Objective:Our previous studies have indicated potentially higher proliferative activity of tumor cells in Chinese patients with mantle-cell lymphoma(MCL)than those in Western.Given the success and tolerability of R-DA...Objective:Our previous studies have indicated potentially higher proliferative activity of tumor cells in Chinese patients with mantle-cell lymphoma(MCL)than those in Western.Given the success and tolerability of R-DA-EDOCH immunochemotherapy in treating aggressive B-cell lymphomas,we designed a prospective,phase 3 trial to explore the efficacy and safety of alternating R-DA-EDOCH/R-DHAP induction therapy for young patients with newly diagnosed MCL.The primary endpoint was the complete remission rate(CRR)at the end of induction(EOI).Methods:A total of 55 patients were enrolled.The CRR at the EOI was 89.1%[95%confidence interval(CI)78%±96%],and the overall response rate was 98.1%(95%CI 90%±100%).Most patients with bone marrow involvement quickly attained minimal residual disease(MRD)negative status,with a 95.7%rate at the EOI.Results:The 3-year progression-free survival(PFS)and overall survival rates were 66.3%and 83.2%,respectively.No patients discontinued treatment because of adverse events.Univariate analysis identified pathologic morphology and TP53 mutations as risk factors for PFS.However,high tumor proliferative activity and certain cytogenetic abnormalities showed no significant adverse prognostic significance.Conclusions:Intensive therapy based on a high cytarabine dose and continuously administered EDOCH achieved a high MRDnegative rate and provides an optional induction choice for young patients with MCL with high-risk factors.展开更多

Large granular lymphocytic leukemia(LGLL)is characterized by the clonal proliferation of cytotoxic T lymphocytes or NK cells.Standard first-line immunosuppressive treatments have limitations,achieving complete remissi...Large granular lymphocytic leukemia(LGLL)is characterized by the clonal proliferation of cytotoxic T lymphocytes or NK cells.Standard first-line immunosuppressive treatments have limitations,achieving complete remission(CR)rates of up to 50%.Immune system dysregulation is implicated in LGLL.Promising results for thalidomide,an immunomodulatory drug,combined with prednisone and methotrexate(TPM),were observed in our pilot study.This multicenter study evaluated the effcacy and safety of a thalidomide,prednisone,and methotrexate(TPM)regimen in 52 symptomatic,methotrexate-and thalidomide-naive LGLL patients from June 2020 to August 2022.Thalidomide(100 mg daily for up to 24 months),prednisone(0.5-1.0 mg/kg every other day,tapered after 3 months),and methotrexate(10 mg/m^(2) weekly for up to 12 months)were administered.The primary objective was to determine the CR rate.The median follow-up duration was 29.0 months(range:4.0-42.0).Forty-seven patients(90.4%)achieved hematological and symptomatic responses.Thirty-nine patients(75.0%)achieved CR.The median time to response was 3.0 months(range:3.0-9.0).The median progression-free survival was 40.0 months(95%confidence interval(Cl):38.0-42.0),and the median duration of response was 39.0 months(95%Cl:36.1-41.9).The most common adverse event was peripheral neuropathy(24.1%),most of which(84.6%)were grades 1-2.Four patients experienced grade≥3 adverse events.In conclusion,the TPM regimen was an effective and safe treatment for symptomatic LGLL patients,with a particularly high CR rate.This trial was registered at www.clinicaltrials.gov(#NCT04453345).展开更多
Real-time thermal sensing through flexible temperature sensors in extreme environments is critically essential for precisely monitoring chemical reactions,propellant combustions,and metallurgy processes.However,despit...Real-time thermal sensing through flexible temperature sensors in extreme environments is critically essential for precisely monitoring chemical reactions,propellant combustions,and metallurgy processes.However,despite their low response speed,most existing thermal sensors and related sensing materials will degrade or even lose their sensing performances at either high or low temperatures.Achieving a microsecond response time over an ultrawide temperature range remains challenging.Here,we design a flexible temperature sensor that employs ultrathin and consecutive Mo_(1−x)W_(x)S_(2) alloy films constructed via inkjet printing and a thermal annealing strategy.The sensing elements exhibit a broad work range(20 to 823 K on polyimide and 1,073 K on flexible mica)and a record-low response time(about 30μs).These properties enable the sensors to detect instantaneous temperature variations induced by contact with liquid nitrogen,water droplets,and flames.Furthermore,a thermal sensing array offers the spatial mapping of arbitrary shapes,heat conduction,and cold traces even under bending deformation.This approach paves the way for designing unique sensitive materials and flexible sensors for transient sensing under harsh conditions.展开更多

Herein, two dual-emissive aggregation-induced emission luminogens(AIEgens)with donor-π-acceptor structures were designed and synthesized, namely QT-1 and QF-2. Both two AIEgens exhibited excitation wavelength-dependence defying the Kasha's rule, and could stain lipid droplets(LDs) and mitochondria in blue and red fluorescence, respectively. Moreover, thanks to the near-infrared emission and abundant reactive oxygen species(ROS) generation efficiency of QT-1, it was chosen as a photodynamic therapy agent to selectively kill cancer cells from normal cells. Upon light irradiation, an obvious decrease of mitochondrial membrane potential(MMP) and serious change of mitochondrial shape in cells were observed, which corresponded to the efficient inhibition of tumor growth in vivo. Rituximab maintenance(RM)prolongs the progression-free survival(PFS)of responding patients with follicular lymphoma(FL),but the maintenance efficacy in different Follicular Lymphoma International Prognostic Index(FLIP...Rituximab maintenance(RM)prolongs the progression-free survival(PFS)of responding patients with follicular lymphoma(FL),but the maintenance efficacy in different Follicular Lymphoma International Prognostic Index(FLIPI)risk group is still confusing.We performed a retrospective analysis of the effect of RM treatments in patients with FL responding to induction therapy based on their FLIPI risk assessment carried out prior to treatment.We identified 93 patients between 2013 and 2019 who received RM every 3 months for≥4 doses(RM group),and 60 patients who did not accept RM or received rituximab less than 4 doses(control group).After a median follow-up of 39 months,neither median overall survival(OS)nor PFS was reached for the entire population.The PFS was significantly prolonged in the RM group compared to the control group(median PFS NA vs 83.1 months,P=.00027).When the population was divided into the 3 FLIPI risk groups,the PFS differed significantly(4-year PFS rates,97.5%vs 88.8%vs 72.3%,P=.01)according to group.There was no significant difference in PFS for FLIPI low-risk patients with RM compared to the control group(4-year PFS rates,100%vs 93.8%,P=.23).However,the PFS of the RM group was significantly prolonged for FLIPI intermediate-risk(4-year PFS rates,100%vs 70.3%,P=.00077)and high-risk patients(4-year PFS rates,86.7%vs 57.1%,P=.023).These data suggest that standard RM significantly prolongs the PFS of patients assigned to intermediate-and high-risk FLIPI groups but not to low-risk FLIPI group,and pending larger-scale studies to validate.展开更多
In order to solve the problems of low sensitivity and complex sample pretreatment in traditional detection,a method was proposed in this study to detect daminozide using terahertz combined with a metamaterial sensor,w...In order to solve the problems of low sensitivity and complex sample pretreatment in traditional detection,a method was proposed in this study to detect daminozide using terahertz combined with a metamaterial sensor,which enables real-time and label-free molecular detection with high sensitivity.The correlation between the transmission frequency shift and absorbance of daminozide solution at different concentrations was analyzed.The simulation and experimental results showed that this metamaterial sensor could achieve highly sensitive sensing of daminozide solutions at 0.05 mg/L.The maximum quality factor(Q_(max))of the two peaks could reach 5.78 and 13.05,and the maximum figure of merit(FOM)of the two peaks can reach 0.82 and 1.72.The maximum sensitivity of two resonance peaks reached 38.148 GHz/(mg·L)and 133.516 GHz/(mg·L)when the concentration of daminozide in the solution was 2000 mg/L.There was an obvious positive correlation between the transmission and transmission,frequency shift and frequency shift,and absorbance and absorbance of the resonance peak of daminozide solutions.Therefore,this platform not only opens up new possibilities for the microanalysis of the chemical composition of substances in solutions but also provides a valuable reference for the design of other metamaterial-based sensors in the field of food safety.展开更多
The external pressure is one of the essential parameters for regulating the structure and energy con-version properties of antiferroelectric AgNbO_(3).For pure AgNbO_(3),however,there has been still a blank of its rea...The external pressure is one of the essential parameters for regulating the structure and energy con-version properties of antiferroelectric AgNbO_(3).For pure AgNbO_(3),however,there has been still a blank of its real lattice structure under the stress field.Here,high-pressure lattice structures and phase transitions of AgNbO_(3)have been explored by spectroscopic experiments and theoretical models.A successive phase transition process from Pbcm to C222_(1) to P2_(1) has been observed at the pressure range of 0-30 GPa,associated with displacive-type characterized by soft-mode kinetics.Note that the paraelectric phase cannot be achieved under high-pressure at room temperature.Significantly,the competition of long-range Coulomb force,short-range interatomic interaction,and covalent interaction in AgNbO_(3)lattice were demonstrated under the stress field.The present work can provide fundamental guidelines to reveal the high-pressure phase transitions of AgNbO_(3),which will open up possibilities for the designing device with functional properties at extremes.展开更多
Waldenstrom macroglobulinemia(WM)is a type of incurable,indolent B-cell lymphoma that is prone to relapse.Over time,treatment strategies have progressed from cytotoxic drugs to rituximab(R)-or bortezomib(V)-based regi...Waldenstrom macroglobulinemia(WM)is a type of incurable,indolent B-cell lymphoma that is prone to relapse.Over time,treatment strategies have progressed from cytotoxic drugs to rituximab(R)-or bortezomib(V)-based regimens,and have now entered into an era of Bruton tyrosine kinase inhibitor(BTKi)-based regimens.However,the optimal treatment for the relapsed patients is still unclear.Herein,we analyzed the outcomes of the first-and second-line therapies in 377 patients with WM to illustrate the optimal choices for second-line therapy.After a median follow-up of 45.4 months,89 patients received second-line therapy,and 53 patients were evaluated for response.The major response rates(MRR)of first-and second-line treatment were 65.1%and 67.9%(P=0.678).The median progression-free survival(PFS)for the second-line therapy(PFS2)was shorter than that for the first-line therapy(PFS1)(56.3 vs 40.7 months,P=0.03).However,PFS2 in targeted drugs group(R-/V-/BTKi-based regimens)was comparable to PFS1(60.7 months vs 44.7 months,respectively,P=0.21).Regarding second-line therapy,patients who underwent sequential treatment escalation—such as transitioning from cytotoxic drugs to R-/V-/BTKi-based regimens or from R-/V-based to BTKi-based regimens(escalation group)—had higher MRR(80.6%vs 47.1%,respectively,P=0.023)and longer PFS2(50.4 vs 23.5 months,respectively,P<0.001)compared to the non-escalation group.Patients in the escalation group also had longer post-relapse overall survival compared with the non-escalation group(median,not reached vs 50.7 months,respectively,P=0.039).Our findings indicate that sequential treatment escalation may improve the survival of patients with WM.展开更多

文摘In order to solve the problems of low sensitivity and complex sample pretreatment in traditional detection,a method was proposed in this study to detect daminozide using terahertz combined with a metamaterial sensor,which enables real-time and label-free molecular detection with high sensitivity.The correlation between the transmission frequency shift and absorbance of daminozide solution at different concentrations was analyzed.The simulation and experimental results showed that this metamaterial sensor could achieve highly sensitive sensing of daminozide solutions at 0.05 mg/L.The maximum quality factor(Q_(max))of the two peaks could reach 5.78 and 13.05,and the maximum figure of merit(FOM)of the two peaks can reach 0.82 and 1.72.The maximum sensitivity of two resonance peaks reached 38.148 GHz/(mg·L)and 133.516 GHz/(mg·L)when the concentration of daminozide in the solution was 2000 mg/L.There was an obvious positive correlation between the transmission and transmission,frequency shift and frequency shift,and absorbance and absorbance of the resonance peak of daminozide solutions.Therefore,this platform not only opens up new possibilities for the microanalysis of the chemical composition of substances in solutions but also provides a valuable reference for the design of other metamaterial-based sensors in the field of food safety.

文摘The external pressure is one of the essential parameters for regulating the structure and energy con-version properties of antiferroelectric AgNbO_(3).For pure AgNbO_(3),however,there has been still a blank of its real lattice structure under the stress field.Here,high-pressure lattice structures and phase transitions of AgNbO_(3)have been explored by spectroscopic experiments and theoretical models.A successive phase transition process from Pbcm to C222_(1) to P2_(1) has been observed at the pressure range of 0-30 GPa,associated with displacive-type characterized by soft-mode kinetics.Note that the paraelectric phase cannot be achieved under high-pressure at room temperature.Significantly,the competition of long-range Coulomb force,short-range interatomic interaction,and covalent interaction in AgNbO_(3)lattice were demonstrated under the stress field.The present work can provide fundamental guidelines to reveal the high-pressure phase transitions of AgNbO_(3),which will open up possibilities for the designing device with functional properties at extremes.

文摘Waldenstrom macroglobulinemia(WM)is a type of incurable,indolent B-cell lymphoma that is prone to relapse.Over time,treatment strategies have progressed from cytotoxic drugs to rituximab(R)-or bortezomib(V)-based regimens,and have now entered into an era of Bruton tyrosine kinase inhibitor(BTKi)-based regimens.However,the optimal treatment for the relapsed patients is still unclear.Herein,we analyzed the outcomes of the first-and second-line therapies in 377 patients with WM to illustrate the optimal choices for second-line therapy.After a median follow-up of 45.4 months,89 patients received second-line therapy,and 53 patients were evaluated for response.The major response rates(MRR)of first-and second-line treatment were 65.1%and 67.9%(P=0.678).The median progression-free survival(PFS)for the second-line therapy(PFS2)was shorter than that for the first-line therapy(PFS1)(56.3 vs 40.7 months,P=0.03).However,PFS2 in targeted drugs group(R-/V-/BTKi-based regimens)was comparable to PFS1(60.7 months vs 44.7 months,respectively,P=0.21).Regarding second-line therapy,patients who underwent sequential treatment escalation—such as transitioning from cytotoxic drugs to R-/V-/BTKi-based regimens or from R-/V-based to BTKi-based regimens(escalation group)—had higher MRR(80.6%vs 47.1%,respectively,P=0.023)and longer PFS2(50.4 vs 23.5 months,respectively,P<0.001)compared to the non-escalation group.Patients in the escalation group also had longer post-relapse overall survival compared with the non-escalation group(median,not reached vs 50.7 months,respectively,P=0.039).Our findings indicate that sequential treatment escalation may improve the survival of patients with WM.
