alexandria Clysmic-Trend in northern Egypt is the main earthquake zone in the country,with a moderate-to-high seismic hazard and a history of significant earthquakes caused by rifting and active faulting.To improve our understanding of the tectonic and seismic processes in this area,more comprehensive imaging of the crustal structure is required.This can be achieved by increasing the number of receiver functions(RFs)recorded by the seismic stations in northern Egypt and the southeastern Mediterranean.Data handling and processing should also be automated to increase process efficiency.In this study,we developed a capsule neural network for automated selection of RFs.The model was trained on a dataset containing RFs(both selected and unselected)from five broadband stations in northern Egypt.Stations SLM,SIWA,KOT,NBNS,and NKL are located in the unstable shelf region of Egypt,where limited knowledge of the deep crustal structure is available.The proposed capsule neural network achieved an average precision of 80%on the test set.The automated selection of RFs using a capsule neural network has the potential to significantly improve the efficiency and accuracy of RF analysis,as demonstrated by the stacking test.This could lead to a better understanding of crustal structure and tectonic processes in northern Egypt and the southeastern Mediterranean.展开更多

It is well known that coarse-grained super-elastic NiTi shape memory alloys(SMAs)exhibit localized rather than homogeneous martensite transformation(MT),which,however,can be strongly influenced by either internal size...It is well known that coarse-grained super-elastic NiTi shape memory alloys(SMAs)exhibit localized rather than homogeneous martensite transformation(MT),which,however,can be strongly influenced by either internal size(grain size,GS)or the external size(geometric size).The coupled effect of GS and geometric size on the functional properties has not been clearly understood yet.In this work,the super-elasticity,one-way,and stress-assisted two-way shape memory effects of the polycrystalline NiTi SMAs with different aspect ratios(length/width for the gauge section)and different GSs are investigated based on the phase field method.The coupled effect of the aspect ratio and GS on the functional properties is adequately revealed.The simulated results indicate that when the aspect ratio is lower than about 4:1,the stress biaxiality and stress heterogeneity in the gauge section of the sample become more and more obvious with decreasing the aspect ratio,which can significantly influence the microstructure evolution in the process involving external stress.Therefore,the corresponding functional property is strongly dependent on the aspect ratio.With decreasing the GS and the aspect ratio(to be lower than 4:1),both the aspect ratio and GS can affect the MT or martensite reorientation in each grain and the interaction among grains.Thus,due to the strong internal constraint(i.e.,the constraint of grain boundary)and the external constraint(i.e.,the constraint of geometric boundary),the capabilities of the functional properties of NiTi SMAs are gradually weakened and highly dependent on these two factors.展开更多
During the initial impoundment period of a canyon-shaped reservoir,the water body fluctuated violently regarding water level,hydrological condition,and thermal stratification.These variations may alter the structure o...During the initial impoundment period of a canyon-shaped reservoir,the water body fluctuated violently regarding water level,hydrological condition,and thermal stratification.These variations may alter the structure of phytoplankton community,resulting in algal blooms and seriously threatening the ecological security of the reservoir.It is of great significance to understand the continuous changes of phytoplankton in the initial impoundment period for the protection of reservoir water quality.Therefore,a two-year in-situ monitoring study was conducted on water quality and phytoplankton in a representative canyonshaped reservoir named Sanhekou and the interannual changes of phytoplankton community and its response to environmental changes during the initial impoundment period were discussed at taxonomic versus functional classification levels.The results showed that the total nitrogen and permanganate index levels were relatively high in the first year due to rapid water storage and heavy rainfall input,and the more stable hydrological conditions in the second year promoted the increase of algae density and the transformation of community,and the proportion of cyanobacteria increased significantly.The succession order of phytoplankton in the first year of the initial impoundment periodwas Chlorophyta-Bacillariophyta-Chlorophyta,or J/F/X1-P/MP/W1-A/X1/MP,respectively.And the succession order in the second year was Cyanobacteria/Chlorophyta-Bacillariophyta-Chlorophyta,or L_(M)/G/P-P/A/X1-X1/J/G.Water temperature,relativewater column stability,mixing depth,and pHwere crucial factors affecting phytoplankton community succession.This study revealed the interannual succession law and driving factors of phytoplankton in the initial impoundment period and provided an important reference for the operation management and ecological protection of canyon-shaped reservoirs.展开更多
Objective:To explore the effect of transarterial chemoembolization(TACE)+CT-guided microwave ablation(MWA)on treating patients with primary liver cancer.Methods:78 primary liver cancer cases were enrolled and divided ...Objective:To explore the effect of transarterial chemoembolization(TACE)+CT-guided microwave ablation(MWA)on treating patients with primary liver cancer.Methods:78 primary liver cancer cases were enrolled and divided into groups according to their assigned surgical plans.The control group was treated with TACE alone,and the observation group was treated with TACE+CT-guided MWA.The efficacy of the treatment and the liver function indicators and follow-up results of the patients of the two groups were compared.Results:The efficacy of the treatment received by the observation group was higher than that of the control group.Besides,the patients in the observation group exhibited better improvement in liver function indicators after 3 months of treatment.Furthermore,the survival rates of 1 and 2 years after surgery of the observation group were all higher than those of the control group(P<0.05).Conclusion:TACE combined with CT-guided MWA is more effective in treating primary liver cancer compared to TACE alone.Besides,it resulted in better improvement of liver function and long-term survival rate.Therefore,this treatment regime should be popularized.展开更多

文摘Piezo2,a mechanosensitive ion channel,serves as a crucial mechanotransducer in dental primary afferent(DPA)neurons and is potentially involved in hypersensitivity to mild mechanical irritations observed in dental patients.Given Piezo2’s widespread expression across diverse subpopulations of DPA neurons,this study aimed to characterize the mechanosensory properties of Piezo2-expressing DPA neurons with a focus on distinct features of voltage-gated sodium channels(VGSCs)and neuropeptide profiles.Using whole-cell patch-clamp recordings,we observed mechanically activated action potentials(APs)and classified AP waveforms based on the presence or absence of a hump during the repolarization phase.Single-cell reverse transcription polymerase chain reaction combined with patch-clamp recordings revealed specific associations between AP waveforms and molecular properties,including tetrodotoxin-resistant VGSCs(NaV1.8 and NaV1.9)and TRPV1 expression.Reanalysis of the transcriptomic dataset of DPA neurons identified correlations between neuropeptides—including two CGRP isoforms(α-CGRP andβ-CGRP),Substance P,and Galanin—and the expression of NaV1.8 and NaV1.9,which were linked to defined AP subtypes.These molecular associations were further validated in Piezo2+DPA neurons using fluorescence in situ hybridization.Together,these findings highlight the electrophysiological and neurochemical heterogeneity of Piezo2-expressing DPA neurons and their specialized roles in distinct mechanosensory signal transmission.

文摘The strategic design and synthesis of photothermal/photocatalytic materials are pivotal to realizing photothermal conversion water evaporation coupled with photocatalytic sewage purification functions.In this work,based on the principle of three primary colors,brick-red g-C_(3)N_(4)/Ag_(2)CrO_(4)composite was loaded onto a green polyurethane(PU)sponge using polyvinyl alcohol(PVA)as the linking agent.The resultant PU/PVA/g-C_(3)N_(4)/Ag_(2)CrO_(4)composite exhibits outstanding performance in simultaneous photothermal/photocatalytic water evaporation,pollutant degradation,sterilization,and thermoelectric generation.Under 1.0 kW m^(-2)irradiation,the water evaporation rate reaches 3.19 kg m^(-2)h-1,while a single thermoelectric module generates a maximum thermoelectric output power of 0.25 W m^(-2).Concurrently,rhodamine B(RhB)at a concentration of 4.0×10^(-4)mol L^(-1)undergoes complete photocatalytic degradation within 40 min.When the light intensity is 2.0 kW m^(-2),the evaporation rate soars to 8.52 kg m^(-2)h^(-1),and the thermoelectric power output increases to 1.1 W m^(-2).Furthermore,this photothermal/photocatalytic material based on the principle of three primary colors has excellent photothermal/photocatalytic antibacterial activity against Escherichia coli.By abandoning black light-absorbing materials,more active sites of the photocatalyst can be exposed.The g-C_(3)N_(4)/Ag_(2)CrO_(4)heterojunction accelerates the separation of photogenerated carriers,while the hydrophilic groups in the photothermal/photocatalytic materials reduce the water evaporation enthalpy.This research provides a novel approach for fabricating multi-function photothermal/photocatalytic materials,which could quicken the development of solution to freshwater and electricity energy shortages as well as environmental pollution issues.

文摘Schwann cell transplantation is considered one of the most promising cell-based therapy to repair injured spinal cord due to its unique growth-promoting and myelin-forming properties.A the Food and Drug Administration-approved Phase I clinical trial has been conducted to evaluate the safety of transplanted human autologous Schwann cells to treat patients with spinal cord injury.A major challenge for Schwann cell transplantation is that grafted Schwann cells are confined within the lesion cavity,and they do not migrate into the host environment due to the inhibitory barrier formed by injury-induced glial scar,thus limiting axonal reentry into the host spinal cord.Here we introduce a combinatorial strategy by suppressing the inhibitory extracellular environment with injection of lentivirus-mediated transfection of chondroitinase ABC gene at the rostral and caudal borders of the lesion site and simultaneously leveraging the repair capacity of transplanted Schwann cells in adult rats following a mid-thoracic contusive spinal cord injury.We report that when the glial scar was degraded by chondroitinase ABC at the rostral and caudal lesion borders,Schwann cells migrated for considerable distances in both rostral and caudal directions.Such Schwann cell migration led to enhanced axonal regrowth,including the serotonergic and dopaminergic axons originating from supraspinal regions,and promoted recovery of locomotor and urinary bladder functions.Importantly,the Schwann cell survival and axonal regrowth persisted up to 6 months after the injury,even when treatment was delayed for 3 months to mimic chronic spinal cord injury.These findings collectively show promising evidence for a combinatorial strategy with chondroitinase ABC and Schwann cells in promoting remodeling and recovery of function following spinal cord injury.

文摘Spinal cord injury necessitates effective rehabilitation strategies, with exercise therapies showing promise in promoting recovery. This study investigated the impact of rehabilitation exercise on functional recovery and morphological changes following thoracic contusive spinal cord injury. After a 7-day recovery period after spinal cord injury, mice were assigned to either a trained group(10 weeks of voluntary running wheel or forced treadmill exercise) or an untrained group. Bi-weekly assessments revealed that the exercise-trained group, particularly the voluntary wheel exercise subgroup, displayed significantly improved locomotor recovery, more plasticity of dopaminergic and serotonin modulation compared with the untrained group. Additionally, exercise interventions led to gait pattern restoration and enhanced transcranial magnetic motor-evoked potentials. Despite consistent injury areas across groups, exercise training promoted terminal innervation of descending axons. In summary, voluntary wheel exercise shows promise for enhancing outcomes after thoracic contusive spinal cord injury, emphasizing the role of exercise modality in promoting recovery and morphological changes in spinal cord injuries. Our findings will influence future strategies for rehabilitation exercises, restoring functional movement after spinal cord injury.

文摘The M1/M2 phenotypic shift of microglia after spinal cord injury plays an important role in the regulation of neuroinflammation during the secondary injury phase of spinal cord injury.Regulation of shifting microglia polarization from M1(neurotoxic and proinflammatory type)to M2(neuroprotective and anti-inflammatory type)after spinal cord injury appears to be crucial.Tryptanthrin possesses an anti-inflammatory biological function.However,its roles and the underlying molecular mechanisms in spinal cord injury remain unknown.In this study,we found that tryptanthrin inhibited microglia-derived inflammation by promoting polarization to the M2 phenotype in vitro.Tryptanthrin promoted M2 polarization through inactivating the cGAS/STING/NF-κB pathway.Additionally,we found that targeting the cGAS/STING/NF-κB pathway with tryptanthrin shifted microglia from the M1 to M2 phenotype after spinal cord injury,inhibited neuronal loss,and promoted tissue repair and functional recovery in a mouse model of spinal cord injury.Finally,using a conditional co-culture system,we found that microglia treated with tryptanthrin suppressed endoplasmic reticulum stress-related neuronal apoptosis.Taken together,these results suggest that by targeting the cGAS/STING/NF-κB axis,tryptanthrin attenuates microglia-derived neuroinflammation and promotes functional recovery after spinal cord injury through shifting microglia polarization to the M2 phenotype.

文摘Very long chain-saturated and-polyunsaturated fatty acids(VLC-SFA and VLC-PUFA, respectively) are a functionally important class of fatty acids containing 28 carbons or more in their acyl chain. They are synthesized by the elongation of very long fatty acids-4(ELOVL4) enzyme, expressed mainly in the brain, retina, skin, testes, and meibomian gland, where these fatty acids are found(Agbaga et al., 2008). Further, these organs exhibit tissuespecific VLC-PUFA and VLC-SFA biosynthesis and incorporation into complex lipids for specific functions. In the brain, skin, and Meibomian glands, the ELOVL4 mainly makes VLC-SFA, which are incorporated into complex sphingolipids. In the retina, the ELOVL4 makes VLC-PUFA that are incorporated into phosphatidylcholine, that are critical for visual function, while in testes and sperm, the VLC-PUFA are incorporated into sphingolipids that are critical for fertility(Yeboah et al., 2021).

文摘BACKGROUND The incidence of malignant gastrointestinal(GI)tumors is increasing,and advancements in medical care have significantly improved patient survival rates.As a result,the number of cases involving multiple primary cancers(MPC)has also increased.The rarity of MPC and the absence of sensitive and specific dia-gnostic markers often lead to missed or incorrect diagnoses.It is,therefore,of vital importance to improve the vigilance of clinicians and the accurate diagnosis of this disease.Patients with GI malignancies face a higher relative risk of deve-loping additional primary malignant tumors compared to those with other systemic tumors.Vigilant monitoring and follow-up are crucial,especially for high-risk groups,which include older adults,men,those with addictions to alcohol and tobacco,those with a family history of tumors,and those who have undergone radiotherapy.CASE SUMMARY In this article,we report three cases of MPC,each involving malignant tumors of the GI tract as the initial primary carcinoma,offering insights that may aid in effectively managing similar cases.CONCLUSION Patients with GI malignancies face a higher MPC risk.Developing screening and follow-up protocols may enhance detection and treatment outcomes.

文摘BACKGROUND Primary biliary cholangitis(PBC)is a chronic autoimmune-mediated cholestatic liver disease.Nanoparticles encapsulating rapamycin(ImmTOR)suppress adaptive immune responses and induce the hepatic tolerogenic immune response.AIM To investigate the effects of ImmTOR in PBC mouse models.METHODS PBC models were induced in C57BL/6 mice by two immunizations of 2-octynoic acid-coupled bovine serum albumin at two-week intervals,and polycytidylic acid every three days.The PBC mouse models were separated into the treatment group and the control group.The levels of alkaline phosphatase(ALP)and alanine aminotransferase in the mice were detected using an automatic biochemical analyzer.Liver and spleen mononuclear cells were analyzed by flow cytometry,and serum anti-mitochondrial antibodies(AMA)and the related cytokines were analyzed by enzyme-linked immunosorbent assay.Liver histopathology was examined by hematoxylin and eosin staining and scored.RESULTS After treatment with ImmTOR,the ALP level was significantly decreased(189.60 U/L±27.25 U/L vs 156.00 U/L±17.21 U/L,P<0.05),the level of AMA was reduced(1.28 ng/mL±0.27 ng/mL vs 0.56 ng/mL±0.07 ng/mL,P<0.001)and the expression levels of interferon gamma and tumor necrosis factorαwere significantly decreased(48.29 pg/mL±10.84 pg/mL vs 25.01 pg/mL±1.49 pg/mL,P<0.0001)and(84.24 pg/mL±23.47 pg/mL vs 40.66 pg/mL±14.65 pg/mL,P<0.001).The CD4+T lymphocytes,CD8+T lymphocytes and B lymphocytes in the liver were significantly reduced,with statistically significant differences(24.21%±6.55%vs 15.98%±3.03%,P<0.05;9.09%±1.91%vs 5.49%±1.00%,P<0.001;80.51%±2.96%vs 75.31%±4.34%,P<0.05).The expression of CD8+T lymphocytes and B lymphocytes in the ImmTOR treatment group also decreased(9.09%±1.91%vs 5.49%±1.00%,P<0.001;80.51%±2.96%vs 75.31%±4.34%,P<0.05).The liver pathology of PBC mice in the treatment group showed reduced inflammation and a decreased total pathology score,and the difference in the scores was statistically significant(4.50±2.88 vs 1.75±1.28,P<0.05).CONCLUSION ImmTOR can improve biochemistry and pathology of liver obvious by inhibiting the expression of CD8+T cells and B cells,and reducing the titer of AMA.

文摘Five-valued Boolean functions play an important role in the design of symmetric cryptography.While the design and properties of single-output almost optimal five-valued spectra Boolean functions have been extensively studied over the past few decades,there has been limited research on the construction of almost optimal five-valued spectra vectorial Boolean functions.In this paper,we present a construction method for even-variable 2-output almost optimal five-valued spectra balanced Boolean functions,whose Walsh spectra values belong to the set{0,±2^(n/2),±2^(n/2+1)},at the same time,we discuss the existence of sufficient conditions in the construction.Additionally,this paper presents a novel construction method for balanced single-output Boolean functions with even variables featuring a special five-valued spectral structure,whose Walsh spectra values are constrained to the set{0,±2^(n/2),±3·2^(n/2)}.These functions provide new canonical examples for the study of Boolean function spectral theory.

文摘Background: Primary non-function(PNF) and early allograft failure(EAF) after liver transplantation(LT) seriously affect patient outcomes. In clinical practice, effective prognostic tools for early identifying recipients at high risk of PNF and EAF were urgently needed. Recently, the Model for Early Allograft Function(MEAF), PNF score by King's College(King-PNF) and Balance-and-Risk-Lactate(BAR-Lac) score were developed to assess the risks of PNF and EAF. This study aimed to externally validate and compare the prognostic performance of these three scores for predicting PNF and EAF. Methods: A retrospective study included 720 patients with primary LT between January 2015 and December 2020. MEAF, King-PNF and BAR-Lac scores were compared using receiver operating characteristic(ROC) and the net reclassification improvement(NRI) and integrated discrimination improvement(IDI) analyses. Results: Of all 720 patients, 28(3.9%) developed PNF and 67(9.3%) developed EAF in 3 months. The overall early allograft dysfunction(EAD) rate was 39.0%. The 3-month patient mortality was 8.6% while 1-year graft-failure-free survival was 89.2%. The median MEAF, King-PNF and BAR-Lac scores were 5.0(3.5–6.3),-2.1(-2.6 to-1.2), and 5.0(2.0–11.0), respectively. For predicting PNF, MEAF and King-PNF scores had excellent area under curves(AUCs) of 0.872 and 0.891, superior to BAR-Lac(AUC = 0.830). The NRI and IDI analyses confirmed that King-PNF score had the best performance in predicting PNF while MEAF served as a better predictor of EAD. The EAF risk curve and 1-year graft-failure-free survival curve showed that King-PNF was superior to MEAF and BAR-Lac scores for stratifying the risk of EAF. Conclusions: MEAF, King-PNF and BAR-Lac were validated as practical and effective risk assessment tools of PNF. King-PNF score outperformed MEAF and BAR-Lac in predicting PNF and EAF within 6 months. BAR-Lac score had a huge advantage in the prediction for PNF without post-transplant variables. Proper use of these scores will help early identify PNF, standardize grading of EAF and reasonably select clinical endpoints in relative studies.

文摘Primary ciliary dyskinesia(PCD)is a clinically rare,genetically and phenotypically heterogeneous condition characterized by chronic respiratory tract infections,male infertility,tympanitis,and laterality abnormalities.PCD is typically resulted from variants in genes encoding assembly or structural proteins that are indispensable for the movement of motile cilia.Here,we identified a novel nonsense mutation,c.466G>T,in cilia-and flagella-associated protein 300(CFAP300)resulting in a stop codon(p.Glu156*)through whole-exome sequencing(WES).The proband had a PCD phenotype with laterality defects and immotile sperm flagella displaying a combined loss of the inner dynein arm(IDA)and outer dynein arm(ODA).Bioinformatic programs predicted that the mutation is deleterious.Successful pregnancy was achieved through intracytoplasmic sperm injection(ICSI).Our results expand the spectrum of CFAP300 variants in PCD and provide reproductive guidance for infertile couples suffering from PCD caused by them.

文摘The Red Sea-Gulf of Suez-Cairo-Alexandria Clysmic-Trend in northern Egypt is the main earthquake zone in the country,with a moderate-to-high seismic hazard and a history of significant earthquakes caused by rifting and active faulting.To improve our understanding of the tectonic and seismic processes in this area,more comprehensive imaging of the crustal structure is required.This can be achieved by increasing the number of receiver functions(RFs)recorded by the seismic stations in northern Egypt and the southeastern Mediterranean.Data handling and processing should also be automated to increase process efficiency.In this study,we developed a capsule neural network for automated selection of RFs.The model was trained on a dataset containing RFs(both selected and unselected)from five broadband stations in northern Egypt.Stations SLM,SIWA,KOT,NBNS,and NKL are located in the unstable shelf region of Egypt,where limited knowledge of the deep crustal structure is available.The proposed capsule neural network achieved an average precision of 80%on the test set.The automated selection of RFs using a capsule neural network has the potential to significantly improve the efficiency and accuracy of RF analysis,as demonstrated by the stacking test.This could lead to a better understanding of crustal structure and tectonic processes in northern Egypt and the southeastern Mediterranean.

文摘Calcium ferrite(CF)is recognized as a potential green and efficient functional material because of its advantages of magnetism,electrochemistry,catalysis,and biocompatibility in the fields of materials chemistry,environmental engineering,and biomedicine.There-fore,the obtained research results need to be systematically summarized,and new perspectives on CF and its composite materials need to be analyzed.Based on the presented studies of CF and its composite materials,the types and structures of the crystal are summarized.In addition,the current application technologies and theoretical mechanisms with various properties in different fields are elucidated.Moreover,the various preparation methods of CF and its composite materials are elaborated in detail.Most importantly,the advantages and disadvantages of the synthesis methods of CF and its composite materials are discussed,and the existing problems and emerging challenges in practical production are identified.Furthermore,the key future research directions of CF and its composite materials have been prospected from the potential application technologies to provide references for its synthesis and efficient utilization.

文摘It is well known that coarse-grained super-elastic NiTi shape memory alloys(SMAs)exhibit localized rather than homogeneous martensite transformation(MT),which,however,can be strongly influenced by either internal size(grain size,GS)or the external size(geometric size).The coupled effect of GS and geometric size on the functional properties has not been clearly understood yet.In this work,the super-elasticity,one-way,and stress-assisted two-way shape memory effects of the polycrystalline NiTi SMAs with different aspect ratios(length/width for the gauge section)and different GSs are investigated based on the phase field method.The coupled effect of the aspect ratio and GS on the functional properties is adequately revealed.The simulated results indicate that when the aspect ratio is lower than about 4:1,the stress biaxiality and stress heterogeneity in the gauge section of the sample become more and more obvious with decreasing the aspect ratio,which can significantly influence the microstructure evolution in the process involving external stress.Therefore,the corresponding functional property is strongly dependent on the aspect ratio.With decreasing the GS and the aspect ratio(to be lower than 4:1),both the aspect ratio and GS can affect the MT or martensite reorientation in each grain and the interaction among grains.Thus,due to the strong internal constraint(i.e.,the constraint of grain boundary)and the external constraint(i.e.,the constraint of geometric boundary),the capabilities of the functional properties of NiTi SMAs are gradually weakened and highly dependent on these two factors.

文摘During the initial impoundment period of a canyon-shaped reservoir,the water body fluctuated violently regarding water level,hydrological condition,and thermal stratification.These variations may alter the structure of phytoplankton community,resulting in algal blooms and seriously threatening the ecological security of the reservoir.It is of great significance to understand the continuous changes of phytoplankton in the initial impoundment period for the protection of reservoir water quality.Therefore,a two-year in-situ monitoring study was conducted on water quality and phytoplankton in a representative canyonshaped reservoir named Sanhekou and the interannual changes of phytoplankton community and its response to environmental changes during the initial impoundment period were discussed at taxonomic versus functional classification levels.The results showed that the total nitrogen and permanganate index levels were relatively high in the first year due to rapid water storage and heavy rainfall input,and the more stable hydrological conditions in the second year promoted the increase of algae density and the transformation of community,and the proportion of cyanobacteria increased significantly.The succession order of phytoplankton in the first year of the initial impoundment periodwas Chlorophyta-Bacillariophyta-Chlorophyta,or J/F/X1-P/MP/W1-A/X1/MP,respectively.And the succession order in the second year was Cyanobacteria/Chlorophyta-Bacillariophyta-Chlorophyta,or L_(M)/G/P-P/A/X1-X1/J/G.Water temperature,relativewater column stability,mixing depth,and pHwere crucial factors affecting phytoplankton community succession.This study revealed the interannual succession law and driving factors of phytoplankton in the initial impoundment period and provided an important reference for the operation management and ecological protection of canyon-shaped reservoirs.
文摘Objective:To explore the effect of transarterial chemoembolization(TACE)+CT-guided microwave ablation(MWA)on treating patients with primary liver cancer.Methods:78 primary liver cancer cases were enrolled and divided into groups according to their assigned surgical plans.The control group was treated with TACE alone,and the observation group was treated with TACE+CT-guided MWA.The efficacy of the treatment and the liver function indicators and follow-up results of the patients of the two groups were compared.Results:The efficacy of the treatment received by the observation group was higher than that of the control group.Besides,the patients in the observation group exhibited better improvement in liver function indicators after 3 months of treatment.Furthermore,the survival rates of 1 and 2 years after surgery of the observation group were all higher than those of the control group(P<0.05).Conclusion:TACE combined with CT-guided MWA is more effective in treating primary liver cancer compared to TACE alone.Besides,it resulted in better improvement of liver function and long-term survival rate.Therefore,this treatment regime should be popularized.

文摘Lonicera japonica(honeysuckle)is a traditional Chinese medicinal food,in which the main active ingredients are phenolic acids,polysaccharides,flavonoids,and volatile oils.They have various biological activities,including antiviral,antibacterial,antioxidant,hypoglycemic and lipid-lowering,and anti-inflammatory effects.This review summarizes the health effects and pharmacodynamic mechanisms of L.japonica extracts and the major active ingredients in these extracts,and the structures,metabolic process in vivo,and biotransformation processes of these compounds.In addition,the current status of the development of L.japonica-related functional foods is summarized.The aim is to provide a theoretical basis and reference for the further development and use of the active ingredients in L.japonica as functional foods for disease prevention and treatment.
