BACKGROUND The use of network pharmacology and blood metabolomics to study the patho-genesis of violent aggression in patients with schizophrenia and the related drug mechanisms of action provides new directions for reducing the risk of violent aggression and optimizing treatment plans.AIM To explore the metabolic regulatory mechanism of olanzapine in treating patients with schizophrenia with a moderate to high risk of violent aggression.METHODS Metabolomic technology was used to screen differentially abundant metabolites in patients with schizophrenia with a moderate to high risk of violent aggression before and after olanzapine treatment, and the related metabolic pathways were identified.Network pharmacology was used to establish protein-protein interaction networks of the core targets of olanzapine.Gene Ontology functional analysis and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis were subsequently performed.RESULTS Compared with the healthy group, the patients with schizophrenia group presented significant changes in the levels of 24 metabolites related to the disruption of 9 metabolic pathways, among which the key pathways were the alanine, aspartate and glutamate metabolism and arginine biosynthesis pathways. After treatment with olanzapine, the levels of 10 differentially abundant metabolites were significantly reversed in patients with schizophrenia.Olanzapine effectively regulated six metabolic pathways, among which the key pathways were alanine, aspartate and glutamate metabolism and arginine biosynthesis pathways.Ten core targets of olanzapine were involved in several key pathways.CONCLUSION The metabolic pathways of alanine, aspartate, and glutamate metabolism and arginine biosynthesis are the key pathways involved in olanzapine treatment for aggressive schizophrenia. 展开更多

BACKGROUND Peer bullying victimization(PBV)is a significant public health issue that adolescents often face, with approximately one-third having experienced PBV.Understanding its interrelationships with mental health problems is crucial for effective intervention. This study aims to examine the longitudinal relationships between internalizing problems(depression and anxiety),externalizing problems(aggression),and PBV among middle school students using cross-lagged panel network analysis.AIM To examine the longitudinal relationships between internalizing problems(depression and anxiety),externalizing problems(aggression),and PBV among middle school students using cross-lagged panel network analysis.METHODS A total of 1260 middle school students(54.6%male)participated in this study.Data were collected at two time points(time 1 and time 2)using self-report questionnaires to assess PBV,depression, anxiety, and aggression.Cross-lagged panel network analysis was applied to examine the bi-directional relationships between these variables over time.RESULTS Depression, particularly a“sad mood,”was the most significant predictor of both PBV and aggression.Cyberbullying victimization also emerged as a key factor influencing depression and anxiety. While anxiety had weaker and less consistent effects on PBV,aggression was predominantly influenced by depressive symptoms and sleep disturbances. The analysis also identified key symptoms, such as a“sad mood”and sleep disturbances,as crucial targets for intervention to mitigate the cycle of PBV,depression, anxiety, and aggression.CONCLUSION This study provides important insights for bullying victimization prevention and intervention strategies:(1)Early identification and intervention targeting depression, particularly a“sad mood,”and sleep disturbances;(2)The importance of addressing cyberbullying as a distinct factor influencing mental health;and(3)The need for comprehensive, longitudinal approaches to understanding and intervening in the interconnected mental health issues among adolescents. 展开更多

Sex-specific neurons play pivotal roles in regulating sexually dimorphic behaviors. In insects, the sex determination gene doublesex(dsx)establishes major sexual dimorphism of the nervous system,in which male-specific dsx^(M)promotes neuronal development, while female-specific dsx^(F)inhibits neuronal development by promoting neuronal apoptosis. In this study,we find that dsx regulates the number of dsx-expressing central neurons in Drosophila in cell-specific manners.Although dsx^(M)overall promotes an increase in the number of dsx neurons,it inhibits the emergence of specific pC1 neurons.dsx^(F)reduces the number of different pC1/pC2 subtypes, but promotes the formation of pC1d.We also find that dsx^(M)and dsx^(F)barely affect the number of some pC2 neurons.Changes in the number of pC1/pC2 neurons alter their roles in regulating different behaviors, including courtship, aggression, and locomotion. Our results illustrate the multifaceted functions of dsx in sexually dimorphic neuronal development and behaviors. 展开更多

This case study explores the efficacy of school-based intervention to address psychosocial challenges faced by an 11-year-old adolescent. The case study aimed to decrease the agression and acting out behavior as result of being victimized at school by the peers. The aim was to assess and manage the child’s aggressive behavior and academic underperformance which played a significant role in the child’s low self-esteem and emotional regulation. A comprehensive assessment was conducted to rule out the difficulties and a multi-faceted intervention strategy was utilized including anger management and structured activity scheduling that helped that child to improve his academic performance as well as to learn to manage his emotional expression. Throughout 16 sessions, the intervention targeted key behavioural indicators such as emotional expression, and aggression;post-assessment results demonstrated a 22% improvement in the child’s behavioral and academic challenges. The findings suggest that a multi-faceted therapeutic approach can be effective in addressing complex issues of aggression and academic underperformance in children, highlighting the importance of integrated psychological and educational interventions. 展开更多

This letter critically reviews a recent longitudinal network study by Bai et al examining the dynamic,symptom-level interplay among peer bullying victimization, depression, anxiety, and aggression in Chinese adolescents. The study highlights that key symptoms, such as persistent sad mood, sleep disturbances, and cyberbullying victimization play a pivotal role in reinforcing the vicious cycle between mental health issues and bullying experiences. While the application of cross-lagged panel network analysis offers a nuanced understanding of these bidirectional relationships, several limitations remain, including the use of selfreported measures and a region-specific sample.Nevertheless, the findings underscore the urgent need for early screening and targeted interventions in school settings, particularly those addressing both emotional symptoms and digital forms of bullying.Moving forward, integrated and culturally sensitive approaches are essential to prevent escalation and break the link between peer victimization and adolescent psychopathology.Future research should incorporate multi-informant data and broaden the cultural context to strengthen generalizability and intervention design. 展开更多

Agitation is a neuropsychiatric syndrome characterized by excessive motor and/or verbal behaviors, with or without aggressive behaviors. The prevalence of agitation in Alzheimer’s disease varies from 5%to over 50%.Multiple factors have been implicated in its pathophysiology, including disease stage, comorbidity with other symptoms(e.g.,psychosis,anxiety/depression),and psychosocial factors.Ruling out delirium and identifying environmental triggers are fundamental steps in the management of agitation in Alzheimer’s disease. For establishing an effective therapeutic plan,it is important to define duration, severity, and potential for harm. While non-pharmacological approaches are considered the first line of intervention, pharmacological agents are frequently used in the treatment of agitation.Antipsychotics are commonly used in acute agitation. For chronic agitation,serotonin-selective reuptake inhibitors, especially citalopram and escitalopram, are often preferred due to safety concerns associated with the longterm use of antipsychotics.Promising novel strategies, such as new compounds and neuromodulation, are likely to be incorporated into agitation therapeutics in the next few years. 展开更多

BACKGROUND Claudin-6(CLDN6),a tight junction protein typically restricted to embryonic tissues,is re-expressed in various cancers. However, its prognostic significance in high-grade endometrial carcinoma(HGEC)remains unclear.AIM To investigate the expression pattern of CLDN6 in HGEC and assess its correlation with clinicopathological parameters and patient survival.METHODS Immunohistochemical analysis of CLDN6 expression was performed on formalinfixed,paraffin-embedded tissues from 80 patients diagnosed with HGEC.Associations between CLDN6 expression and histological subtype, the International Federation of Gynecology and Obstetrics(FIGO)stage, depth of myometrial invasion, lymphovascular space invasion, recurrence, and survival outcomes were statistically analysed.Univariate and multivariate Cox regression models were used to identify independent prognostic factors.RESULTS High CLDN6 expression was detected in a subset of HGEC patients and was significantly associated with nonendometrioid histology(P=0.026),advanced FIGO stage(P=0.015),deep myometrial invasion(P=0.038),and recurrence(P=0.002).While Kaplan-Meier analysis did not reveal a statistically significant difference in disease-free survival or overall survival between the high CLDN6 expression group and the low CLDN6 expression group, multivariate Cox regression revealed that CLDN6 overexpression was an independent predictor of shorter disease-free survival[hazard ratio(HR)=68.98,P=0.022]and overall survival(HR=24.023,P=0.038).CONCLUSION CLDN6 overexpression is associated with aggressive tumor features and poor clinical outcomes in HGEC,suggesting its utility as a prognostic biomarker and potential therapeutic target. 展开更多

BACKGROUND The neural mechanisms underlying aggressive behavior in schizophrenia(SCZ)remain poorly understood. To date,no studies have reported on the event-related potential(ERP)characteristics of aggression in SCZ using the competitive reaction time task(CRTT).Further investigation into the ERP correlates of aggression in SCZ would provide valuable insights into the neural processes involved.AIM To explore the neural mechanism of aggressive behavior in SCZ.METHODS Participants of this study included 40 SCZ patients and 42 healthy controls(HCs).The Reactive Proactive Aggression Questionnaire was used to assess trait of aggression. The Barratt Impulsiveness Scale 11 was used to measure impulsiveness. The Positive and Negative Symptom Scale(PANSS)was used to evaluate psychopathological features and disease severity. All participants were measured with ERP while performing the CRTT.Data of behavior,ERP components(P2,N2,and P3),and feedback-related negativity(FRN)were analyzed.RESULTS Analysis of the behavioral data revealed that compared with HCs,SCZ patients exhibited higher punishment choices.Analysis of ERP components showed that compared with HCs,SCZ patients exhibited higher N2 amplitudes and P2 amplitudes during the decision phase of the CRTT;however,SCZ patients exhibited lower FRN amplitudes and lower P3 amplitudes during the outcome phase of the CRTT.The N2 amplitudes evoked by highintensity provocation were positively related to PANSS-P scores. And the P3 amplitudes evoked in the winning trials were negatively correlated with the PANSS-G scores.CONCLUSION SCZ patients exhibit abnormal ERP characteristics evoked by the CRTT,which suggests the neural correlates of aggressive behavior in SCZ. 展开更多

Intraspecific conflict induced by the innate aggressiveness is one of the main reasons for the extremely low survival rate in mud crab Scylla paramamosain aquaculture, which have impeded the sustainable culture of the species. In this study,we first classified and quantified the aggressive behavior, and established a crab aggressive behavior model, laying the foundation for subsequent research on evaluating combat intensity. The contents of 5-hydroxytryptamine(5-HT),dopamine(DA),and cAMP in the hemolymph of the mud crabs before and after fighting were measured by high-performance liquid chromatography-mass spectrometry(HPLC-MS),and the mud crabs exhibited a significant increase of 5-HT(P<0.05),while the DA and cAMP decreased significantly(P<0.05).In addition,we applied EthoVision to examine the changes of the crab behavior after DA administration. After 0.5 h of injection, the movement speed, distance, duration of aggressive behavior, and intensity of aggression in the high concentration DA group were significantly higher than those in the saline injection group and the untreated control group(P<0.05).The results of real-time quantitative polymerase chain reaction(qRT PCR)analysis showed that the expression of DA 1 in the thoracic ganglia of the mud crabs was significantly down-regulated in the DA injection group, and the aggressive behavior was weakened.Conversely,DA1 expression was up-regulated when aggressive behavior was strengthened.Besides, there were significant differences in the expression levels of receptor expression genes including 5-HT1,5-HT2,and crustacean hyperglycemic hormone(CHH)in different tissues, indicating that the alteration of aggressive behavior of the mud crab after injection with different concentrations of DA could be regulated by changes in the expression levels of corresponding receptor genes. Our results contribute to a deeper analysis of the aggressive behavior mechanism of the mud crabs and provide a theoretical basis for reducing fighting-related mortalities in aquaculture. 展开更多

This paper proposes a fingerprint matching method integrating transfer learning and online learning to tackle the challenges of environmental adaptability and dynamic interference resistance in photovoltaic(PV)array DC arc fault location methods based on electromagnetic radiation(EMR)signals.Initially,a comprehensive analysis of the time–frequency characteristics of series arc EMR signals is carried out to pinpoint effective data sources that reflect fault features.Subsequently,a multi-kernel domain-adversarial neural network(MKDANN)is introduced to extract domain-invariant features, and a feature extractor designed specifically for fingerprint matching is devised. To reduce inter-domain distribution differences,a multi-kernel maximum mean discrepancy(MK-MMD)is integrated into the adaptation layer. Moreover,to deal with dynamic environmental changes in real-world situations, the support-class passive aggressive(SPA)algorithm is utilized to adjust model parameters in real time. Finally,MKDANN and SPA technologies are smoothly combined to build a fully operational fault location model.Experimental results indicate that the proposed method attains an overall fault location accuracy of at least 95%,showing strong adaptability to environmental changes and robust interference resistance while maintaining excellent online learning capabilities during model migration. 展开更多

Objectives:The aim of this study was to verify the causal relationship between trait aggressiveness(TA)and moral disengagement(MD),know more about the growth trajectory of MD,and explore the effects of gender and TA on the growth trajectory. Methods:We used the Buss-Perry Aggression Questionnaire and Moral Disengagement Scale to survey 433 Chinese junior high school students longitudinally three times. Results:The results of the random intercept cross-lagged panel model(RI-CLPM)analysis indicated that TA positively predicted MD,while MD did not predict TA at the within-person level. Thus,TA could be considered an antecedent variable of MD.Furthermore, the unconditional latent growth linear model analysis revealed that MD among junior high school students exhibited an upward trend, characterized by an increasing rate of growth over time.In the conditional latent growth linear model analysis,we found that gender influenced only the initial level of MD but had no effect on its growth rate.Conversely,TA demonstrated both delayed and immediate positive effects on the growth of MD.Conclusions:Our findings suggested a one-way cross-lag effect between TA and MD.Additionally, the growth trajectory of MD among junior high school students was found to be ascending. This growth trajectory was influenced by gender and TA,offering valuable insights for the prevention and intervention of behavioral problems in junior high school students. 展开更多
