In order to elucidate the mechanism of the seed germination in Cistanche deserticola,we investigated the effects of stratification treatment of high/low temperature on seed germination of C.deserticola using the tissu...In order to elucidate the mechanism of the seed germination in Cistanche deserticola,we investigated the effects of stratification treatment of high/low temperature on seed germination of C.deserticola using the tissue culture technique.The effects of high/low temperature stratification and plant hormones on seed germination rate of C.deserticola were investigated using the orthogonal experiment design.Results showed that the seed germination rate was 6.7% at 50 ℃ for 1 h and 1 week at(4 ℃) twice repeated stratification treatment for the seeds,and the germination took 60 d.To the contrast,the seed germination rate was 17.8% at 50 ℃ for 1 h and 1 week at 4 ℃ twice repeated stratification treatment combined with 1.4 μmol·L-16-BA,(2.9 μmol·L)-1 GA3 and 5.8 μmol·L-1 IBA treatment,and the germination needed only 30 d.GA3 was a key plant hormone to the seed germination to promote the germination rate at 50 ℃ and 1 w at 4 ℃.Two repeated stratification treatments and exogenous plant hormone were the essential conditions for the seed germination after being excised from host.展开更多
