Objectives:This study aimed to assess the reliability and validity of the abbreviated Committed Action Questionnaire(CAQ-8)in a cohort of 1635 Chinese university students.Methods:Participants completed the Chinese ver...Objectives:This study aimed to assess the reliability and validity of the abbreviated Committed Action Questionnaire(CAQ-8)in a cohort of 1635 Chinese university students.Methods:Participants completed the Chinese version of the CAQ-8 along with other standardized measures,including the Acceptance and Action Questionnaire-II(AAQ-II),the Valuing Questionnaire(VQ),the Satisfaction with Life Scale(SWLS),the Depression Anxiety Stress Scales(DASS-21),and the World Health Organization Fiveitem Well-Being Index(WHO-5).A retest was conducted one month later with 300 valid responses.Results:Exploratory factor analysis(n=818)identified a 2-factor structure,confirmed through validated factor analysis(n=817),showing good fit indices(CFI=0.990,RMSEA=0.040).Measurement equivalence across genders was established.The CAQ-8 showed significant positive correlations with life satisfaction,mental health,and values,and negative correlations with depression,anxiety,stress,and experiential avoidance.The scale demonstrated good internal consistency(Cronbach’sα=0.76)and retest reliability(ICC=0.70).Network analysis confirmed the robustness of the 2-factor model,with item 4 in CAQ-8 identified as a core item.Conclusion:The CAQ-8 is a reliable and valid tool for measuring committed action within the psychological flexibility model in Chinese populations.展开更多
Human induced pluripotent stem cells(hiPSCs)are invaluable resources for producing high-quality differentiated cells in unlimited quantities for both basic research and clinical use.They are particularly useful for st...Human induced pluripotent stem cells(hiPSCs)are invaluable resources for producing high-quality differentiated cells in unlimited quantities for both basic research and clinical use.They are particularly useful for studying human disease mechanisms in vitro by making it possible to circumvent the ethical issues of human embryonic stem cell research.However,significant limitations exist when using conventional flat culturing methods especially concerning cell expansion,differentiation efficiency,stability maintenance and multicellular 3D structure establishment,differentiation prediction.Embryoid bodies(EBs),the multicellular aggregates spontaneously generated from iPSCs in the suspension system,might help to address these issues.Due to the unique microenvironment and cell communication in EB structure that a 2D culture system cannot achieve,EBs have been widely applied in hiPSC-derived differentiation and show significant advantages especially in scaling up culturing,differentiation efficiency enhancement,ex vivo simulation,and organoid establishment.EBs can potentially also be used in early prediction of iPSC differentiation capability.To improve the stability and feasibility of EB-mediated differentiation and generate high quality EBs,critical factors including iPSC pluripotency maintenance,generation of uniform morphology using micro-pattern 3D culture systems,proper cellular density inoculation,and EB size control are discussed on the basis of both published data and our own laboratory experiences.Collectively,the production of a large quantity of homogeneous EBs with high quality is important for the stability and feasibility of many PSCs related studies.展开更多

The cells act not only by cell replacement in the target organ but also by delivering tr...Background: Numerous studies of tissues’ regeneration have confessed the recovery of damaged liver by hematopoietic stem cells. The cells act not only by cell replacement in the target organ but also by delivering trophic factors that support endogenous liver regeneration. A little is known of how organ-derived signals recruit such committed cells into circulation. Objective: We investigated the roles of noninvasive mechanical percutaneous stress of cirrhotic human liver in numbers fluctuation of trophic, liver-specific alpha-fetoprotein-positive fraction of CD133-positive hematopoietic stem cells in lymphocytes of patients waiting for liver transplantation. Methods: To promote in blood the number of the alpha-fetoprotein-positive fraction of CD133-positive hematopoietic stem cells, committed to liver’ tissue, we activated mechanically the cirrhotic liver of patient by transcutaneous micro vibration received from skin-contacted electro-magnetic vibraphones generated mechanical pulses with amplitude 10 μm and smoothly changing frequency from 0.03 kHz to 18 kHz and back forth during one cycle duration 1 minute. The number of the alpha-fetoprotein-positive fraction of CD133-positive hematopoietic stem cells in lymphocytes of potential recipients was controlled by flow cytometry before and during daily sonication of skin area, which corresponds to liver projection on it. The 15 minutes cyclic sonication of the liver area performed daily for three weeks. Results: The sonication increased significantly averaged number of liver-specific alpha-fetoprotein-positive CD133-positive blood lymphocytes in 2 - 3 times compared to a base lane. The second similar sonication, the same zone after three weeks break showed differences with baseline, but it was statistically insignificant. The result was specifically related to the liver as it showed the control sonication of the backbone’s projection on the skin of a separate group of patients with cirrhotic liver from the waiting list. Conclusion: The stem cells committed to the liver recruit from the bone marrow into circulation, when organ mechanically stresses and secretes specific humoral signals to provoke of lymphopoiesis on host liver repair.展开更多
