旨在探究O-β-N-乙酰葡糖胺(O-linked beta-N-acetylglucosamine,O-GlcNAc)修饰水平变化对牛卵母细胞体外成熟的影响。本研究以牛卵母细胞为研究对象,检测O-GlcNAc转移酶(O-GlcNAc transferase,OGT)、O-GlcNAc糖苷酶(O-GlcNAcase,OGA)及... 旨在探究O-β-N-乙酰葡糖胺(O-linked beta-N-acetylglucosamine,O-GlcNAc)修饰水平变化对牛卵母细胞体外成熟的影响。本研究以牛卵母细胞为研究对象,检测O-GlcNAc转移酶(O-GlcNAc transferase,OGT)、O-GlcNAc糖苷酶(O-GlcNAcase,OGA)及O-GlcNAc蛋白在牛体外成熟卵母细胞中的分布;将卵丘-卵母细胞复合体分别在添加4 mmol·L^(-1) OGT抑制剂BADGP和100μmol·L^(-1) OGA抑制剂PUGNAc的体外成熟液中进行体外成熟,将未添加组作为对照组,分别统计各组卵母细胞第一极体排出率和体外受精胚胎发育率,并采用荧光定量PCR和Western blot检测O-GlcNAc蛋白、OGT、OGA、GFAT和TXNIP的mRNA和蛋白表达。结果表明,OGT和O-GlcNAc蛋白共定位于牛体外成熟卵母细胞的细胞质和细胞核中,而OGA和O-GlcNAc蛋白共定位于体外成熟卵母细胞的细胞质中,且相对集中在卵母细胞的皮质区。与对照组相比,BADGP处理组和PUGNAc处理组卵母细胞第一极体排出率((52.8±5.1)%&(60.9±1.9)%vs.(70.8±5.4)%)和体外受精囊胚发育率((9.6±4.9)%&(10.0±5.8)%vs.(21.5±4.3)%)均显著降低(P<0.05)。BADGP处理显著降低了OGT的表达,使卵母细胞的O-GlcNAc修饰水平发生下调,OGA的表达降低;而在PUGNAc处理组中,卵母细胞OGA的表达显著下调,O-GlcNAc修饰水平升高,OGT的蛋白表达显著减少;抑制OGT显著上调己糖胺生物合成途径关键限速酶GFAT mRNA的表达,而抑制OGA则显著下调GFAT mRNA的表达;此外,O-GlcNAc修饰水平改变显著上调了葡萄糖调控关键因子TXNIP的表达。研究结果表明,O-GlcNAc修饰水平改变会降低牛卵母细胞体外成熟及发育能力,卵母细胞通过反馈调节OGT、OGA、GFAT以及TXNIP的表达,应对O-GlcNAc修饰水平的波动。 展开更多

冷暴露下骨骼肌中O-GlcNAc(O-linkedβ-N-acetylglucosamine)糖基化修饰水平显著升高,作为"应激感受器"调控细胞信号转导及基因转录等;而骨骼肌作为分泌器官通过收缩产生的肌源性IL-6具有免疫作用,更能够调节骨骼肌中糖、脂... 冷暴露下骨骼肌中O-GlcNAc(O-linkedβ-N-acetylglucosamine)糖基化修饰水平显著升高,作为"应激感受器"调控细胞信号转导及基因转录等;而骨骼肌作为分泌器官通过收缩产生的肌源性IL-6具有免疫作用,更能够调节骨骼肌中糖、脂代谢。本试验旨在检测不同时长冷暴露下小鼠成肌细胞系C2C12中O-GlcNAc相关蛋白和IL-6表达变化,初步探究冷暴露下O-GlcNAc与IL-6之间的潜在联系。本试验通过体外培养小鼠成肌细胞系C2C12,并随机分为不做冷暴露处理的对照组(cold 0 h组,也称为control组,37℃±0.5℃)和冷暴露(32℃±0.5℃)3、6、9、12 h处理组,采用Western blot方法检测不同时长冷暴露下O-GlcNAc糖基化水平和OGT、OGA及IL-6表达水平。结果显示,与对照组相比,冷暴露3 h,OGT表达水平显著升高(P<0.05),冷暴露6 h,OGA表达水平显著升高(P<0.05);冷暴露3 h,O-GlcNAc糖基化修饰水平和IL-6表达水平极显著升高(P<0.01)。不同时长冷暴露下,C2C12细胞中O-GlcNAc糖基化修饰水平及IL-6表达水平显著升高,并且,不同时长冷暴露下,O-GlcNAc糖基化修饰和IL-6表达变化趋势高度相似,提示冷暴露下O-GlcNAc糖基化修饰可能参与IL-6表达的调控。 展开更多

Geochronology of oil-gas accumulation (OGA) is a challenging subject of petroleum geology in multi-cycle superimposed basins.By K-Ar dating of authigenic illite (AI) and fluid inclusion (FI) analysis combined with apatite fission track (AFT) thermal modeling,a case study of constraining the OGA times of the Permian reservoirs in northeast Ordos basin (NOB) has been conducted in this paper.AI dating of the Permian oil-gas-bearing sandstone core-samples shows a wide time domain of 178-108 Ma.The distribution of the AI ages presents 2-stage primary OGA processes in the Permian reservoirs,which developed in the time domains of 175-155 Ma and 145-115 Ma with 2-peak ages of 165 Ma and 130 Ma,respectively.The FI temperature peaks of the samples and their projected ages on the AFT thermal path not only present two groups with a low and a high peak temperatures in ranges of 90-78℃ and 125-118℃,respectively corresponding to 2-stage primary OGA processes of 162-153 Ma and 140-128 Ma in the Permian reservoirs,but also appear a medium temperature group with the peak of 98℃ in agreement with a secondary OGA process of c.~30 Ma in the Upper Permian reservoirs.The integrated analysis of the AI and FI ages and the tectono-thermal evolution reveals that the Permian reservoirs in the NOB experienced at least 2-stage primary OGA processes of 165-153 Ma and 140-128 Ma in agreement with the subsidence thermal process of the Mid-Early Jurassic and the tectono-thermal event of the Early Cretaceous.Then,the Upper Permian reservoirs further experienced at least 1-stage secondary OGA process of c.~30 Ma in coincidence with a critical tectonic conversion between the slow and the rapid uplift processes from the Late Cretaceous to Neogene. 展开更多

Responses to oligogalacturonic acid (OGA) were determined in transgenic Arabidopsis thaliana seedlings expressing the calcium reporter protein aequorin. OGA stimulated a rapid, substantial and transient increase in the concentration of cytosolic calcium ([Ca2+]cyt) that peaked after ca. 15 s. This increase was dose-dependent, saturating at ca. 50 μg Gal equiv/ml of OGA. OGA also stimulated a rapid generation of H2O2. A small, rapid increase in H2O2 content was followed by a much larger oxidative burst, with H2O2 content peaking after ca. 60 min and declining thereafter. Induction of the oxidative burst by OGA was also dose-dependent, with a maximum response again being achieved at ca. 50 μg Gal equiv/mL. Inhibitors of calcium fluxes inhibited both increases in [Ca2+]cyt and [H2O2], whereas inhibitors of NADPH oxidase blocked only the oxidative burst. OGA increased strongly the expression of the defence-related genes CHS,GST, PAL and PR-1. This induction was suppressed by inhibitors of calcium flux or NADPH oxidase, indicating that increases in both cytosolic calcium and H2O2 are required for OGA-induced gene expression. 展开更多

利用CRISPR/Cas9系统建立成体心肌细胞特异性Oga基因敲除小鼠模型

目的 利用腺相关病毒9(AAV9)介导的CRISPR/Cas9系统构建成体心肌细胞特异性Oga基因敲除小鼠模型,来研究内源性OGA在成体心脏稳态维持中的功能。方法 首先,筛选靶向Oga编码区的有效sgRNA,构建包装表达有效sgRNA的AAV9病毒。其次,建立心... 目的 利用腺相关病毒9(AAV9)介导的CRISPR/Cas9系统构建成体心肌细胞特异性Oga基因敲除小鼠模型,来研究内源性OGA在成体心脏稳态维持中的功能。方法 首先,筛选靶向Oga编码区的有效sgRNA,构建包装表达有效sgRNA的AAV9病毒。其次,建立心肌细胞特异性SpCas9表达小鼠(α-MHC^(Cas9)),并通过腹腔注射方式将AAV9-sgOga注射到小鼠体内。通过qPCR和Western blot印记杂交实验检测Oga的表达情况,确定Oga敲除是否成功。最后,通过组织学分析心脏结构,以及超声心动图分析心脏功能,来分析Oga对心脏稳态维持的影响。结果 筛选到2条可以有效靶向Oga编码区的sgRNA,通过AAV9递送实现了Oga基因在心脏组织的有效敲除,且导致O-GlcNAcylation表达显著上升;组织学分析和超声心动图分析发现基因敲除小鼠心脏结构及功能在基础水平与对照小鼠无显著变化。结论 利用AAV9介导的CRISPR/Cas9系统成功建立了成体心肌细胞Oga基因敲除的小鼠模型,为研究OGA介导的O-GlcNAcylation清除在心脏稳态维持中的功能和机制提供了有效的小鼠模型。 展开更多

This paper evaluates the place of public relations in the image management strategies of the Nigeria Security and Civil Defence Corps (NSCDC) particularly after the infamous “Oga at the Top” interview by the Lagos State Commandant of the Corps. The paper uses questionnaire and the interview schedule as instruments to gather data from members of the public who are familiar with the “Oga at the Top” incident and the public relations officers (PROs) of the NSCDC respectively. From the data gathered, we conclude that the NSCDC adopted different image management strategies to salvage the corporate image of the Corps;that the image management strategies have engendered a cordial relationship between the media and the Corps;and that the infamous “Oga at the Top” incident brought some fame to the Corps as free advertisement as well as questioned the capacity of its officials to discharge their duties effectively. As a fallout of this evaluation, it is recommended that NSCDC management should endeavour to engage only professionals in the field of public relations who would understand and implement public relations objectives and functions in the organisation and that all ranking officers of the NSCDC should endeavour to involve the public relations unit in all their engagement with the external publics to avoid a repeat of the “Oga at the Top” incident that affected the Corps’ image negatively. 展开更多
