BACKGROUND Irreversible electroporation(IRE)is a novel local tumor ablation approach with the potential to activate the host’s immune system.However,this approach is insufficient to prevent cancer progression,and complementary approaches are required for effective immunotherapy.AIM To assess the immunomodulatory effects and mechanism of IRE combined antiprogrammed cell death protein 1(PD-1)treatment in subcutaneous pancreatic cancer models.METHODS C57BL-6 tumor-bearing mice were randomly divided into four groups:Control group;IRE group;anti-PD-1 group;and IRE+anti-PD-1 group.Tumor-infiltrating T,B,and natural killer cell levels and plasma concentrations of T helper type 1 cytokines(interleukin-2,interferon-γ,and tumor necrosis factor-α)were evaluated.Real-time PCR was used to determine the expression of CD8(marker of CD8+T cells)in tumor tissues of the mice of all groups at different points of time.The growth curves of tumors were drawn.RESULTS The results demonstrated that the IRE+anti-PD-1 group exhibited significantly higher percentages of T lymphocyte infiltration,including CD4+and CD8+T cells compared with the control group.Additionally,the IRE+anti-PD-1 group showed increased infiltration of natural killer and B cells,elevated cytokine levels,and higher CD8 mRNA expression.Tumor volume was significantly reduced in the IRE+anti-PD-1 group,indicating a more pronounced therapeutic effect.CONCLUSION The combination of IRE and anti-PD-1 therapy promotes CD8+T cell immunity responses,leading to a more effective reduction in tumor volume and improved therapeutic outcomes,which provides a new direction for ablation and immunotherapy of pancreatic cancer. 展开更多

Background: Anti-programmed death-1/programmed death-ligand 1(PD-1/PD-L1) immunotherapy has been proved to be effective on gastric cancer in ongoing clinical trials. However, the value of PD-L1 in predicting responses of patients with gastric cancer to anti-PD-1/PD-L1 immunotherapy is controversial. Some studies suggested that intra-and inter-tumoral heterogeneity of PD-L1 expression might explain the controversy.This study aimed to analyze the expression of PD-L1, PD-L2, and PD-1 as well as CD8(+) T-cell density in primary tumors and lymph nodes from patients with stage T1-4 N+M0 gastric adenocarcinoma to explore the heterogeneity of PD-1 signaling pathway molecules.Methods: In primary tumors and metastatic as well as non-metastatic lymph nodes from patients with stage T1-4 N+M0 gastric adenocarcinoma, we detected PD-L1 and PD-L2 expression with immunohistochemistry. CD8(+)T-cell density in primary tumors and PD-1 expression on CD8(+)T cells were detected with immunofluorescence. Univariate analysis was used to determine the prognostic values of them. Cox proportional hazard regression model was used to identify independent risk factors that affect patients' overall survival and disease-free survival.Results: Among 119 eligible patients who had undergone surgical resection, the positive rate of PD-L1 was higher in metastatic lymph nodes than in primary tumors(45.4% vs. 38.7%, P = 0.005); the positive rate of PD-1 on CD8(+)T cells was significantly higher in primary tumors and metastatic lymph nodes than in tumor-free lymph nodes(both P < 0.001). The intensity of PD-1 expression on CD8(+) T cells in primary tumors and in metastatic lymph nodes were stronger than that in tumor-free lymph nodes from the same patient. Beside, the positive rate of PD-L2 did not show any differences between primary tumors and metastatic lymph nodes. In multivariate analysis, PD-L1 expression,PD-L2 expression, a low density of CD8(+) T cells in primary tumors, and PD-1 expression on CD8(+) T cells in primary tumors were associated with poor prognosis.Conclusion: The expression of PD-L1 is heterogeneous in primary tumors and in metastatic lymph nodes from patients with stageT1-4 N+M0 gastric adenocarcinoma, which might explain the inconsistent results in assessing the prognostic value of PD-L1 expression in previous studies. 展开更多

METHODS:Sixty-four patients in different natural stages of chronic HBV infection were enrolled in this study.PD-1 expression in total T cells was detected by flow cytometry.Levels of total CD8+T cell responses and proliferation in relation to PD-1 expression levels were analyzed with intracellular staining and PD-1/ PD-L1 blockage. RESULTS:The PD-1 expression in T cells was dynamically changed during the natural course of chronic HBV infection,did not significantly increase in the immune tolerance phase,and returned to normal in the inactive virus carrier stage.Blockage of the PD-1/PD-L1 pathway could not affect the T-cell response in the immune tolerance and inactive virus carrier stages of chronic HBV infection.However,it could significantly restore the T-cell response in the immune clearance stage of chronic HBV infection.Furthermore,the PD-1 expression level in T cells was associated with the alanine aminotransferase level during the immune clearance stage of chronic HBV infection. CONCLUSION:The PD-l/PD-L1 pathway plays a different role in T-cell response during the natural course of chronic HBV infection. 展开更多

溶瘤痘苗病毒-LZ8治疗淋巴瘤的作用机制研究

目的探讨溶瘤痘苗病毒(OVV)-灵芝(LZ)8治疗淋巴瘤的作用机制。方法利用基因重组技术构建OVV-LZ8和对照病毒OVV-增强绿色荧光蛋白(EGFP),利用流式细胞仪检测OVV-EGFP对淋巴瘤细胞(包括A20、Raji、OCI-LY3、EL4等)的体外感染效率。采用空... 目的探讨溶瘤痘苗病毒(OVV)-灵芝(LZ)8治疗淋巴瘤的作用机制。方法利用基因重组技术构建OVV-LZ8和对照病毒OVV-增强绿色荧光蛋白(EGFP),利用流式细胞仪检测OVV-EGFP对淋巴瘤细胞(包括A20、Raji、OCI-LY3、EL4等)的体外感染效率。采用空斑滴度测定法检测OVV-LZ8在淋巴瘤细胞中的复制能力,3-(4,5-二甲基噻唑-2)-2,5-二苯基四氮唑溴盐法检测OVV-LZ8对淋巴瘤细胞的杀伤能力。将63只已构建A20淋巴瘤模型的BALA/C雌性小鼠按随机数字表法分为磷酸盐缓冲液(PBS)组18只、OVV-EGFP组(注射OVV-EGFP)9只、OVV-LZ8组(注射OVV-LZ8)18只、程序性死亡受体-1(PD-1)抗体组(注射PD-1抗体)9只、联合组(注射OVV-LZ8+PD-1抗体)9只,测量注射药物期间小鼠肿瘤体积,并采用流式细胞术检测注射药物结束后第2天肿瘤组织免疫细胞浸润情况。结果成功构建OVV-LZ8,其在淋巴瘤细胞中的复制能力不受外源性基因LZ8插入的影响。OVV-EGFP能有效感染4种淋巴瘤细胞,其中对A20的感染效率最高,10感染复数感染48 h的EGFP阳性率为23.86%。OVV-LZ8对4种淋巴瘤细胞均有明显的杀伤作用,且这种杀伤作用与浓度有关(均P<0.01)。动物实验结果显示,与PBS组、OVV-EGFP组比较,OVV-LZ8组淋巴瘤的生长得到有效控制(均P<0.05),淋巴瘤组织树突状细胞(DC)、CD8^(+)T细胞和自然杀伤(NK)细胞比例增加(均P<0.05);与OVV-LZ8组比较,联合组淋巴瘤的生长进一步抑制(均P<0.05),淋巴瘤组织CD8^(+)T细胞、CD4^(+)T细胞、NK细胞比例以及肿瘤浸润淋巴细胞、DC数量均增加(均P<0.05),骨髓来源的抑制性细胞数量减少(均P<0.05)。结论本研究构建的OVV-LZ8能在体内外有效抑制淋巴瘤细胞增殖,而且OVV-LZ8联合PD-1抗体能显著提高抗肿瘤效果。 展开更多

天然气压缩因子在天然气工程计算中具有重要的地位,其计算方法有多种,其中就包括由美国燃气协会提出的基于气体组成来计算压缩因子的AGA8-92DC方程。根据GB/T 17747.2-2011标准所给出的AGA8-92DC方法计算过程,用Visual Basic6.0 progra... 天然气压缩因子在天然气工程计算中具有重要的地位,其计算方法有多种,其中就包括由美国燃气协会提出的基于气体组成来计算压缩因子的AGA8-92DC方程。根据GB/T 17747.2-2011标准所给出的AGA8-92DC方法计算过程,用Visual Basic6.0 program编写了计算程序。为了验证程序的正确性,分别将程序计算值与GB/T 17747.2标准提供的计算值和含碳贫气气样实测值进行了对比,且获得了较小的计算误差。由计算值与实测值的对比结果可知,通过Visual Basic6.0 program所编写的AGA8-92DC方法计算程序而得到的计算结果能够满足工程上的要求;对于中高压含碳贫气,AGA8-92DC方法具有较高的计算准确度;当气体温度升高时,AGA8-92DC方法的计算误差将有所减小。 展开更多

背景目前,胃癌组织细胞程序性死亡配体1(PD-L1)表达情况和CD_(8)^(+)肿瘤浸润T淋巴细胞(TILs)密度是否可作为胃癌患者预后的评估指标尚不明确。目的分析胃癌组织PD-L1表达情况、CD_(8)^(+)TILs密度及其与患者临床病理特征和预后的关系... 背景目前,胃癌组织细胞程序性死亡配体1(PD-L1)表达情况和CD_(8)^(+)肿瘤浸润T淋巴细胞(TILs)密度是否可作为胃癌患者预后的评估指标尚不明确。目的分析胃癌组织PD-L1表达情况、CD_(8)^(+)TILs密度及其与患者临床病理特征和预后的关系。方法选择2016年1月至2018年3月在南阳市第一人民医院行手术治疗的胃癌患者125例,采用实时荧光定量反转录聚合酶链反应检测其胃癌组织PD-L1 mRNA相对表达量,采用免疫组化方法检测其胃癌组织CD_(8)^(+)TILs密度。分析胃癌组织PD-L1 mRNA相对表达量、CD_(8)^(+)TILs密度与患者临床病理特征的关系;胃癌组织PD-L1 mRNA相对表达量与CD_(8)^(+)TILs密度的相关性分析采用Pearson相关分析;通过电话随访至术后36个月,以患者死亡或失访为终点事件,绘制Kaplan-Meier曲线以进行生存分析;胃癌患者预后的影响因素分析采用单因素和多因素Cox回归分析。结果所有患者胃癌组织平均PD-L1 mRNA相对表达量为3.1,癌巢内平均CD_(8)^(+)TILs数量为36个,最终归为低水平组73例(PD-L1 mRNA相对表达量<3.1)和高水平组52例(PD-L1 mRNA相对表达量≥3.1),低密度组55例(CD_(8)^(+)TILs数量<36个)和高密度组70例(CD_(8)^(+)TILs≥36个)。低水平组与高水平组患者淋巴结转移、脉管侵犯、腹膜转移情况比较,差异有统计学意义(P<0.05);低密度组与高密度组患者浸润深度、淋巴结转移、脉管侵犯、腹膜转移情况比较,差异有统计学意义(P<0.05)。Pearson相关分析结果显示,胃癌组织PD-L1 mRNA相对表达量与CD_(8)^(+)TILs密度呈负相关(r=-0.412,P<0.001)。中位随访时间为36.0个月,125例患者中预后不良37例,预后良好88例;低水平组与高水平组、低密度组与高密度组患者Kaplan-Meier曲线比较,差异均有统计学意义(P<0.05)。多因素Cox回归分析结果显示,高水平PD-L1 mRNA〔HR=3.021,95%CI(1.632,5.045)〕、低密度CD_(8)^(+)TILs〔HR=2.158,95%CI(1.854,4.632)〕为胃癌患者预后不良的独立危险因素(P<0.05)。结论高水平PD-L1 mRNA、低密度CD_(8)^(+)TILs均是胃癌患者预后不良的独立危险因素,有望成为胃癌患者预后不良的生物标志物。 展开更多

Hepatocellular carcinoma (HCC), chronic hepatitis B (CHB) and chronic hepatitis C (CHC) are characterized by exhaustion of the specific CD8<sup>+</sup> T cell response. This process involves enhancement of negative co-stimulatory molecules, such as programmed cell death protein-1 (PD-1), cytotoxic T-lymphocyte antigen-4 (CTLA-4), 2B4, Tim-3, CD160 and LAG-3, which is linked to intrahepatic overexpression of some of the cognate ligands, such as PD-L1, on antigen presenting cells and thereby favouring a tolerogenic environment. Therapies that disrupt these negative signalling mechanisms represent promising therapeutic tools with the potential to restore reactivity of the specific CD8<sup>+</sup> T cell response. In this review we discuss the impressive in vitro and in vivo results that have been recently achieved in HCC, CHB and CHC by blocking these negative receptors with monoclonal antibodies against these immune checkpoint modulators. The article mainly focuses on the role of CTLA-4 and PD-1 blocking monoclonal antibodies, the first ones to have reached clinical practice. The humanized monoclonal antibodies against CTLA-4 (tremelimumab and ipilimumab) and PD-1 (nivolumab and pembrolizumab) have yielded good results in testing of HCC and chronic viral hepatitis patients. Trelimumab, in particular, has shown a significant increase in the time to progression in HCC, while nivolumab has shown a remarkable effect on hepatitis C viral load reduction. The research on the role of ipilimumab, nivolumab and pembrolizumab on HCC is currently underway. 展开更多

目的 探究染色体结合蛋白8(CBX8)和程序性死亡配体-1(PD-L1)在低龄化宫颈癌患者癌组织中的表达情况,分析其与临床病理特征及预后的关系。方法 获得2016年1月至2018年7月于南京医科大学附属明基医院进行手术治疗的136例≤35岁宫颈癌患者... 目的 探究染色体结合蛋白8(CBX8)和程序性死亡配体-1(PD-L1)在低龄化宫颈癌患者癌组织中的表达情况,分析其与临床病理特征及预后的关系。方法 获得2016年1月至2018年7月于南京医科大学附属明基医院进行手术治疗的136例≤35岁宫颈癌患者(低龄化组)的癌组织及其癌旁正常组织标本,另选同期行手术治疗的52例>35岁患者作为高龄化组。免疫组化分析组织中CBX8、PD-L1的表达情况,分析其与患者的临床病理特征的关系;Kaplan-Meier法分析CBX8、PD-L1表达与患者预后生存的关系;多因素Logistic回归分析影响患者预后的因素,受试者操作特征(ROC)曲线及校准曲线评价模型的预测效能。结果 低龄化组3年生存率(69.12%)明显低于高龄化组(84.62%)(P<0.05)。低龄化患者癌组织的CBX8、PD-L1阳性表达细胞数量明显高于癌旁正常组织(P<0.05);CBX8的表达与肿瘤直径、FIGO分期有关,PD-L1的表达与FIGO分期、肿瘤分化程度、淋巴结转移有关(P<0.05);CBX8低表达患者的3年生存率(76.32%)明显高于高表达患者(60.00%),PD-L1低表达患者3年生存率(78.16%)明显高于高表达患者(53.06%)(P<0.05);宫颈间质浸润深度>4 mm、淋巴结转移、肿瘤宫旁转移、肿瘤脉管浸润、CBX8高表达、PD-L1高表达均是影响患者生存的独立危险因素(P<0.05),模型的区分度较好,准确性较高。结论 CBX8和PD-L1在宫颈癌组织中高表达,与低龄化宫颈癌患者的FIGO分期等临床特征及预后有关。 展开更多

Objective To investigate the expression and regulation of programmed cell death protein 1(PD1),B lymphocyte and T lymphocyte attenuator(BTLA)in peripheral blood of patients with non-small cell lung cancer(NSCLC);to examine the correlation of the mRNA levels between PD and BTLA in NSCLC.Methods Flow cytometry was used to detect the expression of PD1 and BTLA on the surfaces of CD8^+T cells andγδ+T cells in the peripheral blood samples collected from 32 in-patients with stage IV NSCLC and 30 healthy individuals.We compared the expression of PD1 and BTLA on the surfaces ofγδ+T cells in the NSCLC patients with bone metastasis before and after the treatment of zoledronic acid.The correlations of PD1 and BTLA,as well as their ligands were analyzed using Pearson correlation analysis with the cBioPortal data platform.Results The frequency of PD1 on the surfaces of CD8^+T cells was significantly higher than that of theγδT cells in both healthy controls(t=2.324,P=0.024)and NSCLC patients(t=2.498,P=0.015).The frequency of PD1 on CD8^+T cells,rather than onγδ+T cells,was significantly upregulated in advanced NSCLC patients compared with that in healthy controls(t=4.829,P<0.001).The PD1+BTLA+γδT cells of the healthy controls were significantly lower than that of the NSCLC patients(t=2.422,P=0.0185).No differences in percentage of PD1+γδ+and BTLA+γδ+T cells were observed in 7 NSCLC patients with bone metastasis before and after zoledronic acid treatment.PD1 was positively correlated with BTLA in both lung adenocarcinoma(r=0.54;P<0.05)and lung squamous cell carcinoma(r=0.78;P<0.05).Conclusions The upregulation of co-inhibitory molecules occurs on the surfaces of both CD8^+T cells andγδT cells in advanced NSCLC,suggesting that these molecules were involved in regulating the inactivation of CD8^+T cells andγδ+T cells,immune escape and tumor invasion. 展开更多

BACKGROUND Immune checkpoint inhibitors(ICIs)targeting programmed cell death protein 1(PD-1)and T cell immunoglobulin and mucin domain-containing protein 3(TIM-3)are beneficial to the resumption of anti-tumor immunity response and hold extreme potential as efficient therapies for certain malignancies.However,ICIs with a single target exhibit poor overall response rate in hepatocellular carcinoma(HCC)patients due to the complex pathological mechanisms of HCC.AIM To investigate the effects of combined TIM-3 and PD-1 blockade on tumor development in an HCC mouse model,aiming to identify more effective immunotherapies and provide more treatment options for HCC patients.METHODS The levels of PD-1 and TIM-3 on CD4+and CD8+T cells from tumor tissues,ascites,and matched adjacent tissues from HCC patients were determined with flow cytometry.An HCC xenograft mouse model was established and treated with anti-TIM-3 monoclonal antibody(mAb)and/or anti-PD-1 mAb.Tumor growth in each group was measured.Hematoxylin and eosin staining and immunohistochemical staining were used to evaluate T cell infiltration in tumors.The percentage of CD4+and CD8+T cells in tissue samples from mice was tested with flow cytometry.The percentages of PD-1+CD8+,TIM-3+CD8+,and PD-1+TIM-3+CD8+T cells was accessed by flow cytometry.The levels of the cytokines including tumor necrosis factor alpha(TNF-α),interferon-γ(IFN-γ),interleukin(IL)-6,and IL-10 in tumor tissues were gauged with enzyme-linked immunosorbent assay kits.RESULTS We confirmed that PD-1 and TIM-3 expression was substantially upregulated in CD4+and CD8+T cells isolated from tumor tissues and ascites of HCC patients.TIM-3 mAb and PD-1 mAb treatment both reduced tumor volume and weight,while combined blockade had more substantial anti-tumor effects than individual treatment.Then we showed that combined therapy increased T cell infiltration into tumor tissues,and downregulated PD-1 and TIM-3 expression on CD8+T cells in tumor tissues.Moreover,combined treatment facilitated the production of T cell effector cytokines TNF-α and IFN-γ,and reduced the production of immunosuppressive cytokines IL-10 and IL-6 in tumor tissues.Thus,we implicated that combined blockade could ameliorate T cell exhaustion in HCC mouse model.CONCLUSION Combined TIM-3 and PD-1 blockade restrains HCC development by facilitating CD4+ and CD8+T cell-mediated antitumor immune responses. 展开更多

目的通过评估程序性死亡受体配体1(programmed cell death-ligand 1,PD-L1)和分化群8(cluster of differentiation 8,CD8)在肿瘤微环境中的表达情况及其与患者临床病理特征的相关性,明确PD-L1和CD8在判断人表皮生长因子受体2(human epid... 目的通过评估程序性死亡受体配体1(programmed cell death-ligand 1,PD-L1)和分化群8(cluster of differentiation 8,CD8)在肿瘤微环境中的表达情况及其与患者临床病理特征的相关性,明确PD-L1和CD8在判断人表皮生长因子受体2(human epidermal growth factor receptor 2,HER2)阳性乳腺癌患者预后方面的作用。方法选取2016年10月至2018年10月山东省滕州市中心人民医院收治的HER2阳性乳腺癌患者65例,所有患者均为女性。癌组织切片经免疫组织化学方法标记PD-L1和CD8;收集患者的临床病理资料,包括年龄、组织学分级、P53、KI-67、雌激素受体、孕激索受体、肿瘤淋巴结转移分类(tumor node metastasis classification,TNM)分期在内的7项与预后相关的临床病理参数。分别分析PD-L1和CD8的表达是否和上述临床病理参数相关。结果PD-L1阳性表达率为21.5%,PD-L1阳性表达与组织学分级和TNM分期相关,差异有统计学意义(χ^(2)=6.250,P=0.044;χ^(2)=13.730,P=0.001);CD8阳性表达率为21.5%,CD8表达与组织学分级和TNM分期相关,差异有统计学意义(χ^(2)=8.023,P=0.018;χ^(2)=6.117,P=0.047)。结论PD-L1和CD8表达与乳腺癌组织学分级和TNM分期相关,可能是HER2阳性乳腺癌患者重要的预后标志物。 展开更多
