目的探讨创伤性骨折病人血清趋化因子配体3(CCL3)、高迁移率族蛋白Bl(HMGB1)水平与深静脉血栓(DVT)形成的相关性。方法纳入2020年5月至2023年6月于中国人民解放军联勤保障部队第九一〇医院就诊的149例创伤性骨折病人作为研究对象,根据有... 目的探讨创伤性骨折病人血清趋化因子配体3(CCL3)、高迁移率族蛋白Bl(HMGB1)水平与深静脉血栓(DVT)形成的相关性。方法纳入2020年5月至2023年6月于中国人民解放军联勤保障部队第九一〇医院就诊的149例创伤性骨折病人作为研究对象,根据有无DVT发生分为DVT组(31例)和无DVT组(118例)。采用Pearson法分析血清CCL3和HMGB1表达水平的相关性;采用多因素logistic回归分析与创伤性骨折病人并发DVT有关的重要影响因素;采用受试者操作特征曲线(ROC曲线)评估血清CCL3和HMGB1表达对创伤性骨折病人发生DVT的预测效能。结果DVT组病人血清CCL3[(34.66±5.27)ng/L比(29.83±4.32)ng/L]和HMGB1[(10.91±2.89)μg/L比(7.75±1.75)μg/L]表达水平均高于无DVT组(P<0.05);血清中CCL3和HMGB1二者的表达水平呈正相关(r=0.68,P<0.05);DVT组和无DVT组年龄及骨折部位差异有统计学意义(P<0.05);年龄≥60岁(OR=2.33)、骨折部位为下肢(OR=2.00)、CCL3(OR=2.44)和HMGB1(OR=4.03)为创伤性骨折病人发生DVT的危险因素(P<0.05);血清CCL3、HMGB1以及二者联合检测的曲线下面积(AUC)分别为0.71、0.84、0.93,联合高于各自单独检测(Z_(二者联合-CCL3)=3.54、Z_(二者联合-HMGB1)=2.73,P=0.004、0.006)。结论创伤性骨折病人血清CCL3、HMGB1水平与DVT的形成有关,二者联合对创伤性骨折病人发生DVT具有一定的预测效能。 展开更多

目的采用高通量测序技术(RNA-seq)探讨昆布多糖(LAM)对非增生性糖尿病视网膜病变(NPDR)的影响。方法采用链脲佐菌素(STZ)腹腔注射建立糖尿病模型,观察LAM对糖尿病小鼠的影响。将C57BL/6小鼠随机分为对照(Control)、模型(Model)、昆布多... 目的采用高通量测序技术(RNA-seq)探讨昆布多糖(LAM)对非增生性糖尿病视网膜病变(NPDR)的影响。方法采用链脲佐菌素(STZ)腹腔注射建立糖尿病模型,观察LAM对糖尿病小鼠的影响。将C57BL/6小鼠随机分为对照(Control)、模型(Model)、昆布多糖治疗(LAM)组,每组8只小鼠。在建模8周后,LAM组接受为期4周的LAM腹腔注射治疗。记录3组小鼠血糖体质量变化,HE染色检查视网膜病变情况,采用RNA-seq挖掘STZ及LAM作用下糖尿病视网膜病变(DR)的关键差异基因(DEGs)。结果使用STZ可以成功建立DR模型,而LAM可以在一定程度上降低糖尿病小鼠血糖并改善糖尿病小鼠视网膜结构疏松的病理形态。经过RNA-seq对DEG挖掘发现与Control组相比,Model组小鼠视网膜共有214个DEGs,通过富集分析发现DR可通过PI3K-Akt信号通路加重病变程度。Model组与LAM组小鼠视网膜共有DEGs 42个,富集后显示LAM通过中性粒细胞胞外陷阱通路改善其病变程度。早期生长反应因子1(Egr1),FBJ骨肉瘤致癌基因(Fos),核受体亚家族4A成员1(Nr4a1),盐诱导激酶1(Sik1)受STZ调节,而LAM能显著回调其表达,可能与LAM对糖尿病视网膜病变的治疗密切相关。结论DEG可通过PI3K-Akt信号通路加重糖尿病视网膜病变程度。LAM可以通过中性粒细胞胞外陷阱通路减轻糖尿病视网膜病变,Egr1、Fos、Nr4a1、Sik1是LAM治疗STZ诱导DR的关键基因。 展开更多

Microbes play important roles in the transport and transformation of selenium(Se) in the environment, thereby influencing plant resistance to Se and Se accumulation in plant. The objectives are to characterize the bacteria with high Se tolerance and reduction capacity and explore the significance of microbial origins on their Se tolerance, reduction rate and efficiency. Two bacterial strains were isolated from a naturally occurred Se-rich soil at tea orchard in southern Anhui Province, China. The reduction kinetics of selenite was investigated and the reducing product was characterized using scanning electron microscopy and transmission electron microscopy-energy dispersive spectroscopy. The bacteria were identified as Lysinibacillus xylanilyticus and Lysinibacillus macrolides,respectively, using morphological, physiological and molecular methods. The results showed that the minimal inhibitory concentrations(MICs) of selenite for L. xylanilyticus and L. macrolides were 120 and 220 mmol/L, respectively, while MICs of selenate for L.xylanilyticus and L. macrolides were 800 and 700 mmol/L, respectively. Both strains aerobically reduced selenite with an initial concentration of 1.0 mmol/L to elemental Se nanoparticles(SeNPs) completely within 36 hr. Biogenic SeNPs were observed both inside and outside the cells suggesting either an intra-or extracellular reduction process. Our study implied that the microbes from Se-rich environments were more tolerant to Se and generally quicker and more efficient than those from Se-free habitats in the reduction of Se oxyanions. The bacterial strains with high Se reduction capacity and the biological synthesized Se NPs would have potential applications in agriculture, food, environment and medicine. 展开更多

The Ediacaran and early Cambrian black shales are widespread across the South China Craton (Yangtze and Cathaysia blocks). However, the trace element distribution patterns of the Ediacaran and early Cambrian black shales in the Cathaysia Block are still unclear. In this study, thirty- four black shale samples in the Lechangxia Group (Ediacaran) and thirteen black shale samples in the lower Bacun Group (early Cambrian) from Guangning area, western Guangdong Province, South China, were analyzed for major and trace elements concentrations. Compared to the upper continental crust, the Ediacaran black shales exhibit strongly enriched Se, Ga, and As with enrichment factor values (EF) higher than 10, significantly enrichedBi and Rb (10〉EF〉5), weakly enriched Mo, Ba, Cs, V, In, Be, TI, and Th (5〉EF〉2), normal U, Cr, Cd, Sc, Pb, Cu, and Li (2〉EF〉0.5), and depleted Ni, Zn, Sr, and Co. Early Cambrian black shales display strongly enriched Se, Ga, and As, significantly enriched Ba, Bi, and Rb, weakly enriched Mo, Cs, Cd, V, U, Be, In, and TI, normal Sc, Th, Cr, Li, Cu, Ni, and Pb and depleted Co, Zn, and Sr. Moreover, Se is the most enriched trace element in the Ediacaran and early Cambrian black shales: concentrations vary from 0.25 to 30.09 ppm and 0.54 to 5.01 ppm, and averaging 4.84 and 1.72 ppm, with average EF values of 96.87 and 34.32, for the Ediacaran and early Cambrian shales respectively. The average concentration of Se in the Ediacaran black shales is 2.8 times higher than that of early Cambrian black shales. Se contents in the Ediacaran and early Cambrian black shales exhibit significant variation (P = 0.03). Provenance analysis showed that Se contents of both the Ediacaran and early Cambrian black shales were without detrital provenance and volcanoclastic sources, hut of hydrothermal origin. The deep sources of Se and the presence of pyrite may explain the higher Se contents in the Ediacaran black shales. Similar with the Se-rich characteristics of the contemporaneous black shales in the south Qingling Mountain and Yangtze block, the Ediacaran and early Cambrian black shales in Guangning area, Cathaysia, are also enriched in Se, which may provide a clue for looking for the selenium-rich resources in western Guangdong Province. 展开更多

Effect of Astragalus Injection on Levels of Blood Selenium and Immunity Function in Children with Viral Myocarditis 被引量:2

Objective: To observe the effect of Astragalus Injection (AD on levels of blood selenium (Se) and cytokines, and T cellular immune function with viral myocarditis (VM) in children. Methods: Eighty children with VM were randomly divided into 2 groups. The control group consisted of 38 patients, to whom conventional therapy, including energy mixture, vitamin C and coenzyme Q10, etc. were given. The treated group (n = 42), to whom combination therapy of conventional therapy and Al were given. The levels of blood Se and cytokine, including interleukin-1 (IL-1), interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), and also evaluation of T lymphocyte subsets and cardiac function were observed. Results: The results showed that after treatment, the levels of blood Se were significantly higher (P<0.01), while IL-1,IL-6 and TNF-a were significantly lower (P<0.01) than those before treatment in the control group. The left ventricular end diameter (LVED) were significantly decreased (P<0.01), left ventricular ejection fraction (LVEF) and left ventricular fractional shortening (LVFS) were significantly increased than those before treatment in the treated group(P<0.01, P<0.05). T lymphocyte subsets got normalized (P<0.01), and compared with the control group, the difference was significant (P<0.01). Conclusion: Astragalus membranaceus possesses anti-viral effect, adjusts the balance of cytokine and T cellular immunity, and improves the clinical manifestation and cardiac function. It is an effective approach in treating viral myocarditis. 展开更多

To extract selenium(Ⅵ)from the highly caustic leachate of copper anode slime,the Ca-Al-Cl layered double hydroxides(Ca-Al-Cl-LDHs)with a formula of Ca2 Al(OH)6 Cl·2 H2 O by three co-precipitation methods were synthesized.A plate-like morphology and hexagonal crystal structure with typical mineral phases and functional groups were identified by the FESEM,XRD,FTIR,BET and XPS analysis.The forward feeding sample exhibits the best adsorption capacity of Se(Ⅵ).The factor experiments then reveal a favorable adsorption process with low temperature,low NaOH concentration and high adsorbent dosage.Furthermore,the adsorption kinetics and isotherm parameters can be well described by the Langmuir isotherm and the pseudo-second-order models,respectively.Accordingly,the maximum adsorption amount of Se(Ⅵ)onto Ca-Al-Cl-LDHs reaches188.6 mg/g at 50 ℃. 展开更多

In order to enhance the glutathione peroxidase(GPX) catalytic activity of the selenium-containing single-chain variable fragments(Se-scFv), a novel human scFv was designed on the basis of the structure of human antibody and optimized via bioinformatics methods such as homologous sequence analysis, three-dimensional(3D) model building, binding-site analysis and docking. The DNA sequence of the new human scFv was synthesized and cloned into the expression vector pET22b(+), then the scFv protein was expressed in soluble form in Escherichia coli BL21(DE3) and purified by Ni2+-immobilized metal affinity chromatography(IMAC). The serine residue of scFv in the active site was converted into selenocysteine(Sec) with the chemical modification method, thus, the human Se-scFv with GPX activity was obtained. The GPX activity of the Se-scFv protein was characterized. Compared with other Se-scFv, the new human Se-scFv showed similar efficiency for catalyzing the reduction of hydrogen peroxide by glutathione. It exhibited pH and temperature dependent catalytic activity and a typical ping-pong kinetic mechanism. 展开更多

Vacancy engineering is a useful methodology in the development of catalysts and electrode materials.Herein,we report the introduction of Se-vacancy pairs in heteroatom-doped(N,B,and F)CoSe/Mo_(2)CT_(x) MXene(NBF-CoSe/Mo_(2)CT_(x))to enhance the hydrogen evolution reaction(HER)and supercapacitor activities via an ionic liquid-mediated method.Se vacancy pairs and heteroatom doping enable the reallocation of local electron states and add active sites,improving the electrochemical activity of NBF-CoSe/Mo_(2)CT_(x) with high HER activities over a broad range of pH.At a current density of 10 mA cm^(-2),overvoltages of 70 and 81 mV are respectively produced in 0.5 M H_(2)SO_(4)and 1 M KOH.The optimal structure also exhibits outstanding electrochemical performance in an asymmetric supercapacitor with an energy density of 34.2 Wh kg^(-1)at a power density of 15989.6Wkg^(-1).This study opens new avenues for the introduction of Se vacancies and heteroatom doping to improve the application performance. 展开更多

Nutrition is an important aspect of public health because it is linked to many significant diseases and health problems. This work is part of the promotion of traditional foods from the Democratic Republic of Congo in general and in the province of Bandundu, in particular by assessing the selenium content and the antioxidant capacity of wild edible mushrooms. Methanolic extracts from selected mushrooms were characterized for their chemical fingerprint by TLC and their in vitro antioxidant activities using ABTS, DPPH assays. Phytochemical screening revealed the presence of alkaloids, free amines, steroids and terpenes in all species. However, Auricularia delicata and Pleurotus tuberregium contain flavonoids. All extracts displayed a good radical-scavenging activity at the concentration range 1 - 300 μg/mL in the following order: Auricularia delicata > Lentinus cf cladopus > Pleurotus tuberregium > Marasmius buzungolo > Schizophillum commune. Studied mushrooms showed the interest quantity of selenium and L. cf cladodus had the highest concentration. Moderate selenium content of these wild mushrooms associated with their important antioxidant activities could provide health benefits to Bandundu’s populations in protecting against oxidative damage under different conditions including konzo. 展开更多

Forty-eight soil profiles were sampled from different ecological and pedogeochemical areas of China and their available selenium (Av-Se) contents were determined. Results showed that the content of Av-Se in the profiles of soils in China ranged within 2.49-18.10 μg kg--1 ) with a mean value of 10.01 μg kg--1,and was in the sequence of Ferralisols > Luvisols > Isohumisols > Aridisols. The correlation analysis between Av-Se and the soil physical-chemical properties revealed that the correlations between Av-Se and O.M., CEC, Fe2O3,Al2O3, ignition loss, Co and Zn were positive at a highly significant level, but those between Av-Se and pH,base saturation, CaO, Ba and Sr were all negative at a highly significant level. The reason of some diseases related to the Se deficiency might be the leaching loss of selenium in some soils. 展开更多

[目的]本文旨在探究末水条斑紫菜对健康C57BL/6J小鼠肠道微生物及其代谢产物的影响。[方法]对小鼠分别灌胃生理盐水(对照组)、1.15 g·kg^(-1)(低剂量组)和2.30 g·kg^(-1)(高剂量组)剂量的末水条斑紫菜,28 d后取小鼠标本进行病理切片观察、短链脂肪酸含量测定以及肠道菌群16S rDNA高通量测序。[结果]灌胃末水条斑紫菜组小鼠生长性能没有显著性变化。与对照组结肠长度53.50 mm相比,低剂量组小鼠结肠显著增长至65.60 mm;结肠组织病理切片显示,末水条斑紫菜可以增加杯状细胞的数量。通过对结肠内容物中短链脂肪酸含量测定,发现乙酸、丙酸、正戊酸与异戊酸含量均没有显著变化,低剂量组小鼠结肠内容物中正丁酸含量显著增加,高剂量组小鼠结肠内容物中正丁酸与异丁酸含量均显著增加。经16S rDNA高通量测序小鼠结肠内容物发现,灌胃低剂量与高剂量末水条斑紫菜均可降低微生物物种多样性,增加厚壁菌门的相对丰度,降低拟杆菌门的相对丰度。但肠道中杜氏杆菌属(Dobusiella)相对占比显著增加,Norank_f_norank_o_Clostridia_UCG-014、螺杆菌(Helicobacter)等致病菌相对占比减少。[结论]末水条斑紫菜能够促进结肠组织长度变化,调节健康C57BL/6J小鼠肠道菌群,促进有益菌生长以及丁酸的产生,最佳摄入量为1.15 g·kg^(-1)。 展开更多

[Methods] Four kinds of crops (tomato, leaf mustard, peanut, and carrot) were exogenous selenium fertilizer treated, to explore the selenium-enriching effects on these crops. [Results] Exogenous application of microbial nano-selenium can significantly increase the selenium content of the crops, of which the effects on big tomato (Weiyanuo) were the best. After spraying, the selenium content increased by 1.39 times, and at the same time the sugar content of tomato was also increased. [Conclusions] This study is expected to provide references for increase of the selenium content of agricultural products. 展开更多
