High performance liquid chromatography (HPLC) was adopted, using Agilent ZORBAX SB-C18 column (250 mm×4.6 mm, 5 μm) as the chromatographic column, and acetonitrile-0.05 mol/L potassium dihydrogen phosphate solution as the mobile phase in a gradient elution with the flow rate of 1.0 mL/min. Sample solution (10 μL) was injected and was tested at the wavelength of 225 nm for 75 min at the column temperature of 30 ℃, Fingerprint similarity software (2004A version) was used to conduct data analysis. A total of 11 batches of Wuji gastric floating sustained-release tablets were tested and analyzed with HPLC fingerprint. Seventeen common peaks were found and the similarity of the 11 batches of agents was greater than 0.9, indicating that the production process of the agent is stable and feasible. The method is operable and could effectively control the quality of Wuji gastric floating sustained-release tablets.展开更多

Background:To explore the effective components and mechanism of Classic ancient prescription Wuji pill on lung cancer(LC)with the help of network pharmacology and molecular docking technology.Methods:TCMSP database,Ch...Background:To explore the effective components and mechanism of Classic ancient prescription Wuji pill on lung cancer(LC)with the help of network pharmacology and molecular docking technology.Methods:TCMSP database,Chinese Pharmacopoeia(2020 Edition)and related literature were searched to identify the main chemical composition and targets of constituent drugs in Wuji pill,including Houpo(Magnoliae Officinalis Cortex),Huangqin(Scutellariae Radix),Badou(Crotonis Fructus),Huanglian(Coptidis Rhizoma)and Renshen(Ginseng Radix et Rhizoma).Meanwhile,Genecards,OMIM,TTD,Drugbank and PharmGkb databases were used to obtain the main targets of LC.The"chemical compound-target"network was constructed by using Cytoscape 3.8.2 software with the disease drug intersection gene as the research target.The PPI network was constructed by using the string database for protein interaction analysis,and the core target genes were sorted out.The Gene ontology go function enrichment analysis and KEGG signal pathway enrichment analysis were carried out based on the ClusterProfiler of R software.Finally,the Autodock Vina software was used to conduct molecular docking between the core target with high value and the key components.Results:A total of 60 active components,190 corresponding targets and 2516 GO and 163 KEGG pathways of Wuji pill were screened out,including 179 potential targets of LC.The core targets were ESR1,CDKN1A,TP53,NR3C1,MAPK1,AKT1,MAPK8,JUN,RELA,the core components were quercetin,kaempferol,β-sitosterol,stigmasterol,tetrahedron,protoopioid,etc,and the core pathways were PI3K-Akt,MAPK,IL-17,etc.Conclusion:This study preliminarily revealed the mechanism of action of Wuji pill on multi component-multi target-multi pathway of LC,which provided reference for the further clinical development and research of Wuji pill.展开更多

Disruption of the intestinal mucosal barrier caused by gut dysbiosis and metabolic imbalance is the underlying pathology of inflammatory bowel disease(IBD).Traditional Chinese medicine Wuji Wan(WJW)is commonly used to...Disruption of the intestinal mucosal barrier caused by gut dysbiosis and metabolic imbalance is the underlying pathology of inflammatory bowel disease(IBD).Traditional Chinese medicine Wuji Wan(WJW)is commonly used to treat digestive system disorders and showed therapeutic potential for IBD.In this interdisciplinary study,we aim to investigate the pharmacological effects of WJW against experimental colitis by combining functional metabolomics and gut-microbiota sequencing techniques.Treatment with WJW altered the profile of the intestinal microbiota and notably increased the abundance of Lactobacillus,thereby facilitating the conversion of tryptophan into indole-3-acetic acid(IAA)and indoleacrylic acid(IA).These indole derivatives activated the aryl hydrocarbon receptor(AhR)pathway,which reduced colonic inflammation and restored the expression of intestinal barrier proteins.Interestingly,the beneficial effects of WJW on gut barrier function improvement and tryptophan metabolism were disappeared in the absence of gut microbiota.Finally,pre-treatment with the AhR antagonist CH-223191 confirmed the essential role of IAA-mediated AhR activation in the therapeutic effects of WJW.Overall,WJW enhanced intestinal barrier function and reduced colonic inflammation in a murine colitis model by modulating LactobacilluseIAAeAhR signaling pathway.This study provides novel insights into colitis pathogenesis and presents an effective therapeutic and preventive approach against IBD.展开更多
Blood deficiency syndrome(BDS)refers to a pathological state with blood dysfunction and organ dystrophy in traditional Chinese medicine.Danggui Wuji granules(DWG)was developed from a formula containing Angelicae Sinen...Blood deficiency syndrome(BDS)refers to a pathological state with blood dysfunction and organ dystrophy in traditional Chinese medicine.Danggui Wuji granules(DWG)was developed from a formula containing Angelicae Sinensis Radix and Musculus et Os Galli Domestici.Herein,we investigated the mechanism of DWG in treating BDS by modulating gut microbiota.We found that DWG protected mice from BDS by elevating the levels of red blood cell count,hemoglobin,and hematocrit in peripheral blood and increasing the erythrocyte membrane Na+-K+-ATPase activity.Danggui Wuji granules changed the composition and metabolites of colonic flora.Notably,Lactobacillus,Muribaculaceae,and Alistipes were the main genera showing changes after DWG treatment.Our findings revealed that DWG presented a positive therapeutic effect on BDS in mice by regulating the gut microbiota and metabolites.The protective mechanism of DWG was associated with pathways such as metabolic pathways,biosynthesis of secondary metabolites,ABC transporters,ribosome,thyroid hormone synthesis,lysine degradation,galactose metabolism,tyrosine metabolism,etc.展开更多
