Chromatographic fingerprinting has been perceived as an essential tool for assessing quality and chemical equivalence of traditional Chinese medicine.However,this pattern-oriented approach still has some weak points in terms of chemical coverage and robustness.In this work,we proposed a multiple reaction monitoring(MRM)-based fingerprinting method in which approximately 100 constituents were simultaneously detected for quality assessment.The derivative MRM approach was employed to rapidly design MRM transitions independent of chemical standards,based on which the large-scale fingerprinting method was efficiently established.This approach was exemplified on QiShenYiQi Pill(QSYQ),a traditional Chinese medicine-derived drug product,and its robustness was systematically evaluated by four indices:clustering analysis by principal component analysis,similarity analysis by the congruence coefficient,the number of separated peaks,and the peak area proportion of separated peaks.Compared with conventional ultraviolet-based fingerprints,the MRM fingerprints provided not only better discriminatory capacity for the tested normal/abnormal QSYQ samples,but also higher robustness under different chromatographic conditions(i.e.,flow rate,apparent pH,column temperature,and column).The result also showed for such large-scale fingerprints including a large number of peaks,the angle cosine measure after min-max normalization was more suitable for setting a decision criterion than the unnormalized algorithm.This proof-of-concept application gives evidence that combining MRM technique with proper similarity analysis metrices can provide a highly sensitive,robust and comprehensive analytical approach for quality assessment of traditional Chinese medicine. 展开更多

Salvia miltiorrhiza and S.grandifolia are rich in diterpenoids and have therapeutic effects on cardiovascular diseases.In this study,the spatial distribution of diterpenoids in both species was analyzed by a combination of metabolomics and mass spectrometry imaging techniques.The results indicated that diterpenoids in S.miltiorrhiza were mainly abietane-type norditerpenoid quinones with a furan or dihydrofuran D-ring and were mainly distributed in the periderm of the roots,e.g.cryptotanshinone and tanshinone IIA.The compounds in S.grandifolia were mainly phenolic abietane-type tricyclic diterpenoids with six-or seven-membered C-rings,and were widely distributed in the periderm,phloem,and xylem of the roots,e.g.11-hydroxy-sugiol,11,20-dihydroxy-sugiol,and 11,20-dihydroxy-ferruginol.In addition,the leaves of S.grandifolia were rich in tanshinone biosynthesis precursors,such as 11-hydroxy-sugiol,while those of S.miltiorrhiza were rich in phenolic acids.Genes in the upstream pathway of tanshinone biosynthesis were highly expressed in the root of S.grandifolia,and genes in the downstream pathway were highly expressed in the root of S.miltiorrhiza.Here,we describe the specific tissue distributions and mechanisms of diterpenoids in two Salvia species,which will facilitate further investigations of the biosynthesis of diterpenoids in plant synthetic biology. 展开更多

Fe-rich intermetallic phases in recycled Al alloys often exhibit complex and 3D convoluted structures and morphologies.They are the common detrimental intermetallic phases to the mechanical properties of recycled Al alloys.In this study,we used synchrotron X-ray tomography to study the true 3D morphologies of the Ferich phases,Al_(2)Cu phases and casting defects in an ascast Al-5Cu-1.5Fe-1Si alloy.Machine learning-based image processing approach was used to recognize and segment the diff erent phases in the 3D tomography image stacks.In the studied condition,theβ-Al_(9)Fe_(2)Si_(2)andω-Al_(7)Cu_(2)Fe are found to be the main Fe-rich intermetallic phases.Theβ-Al_(9)Fe_(2)Si_(2)phases exhibit a spatially connected 3D network structure and morphology which in turn control the 3D spatial distribution of the Al_(2)Cu phases and the shrinkage cavities.The Al_(3)Fe phases formed at the early stage of solidification aff ect to a large extent the structure and morphology of the subsequently formed Fe-rich intermetallic phases.The machine learning method has been demonstrated as a powerful tool for processing big datasets in multidimensional imaging-based materials characterization work. 展开更多

Gastric ulcer is a widespread disease caused by various etiologies.Dendrobium offi cinale flowers exert several health benefits owing to their rich flavonoid content.In this study,protective effects and possible action mechanisms of D.offi cinale flowers’flavonoid enriched extract(DOFF)were assessed against gastric ulcer.The result of sodium nitrite-aluminum nitrate colorimetry showed that 52.34%of the total extractive was flavonoid,and ultra-high performance liquid chromatography time of flight mass spectrometer(UPLC-QTOF/MS)revealed the presence of 28 components in DOFF of which 14 belonged to flavonoids.In addition,in vivo assay revealed DOFF potential in reducing the formation of ethanol-induced gastric mucosal lesions,with drop-in ulcer index from 64.33±8.76 to 32.00±4.47.Similar results were revealed in human gastric mucosal epithelia cells,with cells viability to increase from 27.2%to 61.6%post DOFF administration.To analyzed the protect effect of DOFF,we used Western blotting and immunofluorometric assay to revealed the expression levels of key proteins in cell pathways.The results showed that DOFF(320μg/mL)could increase the level of oxidation marker protein(HO-1),apoptosis regulatory protein(Bcl-2)and autophagy marker(LC3β)by 50.84%,43.85%,and 59.21%compared with ethanol-treated group respectively.Further analyzed of the mitochondrial activity and apoptosis pathway,we found that DOFF appeared to mitigate against ethanolinduced gastric mucosal injury via AMPK/mTOR/ULK1 and PI3K/AKT autophagy signaling pathways. 展开更多

Nitrogen doping is a promising method for the preparation of functional carbon materials.In this study,a nitrogen-doped porous coral biochar was prepared by using bamboo as raw material,urea as nitrogen source,and KHCO3 as green activator through in-situ pyrolysis.The structure of the obtained biochar was characterized by various techniques including nitrogen adsorption and desorption,Raman spectroscopy,X-ray photoelectron spectrometer,and etc.The adsorption properties of nitrogen-doped biochar were evaluated with phenol and methylene blue probes.The results showed that the nitrogen source ratio had a significant effect on the evolution of pore structure of biochar.Low urea addition ratio was beneficial to the development of pore structures.The optimum specific surface area of nitrogen-doped biochar could be up to 1693 m^2·g^-1.Nitrogen doping can effectively improve the adsorption capacity of biochar to phenol and methylene blue.Biochar prepared at 973.15 K with low urea addition ratio exhibited the highest adsorption capacity for phenol and methylene blue,and the equilibrium adsorption capacity was 169.0 mg·g^-1 and 499.3 mg·g^-1,respectively.By comparing the adsorption capacity of various adsorbents in related fields,it is proved that the nitrogen-doped biochar prepared in this study has a good adsorption effect. 展开更多

Word segmentation is an integral step in many knowledge discovery applications. However, existing word segmentation methods have problems when applying to Chinese judicial documents:(1) existing methods rely on large-scale labeled data which is typically unavailable in judicial documents, and (2) judicial document has its own language features and writing formats. In this paper, a word segmentation method is proposed for Chinese judicial documents. The proposed method consists of two steps:(1) automatically generating some labeled data as legal dictionaries, and (2) applying a hybrid multilayer neural networks to do word segmentation incorporating legal dictionaries. Experiments are conducted on a dataset of Chinese judicial documents showing that the proposed model can achieve better results than the existing methods. 展开更多

Objective: Ferroptosis is a novel cell death process which displays a promising role in cancer treatment.However, clinically available drugs targeting ferroptosis are rarely used, and yet there are no studies reporting on inducing ferroptosis via Chinese herbal extracts. Here we explored the tumor inhibition effects of Ganoderma lucidum(G. lucidum) on oral squamous cell carcinoma(OSCC). Specifically, we aimed to clarify the biological mechanism of components in the dietary, aqueous-soluble sporoderm-removed G. lucidum spore powder(A-GSP).Methods: Preliminary transcriptome analysis revealed the significant enrichment of the ferroptosis pathway.Cellular Fe2+, glutathione(GSH), malondialdehyde(MDA), reactive oxygen species(ROS) and lipid peroxide levels were measured to identify ferroptosis occurrence. Western blotting was used to measure ferroptosis-related proteins. Changes in mitochondria morphology and function were observed with transmission electron microscopy(TEM) and ATP detection assays. Ferroptosis inhibitor ferrostatin-1 was then used to verify the anti-tumor effects of A-GSP. Finally, nude mice xenograft models of oral cancer confirmed that A-GSP inhibited tumor growth.Results: A-GSP promoted ferroptosis in oral cancer cells by inducing Fe2+influx, GSH depletion, as well as lipid peroxide and ROS accumulation. Ferroptosis-related proteins exhibited corresponding changes, particularly Acylco A synthetase long chain family member 4(ACSL4) increase and glutathione peroxidase 4(GPX4) decrease. A-GSP considerably lowered mitochondrial volume and ridge number, while significantly decreasing ATP production. Ferrostatin-1 reversed all of these A-GSP-induced changes. In vivo, A-GSP exerted a ferroptosismediated tumor-suppressing effect without observable adverse reactions.Conclusions: Our findings demonstrate the therapeutic potential of A-GSP for treating patients with OSCC by targeting ferroptosis. 展开更多

Diabetic wounds has a gradually increasing incidence and morbidity.Excessive inflammation due to immune imbalance leads to delayed wound healing.Here,we reveal the interconnection between activation of the NLRP3 inflammatory pathway in endotheliocyte and polarization of macrophages via the cGAS-STING pathway in the oxidative microenvironment.To enhance the immune-regulation based on repairing mitochondrial oxidative damage,a zeolitic imidazolate framework-8 coated with cerium dioxide that carries Rhoassociated protein kinase inhibition Y-27632(CeO_(2)-Y@ZIF-8)is developed.It is encapsulated in a photocross-linkable hydrogel(GelMA)with cationic quaternary ammonium salt groups modified to endow the antibacterial properties(CeO_(2)-Y@ZIF-8@Gel).CeO_(2)with superoxide dismutase and catalase activities can remove excess reactive oxygen species to limit mitochondrial damage and Y-27632 can repair damaged mitochondrial DNA,thus improving the proliferation of endotheliocyte.After endotheliocyte uptakes CeO_(2)-Y@ZIF-8 NPs to degrade peroxides into water and oxygen in cells and mitochondria,NLRP3 inflammatory pathway is inhibited and the leakage of oxidatively damaged mitochondrial DNA(Ox-mtDNA,a damage-associated molecular pattern)through mPTP decreases.Futhermore,as the cGAS-STING pathway activated by Ox-mtDNA down-regulated,the M_(2)phenotype polarization and anti-inflammatory factors increase.Collectively,CeO_(2)-Y@ZIF-8@Gel,through remodulating the crosstalk between macrophage reprogramming and angiogenesis to alleviate inflammation in the microenvironment and accelerates wound healing. 展开更多

The penetration of multi-carrier energy systems in distribution system gains more and more concerns.In this paper,a bi-level transactive energy trading framework is proposed to improve the energy scheduling and operation efficiency for multi-carrier energy systems which are modeled as energy hubs(EHs).In the upper level,each EH in the distribution system not only makes energy scheduling decisions considering supplies and demands of local energy,but also trades energy with each other to further maximize their social welfare.The associated trading payment among EHs is made in a fair manner by applying Nash bargaining theory.We solve the bargaining problem by decomposing it into two subproblems:operation cost minimization problem and payment bargaining problem.Then,based on the trading decision,the nodal equivalent loads of EHs are sent to the distribution system operator(DSO)without publishing trading details.By applying the second-order cone programming(SOCP),DSO reconfigures the network to reduce the transmission loss of the system in the lower level.The network reconfiguration and the trading behavior of EHs interact and iterate until the convergence.Numerical studies on modified IEEE 33-bus distribution system demonstrate the effectiveness of the proposed framework. 展开更多
