Nitrogen(N)is crucial for maize(Zea mays L.)growth,development and yield.Dysfunction of the maize NIN-like protein 5(ZmNLP5)reduces N assimilation,but the precise mechanism by which ZmNLP5 modulates N metabolism and its contribution to agricultural applications are not well understood.In this study,our transcriptomic profiling and chromatin immunoprecipitation followed by sequencing(ChIP-seq)analyses reveal 581 ZmNLP5-modulated target genes.We confirmed that ZmNLP5 physically interacted and transactivated contributors to N assimilation,including ZmNRT1.1D,ZmNIR1.1,ZmNR2.1,ZmGS4,ZmAS1,and ZmLBD6.Overexpression of ZmNLP5 upregulated N metabolism genes,and enhanced the enzymatic activities of nitrate reductase and glutamine synthase.Furthermore,overexpressing ZmNLP5 increased grain yield under both normal-N and low-N conditions.Introgression of the overexpressed ZmNLP5 allele into Zhengdan 958 resulted in comparable yield increases in field trials.Our study unveiled that ZmNLP5 is a potential genetic target for increasing nitrogen use efficiency and grain yield in maize. 展开更多

Objective To investigate the association between long-term glycemic control and cerebral infarction risk in patients with diabetes through a large-scale cohort study.Methods This prospective,community-based cohort study included 12,054 patients with diabetes.From 2006 to 2012,38,272 fasting blood glucose(FBG)measurements were obtained from these participants.FBG trajectory patterns were generated using latent mixture modelling.Cox proportional hazards models were applied to assess the subsequent risk of cerebral infarction associated with different FBG trajectory patterns.Results At baseline,the mean age of the participants was 55.2 years.Four distinct FBG trajectories were identified based on FBG concentrations and their changes over the 6-year follow-up period.After a median follow-up of 6.9 years,786 cerebral infarction events were recorded.Different trajectory patterns were associated with significantly varied outcome risks(Log-Rank P<0.001).Compared with the low-stability group,Hazard Ratio(HR)adjusted for potential confounders were 1.37 for the moderate-increasing group,1.23 for the elevated-decreasing group,and 2.08 for the elevated-stable group.Conclusion Sustained high FBG levels were found to play a critical role in the development of ischemic stroke among patients with diabetes.Controlling FBG levels may reduce the risk of cerebral infarction. 展开更多

Objective The aim of this study was to investigate the prospective association between physical activity(PA),independently or in conjunction with other contributing factors,and osteoporosis(OP)outcomes.Methods The Physical Activity in Osteoporosis Outcomes(PAOPO)study was a community-based cohort investigation.A structured questionnaire was used to gather the participants’sociodemographic characteristics.Bone mineral density(BMD)measurements were performed to assess OP outcomes,and the relationship between BMD and OP was evaluated within this cohort.Results From 2013 to 2014,8,471 participants aged 18 years and older were recruited from Tangshan,China’s Jidong community.Based on their PA level,participants were categorized as inactive,moderately active,or very active.Men showed higher physical exercise levels than women across the activity groups.BMD was significantly higher in the very active group than in the moderately active and inactive groups.Individuals aged>50 years are at a higher risk of developing OP and osteopenia.Conclusion The PAOPO study offers promising insights into the relationship between PA and OP outcomes,encouraging the implementation of PA in preventing and managing OP. 展开更多
