Objective The SOD, POD and APX enzyme activities of tetraploid lines and diploid control line of Chrysanthemum cinerariifolium in test-tube and field were determined in order to provide a reference for the selective breeding of high resistance good tetraploid varieties of C. cinerariifolium. Method The SOD, POD and APX activities of each sample were determined. Every material should have 3 replicates which been determined and calculated mean value to analyze. Result The various kinds of antioxidase activities in protective enzyme system of tetraploid lines were increased generally in comparison with that of diploid control line. And there is certain relativity between the samples in field and in test-tube of these lines. Conclusion The antioxidase activities of each line in test-tube could be considered as reference index to screen high resistance good tetraploid varieties of C. cinerariifolium. 展开更多

[Objective] This study was conducted to determine the physiological and biochemical characteristics of potato tubers infected by Fusarium trichothecioides. [Method] The soluble protein content, SOD activity, POD activity and malondialdehyde (MDA) content of the tubers of two potato varieties Atlantic and Keshan 1 infected by F. trichothecioides were measured 0, 3, 6, 9, 12, 24 and 48 h later. [Result] Com- pared with the control uninfected, the contents of soluble protein and MDA in the tu- bers of both potato varieties infected with F. trichothecioides decreased, while the SOD activity increased, indicating that SOD activity had a certain relationship with the resistance of potato to dry rot. The POD activity in Keshan 1 which is resistant to F. trichothecioides was higher than in Atlantic, which is susceptible to F. trichothe- cioides, indicating that POD activity was closely related with potato's resistance to Fusarium dry rot. [Conclusion] The physiological and biochemical characteristics of potato tubers infected by F. trichothecioides will provide some references for the early waming and prevention of Fusariurn dry rot. 展开更多

The regulatory mechanism of soil cadmium (Cd) on antioxidases in sorghum was studied using 3 sorghum species viz., sweet sorghum [Sorghum bicolor (L.)] Moench. cv. Hunnigreen, sorghum hybrid sudangrass (Sorghum bicolor × S. sudanense, cv. Everlush) and sudangrass [Sorghum sudanense (Piper) Stapf cv. Xinjiang]. The results showed that low concentrations of Cd^2+ (410 mg kg-0 induced the activities of 4 antioxidases in leaves of the 3 species of sorghum, but high Cd^2+ concentrations (≥ 50 mg kg-0 significantly reduced the activities of all the four antioxidases. The glutathione (GSH) contents in leaves of the 3 species of sorghum correlated positively with the Cd^2+ concentrations. The maximum antioxidase activities were observed at the elongation stage of sorghum during the whole growth period. 展开更多

With the potato variety Xishu 1 as the test materials, this paper adopted tissue culture to study the effect of Pb and Cd stress with different mass fractions on potato growth and the antioxidase system. The results showed that under the stress of Pb (200 mg/kg) and Cd (20 mg/kg), growth of potato had no obvious difference with that in control group; under the stress of Pb ( 〉200 mg/kg) and Cd ( 〉20 mg/kg), potato growth suspended, and its physiological indexes such as dry weight, plant height, root number, root length were inhibited obviously. With the increasing mass fraction of Pb, relative electrical conductivity, MDA and proline of potato leaves showed the tendency of increase, and the results in high mass fraction treatments showed significant difference with those in the control group (P 〈 0.05 ), CAT, POD and SOD activity of leaves increased greatly with the increasing mass fraction. In the Pb stress treatment (200 mg/kg), relative electrical conductivity, MDA and proline of potato leaves showed no obvious difference with those in the control group, and the results in high mass fraction treatments were significantly higher than those in the control group ( P 〈 O. 05 ), CAT and SOD activity increased greatly with the increasing mass fraction ( P 〈 0.05 ). Under the stress of Pb and Cd, synthesis of soluble protein in potato leaves was strongly inhibited. The results showed that potato was tolerant to Pb and Cd of lower mass fraction, and damage of potato caused by Pb and Cd stress could be relieved by improving activity of antioxidase and reducing peroxidation of membrane lipid, and the tolerance reduced with the increase of stress. 展开更多

To investigate the effects of ultraviolet blood irradiation and oxygenation (UBIO) on free radicals and antioxidase after spinal cord injury in rabbits Methods Totally, 186 rabbits were used and divided randomly into four experimental groups: control (n=6), blood transfusion (n=24), injured (n=96) and treatment (n=60) groups The relative intensity of free radical (FR) signals, malondialdehyde (MDA) content, as well as the activity of superoxide dismutase (SOD) and glutathione peroxidase (GSH PX) were compared among the four groups at 6, 24, 48, and 72 hours and 6 days after injury Results The relative intensity of FR signals in spinal cord tissue in the injured group increased at 48 hours and showed a striking difference compared with the control group; in the treatment group, it decreased and showed a striking difference compared with the injured group MDA content in blood in the injured group increased and showed a striking difference at 6, 24 and 48 hours and showed a significant difference at 72 hours and 6 days after injury compared with the control group In the treatment group, MDA content in blood decreased and showed a significant difference at 48 hours compared with the injured group MDA content in spinal cord tissue increased in the injured group and showed a striking difference compared with the control group; in the treatment group, it decreased and showed a striking difference compared with the injured group at the corresponding times The activity of SOD in blood and spinal cord tissue decreased in the injured group and showed a striking difference compared with the control group; in the treatment group, it increased and showed a striking difference compared with the injured group at the corresponding times The changes in activity of GSH PX in blood and spinal cord tissue were similar to that in SOD No significant difference was observed between the blood transfusion and control groups Conclusion UBIO can ease free radical damages and elevate the activity of antioxidases after spinal cord injury in rabbits 展开更多

为探究土壤施硒对茶树叶片硒含量、抗氧化酶活性及茶叶品质的影响,研究人员采用随机区组法在茶树种植土壤中施加不同浓度的亚硒酸钠,随后,依据国家相应标准测定茶树老叶及嫩叶中的硒含量、其他矿物质元素含量、抗氧化活性酶的活性以及... 为探究土壤施硒对茶树叶片硒含量、抗氧化酶活性及茶叶品质的影响,研究人员采用随机区组法在茶树种植土壤中施加不同浓度的亚硒酸钠,随后,依据国家相应标准测定茶树老叶及嫩叶中的硒含量、其他矿物质元素含量、抗氧化活性酶的活性以及品质生化成分含量。结果表明:适量的土壤施硒能够显著提高茶树老叶及嫩叶的硒含量,且随着施硒浓度的增加,叶片总硒含量呈显著上升趋势;施硒处理导致茶树老叶中钾、钙和铁的含量显著降低,而对嫩叶中钾、镁和铁的含量无显著影响。嫩叶中锌元素的含量极显著且高于其他处理及对照组。同时,施硒处理显著提升了抗氧化活性酶(过氧化物酶、多酚氧化酶、苯丙氨酸解氨酶和超氧化物歧化酶)的活性,儿茶素、咖啡碱、茶多酚及氨基酸等品质生化成分的含量也有所增加。 展开更多

镁营养缓解植物铝毒机制的研究进展 被引量:1

世界耕地面积的40%以上为酸性土壤,铝(Al)毒害是酸性土壤中限制植物生长和产量的主要障碍因子之一。镁(Mg)是植物的必需营养元素,是植物细胞内含量最丰富的游离二价金属元素,可通过多种途径参与植物非生物逆境的抗性调控过程。Mg^(2+)和... 世界耕地面积的40%以上为酸性土壤,铝(Al)毒害是酸性土壤中限制植物生长和产量的主要障碍因子之一。镁(Mg)是植物的必需营养元素,是植物细胞内含量最丰富的游离二价金属元素,可通过多种途径参与植物非生物逆境的抗性调控过程。Mg^(2+)和Al^(3+)的水化半径相似,这两种离子竞争结合植物离子转运体和其他重要的生物分子。本文基于中国知网(CNKI)和Web of Science核心数据库,共检索到79篇1989—2024年发表的植物镁铝互作研究文献。分析结果表明,外源镁可以有效减轻酸性土壤中的植物铝毒害,其作用机制包括:1)Mg^(2+)与Al^(3+)有效竞争植物质膜上的结合位点;2)有效增加植物有机酸分泌;3)上调铝诱导的镁转运蛋白基因表达,增强对镁的吸收,提高耐铝性;4)显著增强质膜H+-ATPase促进有机酸的分泌;5)增强抗氧化酶活性,减少ROS的产生,降低铝诱导的氧化应激风险;6)提高光合作用和碳氮代谢相关酶活性,缓解铝胁迫导致的光合障碍和源库失衡。除此之外,镁还在植物基本的细胞代谢过程中发挥关键作用,如维持质膜和液泡质体中的质子泵活性,影响一氧化氮生物合成酶活性和相关基因表达,显著提升高铝毒农田土壤中植物产量和品质。有趣的是,在水稻、小麦等单子叶植物中,毫摩尔浓度的Mg^(2+)主要通过降低Al^(3+)在细胞壁和质膜结合位点的饱和活性来减轻土壤铝的毒性。在大豆(Glycine max)、赤小豆(Vigna umbellata)、蚕豆(Vicia faba)等双子叶豆科植物中,微摩尔浓度的Mg^(2+)可以增强有机配体的生物合成,缓解土壤铝毒害。未来的研究工作应着重从以下3个方向展开:一是整合基因组学、转录组学和蛋白质组学等多组学技术,深入解析镁和铝胁迫响应下的基因表达谱及蛋白质组变化,全面揭示镁与铝胁迫互作的分子调控网络,为培育耐铝毒害作物提供理论依据;二是探究外源镁素营养(如镁素肥料)对酸性铝毒障碍土壤中有益微生物群落组装过程及机制的影响;三是开发基于镁基的土壤调理剂以及耐铝作物育种策略,以增强作物对铝毒的耐受性。 展开更多

【目的】油莎豆MYB转录因子基因CeMYB154参与盐胁迫响应过程,克隆及表征油莎豆CeMYB154并分析其耐盐功能,为油莎豆耐盐育种提供分子基础和基因资源。【方法】基于油莎豆参考基因组信息,克隆CeMYB154的全长CDS序列,利用生物信息学软件对... 【目的】油莎豆MYB转录因子基因CeMYB154参与盐胁迫响应过程,克隆及表征油莎豆CeMYB154并分析其耐盐功能,为油莎豆耐盐育种提供分子基础和基因资源。【方法】基于油莎豆参考基因组信息,克隆CeMYB154的全长CDS序列,利用生物信息学软件对其氨基酸序列特征及其启动子上顺式作用元件进行分析,利用分子生物学技术对其进行亚细胞定位和转录激活验证。构建CeMYB154过表达载体,利用农杆菌介导法转化拟南芥,并对过表达株系的耐盐表型和生理指标进行分析。【结果】成功克隆了长度为780 bp的CeMYB154 CDS序列,其编码蛋白序列中2个典型的MYB结构域,属于R2R3型MYB转录因子。蛋白序列比对分析显示,CeMYB154蛋白在多个物种间序列高度保守。启动子区域含有多种激素(如ABA)响应以及MYB响应相关顺式作用元件。此外,亚细胞定位和转录激活分析显示,CeMYB154是一个具有转录激活活性的核转录因子。成功创制了过表达CeMYB154转基因拟南芥,获得了纯合T3转基因阳性株系。在盐胁迫下,与野生型拟南芥相比,转基因植株幼苗生长状态(叶片大小、颜色以及根长)较好;并且,转基因植株中MDA和H2O2含量均显著降低,而CAT、POD和SOD抗氧化酶活性则显著增强,表明过表达CeMYB154基因提高了拟南芥的耐盐性。【结论】油莎豆CeMYB154具有正向调控盐胁迫响应的功能,过表达CeMYB154基因可以增强抗氧化酶活性而提高转基因植株的耐盐性。 展开更多

西瓜成熟过程中果肉水渍化损伤生理响应机制研究

为探讨西瓜成熟过程中果肉水渍化损伤发生的生理响应机制,以果肉水渍化差异显著的西瓜种质07058(耐水渍化)和伊选(易水渍化)为材料,对西瓜成熟过程中5个时期的果肉进行果肉损伤比例、糖酸含量、丙酮酸(PA)、MDA含量、SOD、CAT、POD等抗... 为探讨西瓜成熟过程中果肉水渍化损伤发生的生理响应机制,以果肉水渍化差异显著的西瓜种质07058(耐水渍化)和伊选(易水渍化)为材料,对西瓜成熟过程中5个时期的果肉进行果肉损伤比例、糖酸含量、丙酮酸(PA)、MDA含量、SOD、CAT、POD等抗氧化酶活性,果胶、纤维素等细胞壁结构物质含量及细胞壁降解酶活性的测定。结果表明,在授粉后20 d,果肉易水渍化材料伊选水渍化比例快速上升,授粉后35 d,损伤比例达45%左右,而耐水渍化材料07058无明显水渍化现象。与同时期耐水渍化材料07058相比,伊选在授粉后20 d,蔗糖、葡萄糖及果肉酸含量均高于07058,果糖含量先升高后下降,授粉后25 d含量低于07058;PA、MDA含量快速上升,且显著高于07058。在伊选发生水渍化初期(授粉后20 d),SOD、CAT活性快速升高,活性高于07058,随后又迅速下降,活性低于07058;POD活性先下降后回升,但各时期活性均低于07058。伊选的纤维素含量、总果胶含量整体低于07058,可溶性果胶含量高于07058。在果肉水渍化快速发生阶段(授粉后20~25 d),伊选的纤维素酶(CL)及果胶裂解酶(PL)活性快速上升,并高于07058;在授粉后25 d时,伊选PL活性是07058的1.9倍。相关性分析结果显示,西瓜水渍化损伤与蔗糖含量、MDA含量、CL活性等多个生理指标显著正相关;与果糖含量、SOD活性和纤维素含量等显著负相关。研究表明,西瓜果肉水渍化与糖代谢、膜损伤、氧化应激响应、细胞壁降解等多个生理活动密切相关,是多个生理过程发生变化协同作用的结果。 展开更多

[Objective] The aim of this paper is to investigate the influences of exogenous 5-aminolevulinic acid (5-aLA) application on the growth and mechanism of winter oilseed rape (Brassica napus L.) before winter. [Method] For this purpose a split-plot experimental design with four treatments of seed soaking with 0,25,50 and 75 mg/L ALA solution (CK,A1,A2 and A3) was carried out from October 2009 to January 2010 in order to observe the influences upon the growth and relevant physiological indexes of Zhongshuang 9. [Result] Seed soaking with ALA solution significantly promotes the growth of double-9 before winter,which is probably due to the fact that ALA application enhances its photosynthesis,absorption of nutrients,and its anti-oxidation abilities. The research also finds that ALA treatment notably increases its photosynthetic pigment contents,root activities,leaf nitrate reductase activities,soluble sugar contents,antioxidase activities,and significantly decreases malondialdehyde(MDA) content. [Conclusion] These results indicate that seed soaking with 50 mg/L ALA solution might have a potential application in increasing the productivity of winter oilseed rape in agricultural management. 展开更多

[Method] MDA content was determined by thiobarbituric acid colorimetric method; mitochondrial swelling degree was determined by spectrophotometry; the superoxide anion radical scavenging ability was determined using reduced coenzyme I-tetrazolium-phenazine methosulfate as the superoxide anion generation system. Mice were hypodermically injected in the back and neck with D- galactose, after 50 d, the effects of AEO on the activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and anti-hydroxyl radicals in mouse liver were determined using kits. [Result] AEO can efficiently reduce the liver injury and inhibit mitochondrial swelling induced by Fe2+-L-Cys, which can also scavenge superoxide anion and improve the activities of antioxidant enzymes. [Conclusion] This study provided scientific basis for the development and application of AEO resources. 展开更多

BACKGROUND Fruits of Gardenia jasminoides(G.jasminoides)have extremely high medicinal value.However,the quality and traits of the plants vary significantly based on their provenances.In addition,the behaviour of the known bioactive components,such as geniposide and crocin,has been the primary focus of the research on G.jasminoides.However,the identification of unknown bioactive components and their metabolomics remains underexplored.Therefore,analysing the metabolic differences between gardenias from different sources is essential to provide a comprehensive theoretical basis for the evaluation of G.jasminoides and germplasm resource identification.AIM To systematically evaluate the morphology,secondary metabolites,typical active ingredients,and antioxidant activity of wild G.jasminoides fruits.METHODS Gardenia fruits were collected from different provenances.Metabolites were identified via ultra-high-performance liquid chromatography–tandem mass spectrometry(UPLC-MS/MS).The metabolic differences were compared using hierarchical cluster analysis(HCA).The antioxidant capacity was evaluated using 2,2-diphenyl-1-picrylhydrazyl radical scavenging assays and ferric reducing antioxidant power assays,and its correlation with typical active ingredients was analysed.RESULTS A total of 444 and 240 metabolites were identified using UPLC-MS/MS in positive and negative ion modes,respectively.The HCA results of the flavonoids indicated that the higher content of flavonoids was in the fruits from Lukou.The differential analysis of metabolites in fruits from Shaoyang,Miluo and Lukou showed that the fruit from Miluo had the highest upregulated differential metabolites.CONCLUSION The metabolic characteristics of the Ningxiang and Xiangxi extracts were similar,while those of Lukou,Miluo and Shaoyang extracts differed significantly. 展开更多

三叶草斑潜蝇[Liriomyza trifolii (Burgess)]是中国瓜菜上的重要入侵害虫之一。为进一步探讨三叶草斑潜蝇对温度胁迫的适应性机制,对三叶草斑潜蝇的过氧化氢酶(Catalase,CAT)序列特征、结构和系统发育等进行了生物信息学分析,并利用实... 三叶草斑潜蝇[Liriomyza trifolii (Burgess)]是中国瓜菜上的重要入侵害虫之一。为进一步探讨三叶草斑潜蝇对温度胁迫的适应性机制,对三叶草斑潜蝇的过氧化氢酶(Catalase,CAT)序列特征、结构和系统发育等进行了生物信息学分析,并利用实时荧光定量PCR检测了LtCAT基因在不同发育阶段、温度胁迫下的表达情况。本研究克隆获得了三叶草斑潜蝇CAT基因序列,其开放阅读框长度为1 542 bp,共编码513个氨基酸。系统发育树表明,三叶草斑潜蝇与橘小实蝇(Bactrocera dorsalis)、瓜实蝇(Zeugodacus cucurbitae)、地中海实蝇(Ceratitis capitata)CAT基因亲缘关系更近,聚为一支,置信度达90%。LtCAT基因在幼虫、蛹、成虫均有差异表达,在蛹期表达量最高;温度胁迫处理后,LtCAT基因表达随着温度变化积极响应。本研究为后续深入研究三叶草斑潜蝇CAT基因的功能提供依据。 展开更多

以黑果枸杞、金露梅、蒙古莸、柠条和花棒5种西北旱区植物为材料,采用盆栽试验研究了干旱胁迫对植物叶片光合、水分和生理生化的影响.结果表明:随干旱胁迫时间的延长,5种植物叶片的保水力均升高,叶片相对含水量先升后降,叶绿素含量和叶... 以黑果枸杞、金露梅、蒙古莸、柠条和花棒5种西北旱区植物为材料,采用盆栽试验研究了干旱胁迫对植物叶片光合、水分和生理生化的影响.结果表明:随干旱胁迫时间的延长,5种植物叶片的保水力均升高,叶片相对含水量先升后降,叶绿素含量和叶绿素荧光参数均不同程度地降低,花棒和黑果枸杞的光合系统受到的影响最小,金露梅的光合系统受损最严重.干旱胁迫后金露梅叶片的电导率和丙二醛含量先降低后急剧上升,超氧化物歧化酶(SOD)和过氧化物酶(POD)活性先升高后急剧降低.其余4种植物随土壤含水量的降低电导率先升后降,丙二醛含量先升后降或先降后升,SOD和POD活性总体呈上升趋势,其中蒙古莸SOD活性和花棒POD活性在所有植物中升幅最大,干旱28 d后分别升至为胁迫前的2.1、14.1倍.随胁迫时间的延长,5种植物各有机溶质的积累量存在较大差异,金露梅积累最多的可溶性糖和游离脯氨酸,28 d后分别增至为原来的2.1、23.4倍,花棒积累最多的可溶性蛋白,28 d后增至为原来的2.0倍.隶属函数法表明,植物耐旱能力为花棒>黑果枸杞>柠条>蒙古莸>金露梅.5种旱区植物可通过提高抗氧化酶活性和积累有机溶质对干旱胁迫进行积极的反馈,以减轻细胞受到的伤害.金露梅对水分亏缺的敏感性最高,试验期间其干旱程度已经超出了自身调节能力的阈值. 展开更多

为探讨红壤地玉米-豌豆间作种植模式对铜污染的响应机制,通过盆栽试验,研究了不同铜浓度(0、100、200、400、600 mg· kg-1)对玉米单作和玉米间作豌豆条件下植株生物量、铜含量、玉米抗氧化酶活性[超氧化物歧化酶(SOD)、过氧... 为探讨红壤地玉米-豌豆间作种植模式对铜污染的响应机制,通过盆栽试验,研究了不同铜浓度(0、100、200、400、600 mg· kg-1)对玉米单作和玉米间作豌豆条件下植株生物量、铜含量、玉米抗氧化酶活性[超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和过氧化物酶(POD)]及丙二醛含量(MDA)的影响。结果表明:在高 Cu2+浓度(600 mg·kg-1)胁迫下,间作玉米地上部和地下部干重较单作分别提高了20豫和36.6豫,与单作相比,随着 Cu2+浓度升高(100、200、400、600 mg·kg-1),间作模式下玉米地上部铜含量分别降低了86.81豫、44.57豫、22.01豫、11.11豫,而地下部铜含量则分别提高了78.89豫、24.79豫、35.29豫、13.31豫,且差异均达到显著水平。在不同Cu2+浓度胁迫下,玉米叶和根中的 SOD、POD、CAT 活性和 MDA 含量均随着 Cu2+浓度的增加而提高,仅 Cu2+浓度达到600 mg·kg-1时玉米体内的 CAT 活性有所下降。间作条件下玉米根的 SOD 活性较单作提高了48.07豫~117.27豫,间作玉米叶的 SOD 活性较单作提高了11.30豫~46.90豫。不同种植模式对 Cu2+胁迫条件下的玉米 POD 活性均没有显著影响。在 Cu2+(0~400 mg·kg-1)胁迫下,间作玉米叶的 CAT 活性较单作均显著提高,分别提高了71.37豫、140.40豫、229.80豫和161.75豫,间作玉米根的 CAT 活性与单作无显著差异。间作玉米根的 MDA 含量较单作降低了26.13豫~64.53豫;在100、200 mg·kg-1 Cu2+胁迫下,间作玉米叶的 MDA 含量较单作降低了0.30豫和26.24豫,但在400、600 mg·kg-1 Cu2+胁迫下,间作玉米叶的 MDA 含量较单作提高了32.62豫和93.51豫。综上所述,在一定范围的 Cu2+胁迫条件下,玉米根和叶中的抗氧化酶活性及 MDA 含量均有所提高来维持正常生长,间作模式在 Cu2+胁迫下对玉米根和叶的抗氧化酶系统能起到一定的保护作用。 展开更多
