Objective:To evaluate the influence of extraction solvents on antioxidant and antimicrobial activities of the pulp and seed of Anisophyllea laurina R.Br.ex Sabine fruits.Methods:The antibacterial activities of pulp and seed extracts were tested by using disk diffusion method against eight bacterial strains and three fungal strains.Total phenolic,flavonoid,monomeric anthocyanin and tannin contents,and antioxidant activities were determined by spectrometric methods.Results:The antioxidant analysis of pulp extract revealed the strong radical scavenging capacity and total phenolic content(4 329.66 mg of gallic acid/100 g),while seed extract showed the high antioxidant activity and total tannin content(5 326.78 mg catechin equivalent/100 g).Antibacterial and antifungal activities of methanol and ethanol extracts exhibited potent growth inhibitory activity against Aeromonas hydrophila,Bacillus subtilis,Escherichia coli O157:H7,Pseudomonas aeruginosa,Salmonella Typhimurium and Staphylococcus aureus ATCC 6538 with minimum inhibitory concentration values ranged from 125 to 250 μg/mL.However,seed extract had the strongest potential activity against Aspergillus niger and Candida albicans with minimum inhibitory concentration value of 500 μg/mL compared to pulp extract.Conclusions:Our results therefore demonstrated that ethanol and methanol extractions were more efficient in extracting antioxidants and bioactive compound in pulp and seed.These results support that these plant extracts can be used for the treatment of bacterial infections. 展开更多

In the present study,the antioxidant and anti-human liver cancer(HepG2)cells effects of bioactive peptides from cowhide collagen(BPCC)were evaluated.BPCC exhibited significant scavenging effect on l,1-diphenyl-2-picrylhydrazyl(DPPH)radicals((60.09±3.51)%),2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS)radicals((77.40±3.10)%)and hydroxyl radicals((56.00±2.30)%)as well as strong reducing power(0.320士0.025).Meanwhile,BPCC effectively protected biomacromolecules including proteins,lipids and DNA from oxidative damage induced by Cu^(2+)H_(2)O_(2)and 2,2'-azobis(2-methylpropionamidine)dihydrochloride(AAPH).Moreover,BPCC significantly inhibited cell viability of HepG2 cells in a dose-dependent manner with an estimated IC_(50)of 7.61 mg/mL.The results of 4',6-diamidino-2-phenylindole(DAPI)and acridine:orange/ethidium bromide(AO/EB)staining demonstrated the apoptotic morphological changes and cell mediated death in BPCC treated HepG2 cells.In addition,BPCC induced decrease of mitochondrial membrane potential(MMP)in HepG2 cells.Therefore,the present finding proved that BPCC encompasses significant antioxidant activity and anticancer property on HepG2 cells and can be used as alternative food antioxidants for cancer prevention benefits. 展开更多

【目的】探讨卷枝毛霉中苹果酸酶同工酶V的性质。【方法】克隆卷枝毛霉中编码苹果酸酶同工酶V的mel基因并在大肠杆菌BL21(DE3)中表达,利用His标签纯化获得了高纯度的重组酶BLME1,并进行酶学性质分析。【结果】该重组酶最适pH为8.0,最适... 【目的】探讨卷枝毛霉中苹果酸酶同工酶V的性质。【方法】克隆卷枝毛霉中编码苹果酸酶同工酶V的mel基因并在大肠杆菌BL21(DE3)中表达,利用His标签纯化获得了高纯度的重组酶BLME1,并进行酶学性质分析。【结果】该重组酶最适pH为8.0,最适温度为33℃,在此条件下酶活达到92.8 U/mg,对底物L-苹果酸和NADP^+的米氏常数K_m值为0.74960±0.06129 mmol/L和0.22070±0.01810 mmol/L,最大反应速度V_(max)分别为72.820±1.077 U/mg和86.110±1.665 U/mg。金属离子Mg^(2+)、Mn^(2+)、Co^(2+)、Ni^(2+)可以激活BLME1的活性,而Ca^(2+)、Cu^(2+)对BLME1活性则有抑制作用,中间代谢产物草酰乙酸和α-酮戊二酸也会抑制BLME1的活性,但琥珀酸却对BLME1有激活作用。【结论】本实验调查了卷枝毛霉苹果酸酶同工酶V的最适反应温度和pH、动力学参数,以及各种金属离子和中间代谢产物对酶活力的影响,这为以后深入研究该苹果酸酶的功能提供了理论依据和参考。 展开更多
