Relative tillering rate(RTR)increased linear-ly with the increasing of leaf N concentration(NLV)has been already reported.To testwhether this relationship could be used toquantitatively explain the difference in tilleringamong a wide range of N application,field ex- periments were conducted at the IRRI farm,Los Banos,Laguna,the Philippines.Two in- dica cultivars,IR 72 and IR68284H wereused.For each cultivar,12 treatments includ- ing 4 N levels(0,60,120,and 180kgN·ha)and 3 transplanting spacing(30×20,20×20,and 10×20cm)were arranged in a ran-domized split-plot design with 4 replications.The N treatments were designated as mainplots and spacings as subplots.Fourteen-day-old seedlings were transplanted with 3seedlings per hill.The subplot area was 20m~2.Nitrogen fertilizer was applied as basal,atmidtillering,and at panicle initiation in threeequal splits.P,K,and Zn were applied asbasal at normal dosage.The field was flooded.Plant samples were taken every 7-14 d from 14d after transplanting to flower 展开更多

Effectiveness evaluation of fluorescent compensation in multicolor flow cytometry:A quantitative study

In fluorescence flow cytometry,spectral overlap among multiple fluorescent labels cannot be avoided,and thus detected fluorescent intensities need to be compensated.Although fluorescent compensation in flow cytometry has been widely used for many years,it still lacks quantitative evaluations to validate its effectiveness.Using a home-developed nine-color fluorescence flow cytometer,this study first obtains calibration curves by assaying gradient concentrations of nine different fluorochromes individually,with the fluorescent intensities of the highest concentrations of each fluorochrome being used to obtain a spillover matrix.Mixed fluorescent solutions are analyzed by flow cytometry in which the obtained fluorescent intensities are compensated by the spillover matrix,translated to specific concentrations based on calibration curve and compared with nominal values.Three mixed solutions of Brilliant Violet 650 and Brilliant Violet 711,of Alexa Fluor 488 and PE,and of Pacific Orange,Alexa Fluor 488,and PE are tested,with fluorescent compensation being observed to reduce excessive signals due to spectral overlap.Specifically,concentration deviations(before vs after compensation)in comparison with nominal values for Brilliant Violet 711 and Alexa Fluor 488 are quantified as 40.6%vs 14.9%and 6.7%vs 1.9%,respectively.The results presented here provide a quantitative reference for fluorescent compensation that can be used to effectively address the issue of spectral overlap in fluorescence flow cytometry. 展开更多

Summary: To establish the determination method of dauricine (Dan) concentration in rats' blood and other biological samples, a reverse-phase HPLC method was adopted. Under the given condition, dauricine could be well separated. The retention time (tR) of Dan and its internal standard, daurisoline were 9. 2 and 6.1 respectively. The detection limit was 10-2 mg/ml. The absolute recoveries of all kinds of samples were above 70 %, and the relative ones were over 85%. A good liner relationship has been obtained over the entire range of 0. 030 to 3. 000 rug/L in blood samples and 0. 050 to 5. 000 mg/L in other tissue samples. The intraday and interday coefficients of varia- tion were below 10 %. The results showed that the method can be used for detecting Dan in all kinds of biological samples. 展开更多

In this paper,fluorescence spectra properties of potassium sorbate in aqueous solution and orange juice are studied,and the result shows that in two solution there are many difference in fluorescence spectra of potassium sorbate,but the fluorescence characteristic peak exists in,λex/λem=375/490 nm.It can be seen from the two dimensional fluorescence spectra that the relationship between the fluorescence intensity and the concentration of potassium sorbate is very complex,so there is no linear relationship between them.To determine the concentration of potassium sorbate in orange juice,a new method combining Particle Swarm Optimization(PSO)algorithm with Back Propagation(BP)neural network is proposed.The relative error of two predicted concentrations is 1.83%and 1.53%respectively,which indicate that the method is feasible.The PSO-BP neural network can accurately measure the concentration of potassium sorbate in orange juice in the range of 0.1-2.0g·L^-1. 展开更多
