BACKGROUND Children with critical acute abdominal conditions often undergo intestinal stoma surgery.AIM To explore the impact of a visual mobile terminal-based extended care model on caregiver competence for children with enterostomies.METHODS One hundred twenty children with enterostomies and their caregivers in a children's hospital in Beijing were divided into a control group and a study group.The control group(60 cases)received traditional telephone follow-up for continuity of care,while the study group(60 cases)used a visualization mobile terminal-based care model.The incidence of stoma-related complications,caregiver burden scale,and competence scores of children with stoma were compared between the two groups.RESULTS The primary caregiver burden score in the study group(37.22±3.17)was significantly lower than that in the control group(80.00±4.47),and the difference was statistically significant(P<0.05).Additionally,the caregiving ability score of the study group(172.08±3.49)was significantly higher than that of the control group(117.55±4.28;P<0.05).The total incidence of complications in the study group(11.7%,7/60)was significantly lower compared to the control group(33.3%,20/60;χ2=8.086,P=0.004).CONCLUSION The visual mobile terminal-based care model reduces caregiver burden,improves home care ability,lowers the incidence of complications and readmission rates,and supports successful second-stage reduction surgery for children with enterostomies. 展开更多

BACKGROUND Body composition analysis(BCA)is primarily used in the management of conditions such as obesity and endocrine disorders.However,its potential in providing nutritional guidance for patients with Alzheimer’s disease(AD)remains relatively unexplored.AIM To explore the clinical efficacy of BCA-based dietary nutrition scheme on bone metabolism in AD patients.METHODS This retrospective study included 96 patients with AD complicated by osteoporosis who were admitted to The Third Hospital of Quzhou between January 2023 and December 2024.Based on data from previous similar studies,the patients were randomly assigned to either a routine diet(RD)group(n=48)or a personalized nutrition(PN)group(n=48).The RD group received conventional dietary guidance,while the PN group received individualized diet intervention measures based on human BCA.The intervention period lasted for 12 weeks.Bone mineral density(BMD),body mass index(BMI),muscle mass,mineral content,osteocalcin,25-hydroxyvitamin D,procollagen type I N-terminal propeptide(PINP),beta C-terminal telopeptide of type I collagen(β-CTX),and serum calcium were measured and compared between the two groups before and 12 weeks after the intervention.RESULTS No significant differences were observed between groups in terms of age,sex,height,BMI,or other baseline data(P>0.05).In both groups,BMI did not show significant changes after the intervention(P>0.05),whereas muscle mass and mineral content were significantly increased(P<0.05).After the intervention,BMI in the PN group did not differ significantly from that of the RD group,but muscle mass and mineral content were significantly higher in the PN group(P<0.05).After the intervention,a higher proportion of patients in the PN group had a T score>-1 compared to the RD group(P<0.05).The mini-mental state examination(MMSE)score was similar in both groups before the intervention.However,12 weeks after the intervention,the MMSE score in the PN group was significantly higher than that in the RD group(P<0.05).In both groups,the MMSE score significantly increased 12 weeks post-intervention compared to pre-intervention levels(P<0.05).Before the intervention,the levels of osteocalcin,serum calcium,PINP,β-CTX,and 25-hydroxyvitamin D were not significantly different between the two groups(P>0.05).After 12 weeks of intervention,the PN group exhibited higher levels of osteocalcin,serum calcium,and 25-hydroxyvitamin D,as well as lower levels of PINP andβ-CTX,compared to the RD group(P<0.05).In both groups,osteocalcin,serum calcium,and 25-hydroxyvitamin D levels were significantly higher,while PINP andβ-CTX levels were significantly lower after 12 weeks of intervention compared to baseline(P<0.05).CONCLUSION The human BCA-based dietary nutrition regimen plays a crucial role in improving BMD and bone metabolism,with effects that surpass those of conventional nutrition strategies.The findings of this study provide strong evidence for the nutritional management of AD patients. 展开更多

Background Viral diseases have profoundly influenced the sustainable development of the swine farming industry.With the development of genomics technology,the combination of transcriptome,genetic variation,immune response,and QTL mapping data to illustrate the interactions between pathogen and host immune system,will be an effective tool for identification of disease resistance genes in pigs.The immune system of an organism is the source of disease resistance in livestock,consisting of various immune tissues,as well as the immune cells and cytokines they produced.However,comprehensive systematic studies on transcriptome of porcine immune tissues are still rare.Poly(I:C),as a viral mimic,is commonly used to study immune responses of the body during viral infections,and serves as a valuable tool for investigating immune mechanisms in swine.Results WGCNA analysis identified core immune genes across six immune tissues(bone marrow,jejunum,lymph node,PBMC,spleen,thymus)in Landrace pigs,which are also crucial for the development of PBMCs.The examination of the changes in the proportion of immune cells during three developmental stages(1-month-old,4-month-old,7-month-old)shows a shift from innate immunity to humoral immunity.By integrating different epigenetic genomics datasets,we identified several core immune genes and their causal variants,including IFI44,IFIT5,EIF2AK2 and others,which are closely related to immune development and response.Functional validation studies reveal that the IFI44 gene acts as a negative regulator of the antiviral response;its inhibition effect significantly reduced Poly(I:C)-induced cell necrosis,while enhancing apoptosis to combat viral infections.Conclusion Our study elucidated the fundamental transcriptional program in porcine immune tissues and the immunodynamics underlying development of PBMCs,identifying many core immune genes,including IFI44,which plays a critical negative regulator role in the antiviral response,providing valuable insights for breeding programs aimed at enhancing pig disease resistance. 展开更多

Background Mastitis caused by Staphylococcus aureus(S.aureus)is one of the most intractable problems for the dairy industry,causing significantly reduced milk yields and early slaughter of cows worldwide.MicroRNAs(miRNAs)can post-transcriptionally regulate gene expression and studies in recent years have shown the importance of miRNA-associated gene regulation in S.aureus-induced mastitis.Results In this study,to investigate the role of miR-223 in mastitis,we performed experiments to overexpress and suppress miR-223 in an immortalized bovine mammary epithelial cell line(MAC-T)infected with S.aureus.Overexpression of miR-223 in MAC-T cells repressed cell apoptosis and necrosis induced by S.aureus infection,whereas suppression of miR-223 had the opposite effect.Transcriptome expression profiling with weighted gene co-expression network analysis(WGCNA)and gene set variation analysis(GSVA)showed that miR-223 affects apoptosis and inflammation-related pathways.Furthermore,differentially expressed(DE)genes were evaluated,and genes exhibiting contrasting expression trends in the miR-223 overexpressed and suppressed groups were assessed as potential target genes of miR-223.Potential target genes,including CDC25B,PTPRF,DCTN1,and DPP9,were observed to be associated with apoptosis and necroptosis.Finally,through integrative analysis of genome-wide association study(GWAS)data and the animal quantitative trait loci(QTL)database,we determined that target genes of miR-223 were significantly enriched in single-nucleotide polymorphisms(SNP)and QTLs related to somatic cell count(SCC)and mastitis.Conclusion In summary,miR-223 has an inhibitory effect on S.aureus-induced cell apoptosis and necrosis by regulating PTPRF,DCTN1,and DPP9.These genes were significantly enriched in QTL regions associated with bovine mastitis resistance,underscoring their relevance in genetic regulation of disease resilience.Our findings provide critical genetic markers for enhancing mastitis resistance,particularly S.aureus-induced mastitis,through selective breeding.This work offers valuable insights for developing cattle with improved resistance to mastitis via targeted genetic selection. 展开更多

Neutral oxygen evolution reaction(OER)is a crucial half-reaction for electrocatalytic chemical production under mild condition,but with limited development due to low activity and poor stability.Herein,a tungsten-doped cobalt molybdate(WDCMO)catalyst was synthesized for efficient and durable OER under neutral electrolyte.It is demonstrated that catalyst reconstruction is suppressed by W doping,which stabilizes the Co-O-Mo point-to-point connection in CoMoO_(4) architecture and stimulates to a lower valence state of active sites over the surface phase.Thereby,the surface structure maintains to avoid compound dissolution caused by over-oxidation during OER.Meanwhile,the WDCMO catalyst promotes charge transfer and optimizes*OH intermediate adsorption,which improves reaction kinetics and intrinsic activity.Consequently,the WDCMO electrode exhibits an overpotential of 302 mV at 10 mA cm^(-2) in neutral electrolyte with an improvement of 182 mV compared with CoMoO4 electrode.Furthermore,W doping significantly improves the electrode stability from 50 h to more than 320 h,with a suppressive potential attenuation from 2.82 to 0.29 mV h^(-1).This work will shed new light on designing rational electrocatalysts for neutral OER. 展开更多

目的观察肝经郁热证非哺乳期乳腺炎乳晕部瘘管患者应用清消方联合拔毒生肌散药线挂线治疗的临床疗效。方法选择2018年10月至2020年12月江苏省中医院乳腺外科门诊就诊的非哺乳期乳腺炎乳晕部瘘管患者60例作为研究对象,采用区组随机、临... 目的观察肝经郁热证非哺乳期乳腺炎乳晕部瘘管患者应用清消方联合拔毒生肌散药线挂线治疗的临床疗效。方法选择2018年10月至2020年12月江苏省中医院乳腺外科门诊就诊的非哺乳期乳腺炎乳晕部瘘管患者60例作为研究对象,采用区组随机、临床对照试验方法,按1∶1分为观察组及对照组,每组30例。两组均口服清消方,观察组加用拔毒生肌散药线挂线治疗,对照组加用普通丝线挂线治疗,每隔3~4 d挂线1次,共挂4次。测量瘘管口的大小、红肿范围,填写视觉模拟疼痛评分量表(VAS)。观察两组患者的痊愈时间、瘘管全部挂开时间、VAS评分及安全性指标(血常规及肝肾功能)等,并随访复发情况。结果在挂线治疗第3次时,观察组的瘘管口挂开长度长于对照组,差异有统计学意义(P<0.05)。与对照组比较,观察组瘘管挂开时间[(12.00±2.22)d vs(14.46±3.76)d,t=3.860,P<0.01]、痊愈时间[(19.75±2.94)d vs(23.40±3.22)d,t=4.585,P<0.01]更短。随着挂线治疗次数的增加,瘘管被逐渐挂开,两组患者挂线治疗的疼痛均明显减轻(P<0.05),但组间比较差异无统计学意义(P>0.05)。两组患者均未发生不良反应。结论清消方联合拔毒生肌散药线挂线治疗肝经郁热证非哺乳期乳腺炎乳晕部瘘管的疗效确切,可缩短瘘管挂开时间,加快痊愈时间,伤口感染率和复发率均较低。 展开更多

Resilience traits in pig populations allow animals to deal better with infectious disease and suboptimal production environments.The data on daily weight,feed intake and feed behaviors in pigs are collected in test period by automated feeding stations,which facilitate to evaluate the resilience traits.In this study,we adopted the root mean square error(RMSE)of ordinary least squares(OLS)and the negative residuals of quantile regression(QR)to generate four different novel resilience traits using daily records of feed intake and feed duration between 90 and 180 days of age in a population of commercial Duroc pigs.The genome-wide association studies(GWAS)based on single-and two-trait mixed models were carried out on 550 pigs using 48,603 single nucleotide polymorphisms(SNPs)to identify genomic regions associated with resilience traits in growing pigs.We further focused on the GWAS signals to conduct gene annotation,colocalization with multi-tissue eQTL summary statistics of PigGTEx project and identification of enhancers and promoters using the publicly available data.The genomic heritabilities of four novel resilience traits ranged from 0.09 to 0.41.The pairwise genetic and phenotypic correlations ranged from 0.16 to 0.95 and from 0.05 to 0.36,respectively.Twenty-seven SNPs were identified to be significantly associated with these resilience traits.They were distributed on nine chromosomes(SSC1,SSC2,SSC6,SSC7,SSC8,SSC12,SSC14,SSC16 and SSC17).After annotation,39 QTLs and 49 candidate genes were identified.Several of these are functionally relevant candidate genes including OTUD4,TIFA and CARD14,which are involved in the host immune response,disease susceptibility and signal transduction.Eight unique SNPs were found to be causal in both GWAS and eQTL analyses across 15 tissues.Notably,one SNP(rs80794541)was associated with eQTLs identified concurrently across seven tissues/cell types,including the macrophage cell type.Furthermore,four significant SNPs(rs81467127,rs81356029,rs80794541 and rs81305085)were linked to the function of the primed enhancer,active element,and poised promoter in five pig tissues.Using the porcine fibroblast HiC dataset,SNP(rs81356029)on SSC2 regulates the CARNS1 and SSH3,while SNP(rs80794541)on SSC7 regulates the H2AC6.In this study,we generated four novel resilience traits and identified SNPs significantly associated with these resilience traits in a Duroc pig population.GWAS signals were associated with candidate genes involving in the immune traits,and were linked to the crucial regulatory elements as well.Our findings will contribute to elucidating the genetic mechanism that can enhance genome-enabled breeding and inform further research on resilience in domestic pigs. 展开更多

In this paper,the fracture behaviors and interface stresses of sintered Nd-Fe-B magnets were investigated.It was revealed that cracks propagated along the interfaces of Nd_(2)Fe_(14)B/Nd_(2)Fe_(14)B,Nd_(2)Fe_(14)B/Nd-rich phases and within Nd-rich phases.The misfits between 2:14:1 and Nd-rich phases were quantitatively determined,reflecting the concentration of interface stresses in magnet.Grain boundaries of magnet were reconstructed by diffusing Tb-containing and Tb-free sources,respectively.The Tb-rich shells at extensive layers of 2:14:1 grains caused lattice distortion,i.e.,internal stresses,resulting in the crack mode changing from intergranular to transgranular in near surfaces of Tb-containing diffused magnets.Therefore,the bending strengths were deteriorated although the coercivities were apparently enhanced.In the case of Tb-free Pr-Cu diffused magnet,the Pr and Cu elements were enriched at grain boundaries,and the misfits between 2:14:1 and Nd-rich phases were declined,lowering interface stresses and strengthening grain boundaries.The mechanical and magnetic properties of Pr-Cu diffused magnet were simultaneously improved. 展开更多

目的探讨杞菊地黄丸加减治疗早期乳腺癌术后服用芳香化酶抑制剂(AI)致高血脂症的临床疗效。方法选择正在服用AI且并发高血脂症的绝经后早期乳腺癌术后患者60例,随机分为治疗组30例和对照组30例。对照组口服阿托伐他汀钙片10 mg每日1次,... 目的探讨杞菊地黄丸加减治疗早期乳腺癌术后服用芳香化酶抑制剂(AI)致高血脂症的临床疗效。方法选择正在服用AI且并发高血脂症的绝经后早期乳腺癌术后患者60例,随机分为治疗组30例和对照组30例。对照组口服阿托伐他汀钙片10 mg每日1次,治疗组在口服阿托伐他汀钙片基础上给予杞菊地黄丸加减治疗。2组均以1个月为1个疗程,共治疗2个疗程。结果 2组治疗后TG、TC、LDL-C及HDL-C水平比较差异均无统计学意义(P均> 0. 05)。治疗组治疗后血脂水平改善的总有效率为86. 7%,对照组总有效率为80. 0%,2组比较差异无统计学意义(P> 0. 05)。2组治疗后中医证候评分均明显下降(P均<0. 05),且治疗组中医证候评分较对照组下降程度更明显(P <0. 05)。治疗组治疗后中医证候总有效率为90. 0%,明显高于对照组的46. 7%(P <0. 05)。结论在服用阿托伐他汀钙片的基础上加服杞菊地黄丸加减治疗乳腺癌术后服用AI所致高血脂症患者,可明显降低患者的血脂水平,改善患者的中医证候,提高患者的生活质量。 展开更多

手性无机纳米材料因为具有优异的光物理特性及广泛的应用价值而备受关注。通过采用手性配体对无机纳米材料的表面进行修饰或将无机纳米材料与手性模板进行组装获得的手性结构,可以与光子强烈作用引起偏振态的改变,产生圆偏振光(circular... 手性无机纳米材料因为具有优异的光物理特性及广泛的应用价值而备受关注。通过采用手性配体对无机纳米材料的表面进行修饰或将无机纳米材料与手性模板进行组装获得的手性结构,可以与光子强烈作用引起偏振态的改变,产生圆偏振光(circularly polarized light, CPL)。从产生机理来讲,CPL主要包括圆偏振荧光和圆偏振散射,在一些情况下这两个机理是共存的。本文总结了硫族半导体纳米材料、金属纳米团簇、钙钛矿、镧系配合物及其他复合纳米材料中CPL的研究进展。此外,还讨论了不同的手性无机纳米材料中CPL的主要来源。本综述得出的结论有望在分子水平上实现对CPL活性材料的各向异性因子进行调控,促进其在量子计算、光学数据存储、信息加密、3D显示器和光学传感等多个领域的发展。 展开更多

Those provide references and ideas for the later large-scale production of augmented reality three-dimensional map. The augmented reality three-dimensional map is produced based on skyline software. Including the map browsing, measurement and analysis and so on, the basic function of three-dimensional map is realized. The special functional module including housing management, pipeline management and so on is developed combining the need of residential quarters development, that expands the application fields of augmented reality three-dimensional map. 目的:应用视疲劳调查量表调查调节性视疲劳患者的视疲劳情况及治疗前后评分差异,评价其在该人群中的信度、效度及反应度。方法:对112例上海中医药大学附属曙光医院眼科就诊及线上招募的调节性视疲劳患者,使用视疲劳调查量表进行调查,再... 目的:应用视疲劳调查量表调查调节性视疲劳患者的视疲劳情况及治疗前后评分差异,评价其在该人群中的信度、效度及反应度。方法:对112例上海中医药大学附属曙光医院眼科就诊及线上招募的调节性视疲劳患者,使用视疲劳调查量表进行调查,再随机抽取48例调查对象于1wk后重测,采用多种信度、效度及反应度指标对量表进行评价。结果:量表条目的总体Cronbach’sα系数为0.91;分半信度系数为0.86;量表总分的重复测量相关系数为0.74,前后无差异(P>0.05);验证性因子分析结果表明三因子结构模型绝对拟合指数χ^(2)/df<2.0,RMSEA=0.08;相关分析中各维度总分和量表总分的相关系数分别为0.92、0.90、0.83;标准关联效度分析显示组间有差异(P<0.01)。反应度分析显示治疗前后有差异(P<0.01)。结论:ASS量表用于调节性视疲劳的临床评价具有较高的信度、效度及反应度,是视疲劳人群临床研究或筛检的有效工具。 展开更多

微藻是水体中重要的生产者和环境调节因素,作为饵料和调水剂在水产养殖中起着重要作用。微藻的培养方式多样,探究了微藻Mychonastes afer和小球藻Chlorella sorokiniana在不同营养方式及通气情况的生长与光合能力。在自养、兼养和异养,... 微藻是水体中重要的生产者和环境调节因素,作为饵料和调水剂在水产养殖中起着重要作用。微藻的培养方式多样,探究了微藻Mychonastes afer和小球藻Chlorella sorokiniana在不同营养方式及通气情况的生长与光合能力。在自养、兼养和异养,以及不同浓度二氧化碳(CO;)通气的条件下,分别测定了两株藻的生长曲线、光合电子传递效率、有机碳源利用率、光合放氧和呼吸耗氧速率,并着重对M.afer的油脂组分进行分析。结果表明小球藻C.sorokiniana可以摆脱对光能的依赖,完全依靠有机碳源营异养生长,而在此条件下M.afer几乎不能生长。有机碳源的加入,对两株藻的光合系统活性均产生了一定程度的抑制作用,使光合电子传递效率以及光合放氧水平下降;同时发现在兼养条件下,高浓度CO;最有利于M.afer生长,促进了细胞对葡萄糖的利用效率,且油脂和神经酸产量提高。研究表明两株藻在协同利用外源有机碳源和自身光合系统方面存在显著差异,并探究出一种用于生产天然神经酸的M.afer最佳兼养条件。 展开更多

目的:探讨2型糖尿病合并周围神经病变(DPN)患者的干眼临床特征。方法:前瞻性队列研究。收集2021-07/2022-03上海中医药大学附属曙光医院内分泌科2型糖尿病患者192例,均选右眼为观察眼,其中确诊为DPN患者122例,糖尿病无周围神经病变(NDPN... 目的:探讨2型糖尿病合并周围神经病变(DPN)患者的干眼临床特征。方法:前瞻性队列研究。收集2021-07/2022-03上海中医药大学附属曙光医院内分泌科2型糖尿病患者192例,均选右眼为观察眼,其中确诊为DPN患者122例,糖尿病无周围神经病变(NDPN)患者70例。比较两组患者眼表疾病指数量表(OSDI)评分、泪河高度、泪河宽度、角膜上皮厚度、角膜内皮细胞密度、泪液分泌试验(SⅠt)、角膜敏感度、睑板腺功能状态评分、泪膜破裂试验(BUT)、角膜荧光素钠染色评分及多伦多临床评分系统(TCSS)评分,分析2型糖尿病患者OSDI评分与TCSS评分的相关性。结果:DPN组患者干眼患病率(55眼,45.1%)显著高于NDPN组(20眼,28.6%)(χ^(2)=5.094,P=0.024),BUT和角膜敏感度评分低于NDPN组(P<0.001),角膜染色评分和睑板腺功能评分高于NDPN组(P<0.001)。所有受试者OSDI评分与TCSS评分呈负相关(r_(s)=-0.233,P=0.002),同时DPN组受试者的OSDI评分与TCSS评分呈负相关(r_(s)=-0.511,P<0.001),而NDPN组患者两项评分间无显著相关(r_(s)=0.007,P=0.957)。结论:DPN患者相较NDPN患者更容易患干眼,OSDI评分对于2型糖尿病患者尤其是DPN患者并非是准确的干眼评价指标。 展开更多

目的:比较飞秒激光小切口角膜基质透镜取出术(SMILE)与飞秒激光辅助制瓣的准分子激光角膜原位磨镶术(FS-LASIK)术后角膜光密度的变化,探讨SMILE术后角膜界面混浊对视觉的影响。方法:前瞻性队列研究。选取2020-05/2021-10在南通大学附属... 目的:比较飞秒激光小切口角膜基质透镜取出术(SMILE)与飞秒激光辅助制瓣的准分子激光角膜原位磨镶术(FS-LASIK)术后角膜光密度的变化,探讨SMILE术后角膜界面混浊对视觉的影响。方法:前瞻性队列研究。选取2020-05/2021-10在南通大学附属医院眼科屈光手术中心拟行角膜屈光手术的患者93例186眼,其中SMILE组48例96眼,FS-LASIK组45例90眼,观察两组患者术前和术后1d,1wk,1、3、6mo时角膜光密度、等效球镜、裸眼视力(UCVA)的变化。结果:纳入93例患者均顺利完成手术,术中及术后均未出现相关并发症,无失访病例。FS-LASIK组在术后1d,1wk UCVA分别为0.044±0.064和-0.001±0.065优于SMILE组(0.102±0.077和0.023±0.064)(均P<0.05)。两组间术后各随访时间的等效球镜均无差异(P>0.05)。FS-LASIK组术后1d在距角膜顶点0~2、2~6mm的角膜光密度及总角膜光密度分别为18.0(17.5,18.6)、16.2(15.6,16.7)、16.7(16.1,17.3),均低于SMILE组[18.6(18.1,19.3)、16.8(16.4,17.4)、17.2(16.6,17.8)](均P<0.05);FS-LASIK组术后1wk在距角膜顶点0~2、2~6mm处的角膜光密度及总角膜光密度分别为17.2(16.7,17.6)、15.5(15.0,15.9)、15.9(15.3,16.7),均低于SMILE组[17.6(17.1,18.3)、16.0(15.6,16.5)、16.6(15.9,17.1)](均P<0.05)。结论:SMILE术后出现的一过性界面混浊是导致其早期角膜光密度高于FS-LASIK的原因,界面混浊的出现或是视觉质量的影响因素。 展开更多

目的:探讨山茱萸多糖对乳腺癌MCF-7细胞增殖、凋亡、迁移、侵袭的影响,并通过观察山茱萸多糖对腺苷酸活化蛋白激酶/沉默信息调节因子1(AMPK/SIRT1)通路的调控作用探索其可能的作用机制。方法:体外培养MCF-7细胞,随机分为对照组(正常培... 目的:探讨山茱萸多糖对乳腺癌MCF-7细胞增殖、凋亡、迁移、侵袭的影响,并通过观察山茱萸多糖对腺苷酸活化蛋白激酶/沉默信息调节因子1(AMPK/SIRT1)通路的调控作用探索其可能的作用机制。方法:体外培养MCF-7细胞,随机分为对照组(正常培养基培养)和山茱萸多糖低、中、高剂量组(分别予12.5、25.0、50.0 mg/mL山茱萸多糖培养)以及山茱萸多糖高剂量+AMPK激活剂(AICAR)组(予50.0 mg/mL山茱萸多糖+2.0 mmol/L AICAR培养),采用平板克隆形成试验检测MCF-7细胞增殖情况,流式细胞术检测细胞凋亡情况,划痕试验检测细胞迁移情况,Transwell法检测细胞侵袭情况,蛋白免疫印迹法(Western blot法)检测各组细胞AMPK、磷酸化AMPK(p-AMPK)、SIRT1蛋白相对表达量。选择BALB/c裸鼠,建立荷瘤裸鼠模型,随机分为对照组(灌胃生理盐水)与山茱萸多糖组(每日灌胃800 mg/kg山茱萸多糖溶液),每组6只,连续灌胃28 d。每7 d测量2组裸鼠肿瘤体积并做组间比较,灌胃结束后处死各组裸鼠取移植瘤,采用免疫组化法检测各组裸鼠移植瘤组织中AMPK、p-AMPK、SIRT1蛋白表达。结果:体外细胞学实验结果表明,山茱萸多糖低、中、高剂量组MCF-7细胞集落形成数明显低于对照组(P<0.05),细胞凋亡率明显高于对照组(P<0.05),细胞划痕愈合率、细胞侵袭数明显低于对照组(P<0.05),细胞中p-AMPK、SIRT1蛋白表达明显低于对照组(P<0.05);山茱萸多糖高剂量+AICAR组MCF-7细胞集落形成数明显高于山茱萸多糖高剂量组(P<0.05),细胞凋亡率明显低于山茱萸多糖高剂量组(P<0.05),细胞划痕愈合率、细胞侵袭数明显高于山茱萸多糖高剂量组(P<0.05),细胞中p-AMPK、SIRT1蛋白表达明显高于山茱萸多糖高剂量组(P<0.05);山茱萸多糖各剂量对上述指标的影响表现出显著的剂量依赖性(P<0.05)。荷瘤裸鼠实验结果表明,移植第14、21、28、35 d山茱萸多糖组裸鼠移植瘤体积显著小于对照组(P<0.05);干预结束后,山茱萸多糖组裸鼠移植瘤质量与瘤体组织p-AMPK、SIRT1蛋白阳性表达率均明显低于对照组(P<0.05),AMPK蛋白阳性表达率与对照组比较差异无统计学意义(P>0.05)。结论:山茱萸多糖对乳腺癌MCF-7细胞增殖、迁移、侵袭有一定的抑制作用,并可促进MCF-7细胞凋亡,能抑制裸鼠移植瘤生长,其可能的机制为抑制AMPK/SIRT1通路激活。 展开更多
