Inhibiting GRK2 is a promising mechanism for the treatment of heart failure (HF). Further development and optimization of inhibitors targeting GRK2 are highly meaningful. Therefore, in order to design GRK2 inhibitors with better performance, the most active molecule was selected as a reference compound from a data set containing 4-pyridylhydrazone derivatives and triazole derivatives, and its scaffold was extracted as the initial scaffold. Then, a powerful optimization-based framework for de novo drug design, guided by binding affinity, was used to generate a virtual molecular library targeting GRK2. The binding affinity of each virtual compound in this dataset was predicted by our developed deep learning model, and the designed potential compound with high binding affinity was selected for molecular docking and molecular dynamics simulation. It was found that the designed potential molecule binds to the ATP site of GRK2, which consists of key amino acids including Arg199, Gly200, Phe202, Val205, Lys220, Met274 and Asp335. The scaffold of the molecule is stabilized mainly by H-bonding and hydrophobic contacts. Concurrently, the reference compound in the dataset was also simulated by docking. It was found that this molecule also binds to the ATP site of GRK2. In addition, its scaffold is stabilized mainly by H-bonding and π-cation stacking interactions with Lys220, as well as hydrophobic contacts. The above results show that the designed potential molecule has similar binding modes to the reference compound, supporting the effectiveness of our framework for activity-focused molecular design. The R package and different modules for binary and multi-class classifiers based on generalized linear models were used to model the data. Predictive power was used to evaluate the performance of the model. Receiver operating characteristic curve analysis over pairs of groups was used to identify the best cutoffs differentiating the different groups.RESULTS: We revealed mathematical models, based on a binary classifier, made up of a unique panel of serum proteins that improved the individual performance of AFP in discriminating HCC patients from patients with chronic liver disease either with or without cirrhosis. We discriminated the HCC group from the cirrhotic liver group using a mathematical model (-11.3 + 7.38 × Prot + 0.00108 × sICAM + 0.2574 × β-catenin + 0.01597 × AFP) with a cutoff of 0.6552, which achieved 98.8% specificity and 89.1% sensitivity. Calculated carbon flux distribution for the interval from 1981 to 1997 confirms the existence of a terrestrial carbon sink in mid-high latitude area of North Hemisphere. Strong interannual variability exists in carbon flux patterns, implying a possible link with ENSO and other natural episodes such as Pinatubo volcano eruption in 1991. Mechanism of this possible link was investigated with statistic method. Correlation analysis indicated that in North Hemisphere, climatic factors such as temperature and precipitation, to some extend, could influence the carbon cycle process of land and ocean, thus cause considerable change in carbon flux distribution. In addition, correlation study also demonstrated the possible important role of Asian terrestrial ecosystems in carbon cycle.展开更多

Mathematical models have been developed to simulate, analyse and understand the dynamics of β-cells, insulin and glucose. In this paper, we consider the effect of genetic predisposition to diabetes on dynamics of β-cells, glucose and insulin. We assume that the β-cell dynamics is governed by the differential equation: . The model indicates different behaviours according to the presence or absence of genetic predisposition. In presence of predisposition (ε = 1), the model shows three equilibrium points: a stable physiological equilibrium point (G = 100, I = 20, β = 600), a stable trivial pathological equilibrium point (G = 600, I = 0, β = 0) and a saddle point (G = 250, I = 9.8, β = 129.36). In absence of predisposition (ε = 0), the model has only two equilibrium points: an unstable pathological equilibrium point (G = 600, I = 0, β = 0) and a stable physiological equilibrium point (G = 82.6, I = 23, β = 900). In order to see how physical activity, obesity and other factors affect insulin sensitivity, simulations are carried out with different values of insulin induced glucose uptake rate (c), β-cell maximum insulin secretory rate (d) and environmental capacity (K).展开更多

The Rock-Eval pyrolysis and vitrinite reflectance were executed. Subsequently, the selected parameters were used for source rock evaluation and 1-D Basin Modelling calibration. The upper part of the formation mainly comprises argillaceous limestone with low content of organic matter (0.64% - 1% TOC). By contrast, the lower part is dominated with shale interval and contains high amounts of TOC values (>4% for 1272 - 1278 m) reveling good to very good quality source rock. Accordingly, good to very good hydrocarbon generation potential is suggested for this formation. Organic matter of the Sargelu Formation contains type II and mixed-type II-III kerogen. The values of Tmax and vitrinite reflectance (Ro%) demonstrate that the formation is thermally mature and in the oil zone. In order to construct a thermal history of the formation and determine the timing of hydrocarbon maturation and generation, the 1-D basin modelling PetroMod 2019.1 was used in this study. Based on the 1-D Basin modelling simulation and its outputs, about 3500 m of overburden have been eroded at the study area. The present-day heat flow was found to be 30 mW/m2. The organic matter of Sargelu Formation entered the early oil zone in 64 Ma and reached the main oil zone ca. 5 Ma. The formation is still in the main oil zone at present-day. In well Atrush-2, the highest rate of oil generation for the Sargelu Formation was in the 8.5 Ma, the onset of oil expulsion from Sargelu Formation was in 50 Ma and the expulsion mass has been reached 0.5 Mtons at present-day.展开更多

The aim of this paper is to design a memoryless non-fragile state feedback control law such that the closed-loop system is asymptotically stable for all admissible parameter uncertainties and controller gain variations. A new linear matrix inequality (LMI) based sufficient condition for the existence of such controllers is established. The calculated ozone concentrations and profile at both the Equator and a 60˚N location were found to follow closely with the measured data. The calculated average ozone concentration was within 1% of the measured average, and the deviation of ozone profiles was within 14%. The monthly evolution of stratospheric ozone concentrations and distribution above the Equator was studied with results discussed in details. The influences of slow air movement in both altitudinal and radial directions on ozone concentrations and profile in the stratosphere were explored and discussed. Parametric studies of the influences of gas diffusivities of ozone DO3 and active atomic oxygen DO on ozone concentrations and distributions were also studied and delineated. Having both influences through physical diffusion and chemical reactions, the diffusivity (and diffusion) of atomic oxygen DO was found to be more sensitive and important than that of ozone DO3 on ozone concentrations and distribution. The 2-D ozone model present in this paper for stratospheric ozone and its layer and depletion is shown to be robust, convenient, efficient, and executable for analyzing the complex ozone phenomena in the stratosphere. .展开更多

The parameter uncertainties are assumed to be norm-bounded. A linear matrix inequality (LMI)-based sufficient condition for the existence of delay-dependent g-suboptimal state feedback robust H∞ controllers which guarantees not only the asymptotic stability of the closed-loop system, but also the H∞ noise attenuation g over all admissible parameter uncertainties is established. At a fixed attack angle, pressure distributions and volume fractions of vapor at different cavitation numbers were simulated, and the results on foil sections agreed well with experimental data. In addition, at the various cavitation numbers, the vapor fractions at different attack angles were also predicted. The vapor region moved towards the front of the airfoil and the length of the cavity grew with increased attack angle. The results show that this method of applying FLUENT to simulate cavitation is reliable.展开更多

To get an insight into CO2 miscible flooding performance in oil reservoirs, a multi-compositional non-isothermal CO2 miscible flooding mathematical model is developed. The convection and diffusion of CO2-hydrocarbon mixtures in multiphase fluids in reservoirs, mass transfer between CO2 and crude, and formation damages caused by asphaltene precipitation are fully considered in the model. The governing equations are discretized in space using the integral finite difference method. The Newton-Raphson iterative technique was used to solve the nonlinear equation systems of mass and energy conservation. A numerical simulator, in which regular grids and irregular grids are optional, was developed for predicting CO2 miscible flooding processes. Two examples of one-dimensional (1D) regular and three-dimensional (3D) rectangle and polygonal grids are designed to demonstrate the functions of the simulator. Experimental data validate the developed simulator by comparison with 1D simulation results. The applications of the simulator indicate that it is feasible for predicting CO2 flooding in oil reservoirs for EOR.展开更多

The use of microscopy methods and the measurement of liver-specific functions optimized the model for best cell performances and also proved the superiority of the 3-D LO2 model when compared with the traditional monolayer model. Toxicity tests were conducted using the newly constructed model, finding that four dental castings coated with silver nanoparticles were toxic to human hepatocytes after cell viability assays. In general, the toxicity of both the castings and the coated silver nanoparticles aggravated as time increased, yet the nanoparticles attenuated the general toxicity by preventing metal ion release, especially at high concentrations.展开更多

文摘Interest in understanding the structural behavior of marine floating photovoltaic(FPV)systems has grown significantly over the last decade.Numerical models are the preferred approach for understanding FPV responses under environmental loads, but they require validation.Several methods are commonly used to validate numerical results, such as comparison with analytical, field data, and experimental data. The use of analytical approaches to validate numerical results can sometimes be inaccurate due to the complexity of the problems;nevertheless, field data is commonly restricted and frequently unavailable for numerical model validation. Thus, physical models play a crucial role in validating numerical results. This study focuses on the two-dimensional(2-D)modeling process and sensors development for an FPV system with taut mooring, aiming to investigate wave-structure interaction while considering hydroelastic effects. The model is developed in accordance with the Froude-Cauchy similitude law and is made from composite materials to capture structural stiffness.Structural motions, specifically heave and pitch, are measured using an Inertial Measurement Unit(IMU),while strain gauges measure structural stress and mooring tension. The sensors provide precise measurements for strain and pitch;however, heave,as a result of time-domain integration from acceleration, requires further validation. The motion responses of the model align with reference results.
文摘A 2-dimensional atmospheric transport model is deployed in a simplified CO 2 inverse study. In addition, correlation study also demonstrated the possible important role of Asian terrestrial ecosystems in carbon cycle.
文摘Worldwide, diabetes is affecting 370 million people, causing nearly five million deaths and absorbing more than 471 billion USD per year. In order to see how physical activity, obesity and other factors affect insulin sensitivity, simulations are carried out with different values of insulin induced glucose uptake rate (c), β-cell maximum insulin secretory rate (d) and environmental capacity (K).
文摘In this paper a new approach for PBL simulation, the non-local closure scheme based on the transient turbulence theory has been used. It was set up as an alternative to local closure schemes which physical concept is reasonable and distinct. A 2-D non-local closure model was developed in order to study the PBL structure and simulatesome interesting atmospheric processes over non-ulliform underlying surface, especially under the convective and unique weather conditions, such as sea-land circulation and the TIBL structure. The modelled results show good agreement with field measurement.
文摘The present study focuses on source rock evaluation of the Sargelu Formation by using core chips of rocks collected from well Atrush-2, Duhok, Kurdistan Region-Iraq. In well Atrush-2, the highest rate of oil generation for the Sargelu Formation was in the 8.5 Ma, the onset of oil expulsion from Sargelu Formation was in 50 Ma and the expulsion mass has been reached 0.5 Mtons at present-day.
文摘A key issue of practical importance in the two-dimensional (2-D) discrete system is stability analysis. Linear state-space models describing 2-D discrete systems have been proposed by several researchers. A popular model, called Forna- sini-Marchesini (FM) second model was proposed by Fornasini and Marchesini in 1978. The aim of this paper is to present a survey of the existing literature on the stability of FM second model.
文摘This paper is concerned with the design problem of non-fragile controller for a class of two-dimensional (2-D) discrete uncertain systems described by the Roesser model. The parametric uncertainties are assumed to be norm-bounded. The aim of this paper is to design a memoryless non-fragile state feedback control law such that the closed-loop system is asymptotically stable for all admissible parameter uncertainties and controller gain variations. A new linear matrix inequality (LMI) based sufficient condition for the existence of such controllers is established. Finally, a numerical example is provided to illustrate the applicability of the proposed method.
文摘An engineering system approach of 2-D cylindrical model of transient mass balance calculations of ozone and other concerned chemicals along with fourteen photolysis, ozone-generating and ozone-depleting chemical reaction equations was developed, validated, and used for studying the ozone concentrations, distribution and peak of the layer, ozone depletion and total ozone abundance in the stratosphere. The 2-D ozone model present in this paper for stratospheric ozone and its layer and depletion is shown to be robust, convenient, efficient, and executable for analyzing the complex ozone phenomena in the stratosphere. .
文摘This paper considers the problem of delay-dependent robust optimal H∞ control for a class of uncertain two-dimensional (2-D) discrete state delay systems described by the general model (GM). Furthermore, a convex optimization problem is formulated to design a delay-dependent state feedback robust optimal H∞ controller which minimizes the H∞ noise attenuation g of the closed-loop system. Finally, an illustrative example is provided to demonstrate the effectiveness of the proposed method.
文摘This paper investigates the problem of robust optimal H∞ control for uncertain two-dimensional (2-D) discrete state-delayed systems described by the general model (GM) with norm-bounded uncertainties. A sufficient condition for the existence of g-suboptimal robust H∞ state feedback controllers is established, based on linear matrix inequality (LMI) approach. Moreover, a convex optimization problem is developed to design a robust optimal state feedback controller which minimizes the H∞ noise attenuation level of the resulting closed-loop system. Finally, two illustrative examples are given to demonstrate the effectiveness of the proposed method.
文摘In order to predict the effects of cavitation on a hydrofoil, the state equations of the cavitation model were combined with a linear viscous turbulent method for mixed fluids in the computational fluid dynamics (CFD) software FLUENT to simulate steady cavitating flow. The results show that this method of applying FLUENT to simulate cavitation is reliable.

文摘The injection of fuel-generated CO2 into oil reservoirs will lead to two benefits in both enhanced oil recovery (EOR) and the reduction in atmospheric emission of CO2. The applications of the simulator indicate that it is feasible for predicting CO2 flooding in oil reservoirs for EOR.

文摘This paper is concerned with the problem of stabilization of the Roesser type discrete-time nonlinear 2-D system that plays an important role in many practical applications. First, a discrete-time 2-D T-S fuzzy model is proposed to represent the underlying nonlinear 2-D system. Second, new quadratic stabilization conditions are proposed by applying relaxed quadratic stabilization technique for 2-D case. Third, for sake of further reducing conservatism, new non-quadratic stabilization conditions are also proposed by applying a new parameter-dependent Lyapunov function, matrix transformation technique, and relaxed technique for the underlying discrete-time 2-D T-S fuzzy system. Finally, a numerical example is provided to illustrate the effectiveness of the proposed results.

文摘To address the controversial issue of the toxicity of dental alloys and silver nanoparticles in medical ap- plications, an in vivo-like LO2 3-D model was constructed within polyvinylidene fluoride hollow fiber materials to mimic the microenvironment of liver tissue. In general, the toxicity of both the castings and the coated silver nanoparticles aggravated as time increased, yet the nanoparticles attenuated the general toxicity by preventing metal ion release, especially at high concentrations.
