Background:Diabetic foot,a severe complication of diabetes,is characterized by chronic refractory wounds.Sanhuang Oil,a topical herbal formula, demonstrates significant therapeutic effects including antibacterial,anti-inflammatory,and immunomodulatory activities. However, its active constituents and mechanisms of action against diabetic foot remain to be elucidated. Methods:In this study, the chemical constituents of Sanhuang Oil were identified using UPLC-QE-Orbitrap-MS.Subsequently,the mechanism by which Sanhuang Oil promotes diabetic foot ulcer healing was predicted by integrating network pharmacology and molecular docking.Additionally, diabetic mouse model was established in ICR mice using a combination of a high-fat diet(HFD)and streptozotocin(STZ)chemical induction.A full-thickness skin defect was created on the dorsum of the mice.Wound healing and the healing rate were observed following Sanhuang Oil intervention. The mechanism underlying Sanhuang Oil’s promotion of diabetic ulcer healing was further investigated using transcriptomics and histopathological examination(H&E staining). Results:A total of 97 active ingredients were identified from Sanhuang Oil.Network pharmacology analysis predicted 543 common targets, and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis identified 203 relevant pathways.Molecular docking further confirmed high binding affinity(binding energy≤−5.0 kcal/mol)between specific active components in Sanhuang Oil(e.g.,coptisine,phellodendrine,baicalein)and key targets associated with diabetic foot ulcers(e.g.,EGFR,AKT1,STAT3).In vivo experimental results demonstrated that the wound healing rate was significantly higher in Sanhuang Oil-treated groups compared to the model group(P<0.001).HE staining revealed that the high-dose Sanhuang Oil group exhibited more pronounced epithelial tissue coverage over the wound, reduced inflammatory cell infiltration, and increased collagen deposition and fibroblast proliferation.transcriptomic analysis identified Pdk4,Ttn,Csrp3,Actn2,Myoz2,Tnnc2,Myod1,Myog,Myot,and Myf6 as key regulatory proteins involved in promoting wound healing. Conclusion:Sanhuang Oil promotes wound healing in diabetic ulcer mice, potentially by mitigating inflammation and regulating key targets such as Pdk4 to enhance fibroblast function. These findings provide novel insights into the multi-target,multi-pathway mechanism of Sanhuang Oil for treating diabetic foot ulcers. 展开更多

Ulcerative colitis (UC) is a chronic inflammatory bowel disease characterized byclinical symptoms of diarrhea and mucopurulent bloody stools, and its incidenceis increasing globally. The etiology and pathogenesis of UC remain elusive. Currenttherapeutic approaches, including anti-inflammatory, immunosuppressiveand immunomodulating agents, are often limited in efficacy and frequently associatedwith adverse drug reactions. Therefore, there is an urgent need to developsafer and more effective treatment strategies to address the limitations of existingtherapies. Scutellaria baicalensis Georgi (HQ), a traditional Chinese medicinal herb, has been employed in the treatment of UC for over 2000 years. Recent studieshave demonstrated that HQ contains multiple active components capable oftreating UC through anti-inflammation, immune modulation, intestinal barrierprotection, antioxidant activity, and regulation of the gut microbiota. This paperreviews recent studies on the mechanism of action and clinical trials of HQ intreating UC based on relevant literature, with the aim of providing valuable insightsinto future treatment approaches. 展开更多

Oligodendrocytes and their cell-intrinsic gene regulatory network:Oligodendrocytes(OLs)are the myelinating glial cells of the vertebrate central nervous system. They are responsible for insulating neuronal axons with a lipid-rich myelin sheath, which enables the saltatory conduction of action potentials.During development, oligodendrocyte progenitor cells(OPCs)emerge from neural stem cells in the ventricular zone. They then proliferate, increase their number, and migrate to their final destination where they encounter unmyelinated neuronal axons and differentiate in a stepwise fashion into myelinating oligodendrocytes(mOLs)under the influence of environmental stimuli. 展开更多

A main focus of machine learning research has been improving the generalization accuracy and efficiency of prediction models. However, what emerges as missing in many applications is actionability, i.e., the ability to turn prediction results into actions. Existing effort in deriving such actionable knowledge is few and limited to simple action models while in many real applications those models are often more complex and harder to extract an optimal solution. In this paper, we propose a novel approach that achieves actionability by combining learning with planning, two core areas of AI. In particular, we propose a framework to extract actionable knowledge from random forest, one of the most widely used and best off-the-shelf classifiers. We formulate the actionability problem to a sub-optimal action planning (SOAP) problem, which is to find a plan to alter certain features of a given input so that the random forest would yield a desirable output, while minimizing the total costs of actions. Technically, the SOAP problem is formulated in the SAS+ planning formalism, and solved using a Max-SAT based ap- proach. Our experimental results demonstrate the effectiveness and efficiency of the proposed approach on a personal credit dataset and other benchmarks. Our work represents a new application of automated planning on an emerging and challenging machine learning paradigm. 展开更多

Combined with elastic network model(ENM),the perturbation response scanning(PRS)has emerged as a robust technique for pinpointing allosteric interactions within proteins. Here,we proposed the PRS analysis of drug-target networks(DTNs),which could provide a promising avenue in network medicine. We demonstrated the utility of the method by introducing a deep learning and network perturbation-based framework, for drug repurposing of multiple sclerosis(MS).First, the MS comorbidity network was constructed by performing a random walk with restart algorithm based on shared genes between MS and other diseases as seed nodes. Then, based on topological analysis and functional annotation, the neurotransmission module was identified as the“therapeutic module”of MS.Further, perturbation scores of drugs on the module were calculated by constructing the DTN and introducing the PRS analysis, giving a list of repurposable drugs for MS.Mechanism of action analysis both at pathway and structural levels screened dihydroergocristine as a candidate drug of MS by targeting a serotonin receptor of se-rotonin 2B receptor(HTR2B).Finally,we established a cuprizone-induced chronic mouse model to evaluate the alteration of HTR2B in mouse brain regions and observed that HTR2B was significantly reduced in the cuprizone-induced mouse cortex. These findings proved that the network perturbation modeling is a promising avenue for drug repurposing of MS.As a useful systematic method, our approach can also be used to discover the new molecular mechanism and provide effective candidate drugs for other complex diseases. 展开更多

The evacuation of people under threat is an effective disaster prevention and mitigation measure in response to flash floods and geological hazards, and it is also an essential element of pre-disaster planning. However, the effect of the interactions between perception factors on residents'willingness to evacuate is an urgent problem to be solved. Therefore, this paper introduces risk, stakeholder, and protective action perceptions from the protective action decision model as the main explanatory variables. These three core perceptions are subdivided into affective risk perception, cognitive risk perception, government perception,other-stakeholder perception, resourcerelated attributes, and hazard-related attributes.A questionnaire survey was conducted from June to July 2023 among residents of mountainous communities in nine villages in three towns in Sichuan Province,China.359 cross-sectional data were analyzed using structural equation modeling to explore the effects of six perception factors on evacuation intentions. The results of the study showed that:(1)affective risk perception, government perception,other-stakeholder perception, and hazard-related attributes all directly and positively influence residents'intentions to evacuate;(2)cognitive risk perception is mediated by stakeholder and protective action perceptions, which indirectly and positively affect residents'intentions to evacuate.Based on the hypothesized paths, strategies to improve residents'willingness to evacuate are discussed from the perspective of three core perceptions:strengthening disaster risk education, improving residents'cohesion, and building government credibility. The results of this study can provide theoretical support and practical suggestions for emergency management departments to formulate emergency evacuation strategies, which can aid decision-makers in better understanding residents'intentions to evacuate, optimizing evacuation information dissemination pathways, and strengthening disaster risk management capabilities. 展开更多

In the last few years, scholars have continued to refine the understanding of conservatism through cultural psychology. The application of Symbolic Action Theory, particularly Ernst Boesch’s framework, remains central to discussions, illustrating how conservatism is not merely a political stance but a dynamic system of behaviors that reinforces societal values through symbolic actions(Kölbl,2020).Additionally,recent studies have highlighted the interaction between cultural conservatism and collective identity, with particular focus on how conservatism can manifest in diverse cultural settings(Yilmaz&Alper,2019).Research also underscores the relevance of symbolic politics in shaping both individual attitudes and broader political actions, contributing to the understanding of how symbolic meanings sustain social conservatism in different cultural contexts(Chin&Levey,2022). 展开更多

Video action recognition(VAR)aims to analyze dynamic behaviors in videos and achieve semantic understanding.VAR faces challenges such as temporal dynamics,action-scene coupling, and the complexity of human interactions.Existing methods can be categorized into motion-level,event-level,and story-level ones based on spatiotemporal granularity.However,single-modal approaches struggle to capture complex behavioral semantics and human factors. Therefore,in recent years,vision-language models(VLMs)have been introduced into this field, providing new research perspectives for VAR.In this paper,we systematically review spatiotemporal hierarchical methods in VAR and explore how the introduction of large models has advanced the field.Additionally,we propose the concept of“Factor”to identify and integrate key information from both visual and textual modalities, enhancing multimodal alignment. We also summarize various multimodal alignment methods and provide in-depth analysis and insights into future research directions. 展开更多

The task of student action recognition in the classroom is to precisely capture and analyze the actions of students in classroom videos, providing a foundation for realizing intelligent and accurate teaching. However, the complex nature of the classroom environment has added challenges and difficulties in the process of student action recognition. In this research article, with regard to the circumstances where students are prone to be occluded and classroom computing resources are restricted in real classroom scenarios,a lightweight multi-modal fusion action recognition approach is put forward. This proposed method is capable of enhancing the accuracy of student action recognition while concurrently diminishing the number of parameters of the model and the Computation Amount, thereby achieving a more efficient and accurate recognition performance. In the feature extraction stage, this method fuses the keypoint heatmap with the RGB(Red-Green-Blue color model)image. In order to fully utilize the unique information of different modalities for feature complementarity,a Feature Fusion Module(FFE)is introduced. The FFE encodes and fuses the unique features of the two modalities during the feature extraction process. This fusion strategy not only achieves fusion and complementarity between modalities, but also improves the overall model performance.Furthermore,to reduce the computational load and parameter scale of the model,we use keypoint information to crop RGB images. At the same time, the first three networks of the lightweight feature extraction network X3D are used to extract dual-branch features. These methods significantly reduce the computational load and parameter scale. The number of parameters of the model is 1.40 million, and the computation amount is 5.04 billion floating-point operations per second(GFLOPs),achieving an efficient lightweight design. In the Student Classroom Action Dataset(SCAD),the accuracy of the model is 88.36%.In NTU 60(Nanyang Technological University Red-Green-Blue-Depth RGB+Ddataset with 60 categories),the accuracies on X-Sub(The people in the training set are different from those in the test set)and X-View(The perspectives of the training set and the test set are different)are 95.76%and 98.82%,respectively.On the NTU 120 dataset(Nanyang Technological University Red-Green-Blue-Depth dataset with 120 categories),RGB+Dthe accuracies on X-Sub and X-Set(the perspectives of the training set and the test set are different)are 91.97%and 93.45%,respectively.The model has achieved a balance in terms of accuracy, computation amount, and the number of parameters. 展开更多
