Osteosarcoma(OS)is the most prevalent primary malignant bone tumor affecting children and adolescents.Despite ongoing research efforts,the 5-year survival rate has remained stagnant for many years,highlighting the cri...Osteosarcoma(OS)is the most prevalent primary malignant bone tumor affecting children and adolescents.Despite ongoing research efforts,the 5-year survival rate has remained stagnant for many years,highlighting the critical need for novel drug development to enhance current treatment protocols.ZiyuglycosideⅡ(ZYGⅡ),a triterpenoid saponin extracted from S.officinalis,has recently demonstrated antitumor properties.This study evaluates the antitumor effect of ZYGⅡon osteosarcoma and elucidates its mechanism of action through the co-regulation of p53 and estrogen-related receptor gamma(ESRRG),which inhibits disease progression.The research employs in vitro experiments using multiple established osteosarcoma cell lines,as well as in vivo studies utilizing a nude mouse model of orthotopic xenograft osteosarcoma.Additionally,ESRRG shRNA was used to construct stable ESRRG-reducing OS cell lines to investigate the molecular mechanism by which ZYGⅡexerts its anti-osteosarcoma effects through the co-regulation of ESRRG and p53.Results indicate that ZYGⅡadministration led to decreased OS cell viability and reduced tumor volumes.Furthermore,cell cycles were arrested at the G_(0)/G1 phase,while the proportion of apoptotic cells increased.Expression of p53,ESRRG,p21,Bax,Cleaved Caspase-9,and Cleaved Caspase-3 proteins increased,while expression of CDK4,Cyclin D1,and Bcl-2 proteins decreased.Multiple ZYGⅡand ESRRG docking patterns were simulated through molecular docking.Comparing the pharmacodynamic response of ZYGⅡto OS cell lines with reduced ESRRG and normal expression demonstrated that ZYGⅡinhibits osteosarcoma progression,induces cell cycle arrest,and promotes cell apoptosis through the coordination of p53 and ESRRG.In conclusion,ZYGⅡinhibits osteosarcoma progression,leads to cell cycle arrest,and promotes cell apoptosis through synergistic regulation of p53 and ESRRG.展开更多
This study explored the mediating role of moral courage in the relationship between emotional intelligence and perceptions of patient safety competence among nursing students in clinical practice.The study sample comp...This study explored the mediating role of moral courage in the relationship between emotional intelligence and perceptions of patient safety competence among nursing students in clinical practice.The study sample comprised 220 nursing students from a teaching hospital(female=93.6%;mean age=20.64 years,SD=1.78 years).Nursing students completed standardized measures of Emotional Intelligence Scale,the Moral Courage Scale,and the Health Professional Education in Patient Safety Survey.Following hierarchical regression analysis and bootstrap analysis,the results showed that emotional intelligence and moral courage significantly predicted high levels of the perceptions of patient safety competence among nursing students in clinical practice.Moreover,moral courage partially mediated the relationship emotional intelligence and perceptions of patient safety competence.The results indicate measures focused on improving emotional intelligence and moral courage levels among nursing students in clinical practice will be effective at increasing their perceptions of patient safety competence.展开更多
BACKGROUND Subcutaneous implantable cardioverter-defibrillator(S-ICD)implantation requires effective anesthesia.General anesthesia(GA)carries risks like hemodynamic instability,while ultrasound-guided intercostal nerv...BACKGROUND Subcutaneous implantable cardioverter-defibrillator(S-ICD)implantation requires effective anesthesia.General anesthesia(GA)carries risks like hemodynamic instability,while ultrasound-guided intercostal nerve block(US-ICNB)may offer better pain control.This study hypothesized US-ICNB is superior in perioperative safety and pain management.AIM To compare perioperative outcomes of GA and US-ICNB in S-ICD implantation.METHODS This retrospective single-center study included 64 patients who received S-ICD implantation between February 2021 and December 2024.They were divided into GA and US-ICNB groups based on anesthesia type.Demographic data,perioperative parameters(operation time,pain scores,analgesic usage),and postoperative outcomes(complications,defibrillation events)were collected and analyzed.Statistical tests were used to compare the two groups.RESULTS This study included 64 patients(20 in the GA group and 44 in the US-ICNB group).Baseline left ventricular ejection fraction was significantly lower in the US-ICNB group(39.20%±12.00%vs 56.20%±11.50%in GA,P<0.001),while American Society of Anesthesiologists scores and comorbidities were comparable.US-ICNB showed superior pain control,with significantly lower numeric rating scale scores at 6-48 hours(P<0.001)and fewer patients requiring analgesics(P=0.02).The US-ICNB group had shorter operation times(P<0.001),total hospital stays(P<0.001),and later first analgesia times(P<0.001).No anesthesia-related complications occurred in either group.CONCLUSION Both anesthetic methods were safe in the short term.However,US-ICNB was superior in reducing operation and hospital stay times and alleviating peri-operative pain.It has high safety in S-ICD implantation and deserves further clinical promotion,though large-scale,multi-center,randomized controlled trials are needed to confirm these findings.展开更多
This study explored the role of learning engagement in the relationship between academic self-efficacy and self-directed learning ability among nursing students.Participants were 328 Chinese nursing students(male=11.3...This study explored the role of learning engagement in the relationship between academic self-efficacy and self-directed learning ability among nursing students.Participants were 328 Chinese nursing students(male=11.3%,female=88.7%;mean age=20.86 years;SD=1.75 years).The participants completed surveys on academic self-efficacy(Academic Self-efficacy Scale),learning engagement(Learning Engagement Scale),and self-directed learning ability(Self-directed Learning Instrument).Hayes regression-based PROCESS macro analysis revealed that learning engagement mediated the relationship between academic self-efficacy and self-directed learning ability.The hierarchical regression analysis showed higher academic self-efficacy to be associated with self-directed learning ability.Additionally learning engagement was associated with higher self-directed learning ability.Based on thesefindings,there is a need for interventions to improve students’self-directed learning ability through increasing their academic self-efficacy and enhancing learning engagement.展开更多
To investigate whether transcription of hepatitis B virus (HBV) gene occurs in human sperm, total RNA was extracted from sperm of patients with chronic HBV infection (test-I), from donor sperm transfected with a p...To investigate whether transcription of hepatitis B virus (HBV) gene occurs in human sperm, total RNA was extracted from sperm of patients with chronic HBV infection (test-I), from donor sperm transfected with a plasmid containing the full-length HBV genome (test-2), and from nontransfected donor sperm (control), used as the template for reverse transcription-polymerase chain reaction (RT-PCR). Positive bands for HBV DNA were observed in the test groups but not in the control. Next, to identify the role of host genes in regulating viral gene transcription in sperm, total RNA was extracted from 2-cell embryos derived from hamster oocytes fertilized in vitro by HBV-transfected (test) or nontransfected (control) human sperm and successively subjected to SMART-PCR, suppression subtractive hybridization, T/A cloning, bacterial amplification, microarray hybridization, sequencing and the Basic Local Alignment Search Tool (BLAST) search to isolate differentially expressed genes. Twenty-nine sequences showing significant identity to five human gene families were identified, with chorionic somatomammotropin hormone 2 (CSH2), eukaryotic translation initiation factor 4 gamma 2 (EIF4G2), pterin-4 alpha-carbinolamine dehydratase 2 (PCBD2), pregnancy-specific beta-l-glycoprotein 4 (PSG4) and titin (TTN) selected to represent target genes. Using real-time quantitative RT-PCR (qRT-PCR), when CSH2 and PCBD2 (or EIF4G2, PSG4 and TTN) were silenced by RNA interference, transcriptional levels of HBV s and x genes significantly decreased (or increased) (P 〈 0.05). Silencing of a control gene in sperm did not significantly change transcription of HBV s and x genes (P 〉 0.05). This study provides the first experimental evidence that transcription of HBV genes occurs in human sperm and is regulated by host genes.展开更多

BACKGROUND Endoscopic submucosal dissection(ESD)and transanal endoscopic submucosal dissection(TES)are widely employed surgical techniques.However,the comparative efficacy and safety of both remain inconclusive.AIM To...BACKGROUND Endoscopic submucosal dissection(ESD)and transanal endoscopic submucosal dissection(TES)are widely employed surgical techniques.However,the comparative efficacy and safety of both remain inconclusive.AIM To comprehensively analyze and discern differences in surgical outcomes between ESD and TES.METHODS We conducted a systematic search of the electronic databases PubMed,Embase,Cochrane Central Register of Controlled Trials,Scopus,and CINAHL from inception till August 2023.We analyzed outcomes including recurrence rate,en bloc resection,R0 resection rate,perforation rate,procedure length,and hospital stay length applying a random-effects inverse-variance model.We assessed publication bias by conducting an Egger’s regression test and sensitivity analyses.RESULTS We pooled data from 11 studies involving 1013 participants.We found similar recurrence rates,with a pooled odds ratio of 0.545(95%CI:0.176-1.687).En bloc resection,R0 resection,and perforation rate values were also similar for both ESD and TES.The pooled analysis for procedure length indicated a mean difference of-4.19 min(95%CI:-22.73 to 14.35),and the hospital stay was on average shorter for ESDs by about 0.789 days(95%CI:-1.671 to 0.093).CONCLUSION Both ESD and TES displayed similar efficacy and safety profiles across multiple outcomes.Our findings show that individualized patient and surgeon preferences,alongside specific clinical contexts,can be considered when selecting between these two techniques.展开更多
Objective To evaluate the risk of birth defects in children born following assisted reproductive technology (ART) and spontaneous conceptions. Methods This study carried out an updated systematic review to identify...Objective To evaluate the risk of birth defects in children born following assisted reproductive technology (ART) and spontaneous conceptions. Methods This study carried out an updated systematic review to identify papers published by August 2013 with data relating to birth defects of children conceived using ART (IVF and for ICSI) compared with those spontaneously conceived and also compared birth defects between subgroups of lVF and ICSI. Results Totally 76 studies were identified for review. The individual relative risk (RR) estimated for these studies ranged from 0.44 to 5.51, a signifieantly increased risk of birth defects was observed (RR=l.36, 95%CI=1.25-1.47) in ART compared with the spontaneously conceived group, which was also evident in the subgroup analysis. Among these studies, 16 studies simultaneously gave data of birth defects comparing IVF and ICSI children, which showed no difference in risk of combined effects (RR=0.90, 95%CI=0.80 1.02), but ICSI had a higher risk in subgroups of clinical research (RR=O. 76, 95%CI=0.65-0.89) and crude RR value (RR=O. 78, 95%CI=0.67 0.91). Conclusion Pooled results from all suitable published studies suggested that children born following ART were at increased risk of birth defects compared with spontaneous conceptions. There is no difference in birth defect risk between children conceived by IVF or 1CS1 using a summative analysis, however, 1CS1 had a significant higher risk in birth defect risk comparing with 1VF when using subgroup analyses of sample size and RR value.展开更多

Huge numbers of induced unpropped(IU)fractures are generated near propped fractures during hydraulic fracturing in shale gas reservoirs.But it is still unclear how their fracture space and conductivity evolve under in...Huge numbers of induced unpropped(IU)fractures are generated near propped fractures during hydraulic fracturing in shale gas reservoirs.But it is still unclear how their fracture space and conductivity evolve under in-situ conditions.This paper prepares three types of samples,namely,manually split vertical/parallel to beddings(MSV,MSP)and parallel natural fractures(NFP),to represent the varied IU fractures as well as their surface morphology.Laser scan and reconstruction demonstrate that the initial fracture spaces of MSVs and MSPs are limited as the asperities of newly created surfaces are wellmatched,and the NFPs have bigger space due to inhomogeneous geological corrosion.Surface slippage and consequent asperity mismatch increase the fracture width by several times,and the increase is proportional to surface roughness.Under stressful conditions,the slipped MSVs retain the smallest residual space and conductivity due to the newly sharp asperities.Controlled by the bedding structures and clay mineral hydrations,the conductivity of MSPs decreases most after treated with a fracturing fluid.The NFPs remain the highest conductivity,benefitting from their dispersive,gentle,and strong asperities.The results reveal the diverse evolution trends of IU fractures and can provide reliable parameters for fracturing design,post-fracturing evaluation,and productivity forecasting.展开更多
Essential proteins play a vital role in biological processes,and the combination of gene expression profiles with Protein-Protein Interaction(PPI)networks can improve the identification of essential proteins.However,g...Essential proteins play a vital role in biological processes,and the combination of gene expression profiles with Protein-Protein Interaction(PPI)networks can improve the identification of essential proteins.However,gene expression data are prone to significant fluctuations due to noise interference in topological networks.In this work,we discretized gene expression data and used the discrete similarities of the gene expression spectrum to eliminate noise fluctuation.We then proposed the Pearson Jaccard coefficient(PJC)that consisted of continuous and discrete similarities in the gene expression data.Using the graph theory as the basis,we fused the newly proposed similarity coefficient with the existing network topology prediction algorithm at each protein node to recognize essential proteins.This strategy exhibited a high recognition rate and good specificity.We validated the new similarity coefficient PJC on PPI datasets of Krogan,Gavin,and DIP of yeast species and evaluated the results by receiver operating characteristic analysis,jackknife analysis,top analysis,and accuracy analysis.Compared with that of node-based network topology centrality and fusion biological information centrality methods,the new similarity coefficient PJC showed a significantly improved prediction performance for essential proteins in DC,IC,Eigenvector centrality,subgraph centrality,betweenness centrality,closeness centrality,NC,PeC,and WDC.We also compared the PJC coefficient with other methods using the NF-PIN algorithm,which predicts proteins by constructing active PPI networks through dynamic gene expression.The experimental results proved that our newly proposed similarity coefficient PJC has superior advantages in predicting essential proteins.展开更多
The Warburg effect,characterized by excessive lactate production,and transcriptional dysregulation are two hallmarks of tumors.However,the precise influence of lactate on epigenetic modifications at a genome-wide leve...The Warburg effect,characterized by excessive lactate production,and transcriptional dysregulation are two hallmarks of tumors.However,the precise influence of lactate on epigenetic modifications at a genome-wide level and its impact on gene transcription in tumor cells remain unclear.In this study,we conducted genome-wide profiling of histone H3 lysine 18 lactylation(H3K18la)in T-cell acute lymphoblastic leukemia(T-ALL).We observed elevated lactate and H3K18la levels in T-ALL cells compared to normal T cells,with H3K18la levels positively associated with cell proliferation.Accordingly,we observed a significant shift in genome-wide H3K18la modifications from T cell immunity in normal T cells to leukemogenesis in T-ALL,correlated with altered gene transcription profiles.We showed that H3K18la primarily functions in active transcriptional regulation and observed clusters of H3K18la modifications resembling super-enhancers.Disrupting H3K18la modification revealed both synergistic and divergent changes between H3K18la and histone H3 lysine 27 acetylation(H3K27ac)modifications.Finally,we found that the high transcription of H3K18la target genes,IGFBP2 and IARS,is associated with inferior prognosis of T-ALL.These findings enhance our understanding of how metabolic disruptions contribute to transcription dysregulation through epigenetic changes in T-ALL,underscoring the interplay of histone modifications in maintaining oncogenic epigenetic stability.展开更多
In plant chloroplasts,photosystem II(PSII)complexes,together with light-harvesting complex II(LHCII),form various PSII-LHCII supercomplexes(SCs).This process likely involves immunophilins,but the underlying regulatory...In plant chloroplasts,photosystem II(PSII)complexes,together with light-harvesting complex II(LHCII),form various PSII-LHCII supercomplexes(SCs).This process likely involves immunophilins,but the underlying regulatory mechanisms are unclear.Here,by comparing Arabidopsis thaliana mutants lacking the chloroplast lumen-localized immunophilin CYCLOPHILIN28(CYP28)to wildtype and transgenic complemented lines,we determined that CYP28 regulates the assembly and accumulation of PSII-LHCII SCs.Compared to the wild type,cyp28 plants showed accelerated leaf growth,earlier flowering time,and enhanced accumulation of high molecular weight PSII-LHCII SCs under normal light conditions.The lack of CYP28 also significantly affected the electron transport rate.Blue native-polyacrylamide gel electrophoresis analysis revealed more Lhcb6 and less Lhcb4 in M-LHCII-Lhcb4-Lhcb6 complexes in cyp28 versus wild-type plants.Peptidyl-prolyl cis/trans isomerase(PPIase)activity assays revealed that CYP28 exhibits weak PPIase activity and that its K113 and E187 residues are critical for this activity.Mutant analysis suggested that CYP28 may regulate PSIILHCII SC accumulation by altering the configuration of Lhcb6 via its PPIase activity.Furthermore,the Lhcb6-P139 residue is critical for PSII-LHCII SC assembly and accumulation.Therefore,our findings suggest that CYP28 likely regulates PSII-LHCII SC assembly and accumulation by altering the configuration of P139 of Lhcb6 via its PPIase activity.展开更多
Damaged skin cannot prevent harmful bacteria from invading tissues,causing infected wounds or even severe tissue damage.In this study,we developed a controlled-release antibacterial composite hydrogel system that can ...Damaged skin cannot prevent harmful bacteria from invading tissues,causing infected wounds or even severe tissue damage.In this study,we developed a controlled-release antibacterial composite hydrogel system that can promote wound angiogenesis and inhibit inflammation by sustained releasing Cu-Epigallocatechin-3-gallate(Cu-EGCG)nano-capsules.The prepared SilMA/HAMA/Cu-EGCG hydrogel showed an obvious inhibitory effect on Escherichia coli(E.coli)and Staphylococcus aureus(S.aureus).It could also promote the proliferation and migration of L929 fibroblasts.In vivo full-thickness infected wound healing experiments confirmed the angiogenesis and inflammation regulating effect.Accelerate collagen deposition and wound healing speed were also observed in the SilMA/HAMA/Cu-EGCG hydrogel treated group.The findings of this study show the great potential of this controlled-release antibacterial composite hydrogel in the application of chronic wound healing.展开更多
Coilia nasus is an endangered fish species in the Yangtze River,and there is urgent need to know the genes related to its reproduction and fertility.The DAZ family including boule,daz and dazl,plays an important role ...Coilia nasus is an endangered fish species in the Yangtze River,and there is urgent need to know the genes related to its reproduction and fertility.The DAZ family including boule,daz and dazl,plays an important role in germ cells development.In this study,the cDNA sequence of boule(Cnbol)and dazl(Cndazl)genes were cloned and their expression patterns were characterized in Coilia nasus.RT-PCR showed that the expression of Cnbol and Cndazl mRNAs was restricted in adult gonads.The section in situ hybridization indicated that the temporospatial expression patterns of Cnbol and Cndazl were significantly different.In the testis,Cnbol was mainly detected in spermatocytes and spermatids,while Cndazl was prominently expressed in s permatogonia.In the ovary,Cnbol and Cndazl were highly expressed in the early stages of oocytes.Interestingly,Cndazl was also concentrated in perinuclear speckle and then localized to the Balbiani body in late stages of oocytes.In addition,Cnbol and Cndazl 3′UTR can specifically label medaka Primordial Germ Cells(PGCs).According to our results,Cnbol and Cndazl are marker genes of germ cells and may play a vital role in the development and differentiation of germ cells in Coilia nasus.展开更多
The plasmonic nanogap antenna is an efficient radiating or receiving optical device. The resonance behavior of optical antennas is commonly attributed to the excitation of a localized surface plasmon resonance(LSPR), ...The plasmonic nanogap antenna is an efficient radiating or receiving optical device. The resonance behavior of optical antennas is commonly attributed to the excitation of a localized surface plasmon resonance(LSPR), which can be theoretically defined as the quasi-normal mode(QNM). To clarify the physical origin of the LSPR, we build up an analytical model of the LSPR by considering a multiple scattering process of propagative surface plasmon polaritons(SPPs) on the antenna arms. The model can comprehensively reproduce the complex eigenfrequency and the field distribution of QNMs of the antenna, unveiling that the LSPR arises from a Fabry–Perot resonance of SPPs. By further applying the complex pole expansion theorem of meromorphic functions, the field of the antenna under illumination by a nearby dipole emitter can be analytically expanded with QNMs, which well predicts the frequency response of the enhancement factor of radiation. The present model establishes explicit relations between the concepts of the LSPR and the propagative SPP and integrates the advantages of the Fabry–Perot and QNM formalisms of nanogap antennas.展开更多
The gonad specific expression of insulin-like growth factor 3(igf3)has suggested an important role of igf3 in fish reproduction.In this study,medaka igf3 was isolated and its expression patterns were compared with igf...The gonad specific expression of insulin-like growth factor 3(igf3)has suggested an important role of igf3 in fish reproduction.In this study,medaka igf3 was isolated and its expression patterns were compared with igf1,and germ cell gene vasa in adult medaka ovary.Molecular cloning and sequencing showed that the open reading frame(ORF)of medaka igf3 was comprised of 501 nucleotides and encoded 166 amino acid residues.Reverse transcription polymerase chain reaction(RT-PCR)analysis showed that igf3 RNA was specifically expressed in developing embryos and adult gonads of both sexes.Real time quantitative PCR(q-PCR)indicated that igf3 expression was gradually increased during oogenesis,and reached its highest level at stage V.Using Chromogenic and fluorescent in situ hybridization,igf3 was shown to be present in the germ cells of oogonia and in oocytes at stages I-III,as well as abundant in granulosa cells and theca cells of oocytes at stages IV-V.In addition,igf3 and igf1 were expressed mutually in the outer theca cells of stage V oocytes in ovary.Collectively,we demonstrated that gonad specific igf3 could mark medaka ovarian somatic cells and germ cells.These findings highlight the importance of igf3 during ovarian development.展开更多

文摘Osteosarcoma(OS)is the most prevalent primary malignant bone tumor affecting children and adolescents.Despite ongoing research efforts,the 5-year survival rate has remained stagnant for many years,highlighting the critical need for novel drug development to enhance current treatment protocols.ZiyuglycosideⅡ(ZYGⅡ),a triterpenoid saponin extracted from S.officinalis,has recently demonstrated antitumor properties.This study evaluates the antitumor effect of ZYGⅡon osteosarcoma and elucidates its mechanism of action through the co-regulation of p53 and estrogen-related receptor gamma(ESRRG),which inhibits disease progression.The research employs in vitro experiments using multiple established osteosarcoma cell lines,as well as in vivo studies utilizing a nude mouse model of orthotopic xenograft osteosarcoma.Additionally,ESRRG shRNA was used to construct stable ESRRG-reducing OS cell lines to investigate the molecular mechanism by which ZYGⅡexerts its anti-osteosarcoma effects through the co-regulation of ESRRG and p53.Results indicate that ZYGⅡadministration led to decreased OS cell viability and reduced tumor volumes.Furthermore,cell cycles were arrested at the G_(0)/G1 phase,while the proportion of apoptotic cells increased.Expression of p53,ESRRG,p21,Bax,Cleaved Caspase-9,and Cleaved Caspase-3 proteins increased,while expression of CDK4,Cyclin D1,and Bcl-2 proteins decreased.Multiple ZYGⅡand ESRRG docking patterns were simulated through molecular docking.Comparing the pharmacodynamic response of ZYGⅡto OS cell lines with reduced ESRRG and normal expression demonstrated that ZYGⅡinhibits osteosarcoma progression,induces cell cycle arrest,and promotes cell apoptosis through the coordination of p53 and ESRRG.In conclusion,ZYGⅡinhibits osteosarcoma progression,leads to cell cycle arrest,and promotes cell apoptosis through synergistic regulation of p53 and ESRRG.

文摘This study explored the mediating role of moral courage in the relationship between emotional intelligence and perceptions of patient safety competence among nursing students in clinical practice.The study sample comprised 220 nursing students from a teaching hospital(female=93.6%;mean age=20.64 years,SD=1.78 years).Nursing students completed standardized measures of Emotional Intelligence Scale,the Moral Courage Scale,and the Health Professional Education in Patient Safety Survey.Following hierarchical regression analysis and bootstrap analysis,the results showed that emotional intelligence and moral courage significantly predicted high levels of the perceptions of patient safety competence among nursing students in clinical practice.Moreover,moral courage partially mediated the relationship emotional intelligence and perceptions of patient safety competence.The results indicate measures focused on improving emotional intelligence and moral courage levels among nursing students in clinical practice will be effective at increasing their perceptions of patient safety competence.
文摘BACKGROUND Subcutaneous implantable cardioverter-defibrillator(S-ICD)implantation requires effective anesthesia.General anesthesia(GA)carries risks like hemodynamic instability,while ultrasound-guided intercostal nerve block(US-ICNB)may offer better pain control.This study hypothesized US-ICNB is superior in perioperative safety and pain management.AIM To compare perioperative outcomes of GA and US-ICNB in S-ICD implantation.METHODS This retrospective single-center study included 64 patients who received S-ICD implantation between February 2021 and December 2024.They were divided into GA and US-ICNB groups based on anesthesia type.Demographic data,perioperative parameters(operation time,pain scores,analgesic usage),and postoperative outcomes(complications,defibrillation events)were collected and analyzed.Statistical tests were used to compare the two groups.RESULTS This study included 64 patients(20 in the GA group and 44 in the US-ICNB group).Baseline left ventricular ejection fraction was significantly lower in the US-ICNB group(39.20%±12.00%vs 56.20%±11.50%in GA,P<0.001),while American Society of Anesthesiologists scores and comorbidities were comparable.US-ICNB showed superior pain control,with significantly lower numeric rating scale scores at 6-48 hours(P<0.001)and fewer patients requiring analgesics(P=0.02).The US-ICNB group had shorter operation times(P<0.001),total hospital stays(P<0.001),and later first analgesia times(P<0.001).No anesthesia-related complications occurred in either group.CONCLUSION Both anesthetic methods were safe in the short term.However,US-ICNB was superior in reducing operation and hospital stay times and alleviating peri-operative pain.It has high safety in S-ICD implantation and deserves further clinical promotion,though large-scale,multi-center,randomized controlled trials are needed to confirm these findings.

文摘This study explored the role of learning engagement in the relationship between academic self-efficacy and self-directed learning ability among nursing students.Participants were 328 Chinese nursing students(male=11.3%,female=88.7%;mean age=20.86 years;SD=1.75 years).The participants completed surveys on academic self-efficacy(Academic Self-efficacy Scale),learning engagement(Learning Engagement Scale),and self-directed learning ability(Self-directed Learning Instrument).Hayes regression-based PROCESS macro analysis revealed that learning engagement mediated the relationship between academic self-efficacy and self-directed learning ability.The hierarchical regression analysis showed higher academic self-efficacy to be associated with self-directed learning ability.Additionally learning engagement was associated with higher self-directed learning ability.Based on thesefindings,there is a need for interventions to improve students’self-directed learning ability through increasing their academic self-efficacy and enhancing learning engagement.

文摘To investigate whether transcription of hepatitis B virus (HBV) gene occurs in human sperm, total RNA was extracted from sperm of patients with chronic HBV infection (test-I), from donor sperm transfected with a plasmid containing the full-length HBV genome (test-2), and from nontransfected donor sperm (control), used as the template for reverse transcription-polymerase chain reaction (RT-PCR). Positive bands for HBV DNA were observed in the test groups but not in the control. Next, to identify the role of host genes in regulating viral gene transcription in sperm, total RNA was extracted from 2-cell embryos derived from hamster oocytes fertilized in vitro by HBV-transfected (test) or nontransfected (control) human sperm and successively subjected to SMART-PCR, suppression subtractive hybridization, T/A cloning, bacterial amplification, microarray hybridization, sequencing and the Basic Local Alignment Search Tool (BLAST) search to isolate differentially expressed genes. Twenty-nine sequences showing significant identity to five human gene families were identified, with chorionic somatomammotropin hormone 2 (CSH2), eukaryotic translation initiation factor 4 gamma 2 (EIF4G2), pterin-4 alpha-carbinolamine dehydratase 2 (PCBD2), pregnancy-specific beta-l-glycoprotein 4 (PSG4) and titin (TTN) selected to represent target genes. Using real-time quantitative RT-PCR (qRT-PCR), when CSH2 and PCBD2 (or EIF4G2, PSG4 and TTN) were silenced by RNA interference, transcriptional levels of HBV s and x genes significantly decreased (or increased) (P 〈 0.05). Silencing of a control gene in sperm did not significantly change transcription of HBV s and x genes (P 〉 0.05). This study provides the first experimental evidence that transcription of HBV genes occurs in human sperm and is regulated by host genes.

文摘BACKGROUND Endoscopic submucosal dissection(ESD)and transanal endoscopic submucosal dissection(TES)are widely employed surgical techniques.However,the comparative efficacy and safety of both remain inconclusive.AIM To comprehensively analyze and discern differences in surgical outcomes between ESD and TES.METHODS We conducted a systematic search of the electronic databases PubMed,Embase,Cochrane Central Register of Controlled Trials,Scopus,and CINAHL from inception till August 2023.We analyzed outcomes including recurrence rate,en bloc resection,R0 resection rate,perforation rate,procedure length,and hospital stay length applying a random-effects inverse-variance model.We assessed publication bias by conducting an Egger’s regression test and sensitivity analyses.RESULTS We pooled data from 11 studies involving 1013 participants.We found similar recurrence rates,with a pooled odds ratio of 0.545(95%CI:0.176-1.687).En bloc resection,R0 resection,and perforation rate values were also similar for both ESD and TES.The pooled analysis for procedure length indicated a mean difference of-4.19 min(95%CI:-22.73 to 14.35),and the hospital stay was on average shorter for ESDs by about 0.789 days(95%CI:-1.671 to 0.093).CONCLUSION Both ESD and TES displayed similar efficacy and safety profiles across multiple outcomes.Our findings show that individualized patient and surgeon preferences,alongside specific clinical contexts,can be considered when selecting between these two techniques.
文摘Objective To evaluate the risk of birth defects in children born following assisted reproductive technology (ART) and spontaneous conceptions. Methods This study carried out an updated systematic review to identify papers published by August 2013 with data relating to birth defects of children conceived using ART (IVF and for ICSI) compared with those spontaneously conceived and also compared birth defects between subgroups of lVF and ICSI. Results Totally 76 studies were identified for review. The individual relative risk (RR) estimated for these studies ranged from 0.44 to 5.51, a signifieantly increased risk of birth defects was observed (RR=l.36, 95%CI=1.25-1.47) in ART compared with the spontaneously conceived group, which was also evident in the subgroup analysis. Among these studies, 16 studies simultaneously gave data of birth defects comparing IVF and ICSI children, which showed no difference in risk of combined effects (RR=0.90, 95%CI=0.80 1.02), but ICSI had a higher risk in subgroups of clinical research (RR=O. 76, 95%CI=0.65-0.89) and crude RR value (RR=O. 78, 95%CI=0.67 0.91). Conclusion Pooled results from all suitable published studies suggested that children born following ART were at increased risk of birth defects compared with spontaneous conceptions. There is no difference in birth defect risk between children conceived by IVF or 1CS1 using a summative analysis, however, 1CS1 had a significant higher risk in birth defect risk comparing with 1VF when using subgroup analyses of sample size and RR value.
文摘The taxonomy of Pheidole Westwood,1841 from Fujian,China is discussed.A new species,Pheidole flavigaster sp.nov.,is described.A key to the species of Pheidole based on major worker is provided.This new species is small in size,body color is brown and gaster is yellow.Pheidole flavigaster sp.nov.is similar to Pheidole schoedli Eguchi,Hashimoto&Malsch,2006 from Indonesia,but the propodeal spine is not upward and the hypostoma inner and median teeth are not distinct,making it obvious to distinguish them.The holotype and paratypes are both deposited at the Kunming Natural History Museum of Zoology,Kunming Institute of Zoology(KIZ),Chinese Academy of Sciences,Kunming,Yunnan,China.

文摘Huge numbers of induced unpropped(IU)fractures are generated near propped fractures during hydraulic fracturing in shale gas reservoirs.But it is still unclear how their fracture space and conductivity evolve under in-situ conditions.This paper prepares three types of samples,namely,manually split vertical/parallel to beddings(MSV,MSP)and parallel natural fractures(NFP),to represent the varied IU fractures as well as their surface morphology.Laser scan and reconstruction demonstrate that the initial fracture spaces of MSVs and MSPs are limited as the asperities of newly created surfaces are wellmatched,and the NFPs have bigger space due to inhomogeneous geological corrosion.Surface slippage and consequent asperity mismatch increase the fracture width by several times,and the increase is proportional to surface roughness.Under stressful conditions,the slipped MSVs retain the smallest residual space and conductivity due to the newly sharp asperities.Controlled by the bedding structures and clay mineral hydrations,the conductivity of MSPs decreases most after treated with a fracturing fluid.The NFPs remain the highest conductivity,benefitting from their dispersive,gentle,and strong asperities.The results reveal the diverse evolution trends of IU fractures and can provide reliable parameters for fracturing design,post-fracturing evaluation,and productivity forecasting.

文摘Essential proteins play a vital role in biological processes,and the combination of gene expression profiles with Protein-Protein Interaction(PPI)networks can improve the identification of essential proteins.However,gene expression data are prone to significant fluctuations due to noise interference in topological networks.In this work,we discretized gene expression data and used the discrete similarities of the gene expression spectrum to eliminate noise fluctuation.We then proposed the Pearson Jaccard coefficient(PJC)that consisted of continuous and discrete similarities in the gene expression data.Using the graph theory as the basis,we fused the newly proposed similarity coefficient with the existing network topology prediction algorithm at each protein node to recognize essential proteins.This strategy exhibited a high recognition rate and good specificity.We validated the new similarity coefficient PJC on PPI datasets of Krogan,Gavin,and DIP of yeast species and evaluated the results by receiver operating characteristic analysis,jackknife analysis,top analysis,and accuracy analysis.Compared with that of node-based network topology centrality and fusion biological information centrality methods,the new similarity coefficient PJC showed a significantly improved prediction performance for essential proteins in DC,IC,Eigenvector centrality,subgraph centrality,betweenness centrality,closeness centrality,NC,PeC,and WDC.We also compared the PJC coefficient with other methods using the NF-PIN algorithm,which predicts proteins by constructing active PPI networks through dynamic gene expression.The experimental results proved that our newly proposed similarity coefficient PJC has superior advantages in predicting essential proteins.

文摘The Warburg effect,characterized by excessive lactate production,and transcriptional dysregulation are two hallmarks of tumors.However,the precise influence of lactate on epigenetic modifications at a genome-wide level and its impact on gene transcription in tumor cells remain unclear.In this study,we conducted genome-wide profiling of histone H3 lysine 18 lactylation(H3K18la)in T-cell acute lymphoblastic leukemia(T-ALL).We observed elevated lactate and H3K18la levels in T-ALL cells compared to normal T cells,with H3K18la levels positively associated with cell proliferation.Accordingly,we observed a significant shift in genome-wide H3K18la modifications from T cell immunity in normal T cells to leukemogenesis in T-ALL,correlated with altered gene transcription profiles.We showed that H3K18la primarily functions in active transcriptional regulation and observed clusters of H3K18la modifications resembling super-enhancers.Disrupting H3K18la modification revealed both synergistic and divergent changes between H3K18la and histone H3 lysine 27 acetylation(H3K27ac)modifications.Finally,we found that the high transcription of H3K18la target genes,IGFBP2 and IARS,is associated with inferior prognosis of T-ALL.These findings enhance our understanding of how metabolic disruptions contribute to transcription dysregulation through epigenetic changes in T-ALL,underscoring the interplay of histone modifications in maintaining oncogenic epigenetic stability.

文摘In plant chloroplasts,photosystem II(PSII)complexes,together with light-harvesting complex II(LHCII),form various PSII-LHCII supercomplexes(SCs).This process likely involves immunophilins,but the underlying regulatory mechanisms are unclear.Here,by comparing Arabidopsis thaliana mutants lacking the chloroplast lumen-localized immunophilin CYCLOPHILIN28(CYP28)to wildtype and transgenic complemented lines,we determined that CYP28 regulates the assembly and accumulation of PSII-LHCII SCs.Compared to the wild type,cyp28 plants showed accelerated leaf growth,earlier flowering time,and enhanced accumulation of high molecular weight PSII-LHCII SCs under normal light conditions.The lack of CYP28 also significantly affected the electron transport rate.Blue native-polyacrylamide gel electrophoresis analysis revealed more Lhcb6 and less Lhcb4 in M-LHCII-Lhcb4-Lhcb6 complexes in cyp28 versus wild-type plants.Peptidyl-prolyl cis/trans isomerase(PPIase)activity assays revealed that CYP28 exhibits weak PPIase activity and that its K113 and E187 residues are critical for this activity.Mutant analysis suggested that CYP28 may regulate PSIILHCII SC accumulation by altering the configuration of Lhcb6 via its PPIase activity.Furthermore,the Lhcb6-P139 residue is critical for PSII-LHCII SC assembly and accumulation.Therefore,our findings suggest that CYP28 likely regulates PSII-LHCII SC assembly and accumulation by altering the configuration of P139 of Lhcb6 via its PPIase activity.

文摘Damaged skin cannot prevent harmful bacteria from invading tissues,causing infected wounds or even severe tissue damage.In this study,we developed a controlled-release antibacterial composite hydrogel system that can promote wound angiogenesis and inhibit inflammation by sustained releasing Cu-Epigallocatechin-3-gallate(Cu-EGCG)nano-capsules.The prepared SilMA/HAMA/Cu-EGCG hydrogel showed an obvious inhibitory effect on Escherichia coli(E.coli)and Staphylococcus aureus(S.aureus).It could also promote the proliferation and migration of L929 fibroblasts.In vivo full-thickness infected wound healing experiments confirmed the angiogenesis and inflammation regulating effect.Accelerate collagen deposition and wound healing speed were also observed in the SilMA/HAMA/Cu-EGCG hydrogel treated group.The findings of this study show the great potential of this controlled-release antibacterial composite hydrogel in the application of chronic wound healing.

文摘Coilia nasus is an endangered fish species in the Yangtze River,and there is urgent need to know the genes related to its reproduction and fertility.The DAZ family including boule,daz and dazl,plays an important role in germ cells development.In this study,the cDNA sequence of boule(Cnbol)and dazl(Cndazl)genes were cloned and their expression patterns were characterized in Coilia nasus.RT-PCR showed that the expression of Cnbol and Cndazl mRNAs was restricted in adult gonads.The section in situ hybridization indicated that the temporospatial expression patterns of Cnbol and Cndazl were significantly different.In the testis,Cnbol was mainly detected in spermatocytes and spermatids,while Cndazl was prominently expressed in s permatogonia.In the ovary,Cnbol and Cndazl were highly expressed in the early stages of oocytes.Interestingly,Cndazl was also concentrated in perinuclear speckle and then localized to the Balbiani body in late stages of oocytes.In addition,Cnbol and Cndazl 3′UTR can specifically label medaka Primordial Germ Cells(PGCs).According to our results,Cnbol and Cndazl are marker genes of germ cells and may play a vital role in the development and differentiation of germ cells in Coilia nasus.

文摘The plasmonic nanogap antenna is an efficient radiating or receiving optical device. The resonance behavior of optical antennas is commonly attributed to the excitation of a localized surface plasmon resonance(LSPR), which can be theoretically defined as the quasi-normal mode(QNM). To clarify the physical origin of the LSPR, we build up an analytical model of the LSPR by considering a multiple scattering process of propagative surface plasmon polaritons(SPPs) on the antenna arms. The model can comprehensively reproduce the complex eigenfrequency and the field distribution of QNMs of the antenna, unveiling that the LSPR arises from a Fabry–Perot resonance of SPPs. By further applying the complex pole expansion theorem of meromorphic functions, the field of the antenna under illumination by a nearby dipole emitter can be analytically expanded with QNMs, which well predicts the frequency response of the enhancement factor of radiation. The present model establishes explicit relations between the concepts of the LSPR and the propagative SPP and integrates the advantages of the Fabry–Perot and QNM formalisms of nanogap antennas.

文摘The gonad specific expression of insulin-like growth factor 3(igf3)has suggested an important role of igf3 in fish reproduction.In this study,medaka igf3 was isolated and its expression patterns were compared with igf1,and germ cell gene vasa in adult medaka ovary.Molecular cloning and sequencing showed that the open reading frame(ORF)of medaka igf3 was comprised of 501 nucleotides and encoded 166 amino acid residues.Reverse transcription polymerase chain reaction(RT-PCR)analysis showed that igf3 RNA was specifically expressed in developing embryos and adult gonads of both sexes.Real time quantitative PCR(q-PCR)indicated that igf3 expression was gradually increased during oogenesis,and reached its highest level at stage V.Using Chromogenic and fluorescent in situ hybridization,igf3 was shown to be present in the germ cells of oogonia and in oocytes at stages I-III,as well as abundant in granulosa cells and theca cells of oocytes at stages IV-V.In addition,igf3 and igf1 were expressed mutually in the outer theca cells of stage V oocytes in ovary.Collectively,we demonstrated that gonad specific igf3 could mark medaka ovarian somatic cells and germ cells.These findings highlight the importance of igf3 during ovarian development.
